[Interleukin system in schizophrenics].

Interleukines (IL) levels, mononuclear phagocytes (MP) activity and proliferative T-lymphocytes activity were studied in 6 patients with paranoid (ICD-10 item F20.00) and 20 patients with shift-like (F20.01) types of schizophrenia. Significant (p < 0.05) decrease of MP activity, monocytes-stimulated IL-1 beta production (mostly pronounced in paranoid type) lymphocyte-stimulated IL-2 production (p < 0.05) and proliferate T-lymphocytes activity (p < 0.01) were characteristic of schizophrenia in use of low mitogen doses. A significant increase (p < 0.001) of IL-2 serum concentration was found only in shift-like type. The changes of IL-2 concentration are more frequent in younger patients, aged up to 30 years, with relatively long duration of illness and manifestation stage (over 5 years). An increase of serum IL-2 in the patients was not connected with the changes of soluble IL-2 receptors in the serum as it did not differ from the control values.

[A change of immune profile of patients with schizophrenia during treatment].

Cellular and humoral immunological parameters have been studied in 59 schizophrenic patients and 38 healthy controls. Immunological indices (CIC, autoantibodies to cardiolipin) were found to be significantly elevated in patients in the acute disease stage before the treatment. After olanzapin therapy, a level of these parameters decreased and did not differ from that of controls. In patients, irrespective of clinical condition and treatment, functional activity of immunocompetent cells (phagocyte activity of neutrophils and monocytes, cytotoxic activity of lymphocytes natural killers, interleukin-2, interleukin-10 and gamma-interferon production), was significantly lower both before the treatment as after therapy, i.e. did not change during the whole study (28 weeks). In responders, a level of IL-1B production was higher than in controls before and during the treatment. In non-responders, it was similar to that in controls before the therapy, and increased during the treatment to a higher level.

[Interleukin level in endogenous depression].

Interleukin level and mononuclear phagocytes (MPh) activity have been studied in 19 patients with mono- and bipolar depression. All the patients were examined at admission to a hospital before treatment. The depression was characterized by low MPh activity, a significant (p < 0.05) fall in IL-1 beta production by stimulated monocytes, a trend to reduction of IL-2 production by stimulated lymphocytes, significant (p < 0.05) IL-2 serum concentration increase and significant (p = 0.05) decrease of soluble IL-2 receptor (siL-2R) concentration in the serum. Significant positive correlation (r = 0.76; p < 0.05) between MPh activity and IL-1 beta production has been found. Interleukin alterations were most manifest at the early disease stage (symptoms onset is less than one year). It is suggested that IL-2 accumulation in the blood serum in these patients is strongly influenced by a significant decrease of siL-2R serum concentration. MPh (the main IL-2 producers) activity reduction results in the monocyte IL-1 beta production decrease followed by changes in interleukin (in particular, IL-2) production by lymphocytes and other changes in interleukin system.

[Akatinol memantin in Alzheimer's disease: clinico-immunological correlates].

16 patients with Alzheimer's disease were treated with akatinol memantine. The levels of production of interleukin-1 (IL-1) in lymphocyte culture were evaluated both before and after the treatment that lasted for 2-4.5 months. Before the therapy a significant (p < 0.05) increase in the production of IL-1 was observed in patients with mild and moderate dementia, with early onset of the disease and with the duration of the disease up to 9 years as compared with a control group (24 healthy individuals). After the treatment the production of IL-1 was decreased in patients with moderate dementia, with early onset, and with hereditary form of the disease. Decrease of the level of IL-1 production after the treatment corresponded to improvement of the cognitive functions of the patient. In cases of unsatisfactory treatment (without any improvement of a clinical state) IL-1 production was at higher levels (p < 0.05) as compared with the control values. It is suggested that initial high level of IL-1 production could serve as a predictor of efficiency of therapy with akatinol memantine in patients with early onset of Alzheimer's disease.

[Analyzing correlation between neurophysiological parameters and level of stress hormone cortisol in aging].

A correlation between the serum levels of the stress hormone cortisole, neurophysiological parameters, and age was examined in 23 elderly healthy examinees (whose mean age was 65.7 +/- 2.1 years). The latency of the components of visual evoked potentials (VEP) was found to progressively increase with age. The individuals having high cortisole levels showed an increase in the latency of late VEP components associated with accelerated ageing and/or with the degenerative changes in the brain limbicoreticulocortical pathways. The DC potential level (DCPL) reflecting the intensity of brain energy expenditures correlated with the content of cortisole. Increased DCPL and longer latency of late VEP components were interrelated. It is suggested that high cortisole level produces a neurotoxic action and promotes accelerated ageing of brain regions having glucocorticoid receptors. Imbalance between the significant intensification of catabolic processes, the rise of energy expenditures and the limited increase in anabolism in elderly age is a course of this process.

[Correlation between functional activity of lymphocytes in patients with Alzheimer's dementia and their response to therapy].

16 patients with dementia of Alzheimer's type (DAT) were examined. Functional activity of lymphocytes was determined according to proliferating activity of T-lymphocytes, production of Interleukin-1 (IL-1) and estimation of subpopulations of T-leukocytes (T-helpers, T-suppressors). The patients were observed both before and 1-2.5 months after amiridin therapy. Considerable reduction of both IL-1 production and proliferating activity of T-lymphocytes was observed after the treatment in 8 patients of 11. Their cognitive functioning after the therapy was either improved or stable by the moment of repeated observation. It is quite important that initial functional activity of lymphocytes was considerably higher in this group of patients as compared with the age-matched controls. Change for the worse in cognitive functions by the moment of the second examination was accompanied in 5 patients with an increase in IL-1 production as compared to the initial level. It is proposed that high level of IL-1 production by blood lymphocytes may be used as a criterion in selection of DAT patients for amiridin therapy.

