[Effect of paleocerebellar cortex electrical stimulation on generalized penicillin-induced seizure activity in rats].

It was shown that low-frequency electrical stimulation (ES) (10-12 Hz, 0.5 ms) of paleocerebellar cortex (nodulus, uvula) is followed by activation of spike discharges which were induced via i.p. administration of sodium benzilpenicillin salt to alert Wistar rats (300,000 IU/kg). The facilitation of ictal discharges generation was also seen in the course of such an ES. High-frequency (100-300 Hz, 0.25 ms) ES of the same structure was followed by suppression of spike discharges and prevented the ictal discharges precipitation. Antiseizure action was obvious under condition of relatively low level of epileptic activity generation. In the course of such ES the decreasing frequency and amplitude of spike discharges were noted during interstimuli periods. Besides, the life-span of seizure activity was shortened as well. The repeated paleocerebellar ES made after electrocoagulation of irritated tissue did not cause modulations of seizure activity.

[Effects of L-dopa and transcranial magnetic stimulation on behavioral reactions in kindled rats].

In acute experiment in rats, the chronic epileptogenesis was reproduced in the form of pharmacological kindling induced via repeated picrotoxin administrations (1.0-1.2 mg/kg, intraperitoneally). A reduction of exploratory behavior was shown in the early period of kindling (24 h as of the moment of the last epileptogen administration). The marked alleviation of these disturbances was registered in two weeks from the moment of cessation of kindled irritations. L-DOPA (100 mg/kg, intraperitoneally) and transcranial magnetic stimulation (20 impulses with an induction at the height of their development of 1.5 TI) was followed by the net increasing exploratory, sexual and eating behavior. This is in favour of regarding the activation of dopaminergic system as a mechanism of action of transcranial magnetic stimulation upon kindling-induced behavioral deterioration.

[The effect of the intranigral use of the delta sleep-inducing peptide and its analogs on the motor and seizure activities of rats].

The effects of delta-sleep inducing peptide (DSIP) and its analogues (1-4) administered into substantia nigra pars reticulata on locomotor and seizure activity were estimated in experiments in rats. It was shown that intranigral microinjection of DSIP as well as DSIP-1-DSIP-4 caused decreasing of horizontal, vertical locomotor activity and attendance of central sectors of the "open field". Antiseizure effects, i.e. the first and clonic-tonic picrotoxin-induced convulsive latent period lengthening and their intensity decreasing, revealed DSIP, DSIP-2 and DSIP-3. Authors suppose that changes of DSIP structure lead to changes of effects expression on locomotor and seizure activity in conditions of their administration into substantia nigra reticulata. Obtained data concerning protective effects of studied peptides on experimental seizure syndrome allow to conclude that there is interaction between DSIP-induced hypokinesia and DSIP analogues anticonvulsive effectiveness in case of their intranigral administration which is likelihood is one of the component of nigral-dependent seizure-suppressive mechanism.

[The role of the delta sleep-inducing peptide in the formation of neuropathological syndromes].

The authors considered the pathogenetic role of delta-sleep inducing peptide (DSIP) in different neuropathological syndromes development and manifestation. According to own as well as published in the literature data authors showed the parkinsonian and the rotational syndromes development following DSIP central administration. Briefly, DSIP is a neuropeptide which play significant role in the mechanisms of development of different neuropathological syndromes, namely, epileptic, parkinsonian, withdrawal, rotational and others syndromes.

[The effect of the electrical stimulation of the superior colliculli on focal epileptic activity in the cerebral cortex ].

Acute experiments in rats showed that SC electrical stimulation (ES) (100 Hz, 5 V, 0.25 ms) resulted in suppression of the epileptic activity in contralateral strychnine-induced (0.1%; 1.0%) focus and shortening of the focus existence time. After electrocoagulation of SC ES-zone the EpA in the focus was restored. ES of SC ipsilateral to strychnine-induced (0.1%) focus caused an increase in the spike frequencies but significant part of the antiepileptic system.

[Synthesis and biological properties of new analogs of delta-sleep peptide. I. Antiepileptic effect].

To study structure-functional relationship in the series of DSIP, eleven DSIP analogues varying in positions 1, 2 and 6 were synthesized by the solid-phase method using both Boc- and Fmoc-approaches. The antiepileptic action of these analogues was compared with that of DSIP. The seizure activity was induced by the corazol or picrotoxin i. p. injection. Some analogues proved more efficient as antiepileptic agents than DSIP after their central (50 micrograms/kg in rats) and peripheral (1.0 mg/kg in mice) administration.

