Prevalence of dementia in China.

To obtain prevalence estimates of dementia in China, an analysis of 17 studies published in Chinese from 1990-1999 was carried out. The prevalence rates for the population aged 60 years and older were 1.26% for Alzheimer's disease (AD) and 0.74% for vascular dementia (VD). The prevalence of AD was 2.10% in women and 0.76% in men, while the prevalence of VD was 0.71 and 0.69%, respectively. The prevalence of AD among the three educational levels in our study (illiterate, primary school and high school) were 1.79, 0.45 and 0.15%, respectively, and those of VD were 0.26, 0.58 and 0.26%, respectively. Although the prevalence of AD (2.29%) was higher in urban than in rural areas (1.67%), the difference was not statistically significant. The difference between the prevalence of VD in urban (0.67%) and in rural areas (1.13%) was not significant either. The prevalence of AD increased with age, and gender was found to be associated with Alzheimer's disease. The prevalence of VD also increased with age, but there was no association between VD and gender.

Effects of Rheum tanguticum polysaccharide on TNBS -induced colitis and CD4+T cells in rats.

AIM: To study the effects of Rheum tanguticum polysaccharide(-1) (RTP(-1)) on ulcerative colitis in rats induced by 2, 4, 6-trinitrophene sulphonic acid (TNBS) and their possible mechanism. METHODS: RTP1 (200 mg.kg(-1), i.g.) extracted from Rheum tanguticum Maxim. ex Regel was administrated to rats with colitis induced by TNBS for 5 d, 7 d, 10 d and 14 d, respectively. The effects of RTP1 and dexamethasone (DX, 0.2 mg.kg(-1), i.g.) were contrastively investigated. The MPO level and SOD activity were determined by chromatometry. The expansion and protein expression of CD4+T lymphocytes isolated from colon mucosae and mesenteric lymph nodes of colitis rats were performed by immunohistochemical analysis and Western-blot methods. RESULTS: Treatments of RTP1 (200 mg.kg(-1), i.g.) significantly reduced diarrhea, mortality, colon mass, ulcer areas and MPO level in colon mucosae on days 5, 7, 10 and 14 (5.2+/-1.4, 5.4+/-0.7, 5.2+/-1.8, P<0.05. 3.4+/-0.8, P<0.01. 16.1+/-12.1, P<0.01. 31.8+/-8.6, 17.7+/-5.3, 12.7+/-4.1, P<0.05). The effects of RTP1 were similar to those noted above in DX group, but there were no immunosuppressive effects of DX in RTP(-1) group, such as body mass loss, thymus and spleen atrophy. The decreased number and down-regulated protein levels of CD4+T cells isolated from the colon of colitis rats treated with RTP1 were found. CONCLUSION: RTP1 shows significantly protective effects but lower side effects on rats with colitis induced by TNBS. The mechanism may be due to the resistance to over expansion of CD4.

Cloning of HBsAg-encoded genes in different vectors and their expression in eukaryotic cells.

