Selective inactivation by chlorofluoroacetamides of the major phenobarbital-inducible form(s) of rat liver cytochrome P-450.

Five N-monosubstituted chlorofluoroacetamides have been tested as potential specific irreversible inhibitors of the major phenobarbital-inducible form of rat liver cytochrome P-450 (P450IIB1). In vitro, N-(2-phenethyl)chlorofluoroacetamide was ineffective in causing a time-dependent loss of P450IIB1-mediated androstenedione 16 beta-hydroxylase activity in liver microsomes from phenobarbital-treated rats. However, addition of a nitro or bromo substitutent at the para position of the phenyl group or addition of a second phenyl group at the 1- or 2-position on the phenethyl side chain yielded compounds that caused a selective time-dependent decrease in androstenedione 16 beta-hydroxylase activity relative to four other P-450 form-specific androstenedione or progesterone hydroxylase activities monitored. The two compounds that were the most effective in inactivating P450IIB1 in vitro, N-(2-p-bromophenethyl) and N-(2-p-nitrophenethyl)chlorofluoroacetamide were also administered ip to phenobarbital-treated rats, and inhibition of cytochromes P-450 was assessed by in vitro assays of steroid and R- and S-warfarin hydroxylation in subsequently prepared hepatic microsomes. Both compounds selectively inhibited P450IIB1, and at a dose (200 mg/kg) of N-(2-p-nitrophenethyl)chlorofluoroacetamide that reduced androstenedione 16 beta-hydroxylase activity to approximately one-third of the control level, only two other activities, both attributable to P450IIB1, were decreased. In contrast, steroid and warfarin hydroxylase activities indicative of at least five other cytochromes P-450 were unaffected by the compound. These results indicate the feasibility of an empirical approach to the design of specific cytochrome P-450 inactivators.

Comparison of monooxygenase activities and cytochrome P-450 isozyme concentrations in human liver microsomes.

Concentrations of three human liver microsomal cytochrome P-450 isozymes and 20 different monooxygenase activities were determined in human liver microsomal preparations. The results of correlation analysis suggest that: there are important variations in the amounts of the three cytochrome P-450 isozymes measured, particularly P-450(8) and P-450(9); aldrin epoxidase, d-benzphetamine N-demethylase, and S-warfarin 4-hydroxylase activities are linked to cytochrome P-450(5); aryl hydrocarbon (benzo(a)pyrene) hydroxylase and 4-nitroanisole-O-demethylase activities are linked to P-450(8); hydroxylations at the 4'-, 6-, 7-, and 8-positions of R-warfarin are closely linked to each other but are not correlated with other measured monooxygenase activities or P-450 isozyme levels; and P-450(9) is not related to any of the catalytic activities tested. Thus, certain monooxygenase activities can be attributed to specific cytochrome P-450 isozymes. This approach should be useful in suggesting the roles of different cytochromes P-450 in drug metabolism in man which can be further examined using in vitro and in vivo methods.

Human hepatic cytochrome P-450 composition as probed by in vitro microsomal metabolism of warfarin.

Human liver microsomal fractions from 27 renal donors (tissue obtained post mortem) and from six cancer patients (tissue obtained during surgery) were used to investigate human hepatic cytochrome P-450 isozyme compositions. In vitro microsomal metabolism of the R and S enantiomers of warfarin to dehydrowarfarin and 4'-, 6-, 7-, 8-, and 10-hydroxywarfarin is catalyzed by cytochrome P-450 isozymes and was used as the basis for evaluating similarities and differences between human cytochrome P-450 isozyme compositions. The mean hepatic cytochrome P-450 concentration from postmortem samples was not significantly different from that of surgical patients (0.51 +/- 0.16 vs. 0.35 +/- 0.14 nmol/mg protein), but the NADPH-cytochrome P-450 reductase activity of the former was significantly higher than that of the latter (141 +/- 56 vs. 29 +/- 6 nmol cytochrome c reduced/min/mg protein). In general, the microsomal preparations were overall stereoselective for R warfarin metabolism. The stereoselectivities for formation of the individual metabolites of the R enantiomer were 6-, 8-, and 10-hydroxywarfarin and the S enantiomer were 4'- and 7-hydroxywarfarin. Of the 33 microsomal preparations, 21 exhibited qualitatively similar warfarin metabolite profiles with 6R- and 7S-hydroxywarfarin having the highest formation rates. Some of the preparations exhibited markedly different metabolite profiles, the most notable having 10R-hydroxywarfarin as the major metabolite. Based on the known warfarin metabolite profiles of five purified cytochrome P-450 isozymes, the isozyme composition of the microsomes can be estimated. The majority of the microsomal preparations apparently had similar isozyme compositions but some preparations were markedly different.
