Effect of different kinds of dietary casein on blood cholesterol and triglycerides in pair fed rats.

After casein intake, serum cholesterol levels are dependent on many experimental conditions in rats. The effect of the method of casein preparation was assessed in Wistar rats pair-fed for 4 wk with three identical diets differing only by the type of casein used. Compared with dietary casein and Na caseinate, ingestion of lipid- and vitamin-free casein resulted in higher liver weights (4.35 +/- 0.23 vs. 3.89 +/- 0.30 and 3.90 +/- 0.25 g) and fasting serum cholesterol levels (0.55 +/- 0.13 vs. 0.49 +/- 0.09 and 0.44 +/- 0.09 g/L). Compared with the two other casein diets, ingestion of Na caseinate produced the lowest fasting serum triglyceride levels (0.62 +/- 0.13 vs 0.74 +/- 0.12 and 0.73 +/- 0.13 g/L). Peptic digestibility of caseins used might be a regulating factor of serum cholesterol levels in Wistar rats.

Depletion in choline containing phospholipids of LpB particles in adequately controlled type I insulin-dependent diabetes mellitus.

The aim of the present study was to evaluate the serum and lipoprotein levels of choline--containing phospholipids (lecithin, lysolecithin and sphingomyelin) in a group of type I diabetic patients in fair metabolic control. Forty-eight male adult diabetic patients without macroproteinuria were compared with 48 healthy controls matched for age and body weight. Total cholesterol and phospholipids were assessed in plasma and in apolipoprotein B (apoB) and non apoB containing lipoprotein particles (Lp B and Lp non B particles) separated using precipitation with Concanavalin A. When compared to the control group, diabetic patients have significantly lower serum phospholipids and phospholipids and cholesterol content of LpB particles. All the other parameters studied were similar in the two groups. The phospholipids/cholesterol ratio, calculated in LpB particles is also significantly reduced in the diabetic group. These results strongly suggest a depletion in the choline containing phospholipid content of LpB particles and therefore an alteration of the surface components of atherogenic particles in type I diabetes mellitus.

Computer analysis of ocular results following selective blockade of growth-hormone in diabetic retinopathy. A collaborative study.

Computer storage and analysis of ocular data (visual acuity, fundus photographs and fluorescein angiographies) in diabetic retinopathy (DR) was used before and 1, 3 and 5 years after "Stereo-GIHF". This radiobiological procedure results in the selective inhibition of growth-hormone (GH) release. A series of 107 eyes were analyzed after division into 2 categories A and B (A: progressing background DR, n = 53, B: proliferative DR, n = 54). All patients treated by laser photocoagulation before and/or after stereo-GIHF were excluded from the present study; in contrast complications that impair visual acuity (VA), mainly vitreous hemorrhages, diabetic maculopathy and cataracts, were not discarded. In groups A and B, mean VA was stable showing no significant differences before and 5 years after treatment. In both groups microaneurysms are improved or stable in all cases. Hard exudates were improved in 66% of cases. In group B preretinal new-vessels and neovascularisation on the disc are improved or stable in 90% of cases. The same percentage is observed for extramacular and macular transudations 3 years after Stereo-GIHF. The present data strongly suggest that the selective suppression of GH improves the prognosis of DR. However the prognosis for vision remains poor in macular degenerations; in addition a rapid evolution of new-vessel formations should indicate a combined treatment with argon laser photocoagulation.
