Effect of tamoxifen on pokeweed mitogen stimulated immunoglobulin secretion in vitro.

The influence of Tamoxifen on pokeweed mitogen induced immunoglobulin (Ig) secretion of human blood lymphocytes was tested in vitro. It was observed that pretreatment of T cell enriched preparations with "therapeutic" concentrations of Tamoxifen augmented their capacity to promote IgG but not IgM secretion of untreated autologous B-lymphocytes. Cultures containing Tamoxifen pretreated B cell enriched preparations and untreated T cells exhibited reduced secretion of both IgG and IgM.

Bestatin treatment enhances the recovery of radiation induced impairments of the immunological reactivity of the blood lymphocyte population in bladder cancer patients.

Bestatin, an immunostimulating substance of microbial origin, was examined for its capacity to augment immune responses of blood lymphocytes in bladder cancer patients having received a full course of local irradiation (64 Gy). Following irradiation the patients became lymphopenic and the lymphocytes exhibited impaired mitogenic responses to phytohemagglutinin (PHA) and purified protein derivative of tuberculin (PPD) and reduced poke weed mitogen induced secretion of immunoglobulins in vitro. Patients who were randomized to receive daily oral Bestatin treatment exhibited enhanced recoveries of PHA- and PPD-responses and enhanced recovery of the IgM secreting capacity compared to irradiated patients who did not receive Bestatin. Repopulation of the blood lymphocyte population, however, was not enhanced by Bestatin treatment. It is concluded that Bestatin treatment may enhance the recovery of radiation induced functional defects of the immune system in cancer patients.

Persistent generalized lymphadenopathy: immunological and mycological investigations.

Immunological and mycological investigations were carried out in 21 Swedish homosexual males. One of them had AIDS, one pre-AIDS and 19 lymphadenopathy of whom 18 fulfilled the criteria of persistent generalized lymphadenopathy (PGL) as defined by the Centers for Disease Control, Atlanta, (CDC). The patients were investigated immunologically with respect to their in vitro lymphocyte reactivity to various mitogens. The patients with AIDS and pre-AIDS belonged to the group of 8 patients with low response to mitogens. Blood helper T cell percentages and serum beta 2-microglobulin concentrations correlated with the PHA reactivity. Three patients, with the diagnoses AIDS, pre-AIDS and PGL respectively, had clinical signs of oral candidiasis with rich growth of Candida albicans in culture. These were all low responders to mitogen stimulation. Six cases of tinea pedis were diagnosed and seemed to be distributed among the patients irrespectively of the severity of their immunological disorders.

Further studies on the influence of interferon on pokeweed mitogen induced Ig-synthesis in vitro.

Preincubation with pure interferon beta, but not partially purified interferon alpha, increased immunoglobulin production from non-fractionated peripheral blood mononuclear cells stimulated by pokeweed mitogen. Preincubation of B lymphocytes, but not T lymphocytes or monocytes, with pure interferon beta also increased immunoglobulin synthesis. It is concluded that interferon has a direct effect on B lymphocytes in enhancing immunoglobulin production.

Long-term effects on the immune system following local irradiation for breast cancer. Pokeweed mitogen induced immunoglobulin secretion by blood lymphocytes and serum immunoglobulin levels.

Previous studies have shown that the PWM driven immunoglobulin (Ig) secretion by blood lymphocytes in vitro is reduced shortly after local radiation therapy for breast cancer. In this investigation we have examined the long-term effects of radiation therapy (45 Gy) on this lymphocyte function. A total of 111 disease-free patients treated for operable breast cancer 5-6 and 10-11 years earlier were examined. The Ig classes studied were IgM, IgA and IgG. All patients belonged to a clinical trial where the effect of pre- or post-operative local radiation therapy was evaluated against surgery only. Significant reductions of the amount of spontaneously released IgM in vitro were observed among irradiated patients as compared to the unirradiated 5-6 and 10-11 years after treatment. Such a difference was not observed for IgA and IgG. The PWM induced Ig secretions did not differ significantly between the three patient groups. The long standing reduction of the spontaneous release of IgM by blood lymphocytes in vitro probably lacks clinical significance since the serum levels of this Ig class, as well as IgA and IgG, were similar in irradiated and unirradiated patients.

Single cell protein as an occupational hazard.

Single cell protein (SCP) intended for animal feed purposes was produced in a pilot plant. The SCP consisted of Methylomonas methanolica, a pseudomonas species which is an obligate methanol user. The SCP was cultured in fermenters and later dewatered and dried in a spray-drier. Seven of eight research workers had febrile reactions 6-12 hours after exposure to SCP dust. All workers had high titres of IgG and IgM antibodies against the pseudomonas species as measured with indirect ELISA and passive haemagglutination techniques. The mechanism behind the febrile reaction is judged to be a non-immunological reaction caused by endotoxins. By increasing the particle size of the SCP through using different drying procedures, a product which generated less dust was obtained.

