Electrochemistry of ITO electrode modified by multilayer ultrathin films based on crown-shaped polyoxomolybdate.

The electrochemical multilayer films of crown-shaped polyoxomolybdate Na21{[Na5(H2O)14] intersection[Mo(V)(20)Mo(VI)(26)O134(OH)10(mu-CH3COO)4]}.CH3COONa.90H2O (Mo46) and polyelectrolytes by layer-by-layer assembly were investigated. The stable multilayer films were assembled by alternate adsorption of negatively charged POM and positively charged polyelectrolytes is from their aqueous dispersions. UV-vis spectroscopy and cyclic voltammetry were used to monitor the regular growth of the multilayer films. The multilayer films-modified ITO electrode was used for the detection of electrocatalytic activity toward the reduction of nitrite, bromate, and hydrogen peroxide. The proposed novel immobilized method exhibited good stability, reproducibility and high sensitivity for the determination of electrocatalytic, which is important for practical application.

One-step synthesis of silver nanoparticles, nanorods, and nanowires on the surface of DNA network.

Here, we describe a one-step synthesis of silver nanoparticles, nanorods, and nanowires on DNA network surface in the absence of surfactant. Silver ions were first adsorbed onto the DNA network and then reduced in sodium borohydride solution. Silver nanoparticles, nanorods, and nanowires were formed by controlling the size of pores of the DNA network. The diameter of the silver nanoparticles and the aspect ratio of the silver nanorods and nanowires can be controlled by adjusting the DNA concentration and reduction time.

Observation of the mica surface by atomic force microscopy.

Freshly cleaved mica and a mica surface treated with pure water and dilute-salt solution have been investigated by Atomic Force Microscopy (AFM). On the bare mica surface (after repeated scanning), small dots and islands were observed. The disappearance of these dots and islands has also been captured by AFM. We believe these structures to be condensed water. The water meniscus between AFM tip and mica surface is considered as the source of this water structure. On the mica surface treated with pure water and dilute-salt solution, network structures are frequently observed by AFM.

Significant increase in risk of gastroesophageal cancer is associated with interaction between promoter polymorphisms in thymidylate synthase and serum folate status.

Thymidylate synthase (TS) catalyzes the 5,10-methylene-tetrahydrofolate-mediated conversion of deoxyuridine monophosphate to deoxythymydine monophosphate, a nucleotide required for DNA synthesis and repair. The impaired TS expression has been shown to be related to 28 bp tandem repeats and a G-->C SNP in the 5'-UTR of TS. Folate deficiency has been demonstrated to play a role in gastroesophageal carcinogenesis. This case-control study was to examine the hypothesis that the TS polymorphisms, alone or in combination with serum folate status, may confer susceptibility of the hosts to gastroesophageal cancer. We analyzed TS genotype and serum folate concentration in 324 patients with esophageal squamous cell carcinoma (ESCC), 231 patients with gastric cardia adenocarcinoma (GCA) and 492 controls. It was found that compared with the normal expression TS genotype, the low expression TS genotype alone was significantly associated with increased risk of ESCC [adjusted odds ratio (OR) 1.47; 95% confidence interval (CI) 1.03-2.10] but not GCA (OR=0.98, 95% CI=0.68-1.40). More importantly, a significant interaction between the TS polymorphisms and serum folate status in risk of ESCC and GCA was observed. Among subjects with low serum folate concentration (<3 ng/ml), the ORs of ESCC and GCA for the low expression genotype were 22.63 (95% CI=10.44-49.05) and 4.08 (95% CI=1.94-8.59), which were greater than respective 9.97 (95% CI=5.67-17.53) and 1.88 (95% CI=1.18-3.24) for the normal expression genotype (P=0.002 and 0.029). These results suggest an important role for folate deficiency and impaired TS activity in the etiology of ESCC and GCA.

Effects of nanostructure on catalytic degradation of ethanol on SrCO3 catalysts.

Degradation of ethanol over SrCO3 nanowires and nanoparticles was used as a model reaction to investigate the effect of nanostructure on chemical property. Differences in catalytic degradation activity with nanostructure are evaluated. The results indicated that catalytic activity of SrCO3 particles increases with decreasing of particle size due to high surface area. But this conclusion cannot be applicable to evaluating SrCO3 nanowires and nanoparticles. SrCO3 nanowires have lower ignition temperatures and wider working temperature ranges than SrCO3 nanoparticles, though nanowires had lower surface areas. Besides, ethanol degraded over nanowires in three ways, and the dominating reaction changes with reaction temperature. Consequently, the main degradation products of nanowires differed with temperature. But for nanoparticles, acetaldehyde is the only main product. Since transmission electron microscopy, X-ray diffraction, and bond equilibrium theory analysis demonstrated that nanowires and nanoparticles had similar crystal structure, surface area, and grain size, the differences in catalytic degradation activity between SrCO3 nanowires and nanoparticles can be attributed to different distributions of active sites, as proven by CO2 and ethanol temperature programmed desorption.

