[Use of an immunoperoxidase method in situ for detecting recombinant clones of vaccinia virus containing an expressed gene of influenza viral hemagglutinins].

The vaccine L-IVP strain of vaccinia virus (VV) was used to construct the recombinant viral clones containing the influenza A hemagglutinin gene. The recombinant T plasmid was obtained with HA gene inserted in the vector pGS-20 (B. Moss) under the 7.5 K promoter of VV. A homologous recombination technique was used to insert the gene with the flanking TK sequences into vaccinia virus genome. The recombinant clones were selected by dot-hybridization with [32P]-labeled HA-probe. These recombinants were analysed for HA gene expression by the indirect immunoperoxidase method in situ using the peroxidase conjugate of the staphylococcal A-protein. This technique allows to obtain stable stained preparation and analyse the protein behavior at the ultrastructural level.

[Use of probes containing cloned genes of the Karelian fever virus in detecting viral genetic material in infected cells by a molecular hybridization method].

Two plasmid DNA-probes containing DNA-replicas of KFV genes (clone 1-protein E1 gene, clone 9--proteins E1 and P1 genes of KFV) were used for detection of the genetic material of Karelian fever virus (KFV) in the infected cells and study of the time course of accumulation of virus-specific RNAs in the process of infection. The detection was performed by the method of RNA:DNA dot-hybridization. Both probes were hybridized with KFV and Sindbis virus RNA in equal amounts--5 X 10(2) infected cells at the peak of virus infection (12 hours). None of the probes used could be bound with RNA of Venezuelan equine encephalomyelitis virus. The results obtained by the dot-hybridization method agree with previously published data on the antigenic relationship between Sindbis virus and KFV.

[Mechanism of action of remantadine hydrochloride on Sindbis virus reproduction: inhibition of virus penetration into the cell].

The effect of rimantadine and ammonium chloride on the RNA synthesis of Sindbis virus, added in different times of infection cycle, was studied. When compounds were presented in period of adsorption and first hour post-infection, the maximal inhibitory effect was found. The step blocked by rimantadine was identified as uncoating process (deprotainization). This connection was confirmed by the finding that rimantadine markedly reduced the change of the virus RNA into the nuclease-sensitive form.
