Photochemistry of CH3Mn(CO)5: a multiconfigurational ab initio study.

The electronic spectroscopy of CH3Mn(CO)5 has been investigated by means of ab initio multiconfigurational MS-CASPT2/CASSCF calculations. The absorption spectrum is characterized by a series of Metal-Centered (MC) excited states in the UV energy domain (below 290 nm) that could be responsible for the observed photoreactivity starting at 308 nm. The upper part of the spectrum is overcrowded between 264 and 206 nm and dominated by a high density of Metal-to-Ligand-Charge-Transfer (MLCT) states corresponding mainly to 3d(Mn) --> pi*(CO) excitations. A non-negligible contribution of Metal-to-sigma-Bond-Charge-Transfer (MSBCT) states corresponding to 3d(Mn) --> sigma*(Mn-CH3) excitations is also present in the theoretical spectrum of CH3Mn(CO)5. However, in contrast to other transition metal hydrides and methyl substituted (HMn(CO)5, HCo(CO)4, and CH3Co(CO)4) these MSBCT transitions do not participate to the lowest bands of the spectrum as main contributions. The photochemistry of CH3Mn(CO)5, namely the loss of a CO ligand vs. the metal-methyl bond homolysis, is investigated by means of MS-CASPT2 states correlation diagrams. This study illustrates the complexity of the photodissociation mechanism of this class of molecules, which involves a large number of nearly degenerate electronic states with several channels for fragmentation.

Regulation of Vitamin D hydroxylases gene expression by 1,25-dihydroxyvitamin D3 and cyclic AMP in cultured human syncytiotrophoblasts.

Human placenta synthesizes and metabolizes 1,25-dihydroxyvitamin D(3) [1,25(OH)(2)D(3)/calcitriol] through the activity of 25-hydroxyvitamin D(3)-1alpha-hydroxylase (CYP27B1) and 1,25(OH)(2)D(3)-24-hydroxylase (CYP24A1), the two key enzymes for Vitamin D metabolism. In this study, calcitriol rapidly generated intracellular cAMP accumulation in cultured human syncytiotrophoblast cells, which in turn enhanced hCG secretion, a marker of trophoblast endocrine activity. The effects of 1,25(OH)(2)D(3) upon the expression of CYP27B1 and CYP24A1 were also investigated. 1,25(OH)(2)D(3) and activators of the PKA signaling system decreased the expression of CYP27B1, whereas increased CYP24A1 gene transcription. The use of a selective inhibitor of PKA (H-89) prevented the effects of calcitriol on CYP27B1 gene and hCG secretion, but not on CYP24A1 transcription. Addition of ZK 159222, a Vitamin D receptor (VDR) antagonist, blocked the calcitriol-mediated upregulation of 24-hydroxylase gene expression but did not affect calcitriol-induced downregulation of CYP27B1 gene or hCG stimulation. In addition, our study also demonstrated a role of calcitonin on Vitamin D hydroxylases gene regulation in placenta. The overall data suggest that calcitriol downregulates CYP27B1 expression via a cAMP-dependent signaling pathway, whereas upregulates 24-hydroxylase gene expression through a VDR-dependent mechanism.

A theoretical anharmonic study of the infrared absorption spectra of FHF-, FDF-, OHF-, and ODF- anions.

Anharmonic vibrational frequencies, equilibrium bond lengths, rotational constants, and vibrational absorption spectra have been calculated for the triatomic anions, FHF(-) and OHF(-), and the heavier isotopomers FDF(-) and ODF(-). The triatomic anions are assumed to maintain a collinear configuration throughout all calculations, so only the symmetric (nu(1)) and asymmetric (nu(3)) stretching modes are considered. The two-dimensional permanent dipole surfaces and potential energy surfaces are then constructed along bond coordinates, using high-level ab initio methods. Fundamental and combination bands are obtained from the vibrational eigenfunctions, resulting in anharmonic frequencies, which can be compared with the available theoretical and experimental data. The agreement is very good, especially for the pure symmetric modes, while the asymmetric ones show larger discrepancies, presumably due to the neglected coupling between stretching and bending modes. Strong inverse anharmonicity is found in the level spacing of the asymmetric modes, for both FHF(-) and OHF(-) anions. The calculated mixed modes (nnu(1)+mnu(3), n, m=0-3) also agree reasonably with the few available experimental data, supporting our model. Based on the vibrational eigenfunctions, isotope effects are also rationalized. Infrared absorption spectra are calculated from the dipole autocorrelation function for FHF(-) and FDF(-), and for OHF(-) and ODF(-). Peak locations and relative intensities are assigned in terms of the fundamental and mixed transitions.

