Covalent modification of glassy carbon electrodes with beta-alanine for voltammetric separation of dopamine and ascorbic acid.

beta-Alanine was covalently grafted on a glassy carbon electrode (GCE) by amine cation radical formation in the electrooxidation process of the amino-containing compound. X-ray photoelectron spectroscopy (XPS) and cyclic voltammetry (CV) proved the immobilization of beta-alanine monolayer on GCE. The electrode shows strong electrocatalytic functions to dopamine (DA) and ascorbic acid (AA), reducing the overpotentials by 0.20 V and 0.23 V, respectively. Due to its different catalytic effects toward DA and AA, the modified electrode resolved the overlapping voltammetric responses of DA and AA into two well-defined voltammetric peaks by CV or differential pulse voltammetry (DPV), which can be used for the simultaneous determination of these species in a mixture. The catalytic peak current obtained from DPV was linearly related to DA and AA concentrations in the ranges of 4.0 x 10(-6)-5.0 x 10(-4) mol/L and 2.0 x 10(-5)-6.0 x 10(-3) mol/L with correlation coefficients of 0.997 and 0.995, respectively. The detection limits (3 sigma) for DA and AA were 2.4 x 10(-6) mol/L and 1.2 x 10(-5) mol/L, respectively. The electrode shows good sensitivity, selectivity and stability, and has been applied to the determination of DA and AA simultaneously in samples with satisfactory results.

Risk of developing retinopathy in Diabetes Control and Complications Trial type 1 diabetic patients with good or poor metabolic control.

OBJECTIVE: The study goal was to assess and predict the risk of developing retinopathy in type 1 diabetic patients with extreme metabolic control. RESEARCH DESIGN AND METHODS: Based on material from the Diabetes Control and Complications Trial (DCCT) study (n = 1,441 patients), patients without retinopathy at baseline (DCCT primary cohort) were considered under good or poor metabolic control if the mean HbA(1c) level (until the last visit) fell in the lower or upper 20% of the overall HbA(1c) distribution, respectively. Retinopathy was recorded as either absent or present. Logistic regression was used to predict retinopathy from covariates used in the DCCT retinopathy study. RESULTS: Among the 153 DCCT patients with "good metabolic control" (mean HbA(1c) < or = 6.87%), three-step change retinopathy developed in 15 (9.8%), and 138 (90%) remained free of retinopathy. Conversely, among the 166 patients with "poor metabolic control" (mean HbA(1c) > or = 9.49%), the complication did not develop in 71 (43%) and did develop in 95 (57%). Whereas occurrence of diabetic retinopathy was primarily due to metabolic control (P < 0.0001) and duration of participation in the study (P < 0.0001), two other covariates were found to be significant prognostic factors of the complication: HbA(1c) at baseline (OR 1.37, P < 0.001) and BMI (OR 1.11, P < 0.05). CONCLUSIONS: This study confirms that retinopathy develops in approximately 10% of patients with type 1 diabetes under good metabolic control, whereas > 40% of patients with type 1 diabetes remain free of retinopathy despite poor metabolic control. After adjusting for metabolic control and duration of participation in the study, it was found that previous glycemic exposure (HbA(1c)) and BMI may provide a possible explanation to such paradoxical clinical situations.

Killing effect of TNF-related apoptosis inducing ligand regulated by tetracycline on gastric cancer cell line NCI-N87.

