A refined physical map of the long arm of human chromosome 16.

Mapping of 33 anonymous DNA probes and 12 genes to the long arm of chromosome 16 was achieved by the use of 14 mouse/human hybrid cell lines and the fragile site FRA16B. Two of the hybrid cell lines contained overlapping interstitial deletions in bands q21 and q22.1. The localization of the 12 genes has been refined. The breakpoints present in the hybrids, in conjunction with the fragile site, can potentially divide the long arm of chromosome 16 into 16 regions. However, this was reduced to 14 regions because in two instances there were no probes or genes that mapped between pairs of breakpoints.

Chromosomal location of the human tumor necrosis factor receptor genes.

TNFR1 and TNFR2, the genes encoding the two forms of the human tumor necrosis factor receptor, were localized to normal human chromosomes by in situ hybridization and Southern blot analysis of a series of human x mouse hybrid cell lines. TNFR1 maps to 12p13 and TNFR2 maps to 1p36.

[Pheochromocytoma of the urinary bladder].

Pheochromocytoma is known as a rare tumor present at the urinary bladder. Up to 1988, 145 cases had been reported in literature and 32 cases in our country. Most of them are case reports. 7 cases were collected in this series among 2065 bladder tumors and were studied with light and electron microscopy as well as immunohistochemical staining. According to the clinical features, 5 of them were non-functional tumors. Causes of misdiagnosis were analyzed in these cases. Their histological differential diagnoses are discussed.

Albumin--vitamin D-binding protein haplotypes in Asian-Pacific populations.

We have determined the various haplotypic combinations between alleles as well as restriction fragment length polymorphisms of two linked genetic markers, albumin and vitamin D-binding protein or group-specific component, in a number of Asian-Pacific populations. Using the partial maximum likelihood method, we constructed a phylogenetic network from the haplotype frequencies to assess relationships among the populations sampled. No systematic linkage disequilibrium was detected between most of the combinations, suggesting a lack of operation of any selection pressure at the two loci. The phylogenetic analysis confirmed the known interrelationships among various populations in the Asian-Pacific region. The Australian aborigines clustered closely with the non-Austronesian-speaking highlanders from Papua New Guinea, as expected. Similarly, the Austronesian-speaking Polynesians, Micronesians, and the Southeast Asians branched off together as a separate group. The position of the Austronesian-speaking Tolais from New Britain with respect to other populations from the Southwest Pacific was anomalous. The Tolais revealed a strong affinity with the Australian aborigines, which is inexplicable. The populations from China formed a tight cluster with other populations from the Asian-Pacific region. Genetic interrelationships of these populations with the white Australians were remote, which is in accordance with the known affinities of various human racial groups.

Evolution of beta-globin haplotypes in human populations.

The beta-globin haplotypes of 852 chromosomes from 12 populations in the Asia-Pacific region are described. These data are combined with those from other populations in an investigation of the affinities of regional human populations. Both partial maximum-likelihood and distance Wagner methods indicate that Africans are the most divergent group, with the remaining populations branching in the following order: Australian Aborigines, Highland Melanesians, Lowland Melanesians, Indonesians and Micronesians, Polynesians, east Asians, Indians, and Europeans. This pattern of relationship is consistent with that indicated by other data. Analysis of the evolution and distribution of haplotype occurrence provides some limited support for an origin of modern humans in Africa. Otherwise, however, it was not useful in further elucidating the evolutionary history of human populations.

Erythroid 5-aminolevulinate synthase is located on the X chromosome.

The gene for erythroid 5-aminolevulinate synthase has been mapped to Xpter-Xq26 by Southern blot hybridization analysis of a mouse/human hybrid cell panel. In situ hybridization maps the gene to Xp21-Xq21, with the most likely location being on band Xp11.2. The mapping of the erythroid 5-amino-levulinate synthase gene to the X chromosome suggests that a defect in this gene may be the primary cause of X-linked sideroblastic anemia.

Prostatic carcinoma with endocrine features. A report of a neoplasm containing multiple immunoreactive hormonal substances.

A case of prostatic carcinoma with the cellular patterns of an adenocarcinoma and carcinoid tumor is reported. The tumor contained ultrastructural dense core neuroendocrine granules, and immunoperoxidase staining revealed prostatic acid phosphatase, prostatic-specific antigen, chromogranin, neuron-specific enolase, serotonin, adrenocorticotrophic hormone (ACTH), somatostatin, parathormone, calcitonin, bombesin, and glucagon but no insulin. The patient had exhibited hypercalcemia that may have been related to hormone production by the tumor. The literature on the endocrine aspect of the prostate and its tumor is reviewed.

[Carcinoid of ectopic pancreas tissue--islet cell tumor].

A case of islet cell tumor--carcinoid of the ectopic pancreas is reported. The patient had been operated six times because of recurrent abdominal tumor mass and intermittent hypoglycemic coma. The first five operations were done with the diagnosis of mesotheliosarcoma by pathology. The last operation was done in our hospital with the final diagnosis of carcinoid--islet cell tumor. In addition to the light microscopic view, several diagnostic features were observed as follows: Under electron microscopy, membrane bound secretive granules were present in the tumor cytoplasm. Neither brush-like microvilli nor lumen formation of the mesothelioma were found. By immunohistochemistry, positive granules to immunoperoxidase stain of insulin were observed in some tumor cytoplasm. By laboratory test, levels of blood 5-hydroxytryptamine and urinary 5-hydroxyindolacetic acid were elevated. Sometimes it is difficult to differentiate carcinoid from islet cell tumor only by morphology, because both are derived from the same origin during the embryonic development which is probably related to the endocrine system from the embryonic foregut. The latter arises from the neuro-ectoderm and belongs to APUD system.
