Functional promoter polymorphism in the TBX21 gene associated with aspirin-induced asthma.

Asthma is a phenotypically heterogeneous disorder with many etiologic factors and clinical characteristics. T-bet, a Th1-specific transcription factor of T-box family, has been found to control interferon-gamma (IFN-gamma) expression in T cells. Mice lacking the T-bet gene (tbx21) demonstrate multiple physiological and inflammatory features reminiscent of human asthma. In order to examine whether polymorphisms in the candidate gene, TBX21, located on chromosome 17q21.32, are related to the risk of human asthma phenotypes, we have searched for genetic variations in the human TBX21 gene and identified 24 single nucleotide polymorphisms (SNPs), including five novel SNPs, by direct sequencing in Japanese subjects. Among asthma phenotypes, a promoter -1993T-->C SNP, which is in linkage disequilibrium with a synonymous coding 390A-->G SNP in exon 1, is significantly associated with a risk of aspirin-induced asthma (AIA; P = 0.004, P(c) = 0.016). This association has also been confirmed in additional independent samples of asthma with nasal polyposis (P = 0.008), regardless of aspirin hypersensitivity. Furthermore, our data indicate that the -1993T-->C substitution increases the affinity of a particular nuclear protein to the binding site of TBX21 covering the -1993 position, resulting in increased transcriptional activity of the TBX21 gene. Thus, in addition to the antigen-driven excess Th2 response, increased T-bet (and subsequent IFN-gamma) production in human airways of individuals with the -1993T-->C polymorphism could contribute to the development of certain asthma-related phenotypes, such as AIA.

Effective comparison of two auto-CPAP devices for treatment of obstructive sleep apnea based on polysomnographic evaluation.

BACKGROUND: Automatic continuous positive airway pressure (auto-CPAP) machines differ mainly in algorithms used for respiratory event detection and pressure control. The auto-CPAP machines operated by novel algorithms are expected to have better performance than the earlier ones in the treatment of obstructive sleep apnea syndrome (OSAS). OBJECTIVES: The purpose of this study was to determine the therapeutic characteristics between two different auto-CPAP devices, i.e., the third-generation flow-based (f-APAP) and the second-generation vibration-based (v-APAP) machines, during the first night treatment of OSAS. METHODS: We retrospectively reviewed the polysomnography (PSG) recordings of 43 OSAS patients who were initially performed an overnight diagnostic PSG to confirm the disease and afterwards received the first night auto-CPAP treatment with using either the f-APAP (n=22) or v-APAP (n=21) device under another PSG evaluation. RESULTS: There were 13.6% and 61.9% patients who remained a residual apnea/hypopnea index more than 5 during the f-APAP and v-APAP application, respectively (P<0.005). The f-APAP was more effective than the v-APAP in reducing apnea/hypopnea index (P=0.003), hypopnea index (P=0.023) and apnea index (P=0.007), improving the lowest oxygen saturation index (P=0.007) and shortening stage 1 sleep (P=0.016). However, the f-APAP was less sufficient than the v-APAP in reducing arousal/awakening index (P=0.02). CONCLUSION: These findings suggest that the f-APAP works better than the v-APAP in abolishing breathing abnormities in the treatment of OSAS; however, the f-APAP device might still have some potential limitations in the clinical application.

Effect of probiotics on preventing disruption of the intestinal microflora following antibiotic therapy: a double-blind, placebo-controlled pilot study.

In this pilot-scale, double-blind, placebo-controlled trial, 30 patients with Helicobacter pylori infection were randomised into three groups prior to their 7 days eradication therapy, to study the effects of probiotic supplement comprising Lactobacillus acidophilus and Bifidobacterium bifidum on the intestinal microflora in response to antibiotic therapy. Group I received the placebo product from day 1 to day 15, Group II received placebo from day 1 to day 7 and probiotics from day 8 to day 15 and Group III received probiotics from day 1 to day 15. Patients provided stool samples for analysis on days 1, 7, 12, 17 and 27. For patients in Groups I and II, significant increases in the facultative anaerobe component of the microflora occurred between days 1 and 7. In Group I, the numbers remained elevated to day 27 but in Group II, the numbers decreased significantly between days 7 and 27 back to the starting levels. In Group III, the facultative anaerobe population remained stable throughout. The total anaerobe numbers increased significantly at day 27 than at day 1 for Group I, were unchanged throughout for Group II and decreased significantly for the patients in Group III between days 1 and 7 before reverting to the starting levels by day 27. From these results, it can be seen that probiotic supplementation modulates the response of the intestinal microflora to the effects of antibiotic therapy.

[Land cover classification in Xianghai Nature Reserve].