AIM: To compare the efficiency of different plasmids as DNA vectors by cloning three HBsAg-encoded genes into two eukaryotic expression vectors, pRc/CMV and pSG5UTPL/Flag, and to express HBsAg S, MS, and LS proteins in SP2/0 cells, and to establish monoclone SP2/0 cell strains that are capable of expressing S or S2S proteins stably. METHODS: Segments of S, preS2-S, preS1-preS2-S genes of Hepatitis B virus were amplified by routine PCR and preS1-S fragment was amplified by Over-Lap Extension PCR. The amplified segments were cleaved with restricted endonuclease Hind III/Not I followed by ligation with pRc/CMV, or BamHI/EcoR I followed by ligation with pSG5UTPL/Flag. After the plasmid vectors were cleaved with the correspond enzymes, the amplified segments were inserted into pRc/CMV or pSG5UTPL/Flag plasmid vectors with T4 DNA ligase. KOZAK sequence was added before the initial ATG code of each fragment using specific primer. The inserted segments in the recombinant plasmids were sequenced after subcloning. BALB/c mice myeloma cells (SP2/0 cell line) were transfected with the recombinant plasmids. The expressions of the different recombinants were Compared by Western-blot, using a monoclonal anti-HBs antibody as the primary antibody and peroxidase-labeled multi-linker as the secondary. Stable SP2/0-pRc/CMV-S or SP2/0- pRc/CMV-MS clones were established through clone screening with G418. RESULTS: Fragments with anticipated size were harvested after PCR. After recombination and screening, the sequences of the inserted segments in the recombinants were confirmed to be S, preS2S, preS1-preS2S and preS1S encoding genes, determined by sequencing. The results of Western-blot hybridization were positive for the anticipated proteins. Among them, pRc/CMV-S or pRc/CMV-MS demonstrated the highest expressing their respective antigen. CONCLUSION: Eight recombinant plasmids expressing S, M, L or preS1S proteins are obtained. For hepatitis surface antigen expression in eukaryotic cells, the vector pRc/CMV is superior to pSG5UTPL/Flag, and pRc/CMV-S and pRc/CMV-MS are the most efficient in the pRc/CMV clones. SP2/0 cells stably expressing HBsAg are established, and may be used as target cells for evaluating the CTL activity of a DNA vaccine in vitro.

Effects of KAI1/CD82 on biological behavior of human colorectal carcinoma cell line.

AIM: To investigate the effects of KAI1/CD82 on biological behavior of colorectal carcinoma cells. METHODS: KAI1 cDNA was transfected into highly malignant colorectal carcinoma cell line, LoVo, which had low level of endogenous KAI1 expression, and established stable transfectant clones with high KAI1/CD82 expression. The cell-cell adhesion, cell aggregation, cell-matrix adhesion and cell invasion assay were performed to determine whether KAI1 transfectant could have an effect on proliferation, adhesion and tumor metastasis in comparison with the control transfectant cells. RESULTS: KAI1 expression did not alter in vitro cell proliferation. But the KAI1 transfectant cells exhibited significantly increased homotypic cell-cell adhesion and cell aggregation in comparison with the control transfectant cells(P<0.05). Furthermore, KAI1 expression significantly suppressed the cell adhesion to extracellular matrix components and in vitro cell invasion in KAI1-transfected LoVo cells. The data indicated that KAI1 expression significantly suppressed the metastatic potential of KAI1-transfected LoVo cells. CONCLUSION: Our results suggest that KAI1 might function as a negative regulator of colorectal carcinoma metastasis.

Preparation and characterization of polyclonal antibodies against ARL-1 protein.

AIM: To prepare and characterize polyclonal antibodies against aldose reductase-like (ARL-1) protein. METHODS: ARL-1 gene was inserted into the E. coli expression vector pGEX-4T-1(His)(6)C and vector pQE-30. Recombinant ARL-1 proteins named ARL-(His)(6) and ARL-GST were expressed. They were purified by affinity chromatography. Sera from domestic rabbits immunized with ARL-(His) (6) were purified by CNBr-activated sepharose 4B coupled ARL-GST. Polyclonal antibodies were detected by Western blotting. RESULTS: Recombinant proteins of ARL-(His)(6) with molecular weight of 35.7 KD and ARL-GST with molecular weight of 60.8 KD were highly expressed. The expression levels of ARL-GST and ARL-(His)(6) were 15.1 % and 27.7 % among total bacteria proteins, respectively. They were soluble, predominantly in supernatant. After purification by non-denatured way, SDS-PAGE showed one band. In the course of polyclonal antibodies purification, only one elution peak could be seen. Western blotting showed positive signals in the two purified proteins and the bacteria transformed with pGEX-4T-1(His) (6) C-ARL and pQE-30-ARL individually. CONCLUSION: Polyclonal antibodies are purified and highly specific against ARL-1 protein. ARL-GST and ARL-(His) (6) are highly expressed and purified.

Pathomorphological study on location and distribution of Kupffer cells in hepatocellular carcinoma.