[Epidemiology of cardiovascular risk in Chinese chronic kidney disease patients].

OBJECTIVE: To identify the risk factors associated with cardiovascular disease (CVD) in Chinese chronic kidney disease (CKD) patients. METHODS: As part of a multicenter Chinese cohort study, the clinical data associated with CVD of 1239 patients with CKD (stage 2 - 5) hospitalized in 7 grade 3A hospitals distributed in 5 regions of China 2002 - 2003 were collected. Logistic regression model was used to analyze the association between CVD and the demographic variables, lifestyle, medical history, medication, physical examination, and laboratory variables. RESULTS: (1) Increase of serum C-reactive protein (CRP, cut off > 10 mg/L) was an independent risk factor for development of coronary artery disease (CAD) (OR 2.13; 95% confidence interval [CI], 1.32 - 3.43). 21.5% of the patients in this group showed a value of CRP > 10 mg/L. (2) Being female, anemia, and systolic hypertension were the major determinants of the development of left ventricular hypertrophy (OR 2.99, CI 2.09 - 4.26; OR 2.66, CI 1.19 - 3.57; and OR 1.02, CI 1.00 to -1.02). 54.2% of the patients in this group had their systolic pressure controlled under 140mmHg, and only 15% of the patients in this group had their hemoglobin remain at the level >or= 110 g/L. (3) There was a significant interaction between the calcium-phosphate product and congestive heart failure (CHF) (OR 1.023, CI 1.01 - 1.03). 25.9% of the patients in this cohort had their calcium-phosphate product >or= 55. (4) Hypoalbuminemia (OR 6.01, CI 1.25 - 28.96) and diastolic hypertension (OR 1.05, CI 1.00 - 1.09) played major role in determining cerebrovascular accidents (CVA). In these cohort the prevalence of hypoalbuminemia was 37.3%. (5) Diabetes was associated with CAD (OR 2.34), CHF (OR 1.97), and CVA (OR 4.40), although its prevalence was lower in Chinese CKD patients (20%). Age was the risk factors of CAD (OR 1.04) and CVA (OR 1.22). Hypertension was associated with LVH (OR 1.016), CHF (OR 1.02), and CVA (OR 1.04). CONCLUSION: CKD is associated with nontraditional risk factors for the development of CVD, including chronic inflammation, malnutrition and calcium-phosphate disorders. Particular care must be taken to give optimal treatment for the most important CVD risk factors active in Chinese CKD patients, e.g. anemia and hypertension.

Effects of removal of necrotic blastomeres from mouse cryopreserved embryos on blastocyst formation and hatching.

To evaluate whether the developmental potential of embryos that were partially damaged after freezing and thawing can be improved by removal of necrotic blastomeres. Eight-cell mouse embryos were cryopreserved using 1,2-propanediol and sucrose as cryoprotectant with slow cooling procedure. After thawing, blastocyst formation and hatching of fully intact embryos were compared between no treatment and with laser-assisted hatching. For partially intact embryos, the effects of removal of necrotic blastomeres with micromanipulation were evaluated. Laser-assisted hatching of mouse cryopreserved fully intact embryos significantly increased blastocyst hatching (63.4% versus 48.3%, P<0.05), but had little effect on blastocyst formation (72.0% versus 70.1%, P>0.05). The removal of necrotic blastomeres from partially damaged mouse cryopreserved embryos with micromanipulation significantly increased blastocyst formation (52.9% versus 32.0%, P<0.05) and blastocyst hatching (41.2% versus 22.0%, P>0.05) compared with the control group. The developmental potential of partially damaged cryopreserved embryos can be improved by removal of necrotic blastomeres with micromanipulation.

Electrochemical behavior of poly(ferrocenyldimethylsilane-b-dimethylsiloxane) films.

