Changes in peripheral blood Th1/Th2 cell balance in patients with condyloma acuminatum.

OBJECTIVE: To explore the association of peripheral blood T helper type 1 (Th1) and type 2 (Th2) imbalance with the occurrence of condyloma acuminatum (CA). METHODS: Interferon-gamma(IFN-gamma) and interleukin-4 (IL-4) secreted by CD4(+) T-cells were detected by flow cytometry in 40 patients with CA and 20 healthy volunteers, and significance of Th1/Th2 cell ratio in the etiology of the CA was analyzed. RESULTS: Peripheral blood Th1 cells that secreted IFN-gamma were significantly decreased in number in patients with CA (P <0.05), while Th2 cells producing IL-4 increased (P <0.05). The Th1/Th2 cell ratio was significantly reduced in CA patients as compared with the health control subjects (P <0.01). CONCLUSION: Increases of Th2 cells and decrease of Th1 cells in the peripheral blood of CA patients result in relative Th2 predominance and breach the Th1/Th2 balance, which may play an important role in the etiology and recurrence of CA.

[Spiral computed tomography (CT) and magnetic resonance imaging (MRI) in assessment of the skull base encroachment in nasopharyngeal carcinoma].

BACKGROUND & OBJECTIVE: With the general using of computed tomography (CT) and magnetic resonance imaging (MRI), it is important to determine which method is more sensitive in detecting the skull base encroachment in clinic. This article was designed to investigate the diagnostic value of CT and MRI in detecting the skull base erosion in nasopharyngeal carcinoma patients. METHODS: Sixty-one cases pathologically proven as nasopharyngeal carcinoma were selected from August 1993 to September 2001. three-dimensional reconstruction with spiral CT thin slices scan were performed in 8 cases. CT scan was performed with Elscient CT Twin Flash; axial scan was parallel to the OM line routinely from soft palate to the suprasellar cistern. There were 13 cases with enhancement scan. MRI scan was performed by Philips T5-II super-conducting magnetic resonance imaging system (0.5T). The standard quadrature head coil was used. Routine axial, sagittal, and coronal image with SE sequences were obtained. Scanned field ranged from the soft palate to the suprasellar cistern. After plain scan, enhanced scan was performed in 55 of 61 cases. RESULTS: MRI discovered the skull base encroached more precisely than CT, 17 cases by CT and 26 cases by MRI, respectively. The early bone marrow infiltration was seen at clivus, basilar pterygoid, and basilar sphenoid in 6 cases by MRI scan while CT scan showed no abnormal lesion at these sites. In addition, MRI revealed nasopharyngeal carcinoma tissue infiltrated along the mandibular nerve (3 cases) while CT scan showed no change of these structures. CONCLUSION: Both CT and MRI can reveal that the tumor encroaches on the skull base by either destroying the bony structure or breaking through the natural foramen. MRI is more sensitive than CT in detecting the skull base encroachment. MRI could reveal the early infiltration of the bone marrow and tumor infiltration along the mandibular nerve. MRI confirms the dimension of nasopharyngeal carcinoma more precisely than CT. The three dimension reconstructional spiral CT was directer in discovering the dimension of the tumor.

[Determination of four kinds of cell phenotypes of peripheral blood lymphocytes in patients with colorectal cancer].

