fos-lacZ transgenic mice: mapping sites of gene induction in the central nervous system.

A transgenic mouse line containing a fos-lacZ fusion gene was derived in which beta-galactosidase activity identified cell populations expressing fos either constitutively or after stimulation. Seizures and light pulses induced nuclear lacZ activity in defined populations of neurons in vivo, and an array of neurotransmitters, including glutamate, induced the transgene in primary brain cultures. In unstimulated mice, the major sites of fos-lacZ expression were skin, hair follicle, and bone. fos-lacZ mice provide a new avenue for activity mapping studies based on gene expression.

The RL neurovirulence locus in herpes simplex virus type 2 strain HG52 plays no role in latency.

We have demonstrated that the variant JH2604 of the herpes simplex virus type 2 (HSV-2) strain HG52 is completely avirulent in BALB/c mice following intracranial inoculation, with an LD50 of greater than 10(7) p.f.u./mouse compared to the wild-type LD50 of less than 10(2) p.f.u./mouse. In JH2604, a 1.5 kbp deletion extends from the DR1/Ub junction of the 'a' sequence to 511 bp upstream of the 5' end of IE1 in both long repeats. We have since constructed a second variant (2701) in which only 850 bp are removed from the RL. This deletion lies entirely within the sequences deleted in JH2604 and leaves intact most of a short 189 bp open reading frame (ORF) highly conserved between HSV-1 and HSV-2. Like JH2604, 2701 shows wild-type growth characteristics and is neither host range- nor temperature-restricted. This was most noteworthy in the case of mouse 3T6 cells. 2701 has an LD50 of 5 x 10(5) p.f.u./mouse on intracranial inoculation, a value intermediate between those of HG52 and JH2604. In assays for intracranial replication, JH2604 exhibits no detectable growth with a rapid decline in virus titre, 2701 shows limited growth over the first 24 to 36 h post-inoculation before the titre again declines and HG52 grows rapidly, reaching a high titre until the mice die. Taken together these results suggest that a region of the genome upstream of IE1 encodes a gene product essential for HSV replication in neurons of the central nervous system. It is highly likely that the conserved ORF is in an important region of a polypeptide essential for neurovirulence, although the upstream sequences present in 2701 but absent from JH2604 must also play a role. Although JH2604 and 2701 are avirulent, they both establish latent infection in the dorsal root ganglia of BALB/c mice and reactivate in vitro in a manner indistinguishable from HG52. This suggests a distinct separation of the factors involved in neurovirulence and the establishment of/reactivation from latency.

Campylobacter enteritis associated with consumption of unpasteurised milk.

In October and November 1978 two outbreaks of enteritis occurred in the north of England. Symptoms lasted two to over eight days but in no case necessitated admission to hospital. Faecal specimens from most of the patients were found to contain thermophilic Campylobacter sp. Inquiry disclosed that all patients had consumed unpasteurised milk from local farms. Examination of rectal swabs from the cattle concerned and milk socks yielded strains of Campylobacter sp indistinguishable from those isolated from the patients. It was therefore concluded that, since campylobacters are not known to be excreted in milk, faecal contamination of the milk had probably occurred and had led to these outbreaks. Evidence suggests that thermophilic Campylobacter sp is an occasional contaminant of milk. So long as unpasteurised milk continues to be distributed further outbreaks will probably occur.

Adolescent blood pressure in Richmond, Virginia, schools.

The blood pressure of 3,166 adolescents was measured periodically in Richmond during the period 1974-1977. The subjects were students in public and private schools and ranged in age from 14 to 18 years. Analysis of the readings showed minimal differences relating to age, sex, school and race but a strong correlation with obesity. Only two organic renal causes of hypertension were discovered. A 3 1/2-year followup of 902 students indicated significant tracking, with a correlation coefficient of .385 for systolic and .305 for diastolic pressure. At the conclusion of the study, 340 students (10.7%) were identified as needing followup for hypertension.

Adolescent

A simple method for determining bacterial susceptibility to aminoglycosides.

A method is described of determining the susceptibility of enterobacteria to four amino-glycosides by means of an antibiotic diffusion method. The results are exose obtained by a method that involved titration in agar plates and the use of a multi-point inoculator. A statistical analysis showed that the diffusion method gave accurate reproducible results. The antibiotic can be incorporated into paper disks, paper strips or agar ditches and the calculation converting the measured zones of inhibition to MIC values is easily performed. The appropriate equation is derived from another--obtained from observations on a single strain of known sensitivity--describing the diffusion of the antibiotic from the reservoir.

Anti-Bacterial Agents

Sensitivity of Brucella spp to tetracycline and its analogues.

The sensitivity to eight tetracyclines of 100 strains of brucellae, comprising strains of Brucella abortus, Br. melitensis, and Br. suis, was determined. Demethylchlortetracycline was the most effecitve against all the groups of brucellae, whereas oxytetracycline and chlortetracycline were the least effective. The mean MIC value for demethylchlortetracycline, doxycycline, lymecycline, and tetracycline was less than or equal to 1 mug/ml. Strains of Br. abortus biotype 2 and Br. suis were the most sensitive strains examined.

Brucella

Complete repair of tetralogy associated with sickle cell anemia and G-6-PD deficiency.

The case of a 5-year-old black girl with sickle cell anemia and G-6-PD deficiency is described. The child underwent successful repair of tetralogy of Fallot. Open-heart surgery can be safely performed on these patients if certain guidelines are observed, particularly the avoidance of hypoxia, hypothermia, acidosis, and dehydration. The patients should be prepared for the operation with transfusion of normal red cells. Routine preoperative testing for the above hemoglobinopathologic conditions is urged. The pertinent literature is reviewed.

Serum antibody response in acute brucellosis.

The serum antibody titres in 62 cases of acute brucellosis are presented. The antibody persisiting after infection and the appearance of non-agglutinating anti-human globulin titres were investigated. The criteria for the serological diagnosis of acute and chronic brucellosis are discussed.

Psychopharmacologic investigations in elderly volunteers: Effect of diazepam in males.

Forty male volunteers over the age of 60 took either a daily dose of diazepam (12 mg) or placebo for a two-week period. Differential responses to these compounds partially depended upon age and the initial symptom level. Diazepam produced sedative side effects, a modest antidepressant effect, and a reduction in memory. Placebo had an antianxiety effect in the relatively older subjects, and was associated with decreased fatique, improved memory and improved motor function. Considerable variability was observed in both drug and placebo responses among the elderly. A self-rating depression scale (D30R) was useful for assessing changes in affective states and in discriminating between the effects of active drug and placebo.

Aged

A screening method for the detection of Brucella antibodies in human serum.

The antiglobulin consumption test of Steffen (1954) has been adapted for the detection of Brucella antibodies in human serum. This screening method takes approximately two hours to complete and sera which give a negative result need no further investigations, whilst positive sera should be fully tested by the conventional Brucella serology technique.