The electrochemical behavior of poly(ferrocenyldimethylsilane-b-dimethylsiloxane) (PFDMS-b-PDMS) films deposited on a glassy carbon electrode was investigated by means of cyclic voltammetry (CV). The influences of the solvent, film thickness, temperature, and PDMS block length in PFDMS-b-PDMS on the electrode process were discussed. It was found that in 0.1 M aqueous LiClO(4) the electrochemical processes of the films on a glassy carbon electrode were complex and have a low rate of electron transport and mass diffusion. The kinetic parameters obtained indicated that the electrode process was controlled by both the electrode reaction and mass diffusion.

Comparison of F-actin fluorescent labeling methods in pollen tubes of Lilium davidii.

Fluorescence labeling of F-actin in pollen tubes by various methods has produced inconsistent results in the literature. Here, we report that EGTA, which was always used in fixative buffers in the past and thought to help cytoskeleton stabilization, can significantly affect F-actin distribution and lead to the formation of thick F-actin bundles at the tip of the pollen tube. We also found that vacuum-infiltration for the first 5 min during pollen tube fixation can better preserve normal cytoplasm structure and F-actin distribution. In contrast, m-maleimidobenzoic acid N-hydroxysuccinimide ester (MBS) treatment before chemical fixation resulted in a shortening of the free zone of thick F-actin bundles in the pollen tube tip. Taken together, our results suggest that exclusion of EGTA and MBS from the fixative buffer, in combination with vacuum-infiltration in the first 5 min of fixation, can improve F-actin fluorescence labeling in pollen tubes of Lilium davidii.

Generation and characterization of pluripotent stem cells from cloned bovine embryos.

Bovine embryonic stem (ES) cell lines reported to date vary in morphology and marker expression (e.g., alkaline phosphatase [ALPL], stage-specific embryonic antigen 4 [SSEA4], and OCT4) that normally are associated with the undifferentiated, pluripotent state. These observations suggest that the proper experimental conditions for consistently producing bovine ES cells have not been identified. Here, we report three bovine ES cell lines, one from in vitro-fertilized and two from nuclear transfer embryos. These bovine ES cells grew in large, multicellular colonies resembling the mouse ES and embryonic germ (EG) cells and human EG cells. Throughout the culture period, most of the cells within the colonies stained positive for ALPL and the cell surface markers SSEA4 and OCT4. The staining patterns of nuclear transfer ES cells were identical to those of the blastocysts generated in vitro yet different from most previously reported bovine ES cell lines, which were either negative or not detected. After undifferentiated culture for more than 1 yr, these cells maintained the ability to differentiate into embryoid bodies and derivatives of all three EG layers, thus demonstrating their pluripotency. However, unlike the mouse and human ES cells, following treatment with trypsin, type IV collagenase, or protease E, our bovine ES cells failed to self-renew and became spontaneously differentiated. Presumably, this resulted from an interruption of the self-renewal pathway. In summary, we generated pluripotent bovine ES cells with morphology similar to those of established ES cells in humans and mice as well as marker-staining patterns identical to those of the bovine blastocysts.

[Evaluation the antimycotic activity of itraconazole in human stratum corneum with vitro fungi cultivation].

OBJECTIVE: To establish a new model to evaluate the antifungal activity of antimycotics. METHODS: Twenty-one healthy adult volunteers without clinically significant differences in demographic characteristics, hydration status of the skin, and sebum excretion were randomly divided into two groups: group 1 receiving 200 mg itraconazole bid for 1 week, and group 2 receiving itraconazole 200 mg qd for 1 weeks. Stratum corneum stripping was taken from the forearm skin at days 0, 1, 4, 7, 10, 14, 21, 28, and 35. Spores of selected fungi (Trichophyton rubrum, Trichophyton mentagrophytes, and Microsporum canis) were deposited and cultured on the center of the stratum corneum stripping. After 1-week culture the stratum corneum stripping was staining by PAS, and the area covered by dermatophyte was measured by corneofungimetry and computerized-assisted image analysis so as to calculate the inhibitory effect of the oral antifungal agents administered. RESULTS: All three dermatophytes grew on the stratum corneum stripping, the area covered by the dermatophyte was dense before administration of antifungal agent, and decreased gradually after the administration of drug, and to the least on the seventh day (P < 0.01), however, the area covered by the dermatophyte began to increase since the tenth day, and reach the biggest on the thirty-fifth day. Itraconazole at two different dosages showed significant activity against these 3 dermatophytes without significant difference between the two groups (all P > 0.05). CONCLUSION: The ex vivo growth environment of dermatophyte on human stratum corneum is very similar to that of human milieu interne. Itraconazole displays a high antidermatophyte activity in human stratum corneum. Corneofungimetric bioassay is a new method between in vitro trial and in vivo effect.

