Retinoids induce MMP-9 expression through RARalpha during mammary gland remodeling.

Retinoic acid (RA) is a signaling molecule in the morphogenesis of the mammary gland, modulating the expression of matrix metalloproteinases (MMPs). The aim of this paper was to study the role of RA during weaning, which consists of three events: apoptosis of the secretory cells, degradation of the extracellular matrix, and adipogenesis. CRABP II and CRBP-1 carrier proteins increased significantly during weaning compared with lactating glands but reverted to control values after the litter resuckled. The effects of RA are mediated by the nuclear receptors RARalpha, RARbeta, RARgamma, and RXRalpha, which underwent an increase in protein levels during weaning. In an attempt to elucidate the RARalpha-dependent signaling pathway, ChIP assays were performed. The results showed the binding of RARalpha to the MMP-9 promoter after 24- and 72-h weaning together with its coactivator p300; this fact could be responsible for the increase found in MMP-9 mRNA and protein levels in these conditions. Expression of related MMPs (MMP-2 and MMP-3) was also increased during weaning. Using gelatine zymography, we observed a time-dependent increase in active forms of MMP-9 and MMP-2. On the other hand, the inhibitor of MMPs, TIMP-1, was almost undetectable at 24- and 72-h weaning by Western blot. The role of retinoids in matrix remodeling is reinforced by the fact that administration of an acute dose of retinol palmitate to control lactating rats also induces MMP-9 expression. This emphasizes the importance of retinoids in vivo to regulate mammary gland involution.

The relationship between specific tissue lesions and pain severity in persons with knee osteoarthritis.

INTRODUCTION: Pain is the most common symptom in knee osteoarthritis (OA), a leading cause of chronic disability, and a major source of the disability attributable to OA in general. Pain severity in knee OA is variable, ranging from barely perceptible to immobilizing. The knee lesions that contribute to pain severity have received little attention. OBJECTIVE: To examine whether worse pathology of specific knee tissues - i.e. cartilage, bone (attrition, cysts, bone marrow lesions, and osteophytes), menisci (tears and subluxation), ligaments, and synovium (synovitis/effusion) - is associated with more severe knee pain. METHODS: One hundred and forty-three individuals were recruited from the community with primary (idiopathic) knee OA, with definite tibiofemoral osteophytes in at least one knee, and at least some difficulty with knee-requiring activity. Knee magnetic resonance (MR) images were acquired using coronal T1-weighted spin-echo (SE), sagittal fat-suppressed dual-echo turbo SE, and axial and coronal fat-suppressed, T1-weighted 3D-fast low angle shot (FLASH) sequences. The whole-organ magnetic resonance imaging (MRI) scoring (WORMS) method was used to score knee tissue status. Since summing tissue scores across the entire joint, including regions free of disease, may dilute the ability to detect a true relationship between that tissue and pain severity, we used the score from the worst compartment (i.e. with the poorest cartilage morphology) as our primary approach. Knee pain severity was measured using knee-specific, 100 mm visual analogue scales. In analyses to evaluate the relationship between knee pain severity and lesion score, median quantile regression was used, adjusting for age and body mass index (BMI), in which a 95% CI excluding 0 is significant. RESULTS: The increase in median pain from median quantile regression, adjusting for age and BMI, was significant for bone attrition (1.91, 95% confidence interval (CI) 0.68, 3.13), bone marrow lesions (3.72, 95% CI 1.76, 5.68), meniscal tears (1.99, 95% CI 0.60, 3.38), and grade 2 or 3 synovitis/effusion vs grade 0 (9.82, 95% CI 0.38, 19.27). The relationship with pain severity was of borderline significance for osteophytes and cartilage morphology and was not significant for bone cysts or meniscal subluxation. Ligament tears were too infrequent for meaningful analysis. When compared to the pain severity in knees with high scores for both bone attrition and bone marrow lesions (median pain severity 40 mm), knees with high attrition alone (30 mm) were not significantly different, but knees with high bone marrow lesion without high attrition scores (15 mm) were significantly less painful. CONCLUSION: In persons with knee OA, knee pain severity was associated with subarticular bone attrition, bone marrow lesions, synovitis/effusion, and meniscal tears. The contribution of bone marrow lesions to pain severity appeared to require the presence of bone attrition.

Environmental evidence of fossil fuel pollution in Laguna Chica de San Pedro lake sediments (Central Chile).