AIM: To clone the cDNA fragment of human TRAIL (TNF-related apoptosis inducing ligand) into a tetracycline-regulated gene expression system, the RevTet-On system, transduce expression vectors into a gastric carcinoma cell line-NCI-N87 and examine the effects of controlled expression of TRAIL in vitro on the gastric carcinoma cells. METHODS: The full-length cDNA of TRAIL was inserted into a vector under the control of the tetracycline-responsive element (TRE) to obtain the plasmid pRevTRE-TRAIL, which was transfected into a packaging cell line PT67. In addition, vector pRev-Tet On and pRevTRE were also transfected into PT67 separately. After hygromycin and G418 selection, the viral titer was determined. The medium containing retroviral vectors was collected and used to transduce a gastric carcinoma cell line NCI-N87. The resulting cell line NCI-N87-Tet On TRE-TRAIL and a control cell line, NCI-N87 Tet On-TRE, were established. TRAIL expression in the cell line was induced by incubating cells with doxycycline (Dox), which is a tetracycline analogue. The killing effect on gastric carcinoma cells was analyzed after induction. RESULTS: The recombinant plasmid pRev-TRE-TRAIL was constructed. After hygromycin or G418 selection, the producer cell lines PT67-TRE, PT67-TRE-TRAIL and PT67-Tet On were obtained, with titers of about 10(8)CFU.L(-1). By transducing NCI-N87 cells with retroviral vectors from these cell lines, stable cell lines NCI-N87-Tet-On TRE-TRAIL (NN3T) and control cell line NCI-N87-Tet-On-TRE (NN2T) were established. The growth curves of the selected cell lines were the same with the wild type NCI-N87. When Dox was added, cell death was obvious in the test groups (29%-77%), whereas no difference was observed in control and wild type cell lines. With the addition of a medium from the test group, human leukemia cell line Jurkat was activated till death (83%), indicating the secretion of active TRAIL proteins from the test cells to the medium. CONCLUSION: With the use of the RevTet-On system, a regulated expression system for TRAIL was constructed. Using this system, the selected killing effect of TRAIL on gastric carcinoma cell line NCI-N87 could be observed.

[Effect of a complex tea on reducing blood lipid in rabbits].

In order to observe the effect of a complex tea composed of tea, ginkgo leaf, red koji and fructus lycii on reducing blood lipids in experimental rabbits, 40 rabbits weighted from 1.5 to 2.0 kg were divided randomly into 5 group: (I) normal control group, (II) hyperlipidemia model(positive control), (III) green tea control group, (IV) low dose complex tea group, and (V) high dose complex tea group. Blood triglyceride(TG), cholesterol(TC), lower density lipoprotein(LDL) and high density lipoprotein(HDL) were measured before experiment and 6.12 weeks later. Then the pathological changes of heart, aorta and liver were observed. After 12 weeks of experiment, the results showed that, (1) hyperlipidemia characterized by high TC has been induced by high-fat feeds in group II, (2) the complex tea can reduce body weight and eliminate the lipids deposition in aorta and liver. (3) the complex tea can decrease serum TG, TC,LDL and increase serum HDL in the high complex tea group. The results indicate that the complex tea is more effective than green tea on reducing blood lipids in experimental rabbits.

A-85380 and epibatidine each interact with disparate spinal nicotinic receptor subtypes to achieve analgesia and nociception.

Nicotinic agonists, such as epibatidine (EPI) and A-85380, when administered systemically, elicit analgesia. Intrathecal EPI also produces analgesia accompanied by nociceptive and pressor responses. Since spinal administration of drugs offers a well defined pathway connecting the site of administration with behavioral and autonomic responses, we have compared the responses to intrathecal epibatidine and A-85380 to delineate the role of nicotinic acetylcholine receptors in spinal neurotransmission. Following implantation of intrathecal catheters in rats, we monitored cardiovascular, nociceptive, and antinociceptive responses after administration of various nicotinic receptor agonists. Consistent with A-85380 displacement of epibatidine from isolated spinal cord membranes, A-85380 elicited pressor, nociceptive, and antinociceptive responses similar to EPI. Antinociception was preceded by nociception. Both antinociception and nociception were blocked by mecamylamine, methyllycaconitine, and alpha-lobeline, but dihydro-beta-erythroidine only blocked the antinociceptive response. Whereas prior administration of EPI desensitized the nociceptive and antinociceptive responses to EPI, A-85380 pretreatment only desensitized EPI-elicited nociception and not antinociception. 2-Amino-5-phosphopentanoic acid pretreatment blocked the nociceptive response to A-85380, indicating A-85380 stimulated release of glutamate onto N-methyl-D-aspartate receptors to produce the irritant response of nociception. Intrathecal phentolamine virtually abolished A-85380 antinociception, but had no effect on EPI antinociception. Hence, analgesia can be produced by stimulation of distinct spinal preterminal nicotinic receptor subtypes, resulting in the release of neurotransmitters. In the case of A-85380, these sites primarily appear to be localized on adrenergic bulbospinal terminals. Our data suggest that A-85380 and EPI act at separate preterminal spinal sites as well as on distinct nicotinic receptor subtypes to elicit an antinociceptive response at the spinal level.

