Isolation of two strains (S-R type) of Bacillus circulans and purification of a cyclomaltodextrin-glucanotransferase.

From wild type inoculum of Bacillus circulans DF 9 which produce cyclomaltodextrin-glucanotransferase (EC 2.4.1.19, CGTase), two different kinds of colonies were isolated, which correspond to the classical S-R variation. From the culture medium of both colonies grown together a CGTase was purified about 50 fold with a yield of 54% in two steps. From pure R-cell culture the enzyme was purified by about 38 folds with a yield of 79% in only one step, showing a complete homogeneity as judged by a native PAGE analysis.

Some properties of a cyclomaltodextrin-glucanotransferase from Bacillus circulans DF 9 R type.

The cyclomaltodextrin-glucanotransferase (EC2.4.1.19, CGTase) which was purified to homogeneity from Bacillus circulans strain DF 9, R type, showed a pI of 5.3 determined by disc-isoelectric focusing, a Mw of 78 kDa estimated by SDS-PAGE with a range of pH of optimal enzymatic activity rather large (4.5-7.5). The thermal stability of the enzyme at 55 degrees C was increased 4-5 times when calcium ion (10 to 100 mM) or alpha-cyclodextrins (10 mM) were added to the preincubation mixtures. The alpha: beta: gamma ratio determined by HPLC was about 1:0.9:0.4 and the maximal conversion to cyclodextrins with 5% soluble starch was about 36%.

Transport and metabolism of trehalose in Escherichia coli and Salmonella typhimurium.

The metabolism of trehalose in wild type cells of Escherichia coli and Salmonella typhimurium has been investigated. Intact cells of Escherichia coli (grown on trehalose) accumulated [14C]-trehalose as [14C]-trehalose 6-phosphate. Toluene-treated cells catalyzed the synthesis of the [14C]-sugar phosphate from [14C]-trehalose and phosphoenolpyruvate; ATP did not serve as phosphoryl donor. Trehalose 6-phosphate could subsequently be hydrolyzed by trehalose 6-phosphate hydrolase, an enzyme which catalyzes the hydrolysis of the disaccharide phosphate into glucose and glucose 6-phosphate. Both Escherichia coli and Salmonella typhimurium induced this enzyme when they grew on trehalose. These findings suggest that trehalose is transported in these bacteria by an inducible phosphoenolpyruvate:trehalose phosphotransferase system. The presence of a constitutive trehalase was also detected.

Induction and catabolite repression of L-rhamnose dehydrogenase in Pullularia pullulans.

The growth of Pullularia pullulans on L-rhamnose (6-deoxy-L-mannose) as the sole carbon source induces the synthesis of L-rhamnose dehydrogenase, a nicotinamide adenine dinucleotide-dependent enzyme that catalyzes the oxidation of the deoxy sugar to L-rhamnonolactone. The enzyme induction is inhibited by cycloheximide, suggesting de novo synthesis. The presence of d-glucose (0.2%) or D-galactose (0.2%) simultaneously with the inducer in the induction medium produced 50% repression of dehydrogenase synthesis, but no effect was detected with D-fructose and D-mannose at the same concentration. High levels of D-glucose (2%), under maximal catabolite repression conditions, produced a complete inhibition of enzyme synthesis.

Chitin synthesis in Triatoma infestans and other insects.

In vivo and in vitro synthesis of chitin in Triatoma infestans was studied. For in vivo experiments, [14C] sugars were injected through the abdominal wall. Maximal incorporation of radioactivity into the cuticle was attained immediately after the ecdysis. The identification of in vivo synthesized chitin was performed by the enzymatic hydrolysis of the alkali-insoluble material from the cuticle with Helix chitinase. The main water-soluble compound obtained was N-acetylglucosamine as demonstrated by chromatographic procedures. In vitro synthesis of chitin was carried out with enzymatic crude extracts from Triatoma infestans, and UDP-N-acetylglucosamine was used as "source" of glycosyl moieties. Higher amounts of [14C] N-acetylglucosamine incorporation to chitin than those previously reported by others, were obtained. The identity of the product was confirmed in a similar way as that from in vivo synthesis. Radioactivity was also found in a liposoluble fraction concomitantly with chitin synthesis. This compound had an anionic behavior, was acid labile and had similar chromatographic properties as dolichol pyrophosphate N-acetylglucosamine obtained with pig liver extracts. Knowledge about dolichol phosphate sugars mediated glycoprotein synthesis in eukaryotes, suggests the involvement of this type of N-acetylglucosaminyl-phospholipid in macromolecule "building" even in insects.