AIM: To clarify the location and distribution of Kupffer cells in hepatocellular carcinoma (HCC), and to investigate their role in hepatocarcinogenesis. METHODS: Kupffer cells were immunohistochemically stained by streptavadin-peroxidase conjugated method (S-P). The numbers of Kupffer cells in cancerous, para-cancerous and adjacent normal liver tissues of 48 HCCs were comparatively examined. RESULTS: The mean number of Kupffer cells in cancerous, para-cancerous and adjacent normal liver tissues was 12.7+/-6.8, 18.1+/-8.2 and 18.9+/-7.9 respectively. The number of Kuppfer cells in cancerous tissues was significantly lower than that in para-cancerous tissues (t=2.423, P<0.05) and adjacent normal liver tissues (t=2.521, P<0.05). As tumor size increased, the number of Kupffer cells in cancerous tissues significantly decreased (F=4.61, P<0.05). Moreover, there was also a significant difference in the number of Kupffer cells among well-differentiated, moderately-differentiated and poorly-differentiated cases(F=4.49, P<0.05). CONCLUSION: This study suggests that decrease of Kupffer cells in HCCs may play an important role in the carcinogenesis of HCC, the number of Kupffer cells in HCC is closely related to the size and differentiation grade of the tumor.

Three-dimensional power Doppler imaging in depicting vascularity in hepatocellular carcinoma.

OBJECTIVE: To assess the ability of three-dimensional power Doppler imaging to depict vascularity in hepatocellular carcinoma. METHODS: Forty-three patients with hepatocellular carcinomas were subjected to two- and three-dimensional power Doppler imaging, and 14 of them also underwent angiography. The delineated amounts of intratumoral Doppler signals were compared between 2 methods of power Doppler imaging in 43 patients. In the 14 patients who also underwent angiography, the patterns of blood supply depicted by 2 methods of power Doppler imaging were compared with that depicted by angiography, and the intratumoral vascularity on three-dimensional projection images and angiograms was also quantitatively assessed by calculating the vascularity-area ratio with graphics software. RESULTS: In comparison with two-dimensional power Doppler imaging, three-dimensional power Doppler imaging was subjectively determined as showing more intratumoral Doppler signals in 32 (74.4%) of the 43 lesions. The accuracies in depicting vascular patterns were 64.3% (9 of 14) for three-dimensional projection images and 14.3% (2 of 14) for two-dimensional slices compared with the results of angiography. The vascularity-area ratios on three-dimensional power Doppler imaging projections and angiograms were 46.0% +/- 25.6% and 48.5% +/- 22.5% (mean +/- SD), respectively (P > .05). Three-dimensional projection images correlated significantly with angiograms in quantifying the vascularity (gamma = 0.87; P < .001). CONCLUSIONS: A three-dimensional power Doppler projection image gives a better overall picture of vascular distribution than a two-dimensional slice and correlates with angiography significantly for delineating vascularity in hepatocellular carcinoma.

Comparison of three- and two-dimensional sonography in diagnosis of gallbladder diseases: preliminary experience.

OBJECTIVE: To compare three-dimensional sonography with two-dimensional sonography for diagnosis of gallbladder diseases. METHODS: Sixty-three patients with gallbladder diseases were examined with two- and three-dimensional sonography. The morphologic features presented on both modalities were analyzed, and the diagnostic accuracies of both modalities were evaluated and compared. RESULTS: Both two- and three-dimensional sonography facilitated correct diagnosis in all patients with gallstones. Three-dimensional sonography had no influence on the diagnosis of gallstones compared with two-dimensional sonography. Three-dimensional sonography showed the granular surfaces in 18 (81.8%) of 22 cases of polypoid lesions and the pedunculated fundus in 19 (86.4%) of 22, whereas two-dimensional sonography displayed them in 10 (45.5%) and 3 (13.6%) of 22, respectively (P < .05 and .001). Three- and two-dimensional sonography made correct differential diagnoses between non-neoplastic and neoplastic polyps in 20 (90.9%) and 12 (54.5%) of 22, respectively (P < .05). In gallbladder carcinoma, both three- and two-dimensional sonography accurately showed all the associated gallstones and intrahepatic bile duct dilatation. Two-dimensional sonography could not correctly define the locations of the lesions in 2 (28.6%) and tumor extension in 1 (14.3%) of 7 cases with pathologically proved gallbladder cancer, whereas three-dimensional sonography improved the diagnosis in these cases. CONCLUSIONS: Three-dimensional sonography adds no advantages for diagnosis of gallstones compared with two-dimensional sonography, but it is better than two-dimensional sonography for differential diagnosis of gallbladder polyps and may improve the localization and staging for gallbladder carcinoma; however, additional studies are needed for further determination.