[Correlation between liver fibrosis stage and serum liver fibrosis markers in patients with chronic hepatitis B].

OBJECTIVE: To evaluate the diagnostic value of serum liver fibrosis markers by analyzing the correlation between liver fibrosis stage in patients with chronic hepatitis B and serum liver fibrosis markers, such as hyaluronate, type III procollagen, laminin, and type IV collagen. METHODS: There were 278 patients with chronic hepatitis B in this series. Before liver biopsy, the serum was collected to test hyaluronate, type III procollagen, laminin, and type IV collagen. Data were analyzed statistically. RESULTS: Liver fibrosis stage was correlated to inflammation degree. In patients with moderate or serious liver inflammation and fibrosis, serum hyaluronate was above normal level, and correlated to liver inflammation and fibrosis. For patients with chronic hepatitis B, serum type III procollagen, laminin, and type IV collagen increased apparently. While laminin was correlated to liver fibrosis, type III procollagen and type IV collagen correlated to liver inflammation. CONCLUSIONS: Serum hyaluronate, type III procollagen, laminin, and type IV collagen could reflect the state of liver fibrosis. Hyaluronate may be a useful marker for liver cirrhosis.

[Combustion characteristics of municipal solid waste in fluidized bed].

For the experimental investigation on the combustion characteristics of Municipal Solid Waste(MSW) in Fluidized Bed, a lab scale Fluidized Bed facility was constructed. Single and mixed municipal solid waste burning in Fluidized Bed showed that dry waste can be burnt quickly at the bed temperature of 500 degrees C, and furnace temperature increased about 30-50 degrees C. Many kinds of combustion runs were conducted in this Fluidized Bed combustion facility. The parameters examined were air flow rate(from 5.5 m3/h to 7.5 m3/h), form of fuels(scrap or whole), moisture content and so on. Concentration of CO2, CO, SO2 and NOx in the flue gas were monitored and recorded every 5 seconds. The temperatures along the reactor were recorded every 10 seconds.

Pyrene bioaccumulation, effects of pyrene exposure on particle-size selection, and fecal pyrene content in the oligochaete Limnodrilus hoffmeisteri (Tubificidae, Oligochaeta).

The oligochaete Limnodrilus hoffmeisteri Cleparéde was exposed to sediment-amended pyrene (0-1,196 nmol/g dry wt) for 2, 5, and 10 d to investigate sediment particle-size preference, tissue pyrene bioaccumulation (using biota-sediment accumulation factor, BSAF), fecal pyrene concentrations (using fecal-sediment accumulation factor, FSAF), and pyrene biotransformation. In non-pyrene-amended sediment, L. hoffmeisteri preferentially ingested finer sediment particles (< 3.5 microns). However, pyrene concentrations > 199 nmol/g dry weight resulted in a decreased preference for finer particles, presumably reducing pyrene exposure. Particle selectivity also changed with time such that after 10 d, worms showed an increased preference for finer particles. At high pyrene exposure, tissue pyrene concentrations rapidly attained equilibrium (maximum body residue 4.4 mumol/g wet wt, close to values cited for hydrocarbon critical body residues). Levels of biotransformation were low (mean 14%) and independent of time or pyrene concentration. Biota-sediment accumulation factor showed a negative relationship with sediment pyrene concentration; this trend may be due to reduced availability at high sediment pyrene concentrations or due to reductions in both ingestion rate and selectivity for fine, pyrene-rich particles. Fecal pyrene concentrations were depleted compared to sediment levels, suggesting removal of pyrene during gut passage by absorption. Fecal-sediment accumulation factor showed a negative relationship with increasing sediment pyrene levels.

