Characterization of the mycoplasmal conjunctivitis epizootic in a house finch population in the southeastern USA.

An epidemiological study of the prevalence of mycoplasmal conjunctivitis in the house finch (Carpodacus mexicanus) was conducted in Auburn (Alabama, USA) between March 1998 and February 1999. Clinical disease was observed in 4% of the 1,214 finches trapped and examined. This rate is comparable to the average annual prevalence observed in this population since 1996, although the prevalence of clinical disease observed in the peak months of September through November was lower than in previous years. Clinically ill birds were observed in all months of the study. To estimate the prevalence of recovering and asymptomatic, infected birds, we tested a subset of 334 house finches serologically for exposure to Mycoplasma gallisepticum (MG) using the serum plate agglutination (SPA) assay. The prevalence of clinical disease in this subsample was slightly higher (7%) than in the entire sample, reflecting the fact that the serological survey was initiated in the late summer when the prevalence of MG infection peaks in our study population and a sampling bias for symptomatic birds. The serological survey indicated that 13% of this subpopulation had been exposed to MG. We also tested 46 of 334 finches by polymerase chain reaction (PCR) to detect MG in seropositive, asymptomatic birds. Use of the PCR in conjunction with the SPA detected six asymptomatic, infected birds that may represent potential carriers or birds in the early stages of infection. The decreasing prevalence of clinical disease observed during the peak months suggests a changing host-parasite relationship. Continued surveillance of this population, employing both clinical observation and serological analysis will be useful in characterizing these changes over time.

[Experimental study on bioglass application in extending alveolar bone crest in rabbit].

OBJECTIVE: To observe the effect of Bioglass guiding bone regeneration and forming normal new bone in extending alveolar bone crest by surgery. METHODS: Thirty-six rabbits were randomly divided into three groups. The first group was treated with Bioglass and the second with hudroxyapectite (HA). The third group was control group. At 4 8 12 weeks after surgery, the condition of bone regeneration was observed with radiological and pathological methods. RESULTS: Statistical analysis showed that there was a significant difference between the third group and the other two groups (P<0.05). In addition, Bioglass can guide bone regeneration faster than hydroxyapatite, and it can be substituted by new bone to form completely normal bone. CONCLUSION: Since Bioglass is more effective than HA in guiding bone regeneration, it is an ideal bone graft substitute.

[Separation and determination of purine bases and pyrimidine bases from nucleic acid hydrolysis by HPLC on BDS column].

The hydrolysates of nucleic acid, six purine bases and pyrimidine bases (cytosine, uracil, guanine, hypoxanthine, adenine and thymine) were separated and determined by using HPLC. It is discussed how the column and mobile phase affect the separation. The peaks of cytosine and adenine are tailed on ordinary C18 column, and they are very good on BDS-C18 column. The KH2PO4-H3PO4 buffer can be used in separation the hydrolysates of RNA and DNA, and the NaAc-HAc buffer is only used in DNA. In addition, pH value is a very important factor for separation. With pH value of mobile phase increasing, the retention times of guanine, hypoxanthine and thymine were first increased and then decreased, adenine was increased, and cytosine and uracil were almost constant. The chosen mobile phase was 0.1 mol/L KH2PO4-H3PO4 buffer, with a pH value of 4.05. It was detected at UV 260 nm. The determination was completed within 10 min. The RSDs were all less than 3% and the recoveries were in the range of 82%-114%. The method has been applied to the detection of yeast hydrolysates.

[Using polymerase chain reaction for the detection of gonorrhoea in women and children in Taiyuan].

A polymerase chain reaction (PCR) method was established for the detection of gonococcal gene in samples obtained from women and children with lower genital tract infection. The primers were used for PCR amplification derived from sequence for DNA representing part of the open reading frame (ORF) for the chromosome of N. gonorrhoea. In this study, a total number of 9204 specimens were obtained from women and children with lower genital tract infection from 1986 to 1995 in Taiyuan. The positive rate of N. gonorrhoea for Gram's stain of smears was 26.5% while the rates for culture and PCR were 12.4% and 21.0%, respectively. The results showed that PCR test was simple, rapid sensitive and specific for the detection of gonococcal gene in clinical specimens.

[Building up of an animal model of conditioned immunosuppression and analysis of its possible mechanism].