This paper describes lake sediment spheroidal carbonaceous particle (SCP) profiles from Laguna Chica San Pedro, located in the Biobío Region, Chile (36 degrees 51' S, 73 degrees 05' W). The earliest presence of SCPs was found at 16 cm depth, corresponding to the 1915-1937 period, at the very onset of industrial activities in the study area. No SCPs were found at lower depths. SCP concentrations in Laguna Chica San Pedro lake sediments were directly related to local industrial activities. Moreover, no SCPs were found in Galletué lake (38 degrees 41' S, 71 degrees 17.5' W), a pristine high mountain water body used here as a reference site, suggesting that contribution from long distance atmospheric transport could be neglected, unlike published data from remote Northern Hemisphere lakes. These results are the first SCP sediment profiles from Chile, showing a direct relationship with fossil fuel consumption in the region. Cores were dated using the 210Pb technique.

New limit on the neutrinoless betabeta decay of 130Te.

We report the present results of CUORICINO, a search for neutrinoless double-beta (0nu betabeta) decay of 130Te. The detector is an array of 62 TeO2 bolometers with a total active mass of 40.7 kg. The array is cooled by a dilution refrigerator shielded from environmental radioactivity and energetic neutrons, operated at approximately 8 mK in the Gran Sasso Underground Laboratory. No evidence for (0nu betabeta) decay was found and a new lower limit, T(1/2)(0nu) > or = 1.8 x 10(24) yr (90% C.L.) is set, corresponding to [m(nu)] < or = 0.2 to 1.1 eV, depending on the theoretical nuclear matrix elements used in the analysis.

Role of GSH in the modulation of NOS-2 expression in the weaned mammary gland.

GSH delivery to the lactating mammary gland is essential for the maintenance of lactation as its decrease leads to apoptosis and involution of the mammary gland. In fact, it has already been demonstrated that some of the changes in gene expression found in the lactating mammary gland after forced weaning are reproduced in rats treated with buthionine sulphoximine to deplete GSH levels. An oligonucleotide microarray experiment would give us a better knowledge of the mRNA expression patterns during lactation and after weaning and the possible functions of GSH in the modulation of these events.

The impact of reduction in the benzene limit value in gasoline on airborne benzene, toluene and xylenes levels.

Background benzene, toluene, xylenes (BTX) average concentrations have been measured over the urban agglomeration of Toulouse, France, during both springtime and summer periods of 1999 and 2001. The benzene average amount over the two Toulouse campaigns in 1999 is equal to 2.2 microg/m(3), very close to the French air quality standard and well under the average value of 5 microg/m(3) recommended by European Economic Community countries, recognising that those regulations are given for a whole year. BTX pollution over Toulouse has, in particular, been produced by motor vehicle exhaust gases. For the study conducted during the same periods of 2001, benzene concentrations were within the French quality value in the whole area. This is because the benzene limit value contained in gasoline went from 5% to 1% since 2000 January 1. It will be important to measure benzene over annual periods in order to know its exact values over such a period and to observe its potential seasonal variations.

Assessment of clonal relationships in ipsilateral and bilateral multiple breast carcinomas by comparative genomic hybridisation and hierarchical clustering analysis.

The issue of whether multiple, ipsilateral or bilateral, breast carcinomas represent multiple primary tumours or dissemination of a single carcinomatous process has been difficult to resolve, especially for individual patients. We have addressed the problem by comparative genomic hybridisation analysis of 26 tumours from 12 breast cancer patients with multiple ipsilateral and/or bilateral carcinoma lesions. Genomic imbalances were detected in 25 of the 26 (96%) tumours. Using the genomic imbalances detected in these 26 lesions as well as those previously found by us in an independent series of 35 unifocal breast carcinomas, we compared a probabilistic model for likelihood of independence with unsupervised hierarchical clustering methodologies to determine the clonal relatedness of multiple tumours in breast cancer patients. We conclude that CGH analysis of multiple breast carcinomas followed by unsupervised hierarchical clustering of the genomic imbalances is more reliable than previous criteria to determine the tumours' clonal relationship in individual patients, that most ipsilateral breast carcinomas arise through intramammary spreading of a single breast cancer, and that most patients with bilateral breast carcinomas have two different diseases.

Bacteremia by Streptobacillus moniliformis: first case described in Spain.

Described here is the case of an 87-year-old man who developed fever, chills and discomfort caused by Streptobacillus moniliformis. This pathogen is one of the causes of rat-bite fever, an uncommon bacterial illness transmitted through a bite or scratch from a rodent or the ingestion of food or water contaminated with rat faeces. Cases of rat-bite fever are rarely reported in Spain. The patient reported no history of rat bite or rodent contact, and the only known risk factor was contact with a dog and a cat that were kept as pets. Streptobacillus moniliformis was isolated in two sets of blood cultures. This case represents what is believed to be the first report of bacteremia due to Streptobacillus moniliformis in Spain.

