[CD4+ CD25high regulatory T cells in peripheral blood of patients with B cell non-Hodgkin's lymphoma].

This study was aimed to analyze the proportion of T cell subsets, CD4(+) CD25(high) regulating T cells (Tr) in peripheral blood of B-NHL patients and their change regularity, and to investigate the immunosuppression mechanism and influence of chemotherapy on immunosuppression function of B-NHL patients. The peripheral blood was collected from 42 patients with B-NHL, 36 patients with B-NHL who achieved partial remission (PR) or complete remission (CR) after 4 - 6 cycles of chemotherapy and 15 healthy controls. By using monoclonal antibodies, the blood samples were evaluated with the flow cytometry for lymphocyte subsets and Tr cells. The results showed that the proportion of CD3(+) and CD4(+) T cells, and the ratio of CD4/CD8 in patients with B-NHL group was significantly less than those in the healthy controls, i.e. (68.33 +/- 15.27)% versus (72.06 +/- 9.26)%; (34.47 +/- 12.84)% versus (42.45 +/- 9.2)%; 1.36 +/- 0.26 versus 1.92 +/- 0.20, but the prevalence of the CD4(+) CD25(high) Tr cells was significantly higher than those in the healthy group [(4.10 +/- 1.21)% versus (2.04 +/- 1.03)%, P < 0.001]. The ratio of CD4/CD8 in chemotherapy group was lower than that in control, but the proportion of CD4(+) CD25(high) Treg cells in chemotherapy group was higher than those before chemo-/radio-therapy and the control. It is concluded that the relative increase of CD4(+) CD25(high) Tr cells in peripheral blood of B-NHL patients may be related to immunosuppression and tumor progression.

[Protective effect of mouse 2.5s nerve growth factor on PC12 cells from injury induced by 2, 5-hexanedione].

OBJECTIVE: To explore whether the nerve growth factor has protective effects on PC12 cells from injury induced by 2, 5-hexanedione. METHODS: With PC12 cells as the model of neurons, different concentrations of NGF were added into the culture of PC12 cells. Then cell viability was tested with MTT. The DNA fragment was observed with agarose gel electrophoresis. The apoptosis ratio was tested with flow cytometry (FACS). The p53 protein was detected with western blot. The differences among the groups were compared. RESULTS: Cell viabilities were increased with the increase of the concentrations of NGF (P < 0.05). The DNA fragment, the apoptosis ratio and the expression of p53 were all decreased with the increase of the concentrations of NGF (P < 0.05). CONCLUSION: The NGF might have direct nutritional effects on PC12 cells, and protect them from injury induced by 2, 5 HD. Moreover, it might also have anti-apoptosis effect to some extent.

[Effects of cordyceps sinensis alcohol extractive on serum interferon-gamma level and splenic T lymphocyte subset in mice with viral myocarditis].

AIM: To investigate the effects of early application of cordyceps sinensis alcohol extractive (CSAE) on myocardial injury, level of serum IFN-gamma and splenic T lymphocyte subset in murine viral myocarditis (VMC). METHODS: 100 male BALB/c mice were divided randomly into control group (CG), infectious group (IG) and CSAE therapy group (CTG). Mice in IG and CTG were infected with Coxsackie virus B3 (CVB3). The 14-day survival rates and myocardial histopathology were observed. Serum IFN-gamma level was detected by ELISA and splenic CD3+, CD4+, CD8+ T lymphocytes were detected by flow cytometry. RESULTS: Serum IFN-gamma level and the percentage of splenic T lymphocyte subset in IG were decreased significantly, and CD4+/CD8+ T cell ratio increased significantly as compared with those of CG. Myocardial lesions in CIG were attenuated, while the 14-day survival of mice 85% (vs IG 55%, P<0.05), serum IFN-gamma and the percentage of splenic CD3+, CD8+ T lymphocytes in CTG were significantly higher than those in IG. There were no significant differences of the percentage of T lymphocyte subset and CD4+/CD8+ T cell ratio between CTG and CG. CONCLUSION: CSAE plays an important protective role against viral-induced murine myocarditis by inducing IFN-gamma and regulating T lymphocyte.

[The temporal and spatial expression of CBFalpha1 in rabbit condylar cartilage during forward mandibular positioning].

