Impaired elastic matrix development in the great arteries after ablation of the cardiac neural crest.

The cells that form the aorticopulmonary septum in the avian embryo have been shown to be similar to the cells that form the walls of the great vessels in two ways: both are derived from the cardiac neural crest and both are able to synthesize an elastogenic matrix in the early embryo. Because of these similarities, and because ablation of the cardiac neural crest causes congenital defects of the outflow tract that are related to failure of proper septation, it was hypothesized that such an ablation also would cause the walls of the great vessels to be defective. The purpose of this study was to compare the elastic matrix in the mediae of the great vessels of normal embryos with those from which the cardiac neural crest had been ablated. The results show that the elastic matrix in the great vessels of the experimental embryos was impaired 1) in the rate of downstream propagation of the initiation of elastogenesis among younger embryos, incubation days 4-8 and 2) in the spatial configuration of the elastic matrix among the older embryos, incubation days 16-20. These results may provide a biological explanation for the elastin defect that affects the pulmonary artery of many patients with cyanotic congenital heart defects.

Exogenous glycosaminoglycans modulate chondrogenesis, cyclic AMP level and cell growth in limb bud mesenchyme cultures.

Effects of hyaluronate, heparin and chondroitin-6-sulfate were studied on micromass cultures of chick limb bud mesenchyme (Hamburger and Hamilton stages 23-24). Histochemical, electron microscopical, biochemical and radiochemical investigations of day 4 cultures revealed dose-dependent inhibitory effects of these glycosaminoglycans on chondrogenesis, cyclic AMP level and growth of cells. In addition, hyaluronate with 100 micrograms/ml dose caused a displacement of newly formed proteoglycan from cultures into the medium. It is supposed that exogenous glycosaminoglycans influence ionic equilibrium in the immediate vicinity of cells and disturb the organization of the prechondrogenic extracellular matrix resulting in alterations of cell membrane--cytoskeleton associations. These alterations may provoke a reduction in cyclic AMP level and DNA synthesis. It is suggested that a reduction in cyclic AMP level preceding the expression of cartilage phenotype results in the inhibition of chondrogenesis.

Proteoglycan biosynthesis is stimulated by D-penicillamine in chondrifying high density cell cultures.

Chondrifying high density cell cultures of stage 22-24 chick embryo limb bud mesenchyme were treated with 5, 10 and 15 mmol/l D-penicillamine (DPA) for 4 and 6 days. The cultures were analyzed with morphological and biochemical techniques to learn more about the effect of DPA on the metabolism of cartilage glycosaminoglycans (GAGs). Using light and electron microscopic histochemical reactions for GAG, a considerable increase in the intensity of staining of the cartilage matrix could be detected in cultures treated with DPA as compared to the untreated controls. The uronic acid content of the treated cultures was higher than that of the controls. Liquid scintillation measurements and autoradiography revealed that DPA treatment increased the 35S-sulfate into the cultures. These data suggest that DPA - besides its well known inhibitory effect on collagen crosslink formation - alters the metabolism of sulfated GAGs in differentiating cartilage. It is supposed that DPA stimulates the biosynthesis of these macromolecules.

Energy dispersive X-ray microanalysis of sulfated glycosaminoglycans in cartilage matrix stained with alcian blue 8GX.

X-ray spectra were recorded from 400-700 nm matrix areas of 0.5 micron sections prepared from the articular cartilages of 15- and 23-year-old human cadavers. The X-ray microanalysis was carried out (i) on untreated material; (ii) after removing sulfate group by a methylation procedure; (iii) after staining with a copper containing cationic phatolcyanin dye, alcian blue 8GX, preceded by carboxymethylation. K alpha peaks of sulphur could be detected in methylated (i.e. desulfated) samples. These peaks probably indicated the presence of sulphur-containing amino acids in different matrix proteins. Consequently, the measurements of sulphur despite its general use cannot be recommended for the X-ray microanalysis of sulfated glycosaminoglycans of cartilage matrix. K alpha peaks of copper could be identified after carboxymethylation and staining with alcian blue. After carboxymethylation, alcian blue can only be bound to the dissociated sulfate groups of glycosaminoglycans in the cartilage matrix. According to our spectrophotometric studies, approximately one molecule of alcian blue combined with one sulfate group. These data suggested that this technique could be used for semiquantitative estimation of sulfated glycosaminoglycans in small areas of the cartilage matrix. Using this method, we found a higher occurrence of sulfated glycosaminoglycans in the territorial matrix than in the interterritorial matrix of the intermediate and deep zones of the human articular cartilage.

Effect of vitamin D3 and 25 hydroxyvitamin D3 on glycosaminoglycans in micro high density culture.

Vitamin D3 and 25-OH-D3 were tested for their effect on glycosaminoglycan (GAG) production in micro high density cultures of stage 22-24 chicken limb bud mesenchyme. Vitamin D3 (at concentration of 5 x 10(-6)M) and its biologically active metabolite, the 25-OH-D3 (at concentration of 5 x 10(-8)M) stimulated the synthesis of GAGs indicating the increase of radioactive sulfate incorporation and uronic acid content in a dose-dependent way. The findings obtained after the treatment with 25-OH-D3 are comparable with earlier studies performed on chondrocyte cultures (Corvol et al. 1978, 1980), while the results observed after the vitamin D3 treatment are the first indications that vitamin D3 can also influence the cartilage matrix composition.

Inflammatory fibrous histiocytoma of bone.

