Cross-contamination between processing equipment and deli meats by Listeria monocytogenes.

Contamination of luncheon meats by Listeria monocytogenes has resulted in outbreaks of listeriosis and major product recalls. Listeriae can survive on processing equipment such as meat slicers which serve as a potential contamination source. This study was conducted to determine (i) the dynamics of cross-contamination of L. monocytogenes from a commercial slicer and associated equipment onto sliced meat products, (ii) the influence of sample size on the efficacy of the BAX-PCR and U.S. Department of Agriculture-Food Safety and Inspection Service enrichment culture assays to detect L. monocytogenes on deli meat, and (iii) the fate of L. monocytogenes on sliced deli meats of different types during refrigerated storage. Three types of deli meats, uncured oven-roasted turkey, salami, and bologna containing sodium diacetate and potassium lactate, were tested. A five-strain mixture of L. monocytogenes was inoculated at ca.10(3) CFU onto the blade of a commercial slicer. Five consecutive meat slices were packed per package, then vacuum sealed, stored at 4 degrees C, and sampled at 1 and 30 days postslicing. Two sample sizes, 25 g and contents of the entire package of meat, were assayed. Total numbers of L. monocytogenes-positive samples, including the two sample sizes and two sampling times, were 80, 9, and 3 for turkey, salami, and bologna, respectively. A higher percentage of turkey meat samples were L. monocytogenes positive when contents of the entire package were assayed than when the 25-g sample was assayed (12.5 and 7.5%, respectively). Lower inoculum populations of ca. 10(1) or 10(2) CFU of L. monocytogenes on the slicer blade were used for an additional evaluation of oven-roasted turkey using two additional sampling times of 60 and 90 days postslicing. L. monocytogenes-positive samples were not detected until 60 days postslicing, and more positive samples were detected at 90 days than at 60 days postslicing. When BAX-PCR and enrichment culture assays were compared, 12, 8, and 2 L. monocytogenes-positive samples were detected by both the enrichment culture and BAX-PCR, BAX-PCR only, and enrichment culture only assays, respectively. The number of L. monocytogenes-positive samples and L. monocytogenes counts increased during storage of turkey meat but decreased for salami and bologna. Significantly more turkey samples were L. monocytogenes positive when the contents of the entire package were sampled than when 25 g was sampled. Our results indicate that L. monocytogenes can be transferred from a contaminated slicer onto meats and can survive or grow better on uncured oven-roasted turkey than on salami or bologna with preservatives. Higher L. monocytogenes cell numbers inoculated on the slicer blade resulted in more L. monocytogenes-positive sliced meat samples. In addition, the BAX-PCR assay was better than the enrichment culture assay at detecting L. monocytogenes on turkey meat (P < 0.05).

Comparison of the reveal test, the U.S. Food and Drug Administration culture method, and selective media for recovery of Salmonella enteritidis from commercial egg layer flock environments.

Salmonella is the leading cause of foodborne illnesses in the United States, and Salmonella Enteritidis (SE) is the second most frequently isolated Salmonella serovar. Egg products are most often associated with outbreaks of SE infection. To prevent SE contamination of eggs, many producers are implementing flock inspections for SE at their facilities. A rapid and simple method for detecting SE in poultry environmental samples is critical for effective control of SE. In this study, the Reveal test for SE was compared with the conventional U.S. Food and Drug Administration (FDA) culture method for detecting SE in naturally contaminated environmental samples. The efficacy of two enrichment media, tetrathionate broth (TT) and Rappaport-Vassiliadis medium (RV), and three selective plating media, brilliant green agar with novobiocin (BGN), xylose lysine tergitol 4 agar (XLT4), and bismuth sulfite agar (BS), also were compared for SE isolation. One hundred twenty-eight environmental drag swab samples were collected from two previously identified SE-positive chicken flocks in two U.S. states and analyzed in parallel using the Reveal test and the FDA culture method. Twenty-five samples (19.5%) yielded SE when the Reveal test was used, and 23 samples (18.0%) were positive for SE by the FDA culture method. No significant difference in efficacy (P = 0.527) was found between the two methods. The Reveal test had a sensitivity, specificity, and accuracy of 83, 94, and 92%, respectively. Overall, a significantly greater number of positive samples was obtained after enrichment in RV compared with TT. XLT4 and BGN were more efficient than BS for isolating SE. However, no single method or medium successfully recovered SE from all SE-positive environmental samples.

