Strand bias in targeted gene repair is influenced by transcriptional activity.

Modified single-stranded DNA oligonucleotides can direct nucleotide exchange in Saccharomyces cerevisiae. Point and frameshift mutations are corrected in a reaction catalyzed by cellular enzymes involved in various DNA repair processes. The present model centers on the annealing of the vector to one strand of the helix, followed by the correction of the designated base. The choice of which strand to target is a reaction parameter that can be controlled, so here we investigate the properties of strand bias in targeted gene repair. An in vivo system has been established in which a plasmid containing an actively transcribed, but mutated, hygromycin-enhanced green fluorescent protein fusion gene is targeted for repair and upon conversion will confer hygromycin resistance on the cell. Overall transcriptional activity has a positive influence on the reaction, elevating the frequency. If the targeting vector is synthesized so that it directs nucleotide repair on the nontranscribed strand, the level of gene repair is higher than if the template strand is targeted. We provide data showing that the targeting vector can be displaced from the template strand by an active T7 phage RNA polymerase. The strand bias is not influenced by which strand serves as the leading or lagging strand during DNA synthesis. These results may provide an explanation for the enhancement of gene repair observed when the non-template strand is targeted.

Transcription factor EGR-1 inhibits growth of hepatocellular carcinoma and esophageal carcinoma cell lines.

AIM: The transcription factor EGR-1 (early growth response gene-1) plays an important role in cell growth, differentiation and development. It has identified that EGR-1 has significant transformation suppression activity in some neoplasms, such as fibrosarcoma, breast carcinoma. This experiment was designed to investigate the role of egr-1 in the cancerous process of hepatocellular carcinoma (HCC) and esophageal carcinoma (EC), and then to appraise the effects of EGR-1 on the growth of these tumor cells. METHODS: Firstly, the transcription and expression of egr-1 in HCC and EC, paracancerous tissues and their normal counterpart parts were detected by in situ hybridization and immunohistochemistry, with normal human breast and mouse brain tissues as positive controls. Egr-1 gene was then transfected into HCC (HHCC, SMMC7721) and EC (ECa109) cell lines in which no egr-1 transcription and expression were present. The cell growth speed, FCM cell cycle, plate clone formation and tumorigenicity in nude mice were observed and the controls were the cell lines transfected with vector only. RESULTS: Little or no egr-1 transcription and expression were detected in HCC, EC and normal liver tissues. The expression of egr-1 were found higher in hepatocellular paracancerous tissue (transcription level P=0.000; expression level P=0.143, probably because fewer in number of cases) and dysplastic tissue of esophageal cancer (transcription level P=0.000; expression level P=0.001). The growth rate of egr-1-transfected HHCC (HCC cell line) cells and ECa109 (EC cell line) cells was much slower than that of the controls. The proportion of S phase cell, clone formation and tumorigenicity were significantly lower than these of the controls' (decreased 45.5% in HHCC cells and 34.1% in ECa109 cells; 46.6% and 41.8%; 80.4% and 72.6% respectively). There were no obvious differences between SMMC7721 (HCC) egr-1-transfected cells and the controls with regard to the above items. CONCLUSION: The decreased expression of egr-1 might play a role in the dysregulation of normal growth in the cancerous process of HCC and EC. Egr-1 gene of transfected HHCC and ECa109 cells showed obvious suppression of the cell growth and malignant phenotypes, but no suppression in SMMC7721 (HCC cell line) cells.

Salvia miltiorrhiza monomer IH764-3 induces hepatic stellate cell apoptosis via caspase-3 activation.

AIM: To investigate the effects of IH764-3 on HSC apoptosis and the expression of caspase-3 protein in HSC apoptotic process. METHODS: HSCs were cultured in medium with different IH764-3 doses(10 microg.mL(-1) 20 microg.mL(-1) 30 microg.mL(-1) 40 microg.mL(-1)) and without IH764-3 and HSC proliferation was quantitatively measured by (3)H-thymidine incorporation. The morphological changes of HSCs were observed with transmission electron microscope after exposure to the dose of 40 microg.mL(-1) of IH764-3 for 48 hr. The apoptosis rates were detected by annexin V/PI and TdT-mediated dUTP nick end labeling (TUNEL). The expression of caspase-3 protein was determined by flow cytometry. RESULTS: (1) HSC proliferation rates induced with different IH764-3 doses (10 microg.mL(-1) 20 microg.mL(-1) 30 microg.mL(-1) 40 microg.mL(-1)) were significantly reduced compared with that of the control group (P<0.01). (2)With the doses above,IH764-3 dose-dependently produced HSC apoptosis rates of 6.7%(9.4%) 9.3%(21.6%) 15.1%(27.2%) and 19.0%(28.4%) respectively by annexin V and PI-labeled flow cytometry assay or TUNEL while it was only 2.3%(6.7%) in the control. (3) The expression of caspase-3 protein in IH764-3 groups was significantly higher than that of the control (P<0.05). CONCLUSION: Within the dose range used in present study IH764-3 can inhibit HSC proliferation as well as enhance HSC apoptosis. Furthermore IH764-3 can significantly increase the caspase-3 protein expression.