This paper discussed the application of Fuzzy ARTMAP neural network in land cover classification in Xianghai Nature Reserve. The structure and algorithm of Fuzzy ARTMAP neural network were described in detail, and an automatic adjustment method of vigilance parameter was put forward to solve the problem of searching the optimum value in the selection of vigilance parameter. The results showed that the automatic adjustment method could adjust network parameter automatically, and improve the running speed of network. In comparing with maximum likelihood classification method and traditional Fuzzy ARTMAP neural network, the Fuzzy ARTMAP neural network with the functioning of automatic adjustment depended less on samples and had higher accuracy, and thus, could effectively make the classification of TM image covering Xianghai Nature Reserve. It's shown from the classification that in the Xianghai Nature Reserve, farmland covered large area while grassland and marsh were facing degradation, and hence, corresponding countermeasures should be taken to improve the eco-environment of this Reserve.

Allergic symptoms and microflora in schoolchildren.

We studied 867 junior high school children and administered a questionnaire documenting allergic symptoms and environmental variables, and measured Immunoglobulin E serum levels and the immunoglobulin G titers of serum antibody to microflora. A total of 716 subjects were ultimately used for statistics; those with at least two of the following allergic symptoms: asthma, rhinitis, eczema, or food allergy, showed significantly higher IgG titers to Bactroides vulgatus than other groups. This finding suggests that a species of the Bacteroides genus of the intestinal microflora tends to affect the gut issues, but further studies are needed to clarify this.

[Microsatellite instability and relative gene expressions in sporadic and familial adenomatous polyposis adenomas].

In order to understand the role of mismatch repair (MMR) gene in colorectal carcinogenesis,microsatellite instability (MSI) status of 16 microsatellite loci of 62 adenomas from 59 patients, including sporadic and familial adeonmatous polyposis (FAP) adenomas were detected by microdissection-PCR-SSLP, and protein expressions of beta-catenin, P53, and BAX, etc. were assayed by immunohistochemistry. Results were as following: (1)The overall MSI alteration rate of the 16 loci was 14.4%. Different adenomas from the same patient showed different microsatellite alterations at the same loci; (2)All of the five FAP patients were MSI-L, three of which showed MSI at the locus of hMSH3; (3)The membrane expression rate of beta-catenin in adenomas and accompanied carcinomas was 42.9% and 11.4%, respectively (P<0.001); (4)Microsatellite alterations of the microsatellite loci of TP53, D5S346, TCF4(A)(9), TGFbetaRII(GT)(3) and TGFbetaRII(A)(10) were associated with the changes of their protein expressions. It could be concluded the following: (1)Microsatellite instability existed even in the early stage (adenomas) of colorectal tumorigenesis. The alterations of chromosome 1p, APC genes, and the TGFbeta signal transduction pathway could also be deduced; (2)In the progression of adenoma to carcinoma, the staining of beta-catenin would be transferred from membrane to cytoplasm and then nucleus, and the cytoplasm stain was stronger in carcinoma than that in adenomas. The abnormality of the signal transduction pathway of APC-beta-catenin-TCF4 could be concluded.

[DHPLC analysis of microsatellite instability in gastric cancer].

To establish a fast, simple and solid method of studying microsatellite instability (MSI) in gastric cancer, a panel of 12 microsatellite sites, D1S548, D1S552, D5S346, TP53, IGF II R(G)(8), IGF II R(CT)(5), TGFbetaR II(GT)(3), TGFbetaR II(A)(10), hMSH3(A)(8), hMSH6(G)(8), BAX(G)(8) and Bat26, were detected by denatured polyacrymide gel electrophoresis-silver stain in 28 gastric cancers. Bat26 was also analyzed by denatured high performance liquid chromatograph (DHPLC) at 50 degrees in the DNASep Cartridge. Two MSI-H (7.14%) and 15 MSI-L cancers (53.6%) were identified in 28 gastric cancers. Bat26 was positive only in 2 MSI-H cancers. The alterations of Bat26 and MSI-H status were coincident (P<0.01). The two Bat26+ cancers were also confirmed by DHPLC. Results obtained from DHPLC and gel electrophoresis were completely consistent. Thus, DHPLC analysis of Bat26 site may be a favorable method of detecting MSI-H status in gastric cancer, and be of clinical importance.

Expression of HSP70 in peripheral lymphocytes of the patients with allergic rhinitis.

The expression levels of heat shock protein 70 (HSP70) from peripheral lymphocytes of the patients with allergic rhinitis (AR) and the clinical implication were investigated. In the morning, 3 ml of fasting venous blood was taken out. The lymphocytes were isolated by using Ficoll-Hypaque and the expression of HSP70 in the lymphocytes was detected by using Western blot. In the AR patients the HSP70 level (41.49 +/- 15.77 integrated optical density, IOD) were significantly higher than that in the control group (23.89 +/- 10.13 IOD, P < 0.05). Western blot demonstrated that HSP70 bands in AR patients were more intensive than those in the control group. It was concluded that the elevated HSP70 level in peripheral lymphocytes of the AR patients might contribute to the development of AR.