[A point mutation of protein C gene in a congenital protein C deficiency pedigree].

OBJECTIVE: To study the phenotypes and genotypes of a protein C (PC) deficiency pedigree. METHODS: Immunoassay (ELISA) was used for PC antigen and activated PC (APC) detection, PCR for amplification of the fragment of protein C gene exon II to exon IX, single-strand conformation polymorphism (SSCP) for difference of denatured cDNA and DNA sequencing for gene mutation. RESULTS: Four members in the pedigree were found to be PC antigen levels between 34.3% - 67.8% and PC activity between 22% - 49% which are lower in comparison with normal references (80% - 120% and 70% - 130%, respectively). A G-to-A mutation in exon VII of the protein C gene at 6 219 position was identified in 9 members. This mutation resulted in the substitution of Arg for Gln at 169 amino acid. CONCLUSION: The proband is of heterozygosity. The G6219 A mutation in exon VII of the protein C gene leads to the substitution of Arg 169 Gln. This mutation is reported for the first time in China.

[Proliferation of retrorsine-treated SD rat immortalized hepatocytes after intrasplenic transplantation].

OBJECTIVE: To investigate the effect of retrorsine on the proliferation of SD rat immortalized hepatocytes after intrasplenic transplantation. METHODS: Immortalized hepatocytes from SD rats was prepared by transfecting the primary hepatocytes with a retroviral vector SSR69 expressing the genes encoding Simian virus 40 T large antigen. Twenty-four adult SD rats were randomly assigned to 3 equal groups designated respectively as A, B, and C groups. Intraperitoneal retrorsine injections at the dose of 26 mg/kg.b.w. were performed twice in groups A and B at the interval of two weeks before hepatocyte transplantation. After 20% hepatic lobe resection were completed in all the groups, groups A and C received intraplenic transplant with immortalized hepatocytes, while group B received normal hepatocyte transplant. Post-mortem examinations were performed on the spleen samples with HE staining. Albumin expression in the transplanted hepatocytes was examined by immunohistochemical staining, and the ultrastructure of the transplanted cells was observed under transmission electron microscope. RESULTS: Histological examination demonstrated that the rat spleens in all the 3 groups contained cluster of transplanted hepatocytes from 1 week to 3 weeks after the transplantation, and the proliferation of the transplanted cells were comparable between groups A and B, but both of which showed significant difference from group C. Albumin excretion by the immortalized hepatocyte was identified in all the groups, and the transplanted immortalized cells preserved normal cell ultrastructure. CONCLUSION: Immortalized hepatocytes of SD rat have the normal cell ultrastructure and albumin-excretion function, and retrorsine can promote the proliferation of the transplanted hepatocytes.

[Effect of tumor suppressor gene KAI1 on the biological behaviors of human colorectal carcinoma].

OBJECTIVE: To determine whether the expression of tumor suppressor gene KAI1 can suppress the invasive or metastatic ability of colorectal carcinoma cells. METHODS: KAI1 cDNA was transfected into highly malignant colorectal carcinoma cell line LoVo, which had low levels of endogenous KAI1 expression. The expressions of KAI1 mRNA and protein were determined by immunohistochemistry and in situ hybridization, and the biological behaviors of the KAI1-transfected LoVo cells were investigated by cell-cell adhesion, cell-matrix adhesion and invasion assays. RESULTS: KAI1 expression significantly suppressed the metastatic potential of KAI1-transfected LoVo cells, whose metastasis suppression was correlated with the increased adhesion to homotypic cells and reduced tumor adhesion to extracellular matrix components. KAI1 expression significantly suppressed the in vitro cell invasion in KAI1-transfected LoVo cells. CONCLUSION: KAI1 might function as inhibitor of colorectal carcinoma metastasis.