BACKGROUND & OBJECTIVE: The functional status of lymphocyte is closely correlated with tumor occurrence and development. The multiple and complicated membranous proteins of lymphocytes are molecular biological foundations by which the cell activities such as membranous information transfer, activation, and apoptosis are proceeded. The purpose of this study was to investigate the expression of phenotypes CD8, CD28, CD49b, and CD49d in peripheral blood lymphocytes (PBL) in the patients with colorectal cancer and explore the relationship between these 4 kinds phenotypes and tumor biological characteristics. METHODS: Four kinds of cell phenotypes of PBL were measured in 35 patients with colorectal cancer (cancer group) and 30 patients without cancer (control group) using monoclonal antibody and flow cytometry. RESULTS: The expression of CD8 in cancer and control groups were 30.89+/-6.54(x+/-s, %) and 20.82+/-5.15, respectively. The expression of CD8 in cancer group was significantly higher than that in control group (P< 0.01). The expression of CD28, CD8(+)/CD28(+), CD49b, and CD49d in cancer group were much lower than those in control group(P< 0.01 or P< 0.05). The expression of CD8(+)/CD28(+), CD49b, and CD49b in Dukes D stage and Dukes B stage group were 7.62+/-1.63, 18.53+/-9.59, 59.85+/-16.18 and 10.01+/-2.90, 9.60+/-11.01, 73.12+/-10.86, respectively. The expression of CD8(+)/CD28(+), CD49b, and CD49b in Dukes D stage group were significantly lower than those in Dukes B stage group(P< 0.05). The expression of CD28, CD8(+)/CD28(+), CD49b, and CD49d in lymph node metastasis group and lymph node negative group were 37.62+/-7.38, 9.01+/-2.78, 20.03+/-9.07, 63.19+/-13.10 and 46.63+/-9.04, 13.11+/-2.96, 29.60+/-11.01, 73.12+/-10.86, respectively. The expression of CD28, CD8(+)/CD28(+), CD49b, and CD49d in lymph node metastasis group were much lower than those in lymph node negative group (P< 0.01 or P< 0.05). The expression of CD8 in serosa involved group and no serosa involved group were 34.18+/-3.86 and 28.19+/-5.65, respectively. The expression of CD8 in serosa involved group was higher than that in no serosa involved group(P< 0.01). The expression of CD28, CD8(+)/CD28(+), and CD49d in serosa involved group and no serosa involved group were 39.07+/-7.94, 8.78+/-2.59, 61.32+/-11.95 and 49.27+/-9.54, 12.29+/-3.48, 71.34+/-12.88, respectively. The expression of CD28, CD8(+)/CD28(+), and CD49d in serosa involved group were much lower than those in no serosa involved group (P< 0.01 or P< 0.05). CONCLUSION: Cell phenotypes of CD8, CD28, CD49b, and CD49d of PBL in the patients with colorectal cancer are closely related to tumor invasion and metastasis.

[Treatment of patients with chronic hepatitis B virus infect ion with lamivudine combined with famciclovir].

OBJECTIVE: To evaluate the clinical efficacy of combined treatment with lamivudine and famciclovir on chronic hepatitis B virus (HBV) infection. METHODS: Ninety patients with chronic HBV infection were divided into 3 groups. Group one had 28 patients and was treated with combination of lamivudine (0.1 g/d, PO) and famciclovir (1.5 g/d,PO) for 24 weeks. Group two and three had 30 and 32 cases, respectively, and were treated with lamivudine 100 mg/day PO and famciclovir (1.5 g/d,PO) alone. All the patients had positive markers of HBsAg, HBeAg and anti-HBcAg in serum assayed by ELISA and of HBV DNA tested by PCR. RESULTS: Three strategies of treatment had no different effects on the change of patients' ALT levels. The serum HBV DNA became negative after treatment in 89.3% (25/28) of patients treated with combination of lamivudine and famciclovir, 66.7% (20/30) of patients treated with lamivudine, and 40.6% (13/32) of patients treated with famciclovir. The rate of serum HBeAg loss in 3 groups were 28.6% (8/28), 23.3% (7/30) and 21.9% (7/32), respectively. CONCLUSIONS: The combination treatment of lamivudine and famciclovir for chronic HBV infection is safer than and superior to that of either drug alone.

[Synergetic transactivating effect of HCV core and HBV X proteins on SV40 early promoter/enhancer].

BACKGROUND: To investigate the synergetic transactivating effects of HCV core and HBV X proteins. METHODS: HCV core and HBV X protein-expressing plasmids were constructed with the vector pcDNA3.1(-). The plasmids were transfected into HepG2 cells and cotransfected Hep2 cells with reporter plasmid Psv-lacZ by lipofectamine plus reagents. The virus proteins produced in transient expression system were detected at the transcription and translation levels. The activity of b-galactosidase was detected, which reflected the transactivating function of the proteins. RESULTS: The expression of plasmids were detected in soluble protein cell extracts of transiently transfected HepG2 cells. HCV core protein activated the b-galactosidase expression at a value 4.9 times higher than the control, while HBV X protein activated at a value 3.5 times. It arrived at 9 times transfected with the plasmids simultaneously. The activating effect increased in relation to the amount of plasmids. CONCLUSIONS: The results suggested that the two kinds of virus proteins have transactivating effect on SV40 early promoter/enhancer, and they acted synergistically. These contribute to explain the mechanisms of liver injury or tumorigenesis induced by HCV or/and HBV infection.

[Survey of knowledge, attitude and behavior regarding safe injection among health practitioners in Shandong, China].