A comparison of AT1 angiotensin II antagonists at pre- and postjunctional angiotensin II receptors of the rat tail artery.

A comparison was made of the influence of candesartan, ZD7155, losartan and eprosartan on angiotensin II effects at pre- and postjunctional AT(1) receptors of the rat tail artery. To study the anti-angiotensin II effect at prejunctional receptors, the tissues were preincubated with [(3)H]noradrenaline and then superfused and electrically stimulated (1 Hz, 2 ms, 50 mA, during 5 min); to study the angiotensin II effect at postjunctional receptors, non-cumulative concentration-response curves to angiotensin II were determined in the absence and in the presence of the antagonist. p A(2) values were calculated for competitive antagonists and p D'(2) values for insurmountable antagonists. At the prejunctional level, losartan and eprosartan displayed competitive antagonism with p A(2) values of 6.50 and 8.08, respectively, whereas candesartan and ZD7155 displayed non-competitive antagonism with p D'(2) values of 8.71 and 7.98, respectively. At the postjunctional level, the four antagonists displayed the same kind of antagonism as prejunctionally with p A(2) values for losartan and eprosartan of 8.52 and 8.22, respectively, and p D'(2) values of 10.62 and 9.01, for candesartan and ZD7155, respectively. The ratios between post- and prejunctional potencies were: losartan 101, candesartan 81, ZD7155 11, and eprosartan 1.4. We conclude that, at least functionally, pre- and postjunctional angiotensin II AT(1) receptors are different and propose that the prejunctional receptors in this tissue belong to the AT(1B)-subtype.

Wound Infection due to Vibrio vulnificus in Spain.

Vibrio vulnificus is a gram-negative rod that can cause septicaemia and skin lesions, usually in patients with underlying illnesses such as chronic liver disease or diabetes mellitus. Infections caused by this bacterium are unusual in Spain. A case of skin infection due to Vibrio vulnificus is reported in a patient whose abraded skin on his left leg came into contact with seawater. The patient died suddenly, probably due to septicaemia or bacteraemia caused by this organism. Vibrio vulnificus infection must be considered in the differential diagnosis of septicaemia, skin lesions and wound infections, particularly when a patient reports a history of contact with seawater.

High-speed, multicolor fluorescent two-dimensional gene scanning.

Two-dimensional gene scanning (TDGS) is a method for analyzing multiple DNA fragments in parallel for all possible sequence variations, using extensive multiplex PCR and two-dimensional electrophoretic separation on the basis of size and melting temperature. High throughput application of TDGS is limited by the prolonged time periods necessary to complete the second-dimension electrophoretic separation step--denaturing gradient gel electrophoresis--and the current need for gel staining. To address these problems, we constructed a high-voltage, automatic, two-dimensional electrophoresis system and used this in combination with thinner gels to reduce two-dimensional electrophoresis time about 80%. Instead of gel staining, we used three different fluorophores to simultaneously analyze three samples in the same gel. These improvements greatly increase TDGS speed and throughput and make the method highly suitable for large-scale single-nucleotide polymorphism discovery and genetic testing.

Long-term follow-up and predictors of clinical outcome in obsessive-compulsive patients treated with serotonin reuptake inhibitors and behavioral therapy.

BACKGROUND: The objective of this study was to examine the long-term course of obsessive-compulsive disorder (OCD) in patients treated with serotonin reuptake inhibitors (SRIs) and behavioral therapy and to identify predictors of clinical outcome. METHOD: Sixty outpatients meeting DSM-II-R or DSM-IV criteria for OCD were followed up for 1 to 5 years (mean = 2.5 years). All of them received prolonged pharmacologic therapy with an SRI. RESULTS: Thirty-seven patients (61.7%) completed an adequate behavioral treatment. At long-term assessment, 22 patients (36.7%) exhibited a global Yale-Brown Obsessive Compulsive Scale (Y-BOCS) score greater than 16 or a final reduction in Y-BOCS global score of less than 35% and were considered nonresponders. Patients who completed behavioral therapy showed a significant decrease in Y-BOCS compulsions subscale score (p = .01), whereas no significant differences in either Y-BOCS global or obsessions subscale scores between those who did and those who did not undergo behavioral therapy were detected. Obsessions of sexual/religious content were the unique factor related to a poorer long-term outcome. CONCLUSION: A substantial number of OCD patients showed persistent disabling symptoms at the long-term follow-up in spite of combined pharmacologic and behavioral treatment. Major benefits from behavioral therapy appeared to be the improvement of ritualistic behaviors. Sexual/religious obsessions predicted poorer long-term outcome, whereas short-term response to SRI treatment failed to achieve predictive value in the long-term course of OCD.