[The clinical significance of SSA antigen and its different positive expressions].

OBJECTIVE: To distinguish the difference among the pathogenesis of 60 000 SSA antigen mono-antigen peptides (MAPs), and to discuss the nosogenesis of anti-60 000 SSA antibodies in correlative rheumatic diseases. METHODS: MAPs were artificially synthesized according to the amino acid sequence of 20 positive epitopes and 1 control segment of 60 000 SSA antigen. ELISA against recombinant 60 000 SSA antigen MAPs were done to detect anti-MAPs antibodies in 59 sera with anti-SSA antibodies, and analyzing the relations of anti-MAPs antibodies and organism injuries. RESULTS: Anti-60 000 SSA antibodies are multiple clone autoantibodies. Different patients have different immune response to MAPs. Anti-MAP(1 approximately 21) antibodies have no relation to skin lesion; anti-MAP(2,14,15,21) antibodies have positive relation to salivary gland lesion; anti-MAP(7,16) antibodies have negative relation to kidney lesion; anti-MAP(6) antibodies have negative relation to heart lesion. CONCLUSIONS: The appearance of some anti-MAPs antibodies implies the lesions of some organs, while the appearance of some other anti-MAPs antibodies have the protection to some organs. We concluded that it is maybe the different anti-MAPs antibodies that result in different clinical manifestations.

[Construction and expression of traceable DNA vaccine for prevention of caries].

OBJECTIVE: Streptococcus mutans has been proved as a causative bacteria of human dental caries. The surface protein antigen is one of the important pathogenic factors. The A region of the surface protein antigen pac gene (pacA) can enrich T-cells and B-cells epitope. In this study, a DNA vaccine carrying pacA and gfp gene (a reporter gene) for caries prevention was constructed. The DNA vaccine was liable to be traced in vitro and in vivo. METHODS: The fragment of pacA (1.3 kb) was amplified by PCR with the plasmid pPC41 as template, and inserted into a pEGFP-C1 vector. The recombinant plasmid produced was named as pEGFPC1-pacA. After the COS1 cell line was transfected by the recombinant plasmid, the expression of gfp was detected by observing the green fluorescence and measuring the fluorescence intensity, and the expression of pacA was detected by RT-PCR. RESULTS: Restricted analyzing, sequencing and PCR technique were employed to identify the recombinant plasmid. The phase and orientation of the pacA gene inserted into the vector pEGFPC1 were correct and no changes of their open reading frames were discovered. The transfected COS1 carrying green fluorescent protein (GFP) was observed; the GFP expression level of transfected cells was higher than that of controlled cell. The transcript of pacA gene was confirmed by RT-PCR. CONCLUSION: Construction of the recombinant plasmid was successful. The gfp gene and pacA gene in the plasmid was transcribed and expressed simultaneously in the transfected cells. Moreover, detection of GFP is simple, safe and effective for living cells.

[Discovery of analogous balanced translocation syndrome and its clinical genetics research].

OBJECTIVE: To find out the clinical genetic law of relation between a group of special chromosomal structural abnormality including bit Y, inv(9), S+, No. 1, 9, 16 chromosomal qh+/-, and the balanced translocation. METHODS: By means of binomial distribution method, 980 cases were analyzed for balanced chromosome translocation, big Y, inv(9), S+ and qh+/-, and according to the clinical features the 980 cases were divided into 4 groups: spontaneous abortion, abnormal sexual development, labouring disease and disabled or diseased condition of the patient. RESULTS: In these 980 cases of balanced translocation, big Y, inv(9), S+ and qh+/-, the chief clinical features caused by the various different conditions have a high similarity (P > 0.20-0.50). CONCLUSION: The big Y, inv(9), S+, and qh+/- leading to the occurrence of the chief clinical features belong to the analogous balanced translocation syndrome. This fact indicates that in the process of sexual reproduction, the minute unequivalent exchange in the molecular level of gamete is possible to have genetic effect in the descendants, and may cause a corresponding clinical feature, present the abnormal sex differentiation, in varying degrees, and influence the abnormal phenotype or the health of individuals.