PURPOSE: To identify the relationship between the expression of Core binding factor alpha1 (Cbfalpha1) in condylar cartilage and condylar growth modifications in growing rabbits following forward positioning mandibles with functional appliance. METHODS: Sixty rabbits with age of 8 weeks were randomly divided into the experimental group (n=36) and control group (n=24). Rabbits in the experimental groups were induced to forward their mandibles by a functional appliance. The animals in the two groups were killed after 3 days and 1, 2, 4, 8 and 12 weeks, respectively. The temporal and spatial expression of Cbfalpha1 in condylar cartilage was examined by immunohistochemical staining and measured with Image Computer Analysis Software. Date was statistically analyzed by Student's t test and one-way ANOVA. RESULTS: The expression of Cbfalpha1 was mainly found in the chondrocytes from the transitive zone and hypertrophic zone, and was also observed in the chondrocytes and osteoblasts of the mineralized zone. Compared with those of the age-matched controls, the positive signals for Cbfalpha1 of each zone in the experimental animals were significantly stronger (P<0.05), and the highest expression of transitive zone, hypertrophic zone and mineralized zone was detected after 4 weeks of treatment (71.00+/-1.52, 50.00+/-0.75, 82.00+/-0.39). CONCLUSION: The pattern of Cbfalpha1 expression is concerned with the adaptive remodeling of the condylar cartilage after functional appliance.

[Species diversity of bryophytes in West Tianmu Mountain of Zhejiang Province].

In this paper, an investigation was made on the bryophytes at different altitudes of West Tianmu Mountain, with their species composition, similarity, and alpha- and beta-diversities. The results showed that at altitude 1100 m, the bryophytes under deciduous broad-leaved forest had the highest species number and richness, and the highest similarity with the bryophytes under deciduous broad-leaved shrub at 1300 m. The beta diversity index at altitude 800-1100 m was the largest, suggesting an obvious change and alternation of bryophyte species there. At altitude 1100 m, the species diversity of bryophytes was the highest, where should be the key area for bryophyte diversity conservation in West Tianmu Mountain.

[Effects of prostate water pellets on the serum levels of IL-6 and TNF-alpha in rats with chronic bacterial prostatitis].

OBJECTIVE: To investigate the effect of Prostate Water Pellets (PWP) on serum levels of IL-6 and TNF-alpha in rats with chronic bacterial prostatitis (CBP). METHODS: Fifty healthy, adult, male Wistar rats with the weight of 180 - 220 g were divided into five groups of ten rats each at random: the control group, model group, high dosage of PWP group, low dosage of PWP group and levofloxacin group. The CBP rat model were created by injecting Escherichia coli (0.2 ml/rat, 10(7)/ml) into prostates. A month later after the model creation, high and low dosage of PWP suspension were used by gavage in CBP rats for 30 days, respectively. Levofloxacin tablets were used by gavage as the positive control, and distilled water was used by gavage in the control and model group. After thirty days, serum levels of IL-6 and TNF-alpha were measured with radioimmunoassay. RESULTS: Compared with the model group, serum levels of IL-6 and TNF-alpha of rats in high and low dosage PWP groups were lower and the difference was significant statistically (P < 0.01). CONCLUSION: It has effect to treat CBP rat with the PWP and its mechanism may relate with the decreasing levels of proinflammatory cytokines(IL-6 and TNF-alpha) in blood.

[Modified abdominal radical trachelectomy for treatment of cervical carcinomas].

OBJECTIVE: To investigate the possibility and effect of modified abdominal radical trachelectomy in treating women with early cervical cancers who desired to preserve fertility. METHODS: From October 2002 through September 2004, we performed abdominal radical trachelectomy with a modified Smith's style on 13 patients which were biopsy-proven early stage cervical carcinomas and had a desire to preserve fertility. Pelvic lymphadenectomies were performed in all patients. Radical trachelectomy was performed immediately if lymph nodes were negative. RESULTS: Thirteen women underwent this procedure. The average age was 29.4 years (range 21 - 35 years); 9 were null gravid and 4 were multipara. Mean operative time was 158 minutes (135 - 185 minutes), with a mean blood loss of 120 ml (100 - 180 ml), and an average hospital stay of 9.7 days. There was no intra- or post-operative complication. With an average follow-up of 21.3 months, there have been no recurrences. Five out of 13 patients have tried to get pregnant after operation, 3 patients conceived pregnancy, 1 patient was premature labor at 34 weeks, and the other 2 patients are pregnant at the moment. CONCLUSION: The modified abdominal radical trachelectomy which has less injury and fewer side-effects is safe and effective in treating patients with early cervical carcinoma, and can also give the patients chances to preserve their fertility in the future.