PURPOSE: To assess the knowledge, attitude and behavior of health practitioners (HPs) about safe injection in Shandong, China and to clarify risk factors related to unsafe injection and improper handling of used disposable syringes. METHODS: A cross-sectional study of 497 HPs in three-levels of health units (village clinic, township health center, and county hospital) in two areas of Shandong, China was carried out using a questionnaire. RESULTS: 468 HPs returned completed questionnaires. The proportion that did not receive formal medical education in the village (21.5%) was higher than in the township (9.9%) and in the county (2.5%). Proportions of HPs who only used disposable syringes were 97.0% in the county, 81.1% in the township and 69.0% in the village. Scores for safe injection knowledge among village HPs were significantly lower than those with township and county HPs (P < 0.001). Rate of unsafe injection practice was 6.2%, rate of improper handling of used disposable syringes was 7.6%, and both of which were significantly greater at the village level. Four factors were found to be significantly related to unsafe injection practice (P < 0.05): lower than middle level professional grade; knowledge scores of < 15 (full score 18); thinking that AIDS is not a terrible disease; and thinking that patients would accept the practice of sharing the same syringe for different persons. Four factors were found to be significantly related to improper handling of used disposable syringes (P < 0.05): working in the village; knowledge scores of < 15; thinking that patients would accept the practice of sharing the same syringe for different persons; and unwillingness to properly handle used disposable syringes. CONCLUSIONS: HPs working in village clinics had comparatively high rates of unsafe injection practice and improper handling of used disposable syringes, and a lower level of knowledge about safe injection. Lack of knowledge about safe injection was a risk factor for both unsafe injection and improper handling of used disposable syringe. Therefore, interventions should focus on this insufficient level of safe injection knowledge.

Effects of Salvia miltiorrhiza extracts on rat hypoxic pulmonary hypertension, heme oxygenase-1 and nitric oxide synthase.

OBJECTIVE: To investigate the effects of Salvia miltiorrhiza (SM) extracts on the expression of heme oxygenase-1 (HO-1) and nitric oxide synthase (NOS) in small pulmonary arteries (SPAs) of rats with chronic hypoxia. METHODS: After two weeks of hypoxia, rats were treated with diltiazem, and small, median, and large doses of SM extracts. The lungs were tested for the expression and distribution of HO-1, endothelial NOS (eNOS) and inducible NOS (iNOS) by using immunohistochemistry and Western blot method. RESULTS: Median and large doses of SM extracts significantly reduced hypoxia-induced media thickening in the SPAs (similar with diltiazem), recovered repaired ultrastructure injury, decreased HO-1 and iNOS levels, and increased eNOS expression in the SPAs. CONCLUSIONS: Median and large doses of SM extracts play significant roles in inhibiting structural remodeling in rats with hypoxic pulmonary hypertension. These effects might attribute to the suppression of HO-1 and iNOS, and the promotion of eNOS expression under the conditions of hypoxia.

[Study on expression of ING1, human telomerase reverse transcriptase and telomerase-associated protein 1 genes in human gliomas].

OBJECTIVE: To investigate the relationship between expressions of ING1 gene and genes of human telomerase reverse transcriptase (hTERT) and telomerase-associated protein 1 (hTP1) in human gliomas. METHODS: The expressions of ING1 mRNA and p33(ING1) protein, hTERT mRNA and protein, and hTP1 mRNA and protein in seventy human glioma specimens with different malignant grades were studied using in situ hybridization and immunohistochemistry. RESULTS: All of the 70 gliomas collected expressed hTP1 mRNA and protein and among them, 62 (88.6%) and 58 (82.9%) out of 70 expressed hTERT mRNA and protein respectively. The quantities of the four kinds of positive cells were correlated positively with one another (r = 0.758 - 0.882, P < 0.000 5), and all of them were significantly fewer in gliomas of WHO grade I - II than in grade III gliomas and the most in grade IV gliomas (P < 0.05 approximately 0.01). 66 (94.3%) and 62 (88.6%) out of 70 gliomas expressed ING1 mRNA and p33(ING1) protein respectively. The quantities of their positive cells were also correlated positively with each other (r = 0.831, P < 0.000 5), but the positive cells were more in gliomas of WHO grade I - II than in grade III gliomas and the fewest in grade IV gliomas (P < 0.01). The quantities of positive cells of ING1 mRNA and p33(ING1) protein were correlated negatively with those of hTERT mRNA and protein as well as hTP1 mRNA and protein respectively (r = -0.211 to -0.384, P < 0.05 approximately 0.001). CONCLUSIONS: The results suggest that all of the parameters concerned are valuable in evaluating the biological behavior of gliomas. In glioma cells, overexpressions of hTERT and hTP1 genes might be significant in inhibiting the expression of ING1 gene. The abnormal expressions of the three genes play possibly the important roles in the development and malignant progression of gliomas.