[The effect of the delta sleep-inducing peptide on seizure activity].

The delta-sleep-inducing peptide (DSIP) suppressed seizure activity in the cat cortical strychnine-induced seizure foci. The DSIP delayed development of corazol kindling in rats, prevented seizure induced with bicucullin and other agents in mice. The DSIP effect was shown to be realised through the action upon reticular black substance. The DSIP seems to take part in endogenous control of the brain excitability.

[The effect of activation of the superior colliculus on the convulsive activity during picrotoxin-induced kindling].

Bilateral administration of picrotoxin, bicucullin or benzilpenicillin to the colliculi superior induced high-amplitude hypersynchronous discharges in rats. In these conditions, a suppression of generalized seizures induced with picrotoxin, was observed. The superior colliculi seem to play a significant part in antiepileptic system, the part being realized through generator mechanism.

[Effects of intranasally administered substance P in parkinsonian syndrome].

The effect of intranasal substance P injection on parkinsonian syndrome and the generator of pathologically enhanced excitation (GPEE) in caudate nuclei (CN) was investigated. MPTP or reserpine administration in old rats induced oligokinesia, rigidity and tremor followed by the high amplitude slow and rapid waves in both CN. The bilateral intranasal injection of substance P (25 micrograms/kg) resulted in an increase in motor activity and almost completely abolished the rigidity and tremor. The reduction of extrapyramidal symptoms was considered as a result of the inhibition of GPEE in CN. The possibility of substance P entry from nasal cavity into the brain was discussed. The changes of the substance P balance in nigrostriatal system was suggested to be on of the pathogenetic links of parkinsonian syndrome.

[Effects of neurotropin on seizure activity in picrotoxin kindling].

It is shown that both intracerebral and intraperitoneal neurotropin administration resulted in a decrease of seizure susceptibility of preliminary picrotoxin--kindled rats. On the other hand, neurotropin did not change the course of kindling development. Under conditions of acute picrotoxin--induced seizures it was observed that preliminary cycloheximide (protein-synthesis blocker) administration abolished anticonvulsant properties of neurotropin. It is concluded that anticonvulsant effects of neurotropin are realized via modulation of endogenous peptides synthesis and, in particular, cerulein.

[The role of the opiate mechanisms of the hippocampus and substantia nigra in the behavioral and convulsive disorders in picrotoxin-induced kindling].

It was shown in the experiments on rats that the repeated picrotoxin administration resulted in the kindling of generalized seizures. Generalized convulsions were followed by the development of either postictal depression or explosiveness. The injection of mu-opiate agonist met-enkephalin into hippocampus of kindled rats resulted in the increase in the severity of seizure reactions which were induced by picrotoxin and also in the increase in the number of animals with postictal explosiveness. The injection of dynorphin-A-1-13 (kappa-opiate agonist) into substantia nigra reticulata induced the locomotor depression which was like one in postictal period and resulted in the decrease of picrotoxin-induced seizures severity. It was concluded that mu-opiate system of hippocampus took part in the formation of generator of pathologically enhanced excitation in the structure during kindling and the development of seizure syndrome, providing also the postictal explosiveness. Kappa-opiate system of substantia nigra plays an important role in the activation of the antiepileptic system, limitation of seizures and the development of postictal depression.

[Effects of activation and lesions of the superior colliculi of the lamina quadrigemina on epileptic activity in rats].

In acute experiments on rats it was shown that activation of SC by bilateral microinjection of penicillin (5-15 IU) or bicuculline (25-50 ng) resulted in the increased latency and decreased severity of i.p. picrotoxin-induced seizures (2 mg/kg). The suppression of behavioral convulsions and the decreased epileptic activity in the hippocampus and cerebral cortex were followed by occurrence of spike discharges in SC with an amplitude of 200-500 mcV and frequency of 5-12/sec which testifies to the formation of penicillin- or bicuculline-induced generator excitation in SC. The lesions of SC by kainic acid administration resulted in the decreased seizure threshold and, also, facilitated the development of seizure under conditions of picrotoxin kindling. The conclusion is made that SC activation plays an important role in the realization of functional integrative activity of the antiepileptic system.

[The formation of motor programs during pharmacological kindling].

In Wistar rats with the kindling phenomenon, the number of animals with three or more not stimulus-directed forms of swimming was increased. Obvious changes of swimming behaviour in these animals were also observed after intrastriatal injections of carbachol, apomorphine, haloperidol, muscimol and picrotoxin. These findings are discussed from the standpoint of a latent generator of pathologically enhanced excitation in the striatum which changed the ability of these rats to develop behavioral programs.
