On-line sample extraction and enrichment of non-steroidal anti-inflammatory drugs by pre-column in capillary liquid chromatography mass spectrometry.

A rapid and sensitive analytical method has been developed for the simultaneous determination of 16 non-steroidal anti-inflammatory drugs (NSAIDs) in human plasma by capillary liquid chromatography (LC) and quadrupole mass spectrometry with electrospray ionization operated in the negative ion mode. The sample clean-up and enrichment on a pre-column were accomplished on-line to improve the sensitivity. This method greatly reduced sample preparation time and sample volume compared with off-line sample extraction methods and conventional LC methods, respectively. The recoveries of NSAIDs from human plasma were 56.7-96.9%. The total analytical time for a single analytical run was approximately 15 min. The detection limits of NSAIDs were 0.001-0.075 microg ml(-1) using a selected ion monitoring mode.

Determination of iodide in seawater samples by ion chromatography with chemically-bonded poly(ethylene glycol) stationary phase.

An ion chromatographic method for the rapid and direct determination of iodide in seawater is reported. Poly(ethylene glycol) (PEG) groups were chemically bonded onto silica gel or C30-bonded silica gel via diol groups. PEG-bonded C30 binary phases allowed determination of iodide in seawater samples without any interference. Effects of eluent composition on retention behavior of inorganic anions have been investigated. Both cation and anion of the eluent affected the retention of analyte anions. The retention time of anions increased with increasing eluent concentration. The detection limit for iodide obtained by injecting 0.2 microl of sample was 13 microg l(-1) (S/N=3) while the limit of quantitation was 43 microg l(-1) (S/N=10). The present method was successfully applied to the rapid and direct determination of iodide in seawater with long-term durability.

Development of an on-line immobilized-enzyme reversed-phase HPLC method for protein digestion and peptide separation.

This paper describes use of a novel glass bead-based immobilized-enzyme micro column for simple and swift on-line protein digestion then peptide separation by reversed-phase HPLC. The inexpensive and easily made immobilized-enzyme micro column was prepared from aminopropyl controlled-pore glass that was reacted first with glutaraldehyde then with trypsin in the presence of phosphate buffer. Tryptic digestion of bovine serum albumin (BSA) was achieved simply by passing pretreated protein solution through the laboratory made immobilized-trypsin column; the tryptic fragments were then separated by reversed-phase HPLC. The peptide separation was found to be identical to separation of a sample which had undergone conventional enzymatic protein digestion in solution. Digestion of BSA by the immobilized-trypsin column decreased with increasing flow rate of the solution through the column, and 1.0 muL min(-1) was found to be the optimum flow rate for on-line protein digestion with our system. It was also found that the sample required pretreatment with urea before injection, because of a change in the properties of the protein in the presence of urea, and the immobilized-trypsin column lost its function in the presence of acetonitrile. This on-line proteomics system enables simple and rapid protein digestion and was successfully applied to partially micro two-dimensional (2D) chromatographic separation of proteins.

Rapid and simultaneous determination of nonsteroidal anti-inflammatory drugs in human plasma by LC-MS with solid-phase extraction.

A rapid, selective and sensitive analytical method for the simultaneous determination of 16 nonsteroidal anti-inflammatory drugs (NSAIDs) in human plasma was carried out using Oasis HLB solid-phase extraction (SPE), followed by reversed-phase high-performance liquid chromatography and quadrupole mass spectrometry with electrospray ionization operated in the negative ion mode. The recoveries of NSAIDs from human plasma by SPE were greater than 76.7%. The use of a short column packed with small particles enabled rapid and simultaneous determination within 7 min. The detection limits for the NSAIDs were 0.01-0.9 mug/ml using the selected ion monitoring mode.

Serial separation of cations and anions by ion chromatography using the peak parking technique and sulfosalicylic acid as the eluent.

An improvement of the peak parking technique is described for the serial determination of cations (Na+, NH+4, K+, Mg2+, and Ca2+) and anions (Cl-, SO2-(4), and NO-3) using a single pump, a single eluent and a single detector. The present system used commercially-available unmodified cation exchange and anion exchange columns, which were attached to each switching valve. When 1.75 mM 5-sulfosalicylic acid was used as the eluent, serial separation of the above cations and anions was achieved in less than 20 min. The proposed ion chromatographic method was successfully applied to the serial determination of cations and anions in tap water and river water samples. The limits of detection at S/N=3 for an injection of 20 microl were 16-68 ppb (microg/l) for cations and 15-28 ppb for anions.