[Histological observation of the interface of oral epithelium and titanium implant with titanium dioxide on the surface]

OBJECTIVE: To investigate the healing process at the interface between oral epithelium and a titanium implant with titanium dioxide on the surface and whether a good epithelial barrier could be formed. METHODS: Titanium implants with titanium dioxide on the surface were placed in the mandibular alveolar area of Beagle dogs after their premolars had been removed three months previous. On the 15th day, second month and loading second month after implantation specimens were taken and examined after HE staining as well as by transmission electron microscopy. RESULTS: Fifteen days after implantation the epithelial collar was formed, junctional epithelium actively regenerated and attachment to the matrix on the surface of the implant occurred by hemidesmosomes. Two months after implantation junctional epithelium was increasing, showing three to four layers consistent with each other. Junctional epithelium appeared to be generated when stimulated by inflammation and shaped into epithelium rete pegs after loading for two months. CONCLUSION: Oral junctional epithelium can attach very well to titanium implants with titanium dioxide on the surface which also provides a biological barrier.

Three-dimensional gray scale volume rendering of the liver: preliminary clinical experience.

OBJECTIVE: To investigate the potential clinical usefulness of three-dimensional gray scale volume rendering in the liver. METHODS: Sixty-two patients were enrolled in the study and categorized into 2 groups: group I with ascites and group II without. Two types of volume-rendering modes, i.e., surface and transparent, were used to obtain the three-dimensional images. The data were reviewed to identify the differences between two- and three-dimensional images of the liver in each subject. RESULTS: In group I, three-dimensional sonography was superior to two-dimensional sonography in terms of surface features, edges, overall three-dimensional impression, image clarity, and structural relationships. However, it seemed that three-dimensional sonography in the surface mode was inferior to two-dimensional sonography in showing intrahepatic structures, because it had decreased resolution. In group II, three-dimensional sonography was superior to two-dimensional sonography with respect to the continuity of intrahepatic vessels, overall three-dimensional impression of the vessels, image clarity, and the relationship between lesions and neighboring vessels. However, the resolution of the lesions was decreased in 7 cases of hepatocellular carcinoma. CONCLUSIONS: Our experience suggests that three-dimensional gray scale volume rendering of the liver provides more diagnostic information than two-dimensional sonography; however, further studies are needed to evaluate its clinical importance.

Two novel mutations of the retinitis pigmentosa GTPase regulator gene in two Chinese families with X-linked retinitis pigmentosa.

OBJECTIVE: To detect mutations of the retinitis pigmentosa GTPase regulator (RPGR) gene in two Chinese X-linked retinitis pigmentosa families. METHODS: Fragments of exons 1-19 of the RPGR gene were amplified with intronic primers, using genomic DNA as template. The polymerase chain reaction (PCR) products were analysed by single-strand conformation polymorphism (SSCP) and direct sequencing. Mutations were identified by comparing DNA sequences of the patients with those of the normal controls. RESULTS: Two novel mutations, c1536delC and E332X, were identified in exons 12 and 9 of the RPGR gene in both families. Each mutation was the first mutation found in their respective exons. Both mutations were predicted to cause premature termination, which resulted in truncated proteins without normal functions of the RPGR products. CONCLUSIONS: Both mutations are the genetic basis of the pathogenesis in the respective families. Our data might be helpful in analysing the function of the RPGR protein.

Laboratory study on abutment movement of the combined clasp, the extension clasp and the telescopic crown.

OBJECTIVE: To study the influence of three kinds of retainers on the abutment movement of removable partial dentures (RPDs) for restoring the dentition with unilateral distal-extended tooth missing. METHODS: The combined clasp, the extension clasp and the telescopic crown were designed to retain RPDs. When food chip was masticated between dentitions, the displacement of abutments was measured by a high-sensitive-laser-transferring-detector, and then the data was statistically analyzed. RESULTS: All of abutments showed the inclination, torsion and vertical translocation. Of them, abutments with telescopic crowns had the lowest horizontal torsion (P < 0.01) and the highest vertical translocation (P < 0.01). CONCLUSIONS: Compared with the combined clasp and the extension clasp, the telescopic crowns enable abutments to bear the load more axial and the stress to be distributed more even, so the design of the telescopic crown may be considered more favourable for the health of abutments.

[The influence of N-acetyl-L-cysteine on pulmonary injury and oxygen stress after smoke inhalation injury].

OBJECTIVE: To investigate the influence of N-acetyl-L-cysteine (NAC) on pulmonary injury and oxygen stress caused by smoke inhalation injury. METHODS: Rats inflicted with smoke inhalation injury were employed as the model. WBC in BALF (bronchoalveolar lavage fluid), MPO (myeloperoxidase) activity, hydrogen peroxide (H(2)O(2)) content, GSH (glutathione) content and total antioxidant (TAO) capacity in pulmonary tissue were determined. RESULTS: Postinjury WBC in BALF, MPO activity in pulmonary tissue and H(2)O(2) content decreased obviously after NAC treatment. But the pulmonary tissue contents of GSH and ATO increased evidently after the treatment with NAC. CONCLUSION: NAC treatment could ameliorate pulmonary oxygen stress after smoke inhalation injury. AS a result, the pulmonary antioxidant capacity was improved.