Enhanced formation of non-phenolic androgen metabolites with intrinsic oestrogen-like gene transactivation potency in human breast cancer cells: a distinctive metabolic pattern.

Breast cancer is a sex steroid hormone-dependent malignant neoplasia. The role of oestradiol in this malignancy has been well documented; however, the involvement of androgens has remained controversial. To determine the role of non-phenolic androgen metabolites in human breast cancer, we studied the metabolism of [(14)C] testosterone and [(14)C] androstenedione in oestrogen-dependent MCF-7 cells and non-oestrogen-dependent MDA-MB 231 cells, at different substrate concentrations (1-10 muM) and time periods (30 min-48 h). Cultured non-oestrogen-dependent HeLa and yeast cells served as controls. Metabolites were identified and quantified by reverse isotope dilution. A distinctive pattern of androgen metabolism was identified in MCF-7 cells, being the 5alpha-androstane-3alpha,17beta-diol (3alpha,5alpha-diol) and its 3beta epimer (3beta,5alpha-diol), the major conversion products of testosterone (48.3%), with 5alpha-dihydrotestosterone as intermediary. The formation of 3alpha,5alpha-diol and 3beta,5alpha-diol (diols) was substrate concentration- and time-dependent, and abolished by finasteride. In contrast, very little of any diol formation was observed in MDA-MB 231, HeLa and yeast cell incubations. Additional enzyme gene expression studies revealed an overexpression of 5alpha-steroid reductase type-1 in MCF-7 cells, as compared with MDA-MB 231 cells. The oestrogen-like activities of diols were assessed in HeLa cells co-transfected with expression vectors for alpha or beta subtypes of the human oestrogen receptor (hER) genes and for an oestrogen-responsive reporter gene. The results show that 3beta, 5alpha-diol and to a lesser extent 3alpha,5alpha-diol bind with high relative affinity to hERalpha and hERbeta. Both diols induced hER-mediated reporter gene transactivation in a dose-response manner, similar to that induced by oestradiol, though with lower potency, an effect that was abolished by ICI-182 780. Furthermore, 3beta,5alpha-diol and to lesser extent 3alpha,5alpha-diol induced MCF-7 cell proliferation. The overall results demonstrated that MCF-7 cells exhibit enhanced expression and activity of androgen-metabolising enzymes, leading to rapid and large diol formation, and provide evidence that these androgen metabolites exert a potent oestrogen-agonistic effect, at genomic level, in oestrogen-dependent breast cancer cells. The data suggest that diols may act as in situ intracrine factors in breast cancer and that its formation can be pharmacologically inhibited.

Optical resolution of oriented enantiomers via photodissociation: quantum model simulations for H2POSD.

We demonstrate quantum mechanically how to resolve enantiomers from an oriented racemic mixture taking advantage of photodissociation. Our approach employs a femtosecond ultraviolet (UV) laser pulse with specific linear polarization achieving selective photodissociation of one enantiomer from a mixture of L and R enantiomers. As a result, the selected enantiomer is destroyed in the electronically excited state while the opposite enantiomer is left intact in the ground state. As an example we use H2POSD which presents axial chirality. A UV pulse excites the lowest singlet excited state which has nsigma* character and is, therefore, strongly repulsive along the P-S bond. The model simulations are performed using wavepackets which propagate on two dimensional potential energy surfaces, calculated along the chirality and dissociation reaction coordinates using the CASSCF level of theory.

Deciphering the reaction dynamics underlying optimal control laser fields.

Femtosecond high-resolution pump-probe experiments have been used together with theoretical ab initio quantum calculations and wave packet dynamics simulations to decode an optimal femtosecond pulse that is generated from adaptive learning algorithms. This pulse is designed to maximize the yield of the organometallic ion CpMn(CO)3 while hindering the competing fragmentation. The sequential excitation and ionization of the target ion are accomplished by an optimized field consisting of two dominant subpulses with optimal frequencies and time delays.
