Epithelial tumor suppressor deletion promotes neuroendocrine differentiation in bladder cancer and reveals homoharringtonine as a candidate vulnerability.

Neuroendocrine bladder carcinoma (NEBC) is a highly aggressive malignancy with unresolved lineage determinants and limited preclinical models, hindering mechanistic investigation and therapeutic development. Here, we sought to assess whether bladder epithelial-derived models are competent to acquire neuroendocrine lineage programs under defined tumor suppressor alterations and to identify candidate therapeutic vulnerabilities in these systems. We integrated genomic and transcriptomic analyses of human NEBC with genetically engineered mouse models, epithelial-derived bladder organoids, and patient-derived NEBC models. Human NEBC exhibited dominant RB1 and TP53 alterations and an epithelial transcriptional continuum consistent with lineage plasticity. In vivo, intravesical Adeno-Cre-mediated tumor suppressor deletion predominantly generated sarcoma-like tumors, whereas epithelial-restricted organoid models recapitulated the molecular and neuroendocrine features of human NEBC, supporting epithelial lineage competence for neuroendocrine differentiation. Patient-derived models and human NEBC specimens further supported epithelial identity in NEBC. Using these complementary platforms, drug screening identified homoharringtonine (HHT) as a candidate therapeutic vulnerability in the tested NEBC systems. HHT suppressed neuroendocrine marker expression, induced apoptosis, and attenuated IL6-JAK-STAT3 signaling. Together, these findings describe complementary epithelial-derived NEBC models and support further investigation of HHT as a candidate therapeutic vulnerability.

Palmitate activates AMP-activated protein kinase and regulates insulin secretion from beta cells.

AMP-activated protein kinase (AMPK) is an energy sensor that regulates cellular metabolism. Changes in AMPK activity contribute to the regulation of insulin secretion. Epidemiological evidence links the ingestion of saturated fatty acid with hyperinsulinemia. The aim of the present study was to examine the effects of palmitate on beta cell AMPK activity and insulin secretion. Isolated rat islets and MIN6 beta cells were treated acutely (5-60 min) or chronically (24 h) with palmitate. Insulin secretion, AMPK and acetyl CoA carboxylase phosphorylation were assessed. The acute effects of palmitate included AMPK activation and augmentation in insulin secretion. Activation of AMPK by 24h pretreatment with palmitate suppressed glucose-stimulated insulin secretion, but not the response of insulin secretion to combined stimuli of glucose and palmitate. This study demonstrated that palmitate availability affected beta cell AMPK activity. In beta cells, an increase in AMPK activity may be required for fatty acid-induced fatty acid oxidation and prevention of lipotoxicity.

Polyamidoamine dendrimers-assisted electrodeposition of gold-platinum bimetallic nanoflowers.

Novel Au-Pt bimetallic flower nanostructures fabricated on a polyamidoamine dendrimers-modified surface by electrodeposition are reported. These polyamidoamine dendrimers were stable, and they assisted the formation of Au-Pt bimetallic nanoflowers during the electrodeposition process. These nanoflowers were characterized by field-emitted scanning electron microscopy (FE-SEM), energy-dispersive spectroscopy (EDS), X-ray photoelectron spectroscopy (XPS), X-ray diffraction, and electrochemical methods. FE-SEM images showed that the bimetallic nanoflower included two parts: the "light" and the "pale" part. The two parts consisted of many small bimetallic nanoparticles, which was attributed to the progressive nucleation process. Moreover, the "light" part contained more bimetallic nanoparticles. The morphologies of bimetallic nanoflowers depended on the electrodeposition time and potential and the layer number of assembled dendrimers. The average size of nanoflowers increased with the increase in electrodeposition time. The layer number of assembled dendrimers obviously affected the size and morphologies of the "pale" parts of deposited nanoflowers. EDS and XPS indicated that the content of Au element was higher than that of Pt element in the nanoflowers. The bimetallic nanoflowers-modified electrode had electrochemical properties similar to those of bare gold and platinum electrodes. It also exhibited significant electrocatalytic activities toward oxygen reduction.