In the present study, camphor odor and intraperitoneal (ip) injection of cyclophosphamide (CY) were used as conditional and unconditional stimulus, respectively, in mice. Mice were exposed to camphor odor for 1 h in their cage in a closed area followed by an ip injection of CY (75 mg.kg-1). This association trial session was repeated once on the next day. Delayed type hypersensitivity response (DTH) was induced as follows: six days after the second association trial session the mice were sensitized by smearing dinitrochlorobenezene (DNCB) on their abdominal skin. The mice were challenged by smearing DNCB on the left ear 5 days after the antigen sensitization. The left and right ears were removed 24 h after the challenge and weighed, the weight ratio of left/right ears was calculated for identification of the response. The ratio was 1.30 +/- 0.113 (+/- s, P < 0.001), indicating that the challenged ear was heavier than the other and DTH was induced. In the unconditioned response (UCR) group, CY (75 mg.kg-1) was given 24 h prior to the challenge and the ratio was 1.09 +/- 0.024 (P < 0.001) indicating that DTH was suppressed by unconditional stimulus (CY). In the conditioned response (CR) group mice were reexposed to camphor odor 24 h prior to the challenge and normal saline was injected instead of CY. The ratio was 1.13 +/- 0.074 (P < 0.001), indicating that DTH was also suppressed by conditional stimulus (camphor odor). These results show that a conditioned immunosuppressive response was induced. In the experiment, many other groups, including unconditioned response group, CYE group and camphor control group, were described in more details in the text. In order to further analyse the mechanisms of the conditioned response, the blood from the mice in CR group was obtained 6 h after reexposure to camphor odor and the serum was injected to normal mice 6 h prior to the challenge. DTH was found to be suppressed significantly when compared with the mice injected with normal serum. The conditioned serum was dialyzed against a membrane with a 10,000 molecular weight cut off. The suppressive activity of the conditioned serum disappeared, suggesting that the molecular weight of the suppressive element in the serum was probably less than 10,000 kDa.

Detection of genes for heat-stable enterotoxin in Escherichia coli by biotinylated ST-DNA probes.

Reference strains of enterotoxigenic Escherichia coli (ETEC), non-enterotoxigenic Escherichia coli (non-ETEC), enteropathogenic Escherichia coli (EPEC), enteroinvasive Escherichia coli (EIEC), and other enteropathogenic bacteria were used to prove the reliability of BIO-ST-DNA probe hybridization. In addition, 417 strains of E. coli isolated from children with diarrheal diseases in Shanxi Children's Hospital were examined for BIO-ST-DNA probe hybridization. In the test, BIO-ST-DNA hybridization was compared with suckling mouse assay in identifying ST-ETEC. The results obtained by both methods showed no significant difference. It was found that identification of ST-ETEC using hybridization is a simple, sensitive and more practical method.

Antimalarial activity of new dihydroartemisinin derivatives. 5. Sugar analogues.

A series of dihydroartemisinin derivatives containing a sugar moiety was prepared in the search for analogues with good water solubility and high antimalarial activity. The preparation of the new compounds was achieved by treatment of dihydroartemisinin (2) with chlorotrimethylsilane in pyridine solution at -10 degrees C to give a nearly quantitative yield of 10-O-(trimethylsilyl)dihydroartemisinin (3), which was then condensed with 1-hydroxypolyacetylated sugars 5 to give dihydroartemisinin derivatives 7a-d. Deacetylation of intermediates 7 gave the desired sugar derivatives 8. The resulting derivatives, tested in vitro against Plasmodium falciparum, were found to be more effective against W-2 than D-6 clones and were not cross-resistant with existing antimalarials. Trimethylsilylated compound 3 is more effective than derivatives 7a-d, which possess activity comparable to or better than that of artemisinin itself. Deacetylated compounds 8a-d were substantially less active than 7 in both cell lines. In P. berghei-infected mice, 7a-c showed 5/5, 2/5, and 3/5 cures, respectively, at 320 mg/kg per day x 3, whereas 7d showed no activity at the same dosage. However, 7d did prolong the life span in 3/5 of the infected mice at 640 mg/kg per day x 3 dose level. Trimethylsilylated compound 3 was also the most effective among the compounds studied, with 5/5 cures at 80 mg/kg per day x 3. The deacetylated sugar derivatives 8a-d showed only slight in vivo antimalarial activity.