[Establishment of hepatic metastatic subline of nasopharyngeal carcinoma cell line].

OBJECTIVE: To establish a hepatic metastatic subline of nasopharyngeal carcinoma (NPC) cell line. METHODS: NPC cells metastatic to the liver were isolated from nude mice and the invasion and metastatic ability of the cells was observed in vivo and in vitro. RESULTS AND CONCLUSION: The invasion and metastasis activity of 5-8F-H3B-EGFP (an in vivo isolate with enhanced liver metastatic behaviors) were enhanced obviously in comparison with the parent cell line 5-8F-EGFP. This subline may be useful for cloning genes related to liver metastasis of NPC.

[Experimental study of fast formation of biomimetic apatite coatings on pure titanium surface].

OBJECTIVE: To investigate calcium-phosphate coating on commercial pure titanium substrate using a fast biomimetic procedure. METHODS: The titanium specimens were divided into two groups, group A and group B. The specimens of group A were only treated with the mixture of H2SO4/HCl for 30 min. The specimens of group B were treated with the mixture of H2SO4/HCl for 30 min, immersed into 5 mol/L NaOH solution at 60 degrees C for 24 h, and then heated to 600 degrees C and maintained for 1 h. All specimens were soaked into simulated body fluid-A (SBF) and SBF-B for each day. The surface morphology was observed by field scanning electron microscopy. RESULTS: Dense calcium-phosphate coating deposited on all the titanium surfaces of two groups. The calcium-phosphate coating consisted of spheric structure with a diameter of 1 approximately 3 microm, which was proved to be hydroxycarbonate apatite with the analysis of X-ray diffraction. CONCLUSIONS: Thin hydroxycarbonate apatite coating can deposited on pure titanium using a fast biomimetic procedure.

[An outbreak of human Streptococcus suis serotype 2 infections presenting with toxic shock syndrome in Sichuan, China].

OBJECTIVE: In mid-July 2005, five patients presented with septic shock to a hospital in Ziyang city in Sichuan, China, to identify the etiology of the unknown reason disease, an epidemiological, clinical, and laboratory study were conducted. METHODS: An enhanced surveillance program were established in Sichuan, the following activities were introduced: active case finding in Sichuan of (a) laboratory diagnosed Streptococcus suis infection and (b) clinically diagnosed probable cases with exposure history; supplemented by (c) monitoring reports on meningococcal meningitis. Streptococcus suis serotype 2 infection was confirmed by culture and biochemical reactions, followed by sequencing for specific genes for serotype and virulence factors. RESULTS: From June 10 to August 21, 2005, 68 laboratory confirmed cases of human Streptococcus suis infections were reported. All were villagers who gave a history of direct exposure to deceased or sick pigs in their backyards where slaughtering was performed. Twenty six (38%) presented with toxic shock syndrome of which 15 (58%) died. Other presentations were septicaemia or meningitis. All isolates were tested positive for genes for tuf, species-specific 16S rRNA, cps2J, mrp, ef and sly. There were 136 clinically diagnosed probable cases with similar exposure history but incomplete laboratory investigations. CONCLUSION: An outbreak of human Streptococcus suis serotype 2 infections occurred in villagers after direct exposure to deceased or sick pigs in Sichuan. Prohibition of slaughtering in backyards brought the outbreak to a halt. A virulent strain of the bacteria is speculated to be in circulation, and is responsible for the unusual presentation of toxic shock syndrome with high case fatality.

[Experimental study on protective effects of HupA in the treatment of isocarbophos poisoning].