[The variation of Caspase3 activity in tanshinone induced NB4 cells apoptosis].

OBJECTIVE: To study the variation and role of Caspase3 activity in the process of Tanshinone (Tan II A) induced NB4 cells apoptosis. METHODS: NB4 cell apoptosis induced by Tan II A was demonstrated by cell morphology, DNA content analysis and DNA fragmentation assay. Caspase3 activity was determined by spectrofluorometry, and its inhibitory assay was performed using N-acetyl-Asp-Glu-Val-Asp-aldehyde(AC-DEVD-CHO). RESULTS: Tan II A could induce NB4 cell apoptosis accompanied with increase of caspase3 activity. The induction of NB4 cell apoptosis by use of Tan II A could be partially inhibited by AC-DEVD-CHO. CONCLUSION: The induction of NB4 cell apoptosis by Tan II A could be fulfilled by activating Caspase3.

[Effects of wogonin on inducing apoptosis of human ovarian cancer A2780 cells and telomerase activity].

BACKGROUND & OBJECTIVE: Inducing apoptosis and inhibiting the telomerase activity of tumor cells became a new therapeutic means for tumor. In vivo and in vitro experiments showed that wogonin possesses antioxidant activities and inhibitory effect on tumor cells growth. This study was designed to evaluate the effect of wogonin on telomerase activity and apoptosis of human ovarian carcinoma cell line A2780. METHODS: MTT assay, fluorescent microscopy, and DNA agarose gel electrophoresis were used to determine the role of wogonin on apoptosis of A2780 cells. The telomerase activity of A2780 cells were observed by using TRAP-ELASA method. RESULTS: A2780 cell growth was significantly inhibited by wogonin. The inhibiting effect showed concentration-dependent and time-dependent manners with IC(50) of 85 microg/ml. After treatment with 50 microg/ml and 100 microg/ml wogonin for 48 hours, A2780 cells showed morphological changes associated with the characters of apoptosis under fluorescent microscope. Typical DNA ladder was found using agarose gel electrophoresis. Telomerase activity of A2780 cells was gradually decreased with the increasing of wogonin concentration. When the concentration of wogonin was higher than 200 microg/ml, telomerase activity of A2780 cells was inhibited markedly. CONCLUSION: Wogonin can inhibit proliferation and induce apoptosis of A2780 cells within a certain concentration range(50-250 microg/ml). Anticancer effects of wogonin were associated with the induction of apoptosis and partly with the suppression of telomerase activity.

[Comparison of therapeutic effects between transcatheter arterial chemoembolization and transcatheter arterial-portal venous double chemoembolization in treatment of moderate and advanced stages of primary liver carcinoma].

BACKGROUND & OBJECTIVE: Many clinic data showed that it is difficult to control primary liver carcinoma (PLC) by transcatheter arterial chemoembolization (TACE) alone due to existence of double blood supply of liver by hepatic artery and portal vein. This study was designed to evaluate the efficacy of transcatheter arterial chemoembolization combined with portal venous chemoembolization (PVCE) in the treatment of moderate and advanced stages of PLC. METHODS: Fifty-eight cases of moderate and advanced stages of PLC were treated with a combination of TACE and PVCE (TACE/PVCE group) and 118 patients with moderate and advanced stages of PLC were treated with TACE alone (TACE group). RESULTS: (1)The total response rates were 86.2% in TACE+PVCE group and 69.5% in TACE group. The remarkable response rates were 31.0 % in TACE/PVCE group and 13.6% in TACE group (P< 0.05). (2)The half-year survival rates were 93.1% in TACE/PVCE group and 72.0% in TACE group (P< 0.05). The 1-year survival rates were 43.1% in TACE/PVCE group and 51.7% in TACE group. The 4-year survival rates were 6.9% in TACE/PVCE group and 5.1% in TACE group (P >0.05). (3) Side effects included fever, bellyache, white blood cell drop, hypohepatia, jaundice, and ascites. The degree of liver damage was positively associated with the amount of iodized oil. CONCLUSION: The combination of TACE and PVCE appears to promote the short-term efficacy but not the long-term efficacy, compared with TACE alone for patients with moderate and advanced PLC.