On-line precolumn enrichment of bisphenol A using boronate column in microcolumn liquid chromatography.

The application of a boronate precolumn in the trace determination of 4,4'-isopropylidenediphenol (bisphenol A or BPA) in aqueous solution was investigated by microcolumn liquid chromatography (LC). BPA was enriched by a precolumn packed with TSK(gel) boronate and it was then injected (back-flushed) into an ODS separation column. Comparing to a conventional syringe injection, a better peak shape was obtained via the boronate precolumn injection, and the calculated theoretical plate number improvement was approximately two folds. The relative standard deviations for the retention time, peak area and peak height were 0.12, 3.0 and 1.8%, respectively. The system is linear (R2>0.99) within the working sample concentration and sample-volume ranges, and it presents a detection limit of 0.09 microg L(-1)(S/N=3) when a 0.2 mL sample was enriched. Recovery values of the proposed method were between 107 and 123%, and this method was applied satisfactorily to the determination of BPA in commercially available bottled drink samples. The boronate precolumn was proven to produce better sensitivity than that of an ODS or C30 precolumn as long as the enrichment of BPA in aqueous samples is concerned.

Peak parking technique for the simultaneous determination of anions and cations.

An ion chromatography method is described for the simultaneous determination of anions (Cl-, NO3-, and SO4(2-)) and cations (Na+, NH4+, K+, Mg2+, and Ca2+) using a single pump, a single eluent and a single detector. An anion-exchange column modified with chondroitin sulfate C facilitated the elution of the above three anions using 5 mM tartaric acid as the eluent in isocratic mode, whereas the same eluent facilitated the separation of the above five cations on a commercially-available cation-exchange column. The separation columns were connected in series via two six-port switching valves, so the required cation-exchange or anion-exchange separation could be carried out by selecting the appropriate positions for the switching valves. The separations were completed in 30 min.

Sample enrichment by using monolithic precolumns in microcolumn liquid chromatography.

An on-line sample enrichment system was designed using monolithic precolumns in microcolumn LC. The monolithic ODS capillary columns were prepared via in situ sol-gel processes. The enrichment efficiency of the monolithic columns was tested by using phthalates as the analytes. The relative standard deviations (n = 6) for the retention time, peak area and peak height were between 0.4 and 1.2%, 0.9 and 5.5% and 0.4 and 3.9%, respectively. The system was linear (R2 > 0.99) within the working sample concentration and sample volume ranges. Comparing to 0.2 microl injection with a typical sample injector, the theoretical plate number of a same separation column was increased by 3-6-fold when the precolumn unit was used for sample injection. The recoveries of the analytes were between 88 and 120%, and the sample volume that could be injected into the system was increased up to 5000-fold. The limits of detection were improved by more than 2000-fold and were between 0.21 and 0.87 ng ml(-1) even with a UV absorbance detector. This system was applied to the determination of phthalates contained in laboratory distilled water and tap water samples.

Separation of benzene and deuterated benzenes by reversed-phase and recycle liquid chromatography using monolithic capillary columns.

An alternate pumping-recycle system utilizing a commercially available low dead-volume switching valve was developed for microcolumn LC. The recycle system had two separation columns, and the dead volume of the recycling lines was kept to a minimum by avoiding passage of the sample through the pump chamber, sample injector, and the normal path length of a conventional UV detector. The drawback of the high total back pressure caused by the second column that is placed after the detector was overcome by on-column detection, and this eliminated the need for a high pressure flow cell. The system was used for the separation of an authentic mixture of benzene, benzene-1,3,5-d3, and benzene-d6. Baseline separation was accomplished after six cycles and the calculated theoretical plate number for benzene was 230,000. It was observed that the theoretical plate number (N) increased linearly with increasing number of cycles, and the N per unit time increased with increasing inlet pressure. The separation conditions were optimized and the separation of benzene and benzene-d6 was accomplished within 75 min at 2.5 MPa inlet pressure.

Determination of inorganic anions in natural water by microcolumn ion chromatography with on-column enrichment.

An on-column enrichment method was developed for the rapid determination of inorganic anions in natural water. The system was assembled from a syringe pump, a six-port switching valve with a sample-enrichment loop, a separation column and a UV detector. The enrichment efficiency of the system was tested by using inorganic anions as samples. The limits of detection were between 0.6 and 7.7 microg/L. The system was applied to the determination of anions in river and pond-water samples.
