Assessment of anti-HLA antibodies in sera being tested for platelet reactivity by a platelet lymphocyte immunofluorescence test (PLIFT).

An indirect immunofluorescence test using a platelet mononuclear cell suspension is described. The test is a simple and sensitive method to assess the contamination by anti-HLA antibodies of sera being tested for platelet reactivity and eliminates the need to support two independent test systems such as lymphocytotoxicity and immunofluorescence testing. The test could be of value in routine platelet serology testing and platelet crossmatching.

The Duffy blood group is linked to the alpha-spectrin locus in a large pedigree with autosomal dominant inheritance of Charcot-Marie-Tooth disease type 1.

The alpha-spectrin locus (SPTA) on chromosome 1 maps to 1q22-q25 and alpha-spectrin specific probes detect restriction fragment length polymorphisms (RFLPs) with the endonucleases MspI and PvuII. The Duffy blood group (FY) has been mapped to the 1p21-q23 region. We found positive linkage between the alpha-spectrin and the Duffy loci with a maximal Lod score of 3.81 at theta = 0.0 using the computer program MLINK. This indicates that both loci are very closely linked and probably localized to 1q22-q23.

Absence of linkage with the Duffy blood group in a family with Charcot-Marie-Tooth neuropathy.

We report a large Belgian family with Charcot-Marie-Tooth disease (CMT) or hereditary motor and sensory neuropathy type I (HMSN-I). The pedigree consists of 5 generations with 350 family members comprising 42 patients. The disease is transmitted according to an autosomal dominant inheritance pattern. Several HMSN-I families have been reported to be closely linked to the Duffy blood group marker on chromosome 1. These families were designated HMSN-Ib families, opposed to the HMSN-Ia families which do not show evidence for such a linkage. Therefore we examined our family for the Duffy linkage relationship. We found no evidence for a strong linkage of the disease to the Duffy blood group locus, indicating that this family is of genetic subtype Ia.

Flow cytometric analysis of erythrocytic D antigen density profile.

Red blood cells (RBC) were studied with flow cytometry (FCM) after staining with a directly fluoresceinated anti-D antiserum. The immunofluorescence distribution profiles were slightly asymmetrical, due to a significant, yet small skewness. This skewness was not associated specifically with D antigen positivity, making a homogeneous, normal D antigen distribution on the RBC very likely. With the exception of Du, D-positive RBC could always be distinguished from D-negative ones. FCM was successful in bringing the following phenomena to light: difference in mean fluorescence intensity between the different rhesus phenotypes, the D gene dosage effect, the depressing effect of C antigen on D antigenicity, the nonspecific attachment of IgG on (D-negative) RBC and rhesus mosaicism. The absolute number of D antigen sites could be calculated, using an absolute fluorescence standard.

Binding of serum ferritin to concanavalin A in patients with malignancy.

The percentage non-glycosylated, tissue type ferritin was measured in the serum of 30 cancer patients and 11 normal subjects, as well as in 10 tumour cytosols. As expected, in normal subjects the larger part of serum ferritin corresponded to glycosylated ferritin, and in tumour cytosols the larger part corresponded to non-glycosylated ferritin (tissue ferritin). In all but one cancer patients a significant part of the protein did not bind to Con A, thus behaving as tissue ferritin. The findings obtained suggest release of tissue type, non-glycosylated, ferritin by damaged cells or through abnormal secretion. This relative increase of the non-glycosylated ferritin fraction appears to be very frequently present in cancer, although not specific for it. Assaying for Con A binding may prove to be valuable in the study of malignant disease, and therefore deserves further investigation.

In Belgium, viral hepatitis A is predominantly childhood disease.

Data from medical histories and seroepidemiological surveys of blood donors offer good grounds for regarding viral hepatitis A in Belgium primarily as a childhood disease. The epidemiology of anti-HA has thoroughly altered over the last few decades, which correlates with radical socioeconomic changes and improved hygienic living conditions. The rate of HLA-antigens shows in general no different patterns in groups with or without a recognized history of HAV infections.

Usefulness of apheresis to extract microfilarias in management of loiasis.

Two patients with loiasis in whom treatment with diethylcarbamazine had to be interrupted because of serious side effects underwent apheresis to extract microfilarias from the blood. After three apheresis sessions the number of microfilarias in the blood had dropped from 8980 to 3412/ml in one patient and from 2950 to 1840/ml in the other. Between 40 and 45 X 10(6) microfilarias were removed from each of the patients. After reduction of the parasitaemia the two patients were given diethylcarbamazine; no appreciable side effects occurred, and the patients were cured.

Factors influencing the production of human immune interferon components.

Human interferon (IFN), induced in leukocyte suspensions by mitogens, contains two antivirally active components (45K and 22K), separable from each other by gel filtration. The 45K component was identified as IFN-gamma; the 22K component is serologically related to IFN-beta, but has different physicochemical and biological properties. Relatively large amounts of 22K were produced in leukocyte suspensions isolated from buffy coats after induction with concanavalin A (ConA); induction of plateletpheresis residues with staphylococcal enterotoxin A (SEA) resulted in the production of almost only IFN-gamma (45K). The addition of foetal calf serum (FCS) to the production medium enhanced the yields of IFN-gamma, while the production of 22K remained unchanged. Incubation of the cell suspension in the absence of mitogen gave only small amounts of IFN, containing mostly 22K. Under various conditions tested, the two components had similar kinetics of production. A significant increase of IFN-gamma production was observed when leukocytes were primed with low concentrations of 22K.

HLA-DR and monoclonal gammopathy.

The frequencies of the HLA-DR antigens were compared in 3 groups: a group of 33 patients with multiple myeloma (MM) or Morbus Waldenström (MW): a group of 34 persons with benign monoclonal gammopathy (BMG); and a group of 118 healthy controls. No statistically significant associations were observed.