[Highly sensitive spectrophotometric determination of trace amounts tin in water by Sn(IV)-XO-PVA system].

A new spectrophotometric method for the determination of trace amount of tin (IV) has been developed based on the reaction of Sn (IV) with o-cresolphthalexon (XO), which forms a stable complex in the presence of polyvinyl alcohol (PVA). The complex exhibited a maximum absorbance at 544 nm. The molar absorption coefficient was 4.38 x 10(5) L.mol-1.cm-1. Recovery factor ranged from 97.4%-101.1%. Beers law was obeyed in the range of Sn 0.1-5 micrograms.(25 mL)-1. The complex was stable within 6 h. The suitable conditions for the reaction were investigated. The developed method has been applied to the determination of Sn (IV) in water samples. The result obtained is satisfactory.

[Spectra of the complexes of zinc salts with AMP].

The complexes of ZnCl2 and ZnSO4 with AMP in molar ratios of 1:1 and 1:2 were prepared through reflux in alcohol. The compositions of the complexes were determined to be Zn(AMP)2Cl2(A) and Zn(AMP)SO4(B) by chemical and elemental analyses. It was identified via IR analysis for the complexes that nitrogen atom of amino group and one of nitrogen atoms of pyrimidine coordinated to Zn2+ in bidentate fashion. Cl- in the complex of A and SO4(2-) in the complex of B participated in the coordination to Zn2+ and some feedback characteristic was found in the bond of N-->Zn2+, which was proved by XPS spectra. Study on 1H NMR for the complexes showed that two ligands joined the coordination to Zn2+ in the complex of A while one ligand did in the complex of B. Based on the above analyses, the structures of the complexes were presumed.

[Characterization of CD10 expression and its significance in minimal residual disease detection in childhood B-acute lymphoblastic leukemia].

To observe the expressions of CD10 in childhood B-acute lymphoblastic leukemia (B-ALL) and to define the role of CD10 in minimal residual disease (MRD) detection. 58 cases of childhood B-ALL were studied in this program. Four-color flow cytometry was used to analyze the characteristics of B-ALL phenotypes. The four-color fluorochrome labeled antibody combinations of CD10 with other markers were used to detect MRD. The results showed that CD10 overexpression (CD10(bright)) was detected in 65.5% (38/58) of B-ALL patients and a strong correlation between CD10(bright) and CD34 expression was also observed, i.e. CD10(bright) expression most frequently happened in B-ALL with high percentage of CD34 positive cells. In detection of MRD, CD10(bright), combined with other markers, could effectively distinguish normal cells with leukemic cells, even if there was no any other marker that can be used. It is concluded that CD10(bright) expression correlates with high expression of CD34 in B-ALL, it is a good marker for MRD detection. The combination of CD10 and other markers can be applied in B-ALL MRD detection with flow cytometry.

[Inhibition of antisense-endothelin converting enzyme RNA on interleukin-5 released from dust mite-challenged peripheral blood mononuclear cells in patients with allergic asthma].