Noninvasive diagnosis of cellular and antibody-mediated rejection by perforin and granzyme B in renal allografts.

A major milestone in transplantation would be the use of biomarkers to monitor rejection. We examined the association between perforin and granzyme-B gene expression detected in the peripheral blood of renal allograft recipients with cellular and antibody-mediated rejection. Furthermore, we judged the appropriateness of assigning negative rejection statuses to persons without a biopsy whose grafts were functioning well clinically. Of the 46 patients who completed the study, recipients with cellular rejection had higher perforin and granzyme-B levels compared with nonrejectors (p = 0.006). Interestingly, recipients with antibody-mediated rejection also had higher perforin and granzyme-B levels compared with nonrejectors (p = 0.04). Patients with high levels of granzyme B had a probability of rejecting that was 26.7 times greater than those patients with low levels of granzyme B. Perforin and granzyme B had sensitivities of 50% and specificities of 95% in predicting rejection (cutoff value = 140). Assigning negative rejection statuses to recipients without a biopsy whose grafts were functioning well did not have a major effect on the direction or significance of covariate values. This study suggests that perforin and granzyme-B gene expressions in peripheral blood are accurate in detecting both cellular and antibody-mediated rejection.

Simple online recognition of optical data strings based on conservative optical logic.

Optical packet switching relies on the ability of a system to recognize header information on an optical signal. Unless the headers are very short with large Hamming distances, optical correlation fails and optical logic becomes attractive because it can handle long headers with Hamming distances as low as 1. Unfortunately, the only optical logic gates fast enough to keep up with current communication speeds involve semiconductor optical amplifiers and do not lend themselves to the incorporation of large numbers of elements for header recognition and would consume a lot of power as well. The ideal system would operate at any bandwidth with no power consumption. We describe how to design and build such a system by using passive optical logic. This too leads to practical problems that we discuss. We show theoretically various ways to use optical interferometric logic for reliable recognition of long data streams such as headers in optical communication. In addition, we demonstrate one particularly simple experimental approach using interferometric coinc gates.

Buildup of gold nanoparticle multilayer thin films based on the covalent-bonding interaction between boronic acids and polyols.

A novel method for the fabrication of gold nanoparticle multilayer films based on the covalent-bonding interaction between boronic acid and polyols, poly(vinyl alcohol) (PVA), was developed. The multilayer buildup was monitored by UV-vis absorbance spectroscopy, which showed a linear increase of the film absorbance with the number of adsorbed Au layers and indicated the stepwise and uniform assembling process. The atomic force microscopy (AFM) image showed that a compact gold multilayer thin film was successfully assembled. The residual boronic acid group on the surface of thin film could incorporate glycosylated-protein horseradish peroxidase (HRP), and good catalytic activity for H2O2 could be observed.

[The emission of organic-inorganic single quantum well with organic heterojunction].

Organic-inorganic single quantum well with organic heterojunction ITO\SiO2 (60 nm)\MEH-PPV(40 nm)\Alq3 (40 nm)\SiO2 (60 nm)\Al has been fabricated. With the detailed investigation of photoluminescence and electroluminescence of this novel device, the authors found that the permittivity confinement effect and the quantum size effect have obviously an effect on the optical and electrical properties of organic-inorganic quantum well with organic heterojunction. The electroluminescence of organic-inorganic quantum well with organic heterojunction included three different emissions under alternative-current voltages: 410, 510 and 590 nm, which originated from radiative recombination of MEH-PPV, recombination of Alq3 excitons and MEH-PPV excitons respectively.

[Spectral properties of energy transfer in doubly doped polymer electroluminescence].

