Nerve growth factor receptor immunoreactivity in Meissner and Pacinian corpuscles of the human digital skin.

The presence of nerve growth factor receptors (NGFr) in sensory nerve corpuscles of human digital skin, primarily Meissner and Pacinian corpuscles, was investigated immunohistochemically using two monoclonal antibodies directed against human-NGFr. To ensure the localization of NGFr immunoreactivity (IR) alternative sections to that processed for NGFr detection were assayed for neurofilament protein (NFP) and S-100 protein which selectively label the axon and the periaxonic specialized cells (lamellar cells of Meissner's corpuscles; inner-core cells of Pacinian corpuscles), respectively. Occurrence of NGFr IR was observed in both types of sensory corpuscles. In Meissner's corpuscles NGFr-IR was found in the lamellar cells, whereas in the Pacinian corpuscles the lamellae of the inner core, outer core, and capsule displayed NGFr IR. Moreover, a positive IR was observed in the central axon of some Pacinian corpuscles. However, remarkable differences were encountered among Pacinian corpuscles in the pattern of NGFr IR distribution. Present results demonstrate the presence of NGFr IR in sensory nerve corpuscles of the human digital skin, suggesting that NGFr could be involved in the concentration of NGF and in the conveying of this molecule from the cutaneous sources to the cell body of NGF-dependent primary sensory neurons. However, the mechanisms involved in this process remain to be clarified.

Thymectomy as treatment of autoimmune diseases other than myasthenia gravis.

This paper reviews the different series of thymectomies performed in patients with autoimmune diseases other than myasthenia gravis. It is possible that thymectomy can decrease the activity of T-helper lymphocytes or, alternatively, it may enhance the activity of T-suppressor lymphocytes, whose function is depressed in autoimmune diseases. Thymectomy was performed empirically for systemic lupus erythematosus and rheumatoid arthritis. The therapeutic benefits were questionable. Conflicting results were reported for thymectomy against autoimmune hemolytic anemia. Several trials were conducted to assess the therapeutic value of thymectomy in multiple sclerosis. Benefits were achieved only in relapsing-remitting but not in chronic-progressive multiple sclerosis. The effect of thymectomy in autoimmune diseases associated with myasthenia gravis were also reported. The authors conclude that thymectomy as treatment for autoimmune diseases other than myasthenia gravis is not elective therapeutic choice and it is acceptable only in selected cases.

Immunohistochemical study of sensory nerve formations in human glabrous skin.

The sensory nerve formations (or corpuscles) of normal human glabrous skin from hand and fingers, obtained by punch biopsies, were studied by the streptavidin-biotin method using monoclonal antibodies directed against neurofilament protein (NFP), S-100 protein, glial fibrillary acidic protein (GFAP), cytokeratins, and vimentin. NFP immunoreactivity (IR) was observed in the central axons of most sensory formations, while S-100 protein IR was restricted to non-neuronal cells forming the so-called inner cells core or lamellar cells. Furthermore, vimentin IR was found in the same cells of Meissner's and glomerular corpuscles. None of the sensory nerve formations were stained for GFAP or keratin. The present results suggest that the main nature of the intermediate filaments of the non-neuronal cells of sensory nerve formations from human glabrous skin is represented by vimentin and not by GFAP. Thus, our findings suggest that lamellar and inner core cells of SNF are modified and specialized Schwann cells and not epithelial or perineurial derived cells.

Immunohistochemical study of the sensory formations in the glabrous skin of the rat.

The presence of some cytoskeletal proteins related to the intermediate filaments glial fibrillary acidic protein -GFAP and vimentin) and S-100 protein has been investigated in sensory formations of the glabrous skin of the rat. A positive reaction both for S-100 protein and vimentin was found in the inner core and related cells of glomerular and simple sensory corpuscles; in contrast, no positive reaction was shown for GFAP. The authors discuss these results on the basis of the glial origin of the inner core and related cells in sensory formations.

Immunohistochemical study of Pacinian corpuscles using monoclonal antibodies for neurofilament protein, glial fibrillary acidic protein and S-100 protein.

The presence of neurofilament protein (NFP), glial fibrillary acidic protein (GFAP) and S-100 protein has been investigated in Pacinian corpuscles from cat's mesentery by means of immunohistochemical methods. The NFP-like positivity was found in the central axon of the corpuscles; the GFAP- and S-100 protein-like immunoreactivities were shown in the innermost layers of the differentiated cell of the inner core. No positive reaction was detected in the capsule. The authors discuss these findings.

[Arteriomegaly in the aorto-iliac-femoral area with or without associated aneurysm].

The authors present a group of 199 patients with arteriomegaly, an affection characterized by elongated and distended blood vessels of the arterial system, with or without accompanying aneurysms. Our study on this group of patients, drawn from a large arteriographic series of peripheral abdominal and lower limb arterial disorders, focuses on a comparison between atherosclerotic arteriopathy and arteriomegaly. Small tissue blocks were taken from the arterial wall of patients operated on for megadolichoarteries. Electron microscopic examination of such specimens revealed a specific alteration of the elastic component of the vessel wall. The authors believe that surgical treatment of this condition is indicated in order to prevent thromboembolic complications or aneurysmal rupture.

[Method of biopsy and ultrastructural study of the parathyroid gland in patients with primary hyperparathyroidism].

The authors suggest a new method of biopsy of a seemingly normal human parathyroid gland during surgery for parathyroid adenoma. Such a method provides adequate surgical specimen to rule out a likely diffuse hyperplastic disease associated with the parathyroid adenoma, and for the ultrastructural study of the gland, without any risk of postoperative complication. In all patients, ultrastructural examination showed an abnormal cell population and signs of glandular atrophy as a consequence of the high level of serum calcium due to hormonal hyperfunction of the parathyroid adenoma.