Study of the identification of enterotoxigenic Escherichia coli by LT-DNA gene hybridization.

Reference strains of enterotoxigenic Escherichia coli (ETEC), non-ETEC, and other enteropathogenic bacteria were used to prove the reliability of LT-DNA gene hybridization. In the test, LT-DNA gene hybridization was compared with plate immunohemolytic test (PIHT) in identifying LT-ETEC. The results obtained by both methods showed no significant differences. 791 strains of E. coli isolated from 1,875 children with acute diarrhea in Taiyuan Children's Hospital were examined for LT-ETEC by LT-DNA gene hybridization and PIHT. 289 strains examined by LT-DNA gene hybridization were LT positive, while 205 strains examined by PIHT were LT positive. Three different assays were done: colony hybridization, PIHT and fecal direct blot hybridization on each of 74 fecal specimens from children with acute diarrhea. It was found that identification of LT-ETEC using fecal direct blot hybridization is a simple, sensitive and more practical method.

Comparative evaluation of nine different methods for detecting enterotoxin of Escherichia coli.

Nine different methods for detecting enterotoxin of Escherichia coli were studied and compared. We found rabbit ileal-loop test and suckling mouse assay were both quite accurate and reliable for detecting heat labile toxin (LT) and heat stable toxin (ST) of enterotoxigenic Escherichia coli (ETEC). Mouse ileal-loop test was simple, but its sensitivity and specificity were comparatively low. CHO cell-culture assay might be more sensitive and specific. LT-DNA probe was the most sensitive and specific method. In practical application, PIHT (plate immunohemolytic test), Biken's, SPA-CoA and ELISA methods are recognized as simple, rapid, sensitive and specific methods for detecting ETEC-LT. These methods can be selected for use in clinical laboratory.

Antimalarial activity of new water-soluble dihydroartemisinin derivatives. 3. Aromatic amine analogues.

A series of artemisinin (1) derivatives containing bromo and heterocyclic or aromatic amine functions was prepared in the search for analogues with good water solubility and high antimalarial activity. Treatment of dihydroartemisinin (2a) with boron trifluoride etherate at room temperature gave the key intermediate, 9,10-dehydrodihydroartemisinin (3), which, on reaction with bromine, gave the dibromide 4. The latter was condensed with amines in anhydrous CH2Cl2 at less than -10 degrees C to give the desired products in 25-55% yield. The new derivatives, tested in vitro against Plasmodium falciparum, were found to be more effective against W-2 than D-6 clones and were not cross-resistant with existing antimalarials. Compound 6b, 3-fluoroaniline derivative, was the most active of the series, with the IC50 less than or equal to 0.16 ng/mL, making it several fold more potent than 1. However, no significant in vivo antimalarial activity against Plasmodium berghei was observed in any of the new compounds tested.

Orbital rhabdomyosarcoma. Immunohistochemical studies of seven cases.

An immunohistochemical analysis using a series of mono- and polyclonal antibodies against myoglobin, desmin, vimentin, actin, NSE, S-100 protein and keratin was done on alveolar rhabdomyosarcoma (3 cases), embryonal (3) and pleomorphic rhabdomyosarcoma of the orbit (1). All tumors were stained by monoclonal antibodies to desmin and vimentin, whereas 6 of the 7 cases were stained by myoglobin. The results indicate that desmin and vimentin are highly sensitive immunohistochemical markers for the diagnosis of rhabdomyosarcoma, especially in poorly differentiated ones.

Use of ultrasonography in the diagnosis of dysbaric osteonecrosis.

Comparisons between x-ray and B-mode ultrasonic examinations were carried out in 32 divers in both shoulders and both hips (128 comparisons in all). The x-rays showed dysbaric osteonecrosis (DON) in 46 sites; in 43 of these, the B-mode ultrasonogram result coincided with the x-ray finding. We suggest that B-mode ultrasonograms can depict the location, form, and size of the lesions. It is an easy, efficient, and noninvasive method of diagnosing DON, worthy of further investigation and application. Similar good agreement between x-rays and ultrasonograms was found with the now-obsolete A-mode technique.
