Human salivary arginase and its deficiency in argininaemia.

Arginase from normal human mixed saliva was characterized. The enzyme was completely activated after a preincubation of 20 min at 55 degrees C and a Mn2+ final concentration of 5 mmol/l. The pH optimum was 9.6-9.8, and the Km for L-arginine was 4.2 +/- 0.7 mmol/l. In normal saliva only one form was found, which was chromatographically identical with the cationic form of arginase in liver and blood cells. Salivary arginase was completely precipitated by rabbit antiserum against human liver arginase. Arginase activity was not detectable in the saliva of patients suffering from argininaemia. Enzyme activities in the saliva of the heterozygous parents and the unaffected daughter were 0.08, 0.07 and 0.12 U/mg protein, respectively, whereas the activities in the saliva of 60 healthy adults and 8 children were 0.17 +/- 0.11 and 0.16 +/- 0.06 U/mg protein, respectively.

[Glycosylation of hemoglobin and plasma proteins in petrochemical plant workers].

The concentration of glycosylated hemoglobin and (plasma) proteins has been measured in 111 workers of 6 MZRiP departments in Płock and in 54 healthy people. In all subjects the mean concentrations of glycosylated hemoglobin and glycosylated plasma proteins have been in so called "wide range of normal values". Significant shifts of glycosylated Hb concentrations have been found in two departments--those of ethylenederivatives and distillation. The concentration of glycosylated plasma proteins has been elevated only in workers of the Catalytic Processes Department.

[Usefulness of a modified screening method for the detection of phenol in the saliva in evaluating occupational exposure to this compound].

So far there has not been any quick and simple method to estimate phenol exposure or phenol poisoning directly at the workstation. Our work was aiming at a modification of Lutogniewska's method of determining phenol in the saliva as an indicator of phenol exposure at the workplace. The original method has been improved and modified through essential modification, consisting in a stable borax prealkalization of blotting--paper where the saliva is deposited, which results in a more intense and fast colouring. Apart from original 2,6-dibromchinochlorimid detector, 2,6-dichlorchinochlorimid has been tested. The newly--developed technique has been used in field studies related to 19 workers in one of petrochemical plants and a control group of 40 volunteer students. The proposed method is easy to apply and convenient both for those carrying out the studies and those subjected to them. Nearly in all workers tested, after 6 hours' exposure slight traces of phenol (2 mg/100 ml saliva) have been found in the saliva. On the other hand, in those with no exposure to phenol the test was negative. The method seems to be valuable for the practical use in field studies. The findings of this tentative paper should be checked on more extensive materials.

Intestinal arginase in vertebrates and invertebrates.

1. Arginase was found to be present in the intestine in all species of Annelida, Arthropoda and Chordata studied. 2. The activity of intestinal arginase differs from species to species, the differences reaching two orders of magnitude (100 x). 3. The highest activity of intestinal arginase was observed in the rodents (mouse, rat, hamster). 4. In animals in which the enzyme activity was high or moderately high, arginase activity showed topographical differentiation along the long axis of the intestine.

A simple, rapid and accurate screening test for hyperchlororhoea.

A simple, rapid screening paper test for the quantitative estimation of Cl- concentration in feces is described. The measured time of decolorisation of the Chlorotrex indicator by aqueous extract of feces is inversely proportional to the chloride concentration. The technical details of the test are given. The test is very suitable for laboratory and mass screening.

A new convenient, rapid and accurate method for determination of total lipids in feces.

1. A new, simple and rapid colorimetric micromethod for determination of total lipids in the petroleum ether extract of feces is proposed. The method, based on the sulfophosphovanillin reaction, was found to be of satisfactory precision and accuracy. 2. The concentration of total lipids determined in meconium of 40 healthy newborns in the first five days of life, was 1 to 15 g/100 ml of feces.