OBJECTIVE: To investigate the therapeutic and prophylactic efficiency of HupA in mice with acute isocarbophos poisoning, and the protective effects of the HupA on AChE inhibited by isocarbophos. METHODS: Mice were randomizedly divided into the non-treatment group, the atropine control group, the HupA treatment group and the atropine and HupA combined treatment group. Toxic signs and survival rates were observed and compared among these groups. The AChE activity was monitored in the whole blood, the red cells and brain tissue exposed to isocarbophos in the either treated with HupA or non-treated groups. RESULTS: In HupA treatment group compared with the non-treatment group, toxic signs were significantly decreased and the survival rate was increased. The therapeutic efficiency in the atropine and HupA combined treatment group was better than other groups. After isocarbophos was administered, the AChE activity in the HupA treatment group and the non-treatment group was decreased. However, the AChE activity in the whole blood (1.096 +/- 0.111), (1.262 +/- 0.146), (1.181 +/- 0.353) U/ml, the red cells (0.798 +/- 0.063), (1.000 +/- 0.176), (0.837 +/- 0.331) and the brain tissue (13.739 +/- 2.970), (18.507 +/- 3.466), (10.764 +/- 2.212) U/g in HupA treatment group 0.5, 1 and 2 hours after isocarbophos was administered was significantly higher than those in the non-treatment group (P < 0.05 or P < 0.01). CONCLUSION: HupA has therapeutic effect on mice with acute isocarbophos poisoning. The protective effect of HupA on blood and brain AChE inhibited by isocarbophos may be one of the mechanisms of the therapeutic effect of HupA in acute Isocarbophos poisoning.

[Clinicopathologic features of cardiac myxoma--a report of 47 cases].

BACKGROUND & OBJECTIVE: Myxoma is the most common neoplasm of the heart, and has various morphologic structures, but its biological behaviors and histogenesis are still controversial. This study was to summarize the clinical, histomorphologic, and immunophenotypic features of cardiac myxoma. METHODS: The clinical data and HE sections of 47 cases of cardiac myxoma were reviewed. Special staining and immunohistochemical staining for 10 kinds of antibodies were carried on some cases to observe the morphologic features. RESULTS: Of the 47 patients with cardiac myxoma, 32 were women, the youngest was 3 years old. The volume of tumors ranged from 0.7 cm x 0.7 cm x 0.4 cm to 12 cm x 8 cm x 7 cm. Of the 47 tumors, 42 were pedunculated, the rest had broad fundus without pedicel; 43 located in the left atrium, 3 in the right atrium, and 1 in the left ventricle where near to the apex of heart. Tumor cells were stellate, spindle, round, or irregular, and arranged as masses or stripes, with abundant mucus around them. Tumor cells surrounded arterioles in 4 cases. Pseudo-capsule was formed by hyperplastic fibrous tissue in 4 cases. The mesenchyma presented angiomatous fashion in 2 cases. Sheets of old hemorrhage, hemosiderin and iron salt deposition, and fibrosis were observed in 7 cases. One case had foci adenoid structure, 1 lipoid metaplasia and ossification, 1 was rich of cells. Vimentin and CD34 were found in lining cells and parenchymal cells in 12 cases. Cytokine kinase (CK) and epithelial membrane antigen (EMA) were positive in the region of adenoid differentiation. Proliferating cell nuclear antigen (PCNA) and Ki67 were positive in the cell-rich tumor. None of the 21 patients had tumor recurred after follow-up. CONCLUSIONS: Cardiac myxoma has various secondary morphologic changes, including adenoid metaplasia, old hemorrhage, fibrosis, ossification, and lipoid metaplasia. Whether the cell-rich tumors tend to be malignant needs further study. Cardiac myxoma may come from multipotential mesenchymal cells.

[Effects of first premolar extractions on third molar angulations].

OBJECTIVE: To compare the inclinations of third molars of patients treated orthodontically with and without premolar extractions. METHODS: Records of 37 first premolar extraction patients and 32 nonextraction patients were examined. The pretreatment and posttreatment panoramic radiographs were analyzed. The angles were measured between the long axis of the third molar and the occlusal plane and between the long axis of the third molar and the long axis of the second molar. Changes in third molar angulations from pretreatment to posttreatment for two groups were compared by Mann-Whitney U-test. RESULTS: There were statistically significant difference between the extraction and nonextraction groups in the median angle formed by the long axis of third molar angulation relative to the occlusal plane (P<0.05) and the second molar (P<0.05) in mandible but not in maxillary. In the extraction group, at the end of the orthodontic treatment, the mandibular third molars showed more uprighting than did the maxillary third molars. CONCLUSION: Orthodontic treatment involving premolar extractions improves mandibular third molar angulations.

[Analysis on therapeutic effect of substance-partitioned moxibustion at Guanyuan (CV 4) and shenque (CV 8) for treatment of primary dysmenorrhea of cold-damp type].