Specific and non-specific B cell activation in measles and varicella.

Lymphocytes from eight patients with measles and six patients with varicella were studied during the acute phase (first week) of illness and after recovery for spontaneous and pokeweed mitogen (PWM)-induced production of immunoglobulins (Ig) and viral antibodies by an enzyme linked immunosorbent assay (ELISA). In both infections acute phase lymphocytes showed increased spontaneous in vitro IgM and IgG productions including IgM and IgG antibodies to the aetiological virus as well as IgG antibodies to unrelated viruses (varicella, measles, rubella and mumps) to which the patient had serum antibodies. PWM induced no further Ig synthesis in the acute phase. In the convalescent phase viral antibody production could be demonstrated only in PWM stimulated cultures. In four patients the spontaneous synthesis of antibodies to a non-aetiological virus seemed to precede the production of IgG antibodies to the aetiological virus. All patients showed an increase of ELISA determined serum antibodies to the aetiological virus from the acute to the convalescent phase. Three of seven measles patients also showed a minor but significant increase or decrease of serum IgG antibodies to varicella and one of six varicella patients a significant rise of serum IgG antibodies to measles. Thus both measles and varicella infections were associated with non-specific as well as specific B cell activation. The non-specific B cell activation may be induced by non-specific helper factors from activated T cells.

The effect of radiation on T cell modulation of Ig synthesis by human B cells in vitro.

PWM-induced in vitro Ig synthesis was measured in cell cultures containing B lymphocytes from healthy controls and T lymphocytes collected from breast cancer patients before and after radiotherapy, respectively. The amounts of IgG and IgM did not differ between these two types of cultures. Lymphoid cell suspensions containing 3-5% monocytes were irradiated with 13 Gy and co-cultured with non-irradiated B lymphocytes. The PWM induced production of IgG and IgM in these cultures was decreased as compared to non-irradiated controls.

Effect of radiation therapy and in vitro x-ray exposure on lymphocyte subpopulations and their functions.

Radiation treatment of breast cancer patients (45.0 Gy) profoundly affected the peripheral blood lymphocytes. The number of these cells was markedly reduced with non-T-cells being more extensively depleted than T-cells immediately after radiation. The long-lasting lymphopenia, on the other hand, was mainly due to reduced number of T-cells. Antigen and mitogen stimulability, MLC reactivity, pokeweed (PWM)-induced immunoglobulin (Ig) production in vitro, and different cytotoxic functions decreased. Depletion of lymphocytes largely restored the radiation-depressed lymphocyte reactivity. The effects of in vitro exposure of blood lymphocytes to x-rays were similar to those seen after radiotherapy. Non-T-cells and T-cells with Fc-receptors for IgG were relatively radiosensitive. This latter observation agreed well with demonstrated increase of PWM-induced Ig synthesis after in vitro exposure to x-rays. T-suppressor cells defined by monoclonal antibodies were, however, radioresistant. The cytotoxic functions were reduced. No correlations were found between the pretreatment immunological status or the extent of radiation-induced immunological suppression, respectively, and prognosis.

T lymphocyte subpopulations defined by monoclonal antibodies and FC receptor binding in relation to immunosuppression in vaccine-induced rubella infection.

Lymphocyte stimulation tests and determination of T lymphocyte subpopulations were undertaken before and 7-11 days after rubella vaccination of healthy subjects. The proliferatory response of blood mononuclear cells to phytohemagglutinin (PHA) and purified protein derivate was suppressed after vaccination, as was the immunoglobulin production of pokeweed mitogen (PWM) stimulated cultures. After removal of phagocytic cells the response to PHA was still suppressed after vaccination in most cases, but when the cultures were depleted of non-T lymphocytes the suppression was no longer demonstrable. The proportion of T lymphocytes with receptors for the Fc part of IgG increased after vaccination, whereas there was no significant change of the T lymphocyte subpopulations identified with monoclonal antibodies directed at the suppressor/cytotoxic or helper T cell subsets.

Radiosensitive t cell suppression in relation to the TG cell level in synovial fluid of patients with rheumatic disease.

Radiosensitive T cell suppression was studied in relation to the level of T cells with IgG-Fc receptor (TG cells) in synovial fluid and blood of nine patients with rheumatic diseases. Irradiation of T cells significantly decreased the proportion of TG cells in the blood of patients and healthy controls. The proportion of synovial TG cells was low in all patients before as well as after irradiation. Irradiation enhanced the helper effect of T cells on pokeweed-mitogen induced immunoglobulin production by allogeneic B cells. This increased helper effect was taken as an expression of decreased suppressor activity following irradiation. By this method suppressor activity was demonstrable in synovial T cells of six out of nine patients and in blood T cells of six out of eight patients and six out of eight healthy subjects. Thus the low level of synovial TG cells was in most patients not associated with a lack of radiosensitive synovial suppressor cell activity.