An 11-year-old patient with inflammatory fibrous histiocytoma of the iliac bone is presented. In addition to routine histopathological procedures the lesion was studied by enzyme-histochemical and immuno-histochemical methods. The results of acid phosphatase, alpha-naphthyl-acetate esterase and intracytoplasmic muramidase demonstration in tumour cells supported the histiocytic nature of the presented case.

[Electron microscopic determination of phenoloxidase (EC 1.14.18.1) in eosinophilic granulocytes of the small intestine of white rats].

It is shown that the light microscopic permanent detectable phenoloxidase activity, using dihydroxyphenylalanine as substrate, of the small intestine of white rats is localized in the most cases in eosinophilic granulocytes. The enzyme has been found as well in the cytoplasma as in the granules. The enzyme proof triggered in the granules the so called reverse effect. The results allow to conclude new aspects for the cell mediated immunological defense reaction.

[Behavior of phenoloxidase-containing cells in experimental granuloma].

Using the histochemical method for phenoloxidase (PO) in albino rats, the development of carrageenan granuloma was investigated. It is shown that especially those cells containing the methanol resistent PO activity are the cells which were be found during the formation of the granuloma. These findings support our earlier suggestion that POC are involved in the defence reactions.

Effect of zinc on the local Shwartzman phenomenon.

Local Shwartzman reaction is substantially reduced in intensity by zinc administered locally before and after preparation of the reaction. The endotoxin-induced accumulation of PMN leukocytes is inhibited by zinc rubbed in the skin.

Polarization microscopic investigation of subarachnoid fibrosis after subarachnoid haemorrhage.

The author used the polarization microscope and topo-optical reactions such as phenol reaction, collagen-specific sulphation, and picrosirius red F 3 BA staining to investigate the nature of subarachnoid fibrosis after experimental subarachnoid haemorrhage. It is strongly suggested that the subarachnoid fibrotic deposits--which here analysed in a previous study by scanning electronmicroscopy--contain a high amount of collagen as a main component both three weeks and three months after the SAH.

Ultrastructure of extracellular matrix of embryonic chick limb bud cartilage.

The ultrastructure of the extracellular matrix of chick limb buds at stages 24--36 was investigated with the electron microscope and with polarization microscopic evaluation of topo-optical reactions. Two types of extracellular matrix structures could be distinguished. In the primary matrix of presumptive cartilaginous areas at developmental stage 25, topo-optical reactions revealed polycarboxylated glycosaminoglycans oriented parallel to the cell surface. With the electron microscope few fine filaments and small matrix granules could be found. In the secondary cartilage matrix, from stage 26 on, topo-optical reactions demonstrated glycosaminoglycans and collagen being predominantly ordered parallel to each other. Their amount and the degree of their orientation gradually increased during the developmental period investigated. Electron microscopy demonstrated collagen filaments and matrix granules in increasing number at stages 26--36. While some parallel ordering of filaments was seen in the electron micrographs, no sign of spatial ordering of glycosaminoglycans could be found with this technique.

[Histochemical study of the matrix of calcium-containing urinary calculi by means of topo-optical reactions].

The authors investigated the matrix of calcium-containing urinary calculi of different basic type by means of histochemical and topo-optic methods. They found the glycoprotein reactions of the calcium oxalate calculi by far weaker than in the calcium phosphate and in the much infected struvite-containing calculi. The acid mucopolysaccharides were to be proved particularly in the infected mixed calculi. Collagen could not be found in the calculi.

A critical re-evaluation of the anisotropic index used in polarization microscopy: a study on epiphyseal cartilage.

Topo-optical reactions specific for collagen and glycosaminoglycans were performed on epiphyseal cartilage plates which differed in age, in chemical composition, and in structure. Retardation values were measured with the polarization microscope and anisotropic index values were calculated after each reaction. It was found that the anisotropic index proposed earlier for quantitative evaluation of topo-optical reactions did not refer either to the submicroscopic orientation pattern or to the quantity of cartilage matrix components. It is suggested that retardation values of induced birefringence may be only used to characterize the spatial orientation of these components in the cartilage matrix.

Histological and histochemical investigations of the epiphyseal cartilage in rats after administration of heparin, coumarin as well as coumarin and diphosphonate (EHDP).

Heparin and coumarin (Marcumar) as well as coumarin combined with a diphosphonate (EHDP) were examined histologically and histochemically with regard to their effect on the structure of the epiphyseal cartilage in rats. Topo-optical reactions were used to assess the submicroscopic structural changes in the developing trabeculae of metaphysis. The zone of resting cartilage and the zone of young proliferation cartilage in epiphyseal plates revealed a reduced cell count due to the influence of anticoagulants. With regard to the matrix of the epiphyseal plate, it may be stated that its ultrastructure appeared partly regular and that, on the other hand, large areas of degeneration were detected in the connective-tissue. The negative effect of anticoagulants on the ground substance could not be reduced definitely by means of a diphosphonate (EHDP) in conjunction with Marcumar which was administered simultaneously.

[The participation of phenol-(DOPA-)oxidase containing cells (POZ) with the delayed hypersensitivity. (Preliminary report) (author's transl)].

With the aid of histochemical techniques phenol-(DOPA-)oxidase containing cells were detected in the skin, liver, spleen, small intestine and blood of guinea-pigs. The number of these cells is considerably increased in the connective tissue of the skin during the delayed hypersensitivity reaction which was provoked by an intracutaneous injection of cartilage antigen in guinea-pigs immunized with the same antigen.