[The immune system characteristics during normal aging and in Alzheimer's disease].

The paper presents a review of data available in the literature and the authors' own findings on age-related physiology and pathology of the immune system. Almost all parameters characterizing the functional activity of the immune system in Alzheimer's disease (AD) were demonstrated to significantly differ from those in the healthy elderly, The magnitude of immunological changes clearly correlated with the severity of dementia in AD patients. A hypothesis of the gradual development of immunological changes in an AD patient in relation to his dementia formation is given.

[The neuroimmune relationships during normal aging and in Alzheimer-type dementias].

Relationships between neurophysiological and immune parameters were studied in normal middle-aged and elderly subjects and in patients with dementia of Alzheimer's type (DAT). The rate of such interaction was minimal in normal aging, in DAT neuroimmune relations increased. The correlation between the immune parameters and the characteristics of components of visual evoked potentials and auditory brain-stem responses suggests that in the middle-aged healthy subjects, a primary role in neuroimmune relations belongs to limbicoreticular structures and to the mesencephalon. In normal aging, the influence of limbicoreticular structures on immune structures decreases. In DAT, the modulation of immune processes by limbicoreticular and thalamocortical structures, as well as the pontobulbar part of the brain stem increases. In middle age and in DAT, persons with different functional brain asymmetry (as estimated from electrophysiological criteria) have different lymphocytic functional activities and rates of neuroimmune interactions.

[Changes in the immunological parameters in Alzheimer's disease: their relation to disease severity].

The peculiarities of the immune system in patients with Alzheimer's dementia (AD) and their relations with some clinical parameters were investigated. 21 patients with AD aged 51-70 and 16 age-matched healthy subjects were included in this study. The following parameters were examined: 1) total lymphocyte numbers, 2) the functional activity of T- and B-lymphocytes under conditions of stimulation by nonspecific mitogens phytohemagglutinin (PHA) and pokeweed mitogen (PWM) and 3) the production of interleukin-1 (IL-1) and interleukin-2 (IL-2). A statistically significant decrease (p < 0.01) in lymphocyte number and significant increase of proliferative activity of T-lymphocytes, stimulated by PHA (when the small mitogen doses in the range of 1.25-2.5 mg/ml were used) were found in patients with moderate and severe dementia (p < 0.05, p < 0.01). No significant changes of lymphocyte proliferative activity were detected in patients with mild dementia. Some tendency for higher IL-1 production was found in AD patients, but it was not statistically significant. No changes of functional activity of B-cells and of production of IL-2 in AD patients were found in comparison with the control group.

[Neuroimmune interactions in normal aging and in Alzheimer disease].

The study was undertaken to examine nerve-immune processes and the specific visual and limbic-reticular-cortical systems by analysing visual evoked potentials (VEP), as well as nerve-immune relationships in 21 patients aged 62.2ë1.3 years who had Alzheimer's disease and in 15 healthy individuals of the same age. AD patients were found to have lower lymphocyte count, enhanced proliferative activity of T lymphocytes and to tend to have increased production of interleukin (IL-1) as compared to that in healthy individuals. In AD, latent periods of late EVP components are higher than normal age-specific ones. The relationship between VEP and immunity parameters differ in normal ageing and AD. In health, there is a negative correlation between the latencies of N1, P2, N2 VEP components and the lymphocyte count and between the amplitude of a N1 VEP component and the production of IL-1. There is a prevalent correlation of immune parameters with VEP parameters in the right hemisphere. In AD, the higher proliferative activity of T lymphocytes is associated with more prolonged latent periods of N3 VEP components, suggesting that there are abnormal changes in the limbic-reticular-cortical systems of the brain.

[Changes in the functional activity of T-lymphocytes after administration of stelazine].

It has been established that stelazine administered in the dose corresponding to the therapeutic one is capable of inhibiting the proliferative activity of T lymphocytes and production of IL-2 in experiments in vitro. It is assumed that decrease of these characteristics in schizophrenic patients is related to the systematic intake of neuroleptics.

[Effect of psychotropic preparations on the properties of tubulin in the brain].

Neuroleptics (chlorpromazine, triftazin, haloperidol) inhibited the tubulin polymerization on supernatant of rat brain proteins in vivo at concentrations which corresponded to the therapeutic ones. The electron microscope studies demonstrated that these drugs inhibited the assembly of tubulin in microtubules. As this effect was the same in vivo and in vitro, it was considered specific. Colchicine-binding capacity of tubulin was suppressed only at higher doses of tranquilizers (250 microM). Antidepressant drug melipramine inhibited neither tubulin polymerization in vivo nor its colchicine-binding activity in vivo or in vitro.

[Aminazine--highly effective inhibitor of tubulin polymerization].

Aminasine was found to repress tubulin polymerization into microtubules. This process is associated with the formation of anomalous structures--aggregates which contain, except tubulin, an increased amount of microtubule-associating proteins. The effect of aminasine on microtubules is specific, since it can be observed both in vitro and in vivo. Aminasine seems to be a helpful tool in studying the functions of tubulin and microtubules.
