Case-control association study of Disrupted-in-Schizophrenia-1 (DISC1) gene and schizophrenia in the Chinese population.

Disrupted-in-Schizophrenia-1 (DISC1) has first been identified as a candidate gene for schizophrenia through study of a Scottish family with a balanced (1; 11) (q42.1; q14.3) translocation. Lots of linkage and association studies supported DISC1 as a risk factor for schizophrenia. In this study, we genotyped three SNPs in DISC1 using a set of Han Chinese samples of 560 schizophrenics and 576 controls. No positive association was detected in the whole samples but analysis of allele frequencies in female samples showed weak association between SNP rs2295959 and the disease (chi(2)=6.188, P=0.0135, OR=0.728, 95% CI=0.567-0.935). Our results provide further evidence for sex difference for the effect of the gene on the aetiology of schizophrenia. Our findings also would encourage further studies, particularly family-based association studies with larger samples, to analyze the association between DISC1 and schizophrenia.

Association between the brain-derived neurotrophic factor (BDNF) gene and schizophrenia in the Chinese population.

Brain-derived neurotrophic factor (BDNF) belongs to a family of the neurotrophin which plays important roles in the development of the brain. BDNF has been suggested as a factor that increases the risk of schizophrenia. In this study, we genotyped three single nucleotide polymorphisms (SNPs) in the BDNF gene using a set sample of Han Chinese subjects consisting of 560 schizophrenes and 576 controls. No significant differences were found for either the genotype or allele distribution of analyzed polymorphisms, nor was any gender-specific association found. Thus, our data suggest that the BDNF gene may not be an important factor in susceptibility to schizophrenia.

[Contents and distribution characteristics of heavy metals in water and sediment of intertidalite].

ICP-MS analytical technology was used to analyze the concentration of 13 heavy metals in surface water, pore water in sediment, suspended particulate matter, and sediment of intertidalite of Tianjin. Results show that both total amount and single concentration of heavy metals vary greatly in different media, the biggest and smallest values differ by 4 or 7 magnitudes. While the variation coefficients of the contents of total metals or single metal in the same medium are less than 0.3 and 0.5 respectively except for the pore water. The average concentration of total dissolved heavy metals is 41.2 microg x L(-1), while total contents of heavy metals in original water, pore water, sediment and suspended particles are 62.7 microg x L(-1), 112.7 microg x L(-1), 262.3 mg x L(-1) and 345.0 mg x L(-1) respectively, they are similar to inland natural river but higher then those of ocean. The composition of heavy metals in different media varied greatly, metals prone to dissolve such as Mo account for more than 20% in water, but metals uneasy to dissolve mainly exist in sediment and suspended particles and absorbed by particles. Typical heavy metals, such as Zn, Cu, Pb and Cr, account for 40% - 50% in water and 50% - 80% in other media. The similarity in distribution mode of heavy metals in intertidalite and terrestrial soil suggest that they are originated from terrestrial soil that is contiguity to the sea. Notable correlativity can be seen between concentrations of all heavy metals except for Mo in sediment, which can explain the similarity of their environmental behavior, but only wake correlation between the content of organic carbon and concentrations of heavy metals in suspended particulates and sediment.

[Experimental study of the facial nerve paralysis induced by herpes simplex virus type 1 infection in mice].

OBJECTIVE: To establish an animal model of Bell palsy induced by type I herpes simplex virus (HSV-1) infection and to assess the role and site of HSV-1 in the pathogenesis of facial paralysis. METHODS: Fifty-three female Balb/c mice four-week-old weighted 16-18 g were studied. After scratching the surface of bilateral auricles with a 26-gauge needle, 25 microL HSV-1 with a titer of 6. 7 x 10(7) PFU/ml was inoculated into the right auricle, and the same volume of PBS was placed in the left. As a control, PBS was placed on the bilateral auricles of 4 mice. The HSV-1 DNA in bilateral facial nerve, bilateral brainstem, bilateral trigeminal carrier ganglion, bilateral brain, and blood at different stage was examined with polymerase chain reaction analysis. RESULTS: Thirty-seven animals (75.51%) appeared different degree facial paralysis among the 49 inoculated animals. Fourteen facial paralysis (37.84%) were on the right, 3 (8.11%) on the left, and 20 (54.05%) on the bilateral side. Six animals with facial palsy were recovered during 3-13 days, the average recovery time was 7.83 days. CONCLUSIONS: The existence of HSV-1 in the brainstem and the cerebral cortex is significant for facial paralysis.

[Distribution features of Gleason score and its relationship with clinical stages in 346 prostate cancer patients].

