[Frequency of antibodies to Toxoplasma in pregnant women and their newborn in Sao Paulo City, Brazil (author's transl)].

In 80 apparently healthy pregnant women and their newborns tested for toxoplasmosis through the dye-test (RSF) greater than or equal to 1:16, were found respectively 50% and 38.7% positives. The positive mothers had positive newborns. The major part with the same titers as their mothers or one titer lower. The negative mothers had negative healthy newborns with one exception, considered "false positive reaction". The great majority of positive newborns were found negative at the 8th month, and the other until the 12th month. It's emphazised that the maternal and the newborn serology must be done at the same time. This considerations should be remembered when asymptomatic newborns with positive serology for toxoplasmosis are discussed.

[Itraconazole action on Toxoplasma gondii].

With the purpose to increase our knowledge about the spectrum of therapeutic action of itraconazole, the activity of a new triazolic derivative against Toxoplasma gondii was studied in vivo and in vitro with the aid of an experimental infection model in mice. The trial also aimed at a possible improvement of the therapy of toxoplasmosis in regard to safety and dosing easiness. The present study has not shown significant activity of itraconazole against Toxoplasma gondii, differing from a previous observation that showed it's activity against Trypanosoma cruzi.

[Effect of table salt on Toxoplasma gondii].

UNLABELLED: Results of our inquiry on Toxoplasma gondii frequency in pork sausages from Erechim (RS, Brazil), where the incidence of toxoplasmotic chorioretinitis is high (9% according Silveira, 1987), revealed only one isolation of the parasite among 40 sausage samples. (Martins, 1989). The sausages were prepared with pork and common table salt, and were ingested, frequently, in raw state, during its preparation. In this experiment, table salt action on tachyzoites and cysts of Toxoplasma gondii, was investigated. Albino male mice were intraperitoneally inoculated with 0.5 ml of peritoneal exsudates (tachyzoites) or brain macerates (cysts) from previously infected mice. Some strains were isolated from pork and pork sausages from Erechim. Refined common table salt was added to the inocula at 2.2%, 2.5% and 3.0% concentrations, during 24 and 48 hours, 3-5 and 7 days of exposition time, at 4 degrees C. The results demonstrated that at 2.2%, 2.5% and 3.0% salt concentrations and 24 and 48 hours of time exposition all mice inoculated with tachyzoites were dead, except one, probably not infected. At 3.0% and 3-5 and 7 days of contact, 10 in 37 inoculated mice survived (27%). Of the mice inoculated with cysts plus salt at 3.0%, at 5 and 7 days exposition, 85% survived (17 in 20). All the controls mice inoculated with the same inocula without salt, at the same conditions, died earlier than the test mice. The Toxoplasma was recovered from peritoneal exsudates and brains of recently dead and dying mice, but not from the survivors. CONCLUSION: the table salt has an inactive action on Toxoplasma gondii in 3.0% concentration and at 3 to 7 days exposition.(ABSTRACT TRUNCATED AT 250 WORDS)

[Isolation of Toxoplasma gondii from peritoneal exudate and organs of mice with experimental infection].

UNLABELLED: Peritoneal exudates and organs from 53 male albino mice, experimentally infected by Toxoplasma gondii were studied, 21 at the acute phase of infection, and 32 at the chronic phase. Peritoneal inoculations were made with 0.5 ml of peritoneal exudates (tachyzoites), or brain macerates (cysts) of previously infected mice. Direct examinations of peritoneal exudates (tachyzoites) were realized between 3 to 12 days post-inoculation, and in brain (cysts) after 10 days post-inoculation . Organs macerates were inoculated in new mice, for the parasite recovering, from exudates or from brains. At the acute infection (3 to 12 days) the positivity at the direct examination was: peritoneal exudate 19/19, lung 12/14, muscle 6/9, heart 4/9 and brain 1/3. After inoculation: peritoneal exudate 5/5, heart 9/9, lung 13/13, muscles 14/17 and brain 2/2. Then, there were 9 new positive organs. At the chronic infection, between 10 and 495 days, the positivity was, at direct examination: brain 28/32, heart 0/4 and muscle 0/4. After inoculation: brain 6/6, heart 14/29 and muscle 16/26. After that a new positive mouse was detected, which leads to 29 the positivity for all mice, or 90.6%. Finally the positivity for the acute phase was: peritoneal exudate 19/19 (100%), heart 15/17 (88.5%), muscle 12/14 (85.7%), lung 14/14 (100%) and brain 2/3 (66.6%). For the chronic phase: brain 28/32 (87.5%), muscle 16/28 (57.1%) and heart 14/31 (45.1%). At the end of experience, at the 495th day, the brain still presented large cysts by direct examination and also the heart and muscle were positives after inoculation. CONCLUSIONS: 1st) on mice the Toxoplasma gondii remained for 495 days, mainly on the brain, but also on heart and muscle.(ABSTRACT TRUNCATED AT 250 WORDS)

[Isolation of Toxoplasma gondii from the saliva of patients with acquired immunodeficiency syndrome (AIDS)].

Toxoplasma gondii has been found in saliva of patients with toxoplasmosis. We tried to isolate parasites in saliva in patients with AIDS. As it is known, the finding of antitoxoplasma antibodies is not useful for diagnosis of cerebral abscess in toxoplasmosis occurring in AIDS patients in Brazil where 90% or more adults have serological evidence of past infection with Toxoplasma gondii. In 26 patients with AIDS, including six with tomographical evidence suggesting cerebral abscess caused by Toxoplasma gondii, no parasites were found in the saliva by the use of the mouse inoculation model. This trial that the search for Toxoplasma gondii in the saliva is not an alternative method for the diagnosis of toxoplasmosis in patients with AIDS.

[Simplified techniques for toxoplasmosis studies (author's transl)].

UNLABELLED: Some improvements on laboratory technics for Toxoplasmosis are presented: 1--the use of serum from guinea-pigs as accessory factor (AF) on the dye-test; 2--the use of filter-paper discs for collecting and storing blood of children and small animals for the dye test; 3--conservation of Toxoplasma gondii at various media and temperatures. RESULTS: 1--the pool of serum from 14 guinea-pigs gave good results when diluted in 1/5 Alsever solution at the proportion of 6/10; 2--the blood from digital puncture usually could be collected on filter-paper discs of 15 mm of diameter and stored at the freezer to -23 degrees C for 3-4 weeks, protected with alumen-paper. The dilution with saline 8.5% could be made 1/2 hour before starting the procedure; 3--the Toxoplasma gondii (cysts and trophozoites) could be stored at LIT medium or saline 8.5% at refrigator a +4 degrees C for 15-22 days, with some reduction in virulence.