OBJECTIVE: To observe clinical therapeutic effect of substance-partitioned moxibustion on primary dysmenorrhea (PD). Methods One hundred and thirty-eight cases of PD were randomly divided into two groups, the treatment group (n=78) were treated with substance-partitioned moxibustion at Guanyuan (CV 4) and Shenque (CV 8), once a day; and the control group (n=60) treated with oral administration of Yueyueshu Powder, twice a day, 10 g each time. They were treated for 3 menstrual cycles. RESULTS: The total effective rate and the index of therapeutic effect were 96.1% and 90.8% in the treatment group, and 88.3% and 76.2% in the control group, respectively, with a significant difference between the two groups (P < 0.05). CONCLUSION: Clinical therapeutic effect of substance-partitioned moxibustion on primary dysmenorrhea is obvious.

[Effect of HuperzineA on neurophathologic change of mouse exposed by isocarbophos and phoxim].

OBJECTIVE: In order to assessment the Huperzine A (HupA) on CNS of mice exposed by phoxim and isocarbophos. METHODS: One group was given tween-80 or peanut oil as control. One group was given phoxim or isocarbophos alone as phoxim or isocarbophos group. The other group was given HupA 2h before phoxim or isocarbophos exposure as HupA protective group. Twenty-four hours after exposure, the whole body perfusion in situ technique was performed for tissue fixation. The whole brain was removed and adjacent coronal sections was obtained. The sections of brain were stained with hematoxylin and eosin (HE) and toluidine blue. Under optic microscope images were collected and analysed. RESULTS: The extensive lesions including cortex oedema, pyramidal cell oedema, and nissal body numbers reduced were observed in phoxim or isocarbophos group. The animals of HupA protective group were observed cortex oedema, pyramidal cell oedema, but the lesional degree was lighter obviously than phoxim or isocarbophos group. CONCLUSION: It was suggested that the HupA could have the protective effect of neuropathologic changes exposed by phoxim and isocarbophos.

[Adenovirus-mediated human bone morphogenetic protein 2 gene transferred to rabbit intervertebral disc cells in vitro].

OBJECTIVE: To study the adenovirus-mediated human bone morphogenetic protein-2 gene (Ad-hBMP-2)transferred to the intervertebral disc cells of the New Zealand rabbit in vitro. METHODS: The cells of New Zealand white rabbits were isolated from their lumbar discs. The cells were grown in the monolayer and treated with an adenovirus encoding the LacZ gene (Ad-LacZ) and Ad-hBMP-2 (50, 100, 150 MOI, multiplicity of infection) in the Dulbecco's Modified Eagle Medium and the Ham's F-12 Medium in vitro. Three days after the Ad-hBMP-2 treatment, the expression of hBMP-2 in the cells that had been infected by different doses of MCI was determined by immunofluorescence and the Western blot analysis, and the expression was determined in the cells with the Ad-LacZ treatment in a dose of 150 MDI. Six days after the Ad-hBMP-2 treatment, mRNA was extracted fur the reverse transcription polymerase chain reaction (RT-PCR) and the difference was detected between the control group and the culture group that was treated with Ad-hBMP-2 in doses of 50, 100 and 150 MCI so that the expressions of aggrecan and collagen II mRNA could be observed. RESULTS: The expression of hBMP-2 in the cells was gradually increased after the transfection in an increasing dose, which was observed by immunofluorescence and the Western blot analysis. At 6 days the aggrecan and collagen type II mRNA expressions were up-regulated by Ad-hBMP-2 after the transfection at an increasing viral concentration in the dose-dependent manner. CONCLUSION: The results show that Ad-hBMP-2 can transfect the rabbit intervertebral disc cells in vitro with a high efficiency rate and the expression of hBMP-2 after the infection is dose-dependent in the manner. Ad-hBMP-2 after transfection can up-regulate the expression of aggrecan and collagen IL mRNA at an increasing viral concentration.

[Studies on germination characteristics of Prunella vulgaris seeds].

OBJECTIVE: To understand the effect of illumination, temperature on the germination of the Prunella vulgaris seeds. METHOD: The general germination method. RESULT: Illumination showed no clearly effect on the germination rate of the P. vulgaris seeds under proper temperature condition; However, illumination was the main factor to determine the germination rate of the P. vulgaris seeds when the temperature condition was unsuitable. The germination rate of the P. vulgaris seeds decreased with the time and increased when the marinating time was from 12 to 36 hours. When temperature reached 5-25 9 degrees C, the trend of the germination rate increased at frst and then decreased. CONCLUSION: The optimal germination condition of P. vulgaris seeds is 12 h of marinating time at the temperature of 20 degrees C under illumination.