Influence of Bestatin, a new immunomodulator, on Ig secretion by human lymphocytes in vitro and in vivo.

Bestatin, a chemically well-defined immunomodulator, was tested for its capacity to interfere with PWM-induced Ig synthesis of peripheral lymphocytes from healthy donors. It was observed that secretion of IgA, IgG and IgM was impaired when Bestatin was present in the cultures. Serum Ig concentrations and antibody activities of IgG class to cytomegalovirus, herpes simplex and morbilli virus were examined in cancer patients who were treated with 30 mg of Bestatin daily for 1-21 weeks. These values remained constant during Bestatin treatment indicating that antibody secretion in vivo was not affected.

T cell subpopulations and their functions in vitro. A study in patients with alopecia areata and alopecia universalis.

T lymphocyte subpopulations and their in vitro activities were determined in patients with alopecia areata (AA) and alopecia universalis (AU). The frequencies of T cells with receptors for IgG and T cells with receptors for IgM were determined in 16 cases. The IgG and IgM production of B lymphocytes cocultured with autologous and allogeneic T lymphocytes in the presence of PWM was determined in 12 and 9 cases, respectively, and the antibody-dependent cell-mediated cytotoxicity (ADCC) of unfractionated lymphocytes was examined in 6 cases. Patients with AA and AU had a higher proportion of Tg cells and more pronounced ADCC of unfractionated peripheral lymphocytes than normal persons. Furthermore, PWM-induced Ig production of B lymphocytes in the presence of autologous T lymphocytes seemed to be lower than that of controls.

T lymphocyte subpopulations and pokeweed mitogen-induced immunoglobulin synthesis in vitro by mononuclear cells from psoriasis patients.

T-lymphocyte subpopulations and pokeweed mitogen(PWM)-induced immunoglobulin (Ig) synthesis of mononuclear cells in vitro were studied in patients with psoriasis as well as in age- and sex-matched normal blood donors. The frequences of T cells with receptors for IgG (Tg) and T cells with receptors for IgM (Tm) were examined. The IgG and IgM synthesis in mononuclear cell suspensions in the presence of different amounts of PWM was determined. The percentage of total T cells (rosetting with neuroaminidase-treated sheep red blood cells), including high and low affinity T lymphocytes showed no difference between the patient group and controls. The patients with psoriasis had a significantly higher mean proportion of Tg cells than the normal donors whereas there was no significant difference in the proportions of Tm cells between these two groups. The PWM-induced Ig synthesis in the mononuclear cell suspension seemed lower in patients with psoriasis than in controls, the difference being statistically significant when the results were expressed as ratios of Ig amounts present in the supernatants of PWM-stimulated and non-stimulated cultures (index of stimulation).

ELISA-determined serological reactivity against purified trichophytin in dermatophytosis.

Serological activity in 62 patients wih dermatophytosis was investigated using purified trichophytin preparation as antigen (produced by the ethylene glycol method). Antibodies of IgG and IgM classes were determined by means of ef enzyme linked immunosorbent assay (ELISA). In comparison with 79 controls consisting of groups of healthy blood donors and dermatological outpatients, the dermatophyte-infected patients showed a significantly higher mean IgG response to trichophytin antigen preparation. On the other hand several individual cases were low responders. In a group of 10 control children, no IgG activity against trichophytin could be demonstrated. IgM class antibodies showed no significant differences between the groups investigated. These findings indicate that differences in the amounts of antibodies of IgG class against trichophytin as measured by the ELISA method reflect differences in the degree of mycotic sensitization between groups of individuals having experienced varying degrees of exposure to dermatophytes. Nevertheless, the determination of IgG antibodies in dermatophytosis seems to be of limited clinical value.

Blood lymphocyte subpopulations in Mycosis fungoides and their functions in vitro.

T-lymphocyte subpopulations, PWM stimulated in vitro Ig production by mononuclear cells, and spontaneous cell-mediated cytotoxicity (SCMC) were determined in patients with Mycosis fungoides (MF) as well as in sex- and age-matched normal controls. We were able to confirm our earlier findings of a significantly higher frequency of T-lymphocytes with Fc-receptors for IgG (Tg cells) and a proportion of T lymphocytes with Fe receptors for IgM (Tm cells) not significantly differing from normal controls. As defined by monoclonal antibodies, the proportion of T-helper cells (Leu3A) was significantly lower, whereas the T-suppressors cells (Leu2A) were in the range of normal controls. The PWM stimulated and spontaneous Ig synthesis in vitro was lower in MF patients whereas there was no difference in the SCMC activity, in comparison with normal controls. The lower frequency of T-helper cells in the peripheral blood may be explained by their migration to the skin.