Cocaine-mediated apoptosis in bovine coronary artery endothelial cells: role of nitric oxide.

The present study examined the role of nitric oxide in cocaine-induced apoptosis in bovine coronary artery endothelial cells (BCAECs). Cocaine produced a time-dependent decrease in cell viability and an increase in apoptosis in BCAECs, which were blocked by the nitric oxide donors DETA-NONOate (DETA-NO) and S-nitroso-N-acetyl-penicillamine. In accordance, cocaine decreased nitric oxide production in BCAECs at each time point of the study. Cocaine significantly increased caspase-3 activity that was blocked by the inhibitors of cytochrome c release (cyclosporin A), caspase-3 (Ac-DEVD-CHO), and caspase-9 (Z-LEHD-FMK), respectively. In addition, cocaine activated caspase-9, which was blocked by cyclosporin A and Z-LEHD-FMK. Ac-DEVD-CHO only partially blocked cocaine-induced caspase-9 activity. DETA-NO (20 microM) blocked cocaine-mediated activation of both caspase-9 and caspase-3. Cocaine decreased Bcl-2 protein levels, which was partially blocked by Ac-DEVD-CHO and Z-LEHD-FMK, but not by DETA-NO. Furthermore, cocaine induced a translocation of Bax from the cytosol to the mitochondria in BCAECs, and increased Bax levels in mitochondria by 2.2-fold. In accordance, cytosolic Bax levels decreased about 42%. Neither Ac-DEVD-CHO nor DETA-NO affected cocaine-induced translocation of Bax. We conclude that cocaine-induced Bcl-2 protein down-regulation and Bax translocation to the mitochondria are upstream signals of caspase-9 activation that precedes caspase-3. Cocaine-induced attenuation of nitric oxide plays a key role in the activation of the caspase cascade in BCAECs.

[Expression of delta 6-fatty acid desaturase gene from Mortierella alpina in Saccharomyces cerevisiae].

delta 6-fatty acid desaturase is the rate-limiting enzyme of the desaturation of linoleic acid in the production of an essential fatty acid, gamma-linolenic acid. The 1.4 kb fragment in plasmid pTMACL6 encoding delta 6-fatty acid desaturase from Mortieralla alpina ATCC16266 was subcloned into the yeast-E. coli shuttle vector pYES2.0, thus an expression recombinant plasmid pYMAD6 containing target gene was constructed and obtained in the SC-Ura media. The pYMAD6 was introduced into defective mutant INCSc1 of Saccharomyces cerevisiae by LiAc method. When linoleic acid was provided as an exogenous substrate to the yeast cultures expressing delta 6-fatty acid desaturase activity under appropriate media and temperature condition, the level of gamma-linolenic acid reached 31.6% of the total yeast fatty acids by GC-MS detecting, which is the highest report of delta 6-fatty-acid desaturase gene in S. cerevisiae.

[Progress in studies on the mechanism of substance addiction in central nervous system].

The mechanism of substance addiction in the central nervous system is the key point to solve the problem of drug addiction in clinical prophylaxis and treatment, but it has been obscure so far. Previous researches in this field suggested that the higher brain function of the learning and memory might be pivotal in understanding the mechanism of substance addiction. It is the intent of this review to integrate and analyze the substance addiction and the learning and memory in an attempt to shed light on the strategies to conduct further studies in this field.

Protein adducts of 1,4-benzoquinone and benzene oxide among smokers and nonsmokers exposed to benzene in China.

