A gene that modifies the sex ratio in a bisexual strain of Sciara ocellaris.

The influence of an X-linked recessive mutation, sepia, on the sex determination of a bisexual strain of Sciara ocellaris was studied. It induces an alteration in the sex ratio, especially in the progeny of heterozygous females, increasing the proportion of males. These results cannot be explained by differential fecundity of the female parents of different genotypes nor by differential mortality between the sexes. The occurrence of gynandromorphs indicates that the mutant probably interferes with the processes of X-chromosome elimination. Observations that heterozygous females which received the sepia allele from their mothers produced a higher frequency of gynandromorphs than females which received the mutated allele from their male parents, suggest that the mutation is interfering with the mechanism of chromosome elimination present in the egg cytoplasm.

Modified Jendrassik--Grof method for bilirubins adapted to the Abbott Bichromatic Analyzer.

I describe a semi-micro method for use with a discrete analyzer, the Abbott ABA-100, requiring 75 microliter of sample for determination of both the total and direct bilirubins. This method utilizes a serum blank. It is simple and reasonably rapid. The azobilirubin formed by both the total and the direct bilirubins produces a reddish-violet color, which is measured at 550 nm. Values obtained are comparable to those by the automated Jendrassik and Grof procedure adapted by Gambino and Schreiber (total r = 0.999, direct r = 0.990). The method is linear to 300 mg/liter. Day-to-day precision (CV) for the total bilirubin was 8.8% for a 8 mg/liter sample (n = 45) and 4.7% for a 57 mg/liter sample (n = 46). The absorbance of hemoglobin and its derivatives (540 nm) caused a false decrease in the measured bilirubin content, the direct being more affected than the total. This problem was alleviated by the use of caffeine and acid blanks.

D1 versus D2 pancreatoduodenectomy in surgical therapy of pancreatic head cancer.

BACKGROUND/AIMS: The aim of this study was to evaluate the influence of standard pancreatoduodenectomy versus pancreatoduodenectomy with extended lymphadenectomy and the role of adjuvant therapy on survival in patients with ductal adenocarcinoma of the pancreatic head. In addition the problems related to resection are discussed. METHODOLOGY: A total number of 124 pts operated on between 1985 and 1999 were divided into three groups according to our different strategies. Standard resection (D1) was performed on 48 patients (group A), extended resection (D2) on 45 patients (group B) and combined treatment (extended resection plus adjuvant therapy) on 31 patients. The outcome of these three groups was compared with regard to postoperative morbidity and survival. RESULTS: There was no significant difference in terms of survival between group A and B, while adjuvant therapy (group C), achieved statistical significance as factor influencing survival, together with tumor stage. CONCLUSIONS: Our data suggest that no further improvement can be obtained on long-term survival by extended retroperitoneal dissection while chemoradiotherapy showed a doubling of median survival.

Immunity to glycolipid antigens in microbial infections.

T cells recognize ligands of different chemical structures. Recently, it has become clear that also self glycosphingolipids and bacterial lipoglycans may act as T cell stimulatory ligands. This type of antigen recognition is restricted by the non-polymorphic CD1 molecules, which have a structure resembling that of classical MHC molecules. Glycolipids insert their hydrophobic lipid tails in two pockets below the antigen-binding groove and position their hydrophilic heads on the external part of CD1 molecules. TCR interacts with these carbohydrates and discriminates their structural variations. Glycolipid-specific T cells may provide protection during bacterial and parasite infection probably with different mechanisms: by secreting pro-inflammatory lymphokines, by the direct killing of infected target cells, and by helping specific B cells in Ig production. Lipoglycans represent excellent candidates for new anti-microbial vaccines due to their wide distribution in the microbial world and their structural composition which does not change and thus cannot give rise to escape mutants. Moreover, these vaccines might induce anti-microbial protective T cell responses in the whole population due to the non-polymorphic nature of CD1 presenting molecules.

Vibrio parahaemolyticus: aetiological agent of food poisoning.

After mentioning the principal morphological, cultural, physiological, biochemical, enzyme and serological characteristics of Vibrio parahaemolyticus (a facultative halophilic bacterium and the aetiological agent of food poisoning caused by the consumption of fish products) a description is given of its ecological aspects, geographical distribution, and potential enteropathogenicity for man, with general information on its isolation and identification.