Comparison of media and sampling locations for isolation of Listeria monocytogenes in queso fresco cheese.

Listeriosis associated with Hispanic-style soft cheese is an ongoing public health concern. Although rapid detection methods based on molecular and immunological technologies have been applied successfully for detecting Listeria monocytogenes in foods, obtaining isolates of the pathogen is a critical procedure for epidemiologic studies and regulatory analysis. Oxford agar, a medium recommended by the U.S. Food and Drug Administration Bacteriological Analytical Manual (BAM) to isolate L. monocytogenes from cheese, is unable to differentiate L. monocytogenes from other Listeria species. Hence, two selective isolation media, L. monocytogenes blood agar (LMBA) and Rapid 'L. mono agar (RLMA), were compared with Oxford agar for isolating L. monocytogenes from cheese. Queso fresco cheese was inoculated at 10(0) or 10(1) CFU/g with a five-strain mixture of L. monocytogenes or with the five-strain L. monocytogenes mixture and Listeria innocua. Cheese samples were stored at 21, 12, and 4 degrees C and Listeria counts were determined at 3, 7, and 10 days; 7, 10, 14, 21 days; and 2, 4, 8, and 12 weeks postinoculation, respectively. Surface and interior cheese samples as well as liquid exudate produced during storage were assayed individually to determine differences in Listeria contamination at different sampling locations. L. monocytogenes was more easily differentiated from L. innocua on RLMA than LMBA and Oxford agar. Similar L. monocytogenes counts (ca. 10(4) CFU/g) were obtained on the last sampling day on the surface and interior of cheese samples (P > 0.05) for all storage temperatures and both initial inoculation levels, but smaller cell numbers were detected in the exudate produced during storage. In addition, simultaneous inoculation of L. innocua with L. monocytogenes did not affect the final L. monocytogenes counts in the cheese. The amount of exudate released from the cheese and decrease of pH correlated with storage temperature. More exudate was produced and a greater decrease of pH occurred at 21 degrees C than at 12 or 4 degrees C. Our results indicate that RLMA is a suitable medium for isolating L. monocytogenes from queso fresco cheese. Higher counts of L. monocytogenes were obtained from surface and interior samples of cheese than from the exudate of the cheese during storage. In addition, pH may be a useful indicator of improperly stored queso fresco cheese.

[Effect of nitric oxide on Toll-like receptor 2/4 gene expression in the liver in acute hemorrhagic necrotizing pancreatitis in rats].

OBJECTIVE: To investigate the effect of nitric oxide (NO) on Toll-like receptors 2 and 4 (TLR 2/4) gene expression in the liver in acute hemorrhagic necrotizing pancreatitis (AHNP) in rats. METHODS: Seventy SD male rats were randomly divided into sham-operated group (n=10), AHNP group (n=30) and L-arginine (L-Arg) treatment group (n=30). Blood samples and liver tissues were obtained at 6 hours in sham-operated group, and 3, 6, 12 hours respectively in AHNP group and L-Arg-treated group. Alanine aminotransferase (ALT), aspartate aminotransferase (AST), amylase in serum, NO, tumor necrosis factor-alpha (TNF-alpha) in liver tissue were determined. TLR2/4 mRNA expressions in the liver tissue were measured by reverse transcription polymerase chain reaction (RT-PCR). RESULTS: TLR2/4 mRNA could be detected in the liver with low values in sham-operated group [(1.150+/-0.725)x10(-6), (11.450+/-1.724)x10(-4)], but they were markedly upregulated at 3 hours in AHNP group [(1.970+/-0.362)x10(-3), (175.000+/-0.111)x10(-3)], and peaked at 12 hours [(29.400+/-3.155)x10(-4), (267.300+/-8838)x10(-2), P<0.01). At the same time serum levels of amylase, ALT and AST increased, hepatic injuries were aggravated, the levels of TNF-alpha in the liver were increased and levels of NO in the liver were lowered (P<0.05 or P<0.01). Treatment with L-rg could effectively inhibit TLR2/4 mRNA expression [3 h: (3.510+/-1.528)x10(-4), (13.500+/-2.231)x10(-2); 6 h: (21.000+/-5.346)x10(-4), (18.700+/-2.685)x10(-2); 12 h: (26.200+/-2.076)x10(-4), 1.959+/-0.270, P<0.05 or P<0.01] and alleviate hepatic injuries. The levels of serum amylase, ALT and AST lowered, and the levels of TNF-alpha in the liver were lowered and the levels of NO were markedly increased (P<0.05 or P<0.01). CONCLUSION: These findings suggest that the expression of TLR2/4 mRNA is increased in the liver in AHNP, and the hepatic injuries are aggravated. NO could markedly inhibit TLR 2/4 mRNA gene expression in the liver in AHNP.