The authors report a cascade energy transfer based on the long-range dipole-dipole Förster energy transfer between polymer and two fluorescent dyes in solid thin films. The authors studied the fabrication and emission characteristics of a new doubly doped system for color purity and energy transfer efficiency, in which two fluorescent dyes, i. e. TPB and Rubrene were doped into PVK. In the blend film, PVK acted as the energy donor, Rubrene as the fluorescent dye, and the assistant dopant TPB was used to facilitate the energy transfer from PVK to the dye Rubrene. Orange emission comes from Rubrene via a cascade energy transfer from PVK toTPB and then from TPB to Rubrene.

[The influence of holes buffer layer doped with different one-dimensional nanomaterials on the performance of polymer light-emitting diodes].

The effect of holes buffer layer PEDOT doped with one-dimensional nanomaterials on the performance of polymer light-emitting diodes was investigated. The photoluminescence efficiency of double layer sample PEDOT/MEH-PPV was enhanced by doping one-dimensional nanomaterials (TiO2 nanotube and ZnO nanorod) into PEDOT layer. Stronger interaction of one-dimensional nanomaterials and PEDOT suppressed the creation of defect states which can quench radiative recombination of excitons. The electroluminescence efficiency of device was improved by 2 and 2.5 folds by doping TiO2 nanotubes and ZnO nanorods into holes buffer layer PEDOT respectively.

[Optical and morphological investigation on the interaction of dual dopants in poly (N-vinylcarzole)].

The effect of optical and electrical properties of poly(N-vinylcarzole) (PVK) doped with two dyes, i. e. 8-tris-hydroxyquinoline (Alq3) and 4-(dicyanomethylene)-2-tert-butyl-6 (1, 1, 7, 7-tetramethyljulolidyl-9-enyl)-4H-pyran (DCJTB), on the energy transfer and charge trapping processes was investigated. The phase separation in blends film at different doping concentration was also studied. More homogeneous dispersion of dyes in PVK with increasing doping concentration was showed. The results indicate that there is a certain interaction of Alq3 and DCJTB in this dual-doped system. It is the incorporation of DCJTB that untangled the aggregation of Alq3 owing to the interaction of DCJTB and Alq3. But for higher doping concentration, DCJTB results in an isolated charge transport channel that decreases the electroluminescence (EL) operating voltage.

Automated measurement of cell motility and proliferation.

BACKGROUND: Time-lapse microscopic imaging provides a powerful approach for following changes in cell phenotype over time. Visible responses of whole cells can yield insight into functional changes that underlie physiological processes in health and disease. For example, features of cell motility accompany molecular changes that are central to the immune response, to carcinogenesis and metastasis, to wound healing and tissue regeneration, and to the myriad developmental processes that generate an organism. Previously reported image processing methods for motility analysis required custom viewing devices and manual interactions that may introduce bias, that slow throughput, and that constrain the scope of experiments in terms of the number of treatment variables, time period of observation, replication and statistical options. Here we describe a fully automated system in which images are acquired 24/7 from 384 well plates and are automatically processed to yield high-content motility and morphological data. RESULTS: We have applied this technology to study the effects of different extracellular matrix compounds on human osteoblast-like cell lines to explore functional changes that may underlie processes involved in bone formation and maintenance. We show dose-response and kinetic data for induction of increased motility by laminin and collagen type I without significant effects on growth rate. Differential motility response was evident within 4 hours of plating cells; long-term responses differed depending upon cell type and surface coating. Average velocities were increased approximately 0.1 microm/min by ten-fold increases in laminin coating concentration in some cases. Comparison with manual tracking demonstrated the accuracy of the automated method and highlighted the comparative imprecision of human tracking for analysis of cell motility data. Quality statistics are reported that associate with stage noise, interference by non-cell objects, and uncertainty in the outlining and positioning of cells by automated image analysis. Exponential growth, as monitored by total cell area, did not linearly correlate with absolute cell number, but proved valuable for selection of reliable tracking data and for disclosing between-experiment variations in cell growth. CONCLUSION: These results demonstrate the applicability of a system that uses fully automated image acquisition and analysis to study cell motility and growth. Cellular motility response is determined in an unbiased and comparatively high throughput manner. Abundant ancillary data provide opportunities for uniform filtering according to criteria that select for biological relevance and for providing insight into features of system performance. Data quality measures have been developed that can serve as a basis for the design and quality control of experiments that are facilitated by automation and the 384 well plate format. This system is applicable to large-scale studies such as drug screening and research into effects of complex combinations of factors and matrices on cell phenotype.