Independent spectral representations of images for recognition.

In recent years, studies have shown that independent components of local windows of natural images resemble the receptive fields of cells in the early stages of the mammalian visual pathway. However, the role of the independence in visual recognition is not well understood. We argue that the independence resolves the curse of dimensionality by reducing the complexity of probability models to the linear order of the dimension. In addition, we show empirically that the complexity reduction does not degrade the recognition performance on all the data sets that we have used with an independent spectral representation. In this representation, an input image is first decomposed into independent channels given by the estimated independent components from training images, and each channel's response is then summarised by using its histogram as an estimate of the underlying probability model along that dimension. We demonstrate the sufficiency of the proposed representation for image characterization by synthesizing textures and objects through sampling and for recognition by applying it to large data sets. Our comparisons show that the independent spectral representation often gives improved recognition performance.

Aberrant crypt foci as microscopic precursors of colorectal cancer.

Since the first detection of aberrant crypt foci (ACF) in carcinogen-treated mice, there have been numerous studies focusing on these microscopically visible lesions both in rodents and in humans. ACF have been generally accepted as precancerous lesions in regard to histopathological characteristics, biochemical and immunohistochemical alterations, and genetic and epigenetic alterations. ACF show variable histological features, ranging from hyperplasia to dysplasia. ACF in human colon are more frequently located in the distal parts than in the proximal parts, which is in accordance with those in colorectal cancer (CRC). The immunohistochemical expressions of carcinoembryonic antigen (CEA), beta-catenin, placental cadherin (P-cadherin), epithelial cadherin (E-cadherin), inducible nitric oxide synthase (iNOS), cyclooxygenase (COX-2), and P16INK4a are found to be altered. Genetic mutations of K-ras, APC and p53, and the epigenetic alterations of CpG island methylation of ACF have also been demonstrated. Genomic instabilities due to the defect of mismatch repair (MMR) system are detectable in ACF. Two hypotheses have been proposed. One is the "dysplasia ACF-adenoma-carcinoma sequence", the other is "heteroplastic ACF-adenoma-carcinoma sequence". The malignant potential of ACF, especially dysplastic ACF, makes it necessary to reveal the nature of these lesions, and to prevent CRC from the earliest possible stage. The technique of magnifying chromoscope makes it possible to detect "in vivo" ACF, which is beneficial to colon cancer research, identifying high-risk populations for CRC, and developing preventive procedures.

[Expression of main heat stress proteins and p53 in nasopharyngeal carcinoma].

OBJECTIVE: To investigate the expression and the significance of main heat stress proteins (HSPs) and p53 in nasopharyngeal carcinoma. METHOD: Tissues homogenate and western blot were used to determine the level of HSPs and p53. RESULT: HSP27 and p53 elevated remarkably (P < 0.05) while HSP90 and HSP70 showed no significance between nasopharyngeal carcinoma and control tissues. Elevated level of HSP27 and p53 don't correlated with histological type and clinical stage. CONCLUSION: Expression of HSPs and p53 show some difference in nasopharyngeal carcinoma tissue. These data suggest that HSPs and p53 may play a role in the development of nasopharyngeal carcinoma.

[Express of plasma ROS, SOD and GSH-PX in patients with nasopharyngeal carcinoma].

OBJECTIVE: To investigate the levels of plasma reactive oxygen species(ROS), superoxide dismutase (SOD) and glutathione peroxidase (GSH-PX) in patients with nasopharyngeal carcinoma (NPC) and to observe the state of metabolism of free radical in patients with NPC and the effects of them on nasopharyngeal carcinoma. METHOD: The levels of plasma ROS, SOD and GSH-PX in 40 patients with NPC and 20 normal controls were identified by Fenton reaction method, xanthine oxidase method and dithiocarbamate method respectively. RESULT: Plasma ROS activity[(11,327.835 +/- 483.777)/(U.ml-1)] in NPC increased significantly in comparison with the control group [(10,262.712 +/- 525.287)/(U.ml-1)] (P < 0.05). Plasma SOD and GSH-PX activities [(76.619 +/- 22.283)/(NU.ml-1), (98.653 +/- 46.374)/AU] in NPC decreased significantly in comparison with the control group [(154.603 +/- 27.241)/(NU.ml-1), (307.872 +/- 116.275)/AU] (P < 0.05). But there was no significant difference of the levels of plasma ROS[(10,987.235 +/- 469.340)/(U.ml-1), (10,081.53 +/- 488.409)/(U.ml-1), (12,085.044 +/- 564.721)/(U.ml-1)], SOD[(79.517 +/- 20.368)/(NU.ml-1), (67.975 +/- 24.202)/(NU.ml-1), (58.692 +/- 29.946)/(NU.ml-1)] and GSH-PX[(101.362 +/- 59.859)/AU, (89.338 +/- 42.259)/AU, (101.890 +/- 33.559)/AU] in different pathological kinds of NPC. CONCLUSION: The metabolism of free radical is disturbed and the ability of anti-oxidation injury decreases significantly in patients with NPC, so anti-oxidation treatment should be given to suppress the development of NPC.