[Combined free flap and pedicled pectoralis major myocutaneous flap in reconstruction of extensive composite defects in head and neck region: a review of 9 consecutive cases].

OBJECTIVE: To analyze the value and reliability of combined free flap and pedicled pectoralis major myocutaneous flap in the reconstruction of extensive composite head and neck defects. METHODS: Nine consecutive cases of combined free flap and pedicled pectoralis major myocutaneous flap transfers for extensive head and neck defects from March 2002 to April 2005 were reviewed. Data concerning the operation included defect description, type of free flap, recipient vessel and complications. RESULTS: There were 9 cases in this group, with 7 males and 2 females. Among the 9 free flaps, there were 6 radial forearm flaps, two fibula flaps, and one anterolateral thigh flap. The overall flap survival rate was 100% (all of 18 flaps), without partial or total flap necrosis. One radial forearm flap developed venous thrombosis 24 hours after operation, but salvaged by emergent exploration and reanastomosis of veins. The overall complication rate was 44.4%. CONCLUSION: In selected cases, the combined free flap and pedicled pectoralis major myocutaneous flap method provided satisfactory reconstruction for extensive head and neck defect, and simplified the double free flap method.

[Brain activities during maximum voluntary clenching with and without soft splint in patients with hemimasticatory spasms by functional magnetic resonance imaging].

OBJECTIVE: Functional magnetic resonance imaging (fMRI) was used to detect the cerebral cortical somatotopy during maximum voluntary clenching with and without soft splint in patients with hemimasticatory spasms (HMS) and the central mechanisms of HMS and the rule of the splint therapy. METHODS: Four HMS patients were selected and the spasms sides were right in two cases and left in the other two cases. FMRI images were obtained on Elscint/GE 2.0 Tesla MR system. Block design was used and the movement pattern was the onset of spasms after maximum voluntary clenching with and without soft splint. The fMRI data were analyzed by SPM99 software. RESULTS: With the onset of spasms after maximum voluntary clenching, the activation of motor cortex in 3 HMS patients was found lateral dominance on the left side, and the other one showed bilateral activation. All the 4 patients were found activation in cingulate area. With the onset of spasms after maximum clenching wearing soft splints, the activation of motor cortex showed no lateral dominance on the left side, and 3 patients were not found activation in cingulate area. CONCLUSIONS: The changes of the activation in motor cortex and cingulate area during the onset of spasms after clenching with and without soft splint might be the central mechanisms of the rule of splint therapy, through which the soft splint might function in alleviating muscle pain.

Analysis of intraspecific variation of Chinese Carthamus tinctorius L. using AFLP markers.

AIM: To investigate the intraspecific variation of Carthamus tinctorius L. (safflower) and establish foundation for further breeding of safflower germplasm resource and screening the quality correlation genes. METHODS: Amplified fragment length polymorphism (AFLP) was carried out to analyze genetic variation of 28 safflower populations collected in China. Unweighed pair-group method of with arithmetical averages (UPGMA) cluster analysis was used to construct a dendrogram and to estimate the genetic distances among the populations. RESULTS: All populations could be uniquely distinguished using 12 selected primer combinations. Similarity coefficients ranged from 0.48 to 0.96 among the populations. Dendrogram revealed distinct segregation of all the cultivars into three main groups and one midst group. CONCLUSION: Limited genetic diversity exists within the tested 28 collections at intra specific level and AFLP-based phyiogeny was not absolutely consistent with that based on morphological characters may be due to the interaction effect between genotype and environment.

[Contaminative features of heavy metals for tidal sediment cores in Tianjin Bohai Bay].