Hemoglobin (Hb) and albumin (Alb) adducts of the benzene metabolites benzene oxide (BO) and 1,4-benzoquinone (1,4-BQ) were analyzed by gas chromatography-mass spectrometry in 43 exposed workers and 44 unexposed controls from Shanghai, China, as part of a larger cross-sectional study of benzene biomarkers. When subjects were divided into controls (n = 44) and workers exposed to </=31 ppm (n = 21) and >31 ppm (n = 22) of benzene, median 1,4-BQ-Alb adducts were 2110, 5850, and 13,800 pmol/g Alb, respectively (correlation with exposure: Spearman r = 0.762; P < 0.0001); median BO-Alb adducts were 106, 417, and 2400 pmol/g Alb, respectively (Spearman r = 0.877; P < 0.0001); and median BO-Hb adducts were 37.1, 50.5, and 136 pmol/g Hb, respectively (Spearman r = 0.757; P < 0.0001). To our knowledge, this is the first observation that adducts of 1,4-BQ are significantly correlated with benzene exposure. When compared on an individual basis, Alb adducts of 1,4-BQ and BO and Hb adducts of BO were highly correlated with each other and with urinary phenol and hydroquinone (P < 0.0001 for all of the comparisons). Although detectable in the assays, Hb adducts of 1,4-BQ and both Hb and Alb adducts of 1,2-BQ produced erratic results and are not reported. Interestingly, cigarette smoking increased Alb adducts of 1,4-BQ but not of BO, suggesting that benzene from cigarette smoke was not the primary contributor to the 1,4-BQ adducts.

[Effect of pH on copper sorption by the loess and its species].

To study the effect of pH on copper sorption and species, loess was taken as sorbent in this research, and control experiments were conducted with quartz. All the curves of sorption amount-pH showed three regions, namely, sorption amount increased slowly at low pH, then increased quickly with the rise of pH, and reached the maximum amount at high pH. Copper of sorbed, exchangeable and bound to carbonate decreased with the increase in sediment concentration at constant Cu concentration, and increased with the increase in Cu concentration at constant sediment concentration. The MINTEQA2 model results showed that the proportions of precipitates and their compositions were different for loess and quartz, and the proportions of precipitates decreased with the concentrations of loess.

[Relationship between serum matrix metalloproteinase-2 and metastasis and recurrence following radical hepatic resection in hepatocellular carcinoma].

OBJECTIVE: To study the relationship between serum matrix metalloproteinase-2 (MMP-2) and metastasis and recurrence following radical hepatic resection in hepatocellular carcinoma (HCC). METHODS: Cox's proportional hazard model was used to determine the significant clinical factor of prognosis. Meanwhile, enzyme linked immunoadsordent assay (ELISA) and northern blot analysis were adopted to detect serum and tumor tissue levels of MMP-2 respectively in HCC patients. RESULTS: Cox's proportional hazard model showed that tumor size and tumor number were significant prognostic factors of HCC patients following hepatic resection (P=0.022 and P=0.040). According to this result, we divided HCC patients into high-tendency to metastasis recurrence and low-tendency to metastasis recurrence groups. Serum levels of MMP-2 in high-tendency to metastasis recurrence following radical hepatic resection group were significantly higher than those in low-tendency group (26.39ng/m+/-2.64ng/ml vs 24.86ng+/-1.95ng/ml, P<0.05). Serum levels of MMP-2 in HCC patients performed non-radical hepatic resection group were also significantly higher than those in HCC patients performed radical hepatic resection group (29.43ng+/-3.12 ng/ml vs 25.72ng+/-2.45ng/ml, P<0.01). Northern blot analysis revealed that the changes of MMP-2 level in the serum were consistent with those of its mRNA level in the tumor tissue of HCC patients. CONCLUSIONS: Serum MMP-2 level is an indicator for metastatic recurrence following radical hepatic resection in HCC. The high levels of MMP-2 in the serum are due to high MMP-2 mRNA expression in the tumor tissue of HCC patients.