[Studies on improving crop nutritional quality through gene engineering].

This paper summarizes the studies on improving crop nutritional quality including protein, saccharide and lipid through gene engineering in recent 10 years. Special emphasis is laid upon the improvement of protein contains and amino acid components. The food safety caused probably by gene engineering and some ways to solve the problem are introduced briefly.

[Effect of FACE on competition between a C3 crop (rice, Oryza sativa) and a C4 weed (barnyardgrass, Echinochloa crusgalli)].

The growth and competition of a C3 crop rice and a C4 weed, barnyardgrass under FACE(free-air carbon dioxide enrichment) conditions were studied by field experiment. The results showed that under FACE condition, the biomass and yield, leaf number, tiller number, and leaf area index of rice increased, while those of barnyardgrass decreased. Both of the leaf area indexed of rice and barnyardgrass were decreased, while their net assimilation rate increased. When the ratio of planting density of rice and barnyardgrass was 1:1, all of the ratio of biomass, yield, leaf area index, tiller and net assimilation rate were increased under FACE condition. It indicated that the competition between rice and barnyardgrass changed under FACE condition. The competition ability of rice(C3 crop) was enhanced relatively, while that of barnyardgrass weakened.

[Responses of agricultural crops of free-air CO2 enrichment].

Over the past decade, free-air CO2 enrichment (FACE) experiments have been conducted on several agricultural crops: wheat(Triticum aestivum L.), perennial ryegrass (Lolium perenne), and rice(Oryza sativa L.) which are C3 grasses; sorghum (Sorghum bicolor (L.) Möench), a C4 grass; white clover (Trifolium repens), a C3 legume; potato (Solanum tuberosum L.), a C3 forb with tuber storage; and cotton (Gossypium hirsutum L.) and grape (Vitis vinifera L.) which are C3 woody perennials. Using reports from these experiments, the relative responses of these crops was discussed with regard to photosynthesis, stomatal conductance, canopy temperature, water use, water potential, leaf area index, shoot and root biomass accumulation, agricultural yield, radiation use efficiency, specific leaf area, tissue nitrogen concentration, nitrogen yield, carbohydrate concentration, phenology, soil microbiology, soil respiration, trace gas emissions, and soil carbon sequestration. Generally, the magnitude of these responses varied with the functional type of plant and with the soil nitrogen and water status. As expected, the elevated CO2 increased photosynthesis and biomass production and yield substantially in C3 species, but little in C4, and it decreased stomatal conductance and transpiration in both C3 and C4 species and greatly improved water-use efficiency in all the crops. Growth stimulations were as large or larger under water-stress compared to well-watered conditions. Growth stimulations of non-legumes were reduced at low soil nitrogen, whereas elevated CO2 strongly stimulated the growth of the clover legume both at ample and under low N conditions. Roots were generally stimulated more than shoots. Woody perennials had larger growth responses to elevated CO2, while at the same time, their reductions in stomatal conductance were smaller. Tissue nitrogen concentrations went down while carbohydrate and some other carbon-based compounds went up due to elevated CO2, with leaves and foliage affected more than other organs. Phenology was accelerated slightly in most but not all species. Elevated CO2 affected some soil microbes greatly but not others, yet overall activity appears to be stimulated. Detection of statistically significant changes in soil organic carbon in any one study was impossible, yet combining results from several sites and years, it appears that elevated CO2 did increase sequestration of soil carbon. Whenever possible, comparisons were made between the FACE results and those from prior chamber-based experiments reviewed in the literature. Over all the data and parameters considered in this review, there are only two parameters for which the FACE- and chamber-based data appear to be inconsistent. One is that elevated CO2 from FACE appears to reduce stomatal conductance about one and a half times more than observed in prior chamber experiments. Similarly, elevated CO2 appears to have stimulated root growth relatively more than shoot growth under FACE conditions compared to chamber conditions. Nevertheless, for the most part, the FACE- and chamber-based results have been consistent, which gives confidence that conclusions drawn from both types of data are accurate. However, the more realistic FACE environment and the larger plot size have enabled more extensive robust multidisciplinary data sets to be obtained under conditions representative of open fields in the future high-CO2 world.