Three sediment cores were collected in Nov. of 2003 from Dagu estuary to Qikou estuary. The grain size, TOC and heavy metal contents of core sediments were analyzed in order to study the geochemical characteristics, contamination features and the spatial and vertical distribution characters for heavy metals of tidal zones in Bohai Bay. The grain size of tidal sediments becomes finer from north to south. Ultrafine, fine and power sand are the main compositions in the sediment of Dagu and Duliujian estuary. The sediment of Qikou estuary is mainly composed by power and ultrafine sand. The vertical distribution trends of three sediment cores indicate that the grain-size becomes bigger from the bottom to the upper. The distribution of Fe, Al, Mn contents has distinct negative correlation with the grain-size of sediment, that is to say, the fine-grained sediments have higher contents of Fe, Al, Mn. Pb, Zn and Cd are the dominating contaminative elements in tidal sediments of Bohai Bay. Their contents are higher than the corresponding environmental background values, indicating of the anthropogenic enrichment.

[Biological characteristics of Metarhizium anisopliae var. major and its virulence to white grubs].

With Beauveria brongniartii and Metarhizium anisopliae var. anisopliae as the reference, this paper studied the biological characteristics and invading ability of Metarhizium anisopliae var. major, a new variety separated from white grubs. The results showed that the optimal culture condition of M. anisopliae var. major was similar with that of other entomopathogenic species, and the optimal temperature was 25 degrees C. Among the three test media, PPDA was the most appropriate one, followed by SDAY and Czapek. M. anisopliae var. major had a stronger virulence than B. brongniartii at lower temperature, with an accumulative mortality of white grubs being 88.23% and 76.47%, respectively, after treated with soil and through spraying, suggesting that M. anisopliae var. major had a good prospect to widely apply in field.

[Bcl-XL antisense oligodeoxynucleotide sensitizes human esophageal cancer cell line to 5-fluorouracil].

OBJECTIVE: To investigate the effects of the Bcl-XL antisense oligodeoxynucleotides (ASODN) in suppressing the Bcl-XL expression and increasing the sensitivity of esophageal cancer cell line EC9706 to 5-fluorouracil (5-Fu). METHODS: The proliferation inhibitory rate of EC9706 was assessed by MTT, the expression of Bcl-XL was detected by RT-PCR and Western blot, and the apoptotic changes were examined by acridine orange (AO) fluorescent staining and flow cytometry, respectively. RESULTS: In the group of ASODN combined with 5-Fu, the proliferation inhibitory rate of esophageal cancer cells was 71.58%, the expression inhibitory rate of Bcl-XL mRNA was 81.25%, the expression of Bcl-XL protein was decreased significantly. The apoptosis rates detected by AO fluorescent staining and flow cytometry were 69.5% and (63.32 +/- 9.23)%, respectively. There were significant differences as compared with the cell control group, the vacuity control group, the N-ODN group, the ASODN group and the 5-Fu group, respectively (P < 0.05). CONCLUSION: Bcl-XL ASODN combined with 5-Fu can effectively inhibit the proliferation of esophageal cancer cells in vitro. Bcl-XL ASODN can significantly increase the sensitivity of esophageal cancer cells to 5-Fu through suppressing the expression of Bcl-XL.

[Inhibitory effects of natural plant extracts on Verticillium albo-atrum].

This paper studied the inhibitory effects of 54 kinds of ethanol-extracted plant solutions on Verticillium albo-atrum. The results showed that 15 kinds of these extracts could inhibit the growth of verticillium albo-atrum mycelium, with an inhibitory rate more than 50%, among which, the inhibitory rate of Asarum sieboldii, Coptis chinensis, Magnolia officinalis, Acacia catechu, Sophora flavescens, Glycyrrhiza uralensis, Cnidium monnieri, Platycodon grandiflorum and Allium. cepa. extracts was higher than 65%, and that of Cnidium monnieri extract reached 86.84%. A total of 16 kinds of plant extracts decreased the spore germination of V. albo-atrum, with the inhibitory rate higher than 70%, and there were 7 kinds of plant extracts whose inhibitory rate reached 95%. Almost no spore bourgeon was found after treated with Asarum sieboldii, Coptis chinensis, and Magnolia officinalis extracts. The extract of Acacia catechu did not inhibit the growth of mycelium, but restrained the spore germination by 100%. Cnidium monnieri extract could strongly inhibit the growth of mycelium, but had a less effect on spore germination rate (only by 11.3%). The active substances found in natural plant extracts which had the inhibitory effects on pathogen brought us a new and promising method to deal with V. albo-atrum and other vegetable diseases.