Imaging DNA molecules on mica surface by atomic force microscopy in air and in liquid.

DNA molecules immobilized on mica surface by various methods have been observed by atomic force microscopy both in air and in liquid. Divalent cations and 3-aminopropyltriethoxysilane (APTES) modified mica surface have been used to immobilize the DNA molecules. Optimal DNA and divalent cations concentration for AFM imaging are presented. Among the different methods of modifying mica surface with APTES, the water solution modifying method appears to get the best results. When using high DNA concentration for AFM imaging, DNA networks can be formed. A simple method to extend long DNA molecules is demonstrated. The optimal imaging conditions and AFM operating techniques are discussed. Different DNA immobilizing methods have been compared and evaluated.

Lysophosphatidylcholine-induced surface redistribution regulates signaling of the murine G protein-coupled receptor G2A.

Intracellular trafficking and spatial dynamics of membrane receptors critically regulate receptor function. Using microscopic and subcellular fractionation analysis, we studied the localization of the murine G protein-coupled receptor G2A (muG2A). Evaluating green fluorescent protein-tagged, exogenously expressed as well as the endogenous muG2A, we observed that this receptor was spontaneously internalized and accumulated in endosomal compartments, whereas its surface expression was enhanced and stabilized by lysophosphatidylcholine (LPC) treatment. Monensin, a general inhibitor of recycling pathways, blocked LPC-regulated surface localization of muG2A as well as muG2A-dependent extracellular signal-regulated kinase (ERK) activation and cell migration induced by LPC treatment. Mutation of the conserved DRY motif (R-->A) enhanced the surface expression of muG2A, resulting in its resistance to monensin inhibition of ERK activation. Our data suggest that intracellular sequestration and surface expression regulated by LPC, rather than direct agonistic activity control the signaling responses of murine G2A toward LPC.

[Cardiovascular disease in Chinese chronic renal insufficiency patients-epidemiology survey].

OBJECTIVE: Cardiovascular (CV) disease (CVD) is the single most important cause of death among Chinese dialysis patients, accounting for 51% of overall mortality. The study was performed to investigate the prevalence and the spectrum of CVD in Chinese chronic kidney disease (CKD) patients. METHODS: The multicenter Chinese cohort study examined 1239 CKD patients from 7 main medical centers (distributed in 5 regions of China) who were hospitalized between 2002 and 2003. RESULTS: (1) The most prevalent pathological form of CVD was left ventricular (LV) hypertrophy (LVH), accounting for 58.5% of total patients. The prevalence of coronary artery disease (CAD), congestive heart failure (CHF), and cerebrovascular accidents (CVA) was 16.5%, 27.7% and 5.6%, separately. (2) The cohort with minor renal dysfunction (stage 2-3) had higher prevalence of CAD (5.9%) and CVA (1.0%) compared with general population in the same regions. Up to 41.2% of minor CKD patients were complicated with LVH, and 13.8% of them had clinical evidence of CHF. The prevalence of CAD, LVH and CHF increased as glomerular filtration decline. (3) The prevalence of CAD (20.0%) was much lower and the prevalence of CVA (5.4%) was higher in Chinese dialysis patients than that in American dialysis population. There was significant geographical variations in CAD prevalence, but it was not different between genders. CONCLUSION: The CV risk is significantly increased in patients with CKD. Even minor CKD has a major impact on the CV risk. The prevalence of CAD in Chinese dialysis patients is markedly lower than that in American dialysis population.

A central kinase domain of type I phosphatidylinositol phosphate kinases is sufficient to prime exocytosis: isoform specificity and its underlying mechanism.