[Effect of jaw shape on stresses at implant-bone interface: a three-dimensional finite element analysis].

OBJECTIVE: To study the influence of different shape of lower jaw on stress distribution at bone interface in the mandibular molar region of implant restoration models. METHODS: Six models with different lower jaw shapes were constructed by using three-dimensional finite element method. Implant-bone interface stresses in these models were studied under vertical and lateral loads. RESULTS: No matter in the condition of vertical loading of lateral loading, stress distribution patterns were similar in variant models and differences of stress values were not statistically significant. The maximal differences in Von-Mises stress, compressive stress and tensile stress among the six models were 6.4%, 2.8% and 6.2%respectively. CONCLUSION: It is rational to simplify the lower jaw shape in finite element analysis.

[Construction of recipient strain of expression-secretion system in filamentous fungi].

Glucoamylase overproducing A. niger T21 was mutated by UV mutagensis. An extracellular acid protease-deficient mutant, A. niger T21-201, which produced only 0.76% extracellular acid protease activity of the parent strain, was screened by casein-degradating plate and determination of protease activity. Moreover, the growth properties and the ability to secrete glucoamylase of A. niger T21-201 are identical to these of starting strain T21. The comparison of expression-secretion levels of heterologous gene in A. niger T21-201 and T21 was carried out with bacterial vhb as reporter, the level of expression-secretion of VHb in A. niger T21-201 was 6-7 times higher than that in T21, but the transcriptional levels of vhb gene in both strains were similar revealed by Northern blot. Therefore, it was demonstrated that the deficiency of acid protease of recipient T21-201 has significant effect on the protection of heterologous protein.

[Alteration of the Cell Cycle during the Differentiation of HL-60 Cells by Induction of Retinoic Acid]

To study the alteration of the cell cycle during the differentiation of human myeloid leukemia cell line HL-60 induced with all trans-retinoic acid (RA), the flow cytometry was used to assay the various phases of cell cycle in HL-60 cells treated with RA. The results showed: (1) S + G(2)/M phase proportion kept relative invariability during the 48 hours incubation of HL-60 cells with RA, however, the proportion alteration of S-phase cells was associated with the RA concentrations. At 10(-6) mol/L RA, the proportion of S-phase cells appeared a temporarily increasing peak followed by persistent decrease of S-phase proportion. At 10(-5) mol/L RA, S-phase cell proportion only appeared the persistent decreasing tendency. (2) Re-culture of HL-60 cells without RA showed the decrease of S + G(2)/M and S-phase cells was associated with the increase of differentiated cells, but not all HL-60 cells were triggered into differentiation at the same time. Once the cells start to differentiate, even if there is no RA presence, the HL-60 cells were still differentiated until maturation. In conclusion, HL-60 cells are able to differentate and maturate after exposure to RA for a period of time. S-phase proportion is related to the concentrations of RA. Once the cells start to differentiate, even if there is no RA presence, the HL-60 cells still differentiatiated until maturation.

DNA-based vaccination induces humoral and cellular immune responses against hepatitis B virus surface antigen in mice without activation of C-myc.

AIM:To develop a safe and effective DNA vaccine for inducing humoral and cellular immunological responses against hepatitis B virus surface antigen (HBsAg).METHODS:BALB/c mice were inoculated with NV-HB/s, a recombinant plasmid that had been inserted S gene of hepatitis B virus genome and could express HBsAg in eukaryotes. HBsAg expression was measured by ABC immunohis tochemical assay, generation of anti-HBs by ELISA and cytotoxic T lymphocyte (CTL), by MTT method, existence of vaccine DNA by Southern blot hybridization and activation of oncogene C-myc by in situ hybridization.RESULTS:With NV-HB/s vaccination by intramuscular injection, anti-HBs was initially positive 2 weeks after inoculation while all mice tested were HBsAg positive in the muscles. The titers and seroconversion rate of anti-HBs were steadily increasing as time went on and were dose dependent. All the mice inoculated with 100&mgr;g NV-HB/s were anti-HBs positive one month after inoculation, the titer was 1 1024 or more. The humoral immune response was similar induced by either intramuscular or intradermal injection. CTL activities were much stronger (45.26%) in NV-HB/s DNA immunized mice as compared with those (only 6%) in plasma-derived HBsAg vaccine immunized mice. Two months after inoculation, all muscle samples were positive by Southernblot hybridization for NV-HB/s DNA detection, but decreased to 25% and all were undetectable by in situ hybridiza-tion after 6 months. No oncogene C-myc activation was found in the muscle of inoculation site.CONCLUSION:NV-HB/s could generate humoral and cellular immunolo-gical responses against HBsAg that had been safely expressed in situ by NV-HB/s vaccination.