[A clinical study of Periocline on the treatment of severe chronic periodontitis].

PURPOSE: To evaluate the clinical effect of subgingivally administering Periocline as an auxiliary treatment of chronic periodontitis. METHODS: A randomized, single-blind, self-control study was designed.Following initial therapy ,Periocline was administered into the periodontal pocket of treatment sides at baseline and week 1 to week 4, but the control sides received no drug. For both groups, standard clinical indices including plaque index (PLI), gingival index(GI), probing depth (PD) and response to the ointments were evaluated at baseline week 1,week 4 and week 8. The plaque index and gingival index were analyzed by Wilcoxon' test for each treatment. The probing depth was analyzed by Student's t test. RESULTS: At the baseline, there was no significant difference in the indexes between the two groups (P>0.05). At 1 week after treatment, both groups showed significant improvement in their examined indexes (P<0.01). At week 1 the PLI was significantly lower in the experimental group than that in the control group; at week 4 and week 8 there was no significant difference between the groups. At 1 week after the treatment, the GI had no significant difference between the two groups (P>0.05); but there was difference 4 weeks and 8 weeks after treatment. At 1 week after treatment, PD was significantly different between the two groups (P<0.05), while at week 4 and week 8, were significant difference between the two groups (P<0.01). CONCLUSION: Administration of the local drug delivery system of Periocline ointment as the auxiliary treatment of severe chronic Periodontitis could get good clinical results.

Model-based analysis of matrix metalloproteinase expression under mechanical shear.

We report the development of a model-based analysis for identification of the role of transcription factor (TF) binding motifs. A nonlinear mathematical model was formulated to establish the quantitative relationship between the temporal expression profiles and the distribution of known TF binding motifs on regulatory DNA regions. In order to evaluate biological meaning of the nonlinear model, the role of TF binding motifs predicted by the model was examined by a promoter competition assay where specific TF binding motifs were inactivated by a transient transfer of the DNA fragments consisting of the TF binding motifs. Using the shear stress responses of a family of matrix metalloproteinases in human synovial cells as a model system, we showed that the nonlinear formulation was able to approximate the experimentally observed expression profile better than the linear formulation, and the stimulatory and inhibitory roles of TF binding motifs extracted from the model were validated by the competition assay. The results support that an integrated usage of the nonlinear model and the biochemical evaluation assay would contribute to identifying critical regulatory DNA elements in mechanical responses in connective tissue.

Systems analysis of matrix metalloproteinase mRNA expression in skeletal tissues.

The availability of human genome sequences provides life scientists and biomedical engineers with a challenging opportunity to develop computational and experimental tools for quantitatively analyzing biological processes. In response to a growing need to integrate experimental mRNA expression data with human genome sequences, we present here a unique analysis named "Promoter-Based Estimation (PROBE)" analysis. The PROBE analysis is "systems analysis" of transcriptional processes using control and estimation theories. A linear model was built in order to estimate the mRNA levels of a group of genes from their regulatory DNA sequences. The model was also used to interpret two independent datasets in skeletal tissues. The results demonstrated that the mRNA levels of a family of matrix metalloproteinases can be modeled from a distribution of cis-acting elements on regulatory DNA sequences. The model accurately predicted a stimulatory role of cis-acting elements such as AP1, NFY, PEA3, and Sp1 as well as an inhibitory role of AP2. These predictions are consistent with biological observations, and a specific assay for testing such predictions is proposed. Although eukaryotic transcription is a complex mechanism, the two examples presented here support the potential use of the described analysis for elucidating the functional significance of DNA regulatory elements.