OBJECTIVE: To investigate the effect of antisense endothelin converting enzyme (ECE) RNA on levels of cytokines released from CD(4)(+) lymphocytes in patients with allergic diseases responsive to house dust mites. METHODS: Peripheral blood mononuclear cells (PBMCs) were separated from 21 patients who were sensitive to dust mites. PBMCs from those patients were divided into two groups. No stimulation group (A group) induded A(1) group (anti-ECE epithelial cells + PBMCs) and A(2) group (control cells + PBMCs). Stimulation group (B group) included B(1) group (anti-ECE epithelial cells + PBMCs + dust mites extract) and B(2) group (control cells + PBMCs + dust mites extract). House dust mite extract (20 microg/ml) was added to the culture of stimulation group as described above. After 72 hours, supernatants from both groups were collected and the levels of IL-5 and IFN-ggr; released into the supernatants were detected by enzyme-linked immunoabsorbent assay. RESULTS: IL-5 levels were increased significantly after treatment with dust mite in twelve of 21 cases. No significant differences of IL-5 were found between the groups of A(1)[(6.0 +/- 1.3) x 10(-9) g/L] and A(2) [(7.5 +/- 1.1) x 10(-9) g/L] before house dust mite stimulation in the 12 cases (P > 0.05), and no significant differences in IFN-ggr; were found between the groups of A(1) [(63 +/- 26) x 10(-9) g/L] and A(2) [(70 +/- 52) x 10(-9) g/L] before house dust mite stimulation (P > 0.05). IL-5 level was increased in both groups after stimulation but it was significantly lower in the B(1) group [(8.2 +/- 1.6) x 10(-9) g/L] than that in the B(2) [(12.0 +/- 1.8) x 10(-9) g/L] (P = 0.047). It seemed that increased IFN-ggr; level after stimulation was higher in B(2) [(153 +/- 71) x 10(-9) g/L] than that in the B(1) group (100 +/- 41) x 10(-9) g/L], but there was no statistic significance (P > 0.05). In addition, our results also showed that the release of IL-5 was significantly increased in those cases with asthma, or asthma plus allergic rhinitis after dust mites stimulation [(44 +/- 15)%] compared with that in those with urticaria [(7 +/- 4)%] (P = 0.047). CONCLUSIONS: Antisense-ECE downregulated the IL-5 secretion from Th2 lymphocytes in patients with allergic asthma after being challenged with dust mites. It is indicated that ET-1 is an important cytokine involved with allergic airway inflammation. Antisense-ECE RNA management in airways may be of value in treating allergic asthma.

[Inhibitive effects of recombinant adenovirus-mediated human endostatin on the growth of human hepatocellular carcinoma xenograft in nude mice].

OBJECTIVE: To investigate the inhibitive effect on the growth of hepatocellular carcinoma (HCC) xenografted in nude mice by adenovirus-mediated human endostatin gene. METHODS: The expression efficiency of endostatin was examined after ECV-304 cells infected with Ad/hEndo by western blot. The hepatoma BEL-7402 cells were injected into Balb/c nude mice to detect the inhibition of Ad/hEndo on the growth of HCC xenografted in nude mice. The expression of endostatin mRNA in tumor tissue was analyzed with RT-PCR, and its distribution in vivo was also analyzed. RESULTS: High level expression of endostatin achieved in infected ECV-304 cells by western blot. Ad/hEndo significantly inhibited the growth of xenografted BEL-7402 tumors (F=4.061, P<0.05). The intratumoral microvessel density (MVD) decreased significantly in the treated mice (6.88+/-1.08 vs 13.60+/-1.71, t=9.216, P<0.01). The expression of endostatin mRNA in tumor tissue was detected by RT-PCR in 3 days after administration intratumorally with Ad/hEndo and almost disappeared in 7 days. Endostatin mRNA was mainly located in tumor tissue with a higher concentration than that in heart, lung, spleen and liver after Ad/hEndo administration. CONCLUSION: Adenovirus-mediated human endostatin gene can be expressed efficiently in vitro and in vivo, and significantly inhibit the growth of BEL-7402 xenografted tumors in nude mice.

[Treatment of one case of adult chronic myelogenous leukemia by two units of unrelated umbilical cord blood transplantation].

To explore the hematopoietic reconstitution and transplantation-related complications of two units of unrelated umbilical cord blood combined transplantation for the treatment of adult hematologic malignancies, one adult patient with chronic myelogenous leukemia received two units of unrelated umbilical cord blood combined transplantation. The conditioning regimen was busulfan and cyclophosphamide (Bu-Cy). GVHD prophylaxis regimen consisted of mycophenolate mofetil (MMF), cyclosporine A (CsA) and methotrexate (MTX). The patient received total nucleated cells 4.63 x 10(7)/kg with CD34+ cells 8.34 x 10(5)/kg. Engraftment was documented by the analysis of short tandem repeat with polymerase chain reaction (STR-PCR). The results showed that the STR-PCR analysis for peripheral blood at day 31, 46 and 71 after transplantation suggested that one of two units of cord blood were completely engrafted. The ANC > 0.5 x 10(9)/L in the patient occurred at day 23, blood platelet counts > 20 x 10(9)/L at day 33 and > 50 x 10(9)/L at day 47. The Philadelphia chromosome and bcr/abl fusion gene of the patient also turned to negative after engraftment. Acute GVHD grade II occurred at day 13 and cured after treatment. It is concluded that umbilical cord blood can be used in adult hematopoietic stem cell transplantation. Two or more units umbilical cord blood combined transplantation might be the way to solve the problem of the low counts of nucleated cells when be used for adult.