Exocytosis, a critical process for neuronal communication and hormonal regulation, involves several distinct steps including MgATP-dependent priming (which involves the synthesis of phosphatidylinositol 4,5-bisphosphate). Type I phosphatidylinositol phosphate kinases (PIPKIs) were purified biochemically as a priming factor. PIPKI consists of three domains: the N-terminal region, the central kinase domain, and the C-terminal region. Three isoforms (alpha, beta, and gamma) of PIPKI have been identified, and each is alternatively spliced at the C-terminal region. In the present study, we conducted a structure/function analysis of PIPKIs in the priming of exocytosis, and we found that recombinant PIPKIalpha and PIPKIgamma had priming activity. However, an unexpected finding of these results was that PIPKIbeta did not prime exocytosis. The N- or C-terminal region of PIPKIalpha and PIPKIgamma was not required for priming, which indicates that the central kinase domain is sufficient for this process. Alternative splicing in each isoform did not affect the isoform specificity in priming. Priming activity by isoforms is strongly correlated with their phosphatidylinositol phosphate kinase activity because PIPKIalpha and PIPKIgamma had higher kinase activity than PIPKIbeta. These results suggest that PIPKIalpha and PIPKIgamma are the critical priming factors for exocytosis; it also suggests that the levels of phosphatidylinositol phosphate kinase activity in producing phosphatidylinositol 4,5-bisphosphate specify the function of PIPKI isoforms in priming.

A comparative study of different references for EEG spectral mapping: the issue of the neutral reference and the use of the infinity reference.

Based on EEG data recorded from 11 subjects with eyes open and the left mastoid (M) reference, three data sets were generated by re-referencing to the conventional linked mastoids (L), average (A) and the new 'infinity' (I) reference provided by the reference electrode standardization technique (REST, Yao 2001 Physiol. Meas. 22 693-711). The EEG power in the alpha frequency band with the four different references was calculated and compared with respect to the total energy and spatial amplitude weight centre (AWC) coordinates, to compare the effects of different references on power mapping in the frequency domain. Compared with the I reference, the AWCs of the EEG with the M reference show significant shifts to the right, frontal and superficial positions, the L reference significant shifts to frontal and superficial positions, and the A reference shifts the AWC significantly to a deeper position. Furthermore, the power maps of the M and L references have larger total power than the I reference, while that of the A reference has the smallest total power. These results confirm that different choices of reference electrodes result in systematic changes in the distribution of EEG frequency power, and in order to reduce the effect of such systematic shifts on the explanation of EEG mappings, a common reference is necessary for EEG research. We recommend the I reference for objective use in cross-laboratory studies and clinical practices, as it is far from all the other electrodes and can act as a neutral reference.

Knockdown of survivin gene expression by RNAi induces apoptosis in human hepatocellular carcinoma cell line SMMC-7721.

AIM: To investigate the survivin gene expression in human hepatocellular carcinoma cell line SMMC-7721 and the effects of survivin gene RNA interference (RNAi) on cell apoptosis and biological behaviors of SMMC-7721 cells. METHODS: Eukaryotic expression vector of survivin gene RNAi and recombinant plasmid pSuppressorNeo-survivin (pSuNeo-SVV), were constructed by ligating into the vector, pSuppressorNeo (pSuNeo) digested with restriction enzymes Xba I and Sal I and the designed double-chain RNAi primers. A cell model of SMMC-7721 after treatment with RNAi was prepared by transfecting SMMC-7721 cells with the lipofectin transfection method. Strept-avidin-biotin-complex (SABC) immunohistochemical staining and RT-PCR were used to detect survivin gene expressions in SMMC-7721 cells. Flow cytometry was used for the cell cycle analysis. Transmission electron microscopy was performed to determine whether RNAi induced cell apoptosis, and the method of measuring the cell growth curve was utilized to study the growth of SMMC-7721 cells before and after treatment with RNAi. RESULTS: The eukaryotic expression vector of survivin gene RNAi and pSuNeo-SVV, were constructed successfully. The expression level of survivin gene in SMMC-7721 cells was observed. After the treatment of RNAi, the expression of survivin gene in SMMC-7721 cells was almost absent, apoptosis index was increased by 15.6%, and the number of cells was decreased in G2/M phase and the cell growth was inhibited. CONCLUSION: RNAi can exert a knockdown of survivin gene expression in SMMC-7721 cells, and induce apoptosis and inhibit the growth of carcinoma cells.

Brahma links the SWI/SNF chromatin-remodeling complex with MeCP2-dependent transcriptional silencing.

Transcriptional repression of methylated genes can be mediated by the methyl-CpG binding protein MeCP2. Here we show that human Brahma (Brm), a catalytic component of the SWI/SNF-related chromatin-remodeling complex, associates with MeCP2 in vivo and is functionally linked with repression. We used a number of different molecular approaches and chromatin immunoprecipitation strategies to show a unique cooperation between Brm, BAF57 and MeCP2. We show that Brm and MeCP2 assembly on chromatin occurs on methylated genes in cancer and the gene FMR1 in fragile X syndrome. These experimental findings identify a new role for SWI/SNF in gene repression by MeCP2.