OBJECTIVE: To analyze the distribution features of Gleason score and evaluate the relationship between Gleason score and clinical stages in patients with prostate cancer. METHODS: Surveys were made of the inpatients with prostate cancer diagnosed by pathology from January 1992 to June 2005 in our hospital. Gleason score and clinical stages were determined on the basis of pathological examination and clinical data of the prostate cancer patients. The patients were divided into three groups (1992-1999, 2000-2002 and 2003-2005). The Chi-square test was used to evaluate the distribution and differences of Gleason score among the three groups. Spearman rank correlation was applied to the evaluation of the relationship between Gleason score and clinical stages. RESULTS: We found a statistically significant shift in the distribution of Gleason score (chi2 = 17.703, P < 0.01), and a slight increase in the mean Gleason score. The proportion of moderately differentiated tumor increased (chi2 = 10.736, P < 0.01). There was little change in the proportion of Gleason score 7, 8, 9 and 10 (chi2 = 4.038, P > 0.05). Gleason score had a significant positive correlation with clinical stages in the 346 cases of prostate cancer (r = 0.452, P < 0.01). Significant difference was observed between Gleason score 2-6 and 7 or 8-10 (chi2 = 8.786, P < 0.01, chi2 = 22.956, P < 0.01), but not between the latter 2 groups (chi2 = 0.787, P > 0.05) in prediction of organ-confined disease. CONCLUSIONS: Gleason score 7 shows the similar value to Gleason score 8-10 in predicting the progression of the disease. Gleason score was significantly correlated with clinical stages, which suggests that Gleason score is also an important indicator for the prognosis of prostate cancer.

Mesenchymal hamartoma of liver: magnetic resonance imaging and histopathologic correlation.

AIM: To describe the imaging features of hepatic mesenchymal hamartoma with emphasis on magnetic resonance imaging (MRI) compared to histopathologic results. METHODS: Spin-echo sequence(SE),fast spin-echo sequence(FSE) were performed in 12 children (7 males,5 females) with mesenchymal hamartoma of the liver (MHL), aged 1.2 months to 12 years;( mean age, 6.3 years) axial, saggital, coronary plain imaging with an Elscint 2.0T MR equipment. Their main symptoms were abdominal mass (5 cases), enlarged liver (8 cases), abdominal pain (1 case) and anemia (2 cases), and negative alphafetoprotein. Dynamic enhancement examination was added in 2 cases. RESULTS: Six cases had single mass type of MHL, in which 3 cases had solid masses showing slight low-signal-intensity in T1WI, and irregular high-signal-intensity in T2WI, 1 case had a cystic-solid mixed mass showing several border-clear cysts in a solid mass, 2 cases had cystic masses with multi-septa. Five cases had diffuse and multifocal lesions type of MHL with its signal intensity being similar to that of the solid mass. One case had a combined diffuse and single cystic mass. In the early dynamic enhancement examination, the lesions were slightly circum-enhanced , and the center was enhanced in the later scan images. Inner hepatic vessels were compressed in 5 cases, vena cava and abdominal aortae were compressed in 3 cases. Pathological findings included fiber hyperplasia, hyaline degeneration, biliary duct hyperplasia, lobule-like array. CONCLUSION: MR imaging is a better way to differentiate and diagnose MHL. MHL may be recognized by its characteristic occurrence in infancy and MR imaging features.

Diagnostic analysis of hepatic angiomyolipoma.

BACKGROUND: Hepatic angiomyolipoma (HAML) is a rare tumor containing a variable amount of fat, vessels and smooth muscle. We report the image findings on ultrasonography and computed tomography (CT) of huge HAML. METHOD: The clinical, imaging and pathological data of a case of HAML were retrospectively collected and analyzed. RESULTS: A huge heterogeneous hyperecho mass with anecho and hypoecho areas inside was found in the left hepatic lobe on ultrasonography. Color Doppler showed blood flow and arterial spectrum in it. CT scan showed a huge heterogeneous solid mass in the left lobe of the liver, with a low density and hypervascular area in arterial phase. The serum tumor marks were all negative. Ultrasound-guided biopsy was taken twice before resection and both showed necrosis tissue and reaction of inflammatory cells. Postoperative pathological results showed that the tumor was composed of epithelioid smooth muscle cells, thick-walled blood vessels and a few adipose cells with necrosis. The immunohistochemistry results showed appearance of typical HAML, with HMB-45 positive and alpha fetoprotein (AFP) negative. CONCLUSIONS: Preoperative diagnosis of HAML relies on combination of CT, MRI and ultrasonography. Our case of HAML showed heterogeneous hyperecho image on ultrasonography. Ultrasound-guided biopsy combined with morphological manifestation and specimen examination for HMB-45 may be helpful in the diagnosis of HAML.

[Age and pathological features of 481 prostate cancer patients].

OBJECTIVE: To investigate the age and pathological features of prostate cancer patients in recent years. METHODS: An analysis was made of the age and pathological features of 481 cases of prostate cancer pathologically diagnosed from January 1998 to April 2004, 39 cases in 1998, 69 in 1999, 73 in 2000, 68 in 2001, 72 in 2002, 121 in 2003, and 39 in the first four months of 2004. RESULTS: The patients ranged in age from 40 to 91 years, averaging 72, 95% between 55 and 84, and 84.2% over 65 years. Pathologically, 14 cases were well, 29 moderately, and 83 poorly differentiated according to the three-grade system (WHO, the Mostofi system), with 355 cases ungraded. Forty cases (8.3%) were microcarcinoma (< 1 cm), and 20 cases (4.2%) incidental carcinoma. Of the total number, 473 cases (98.1%) were pathologically diagnosed as adenocarcinoma, 1 endometrioid adenocarcinoma, 1 squamous cell carcinoma, 1 signet ring cell carcinoma, 1 adenosquamous cell carcinoma, 1 small cell carcinoma, 1 mucinous adenocarcinoma, 1 adenoid cystic carcinoma, and 1 transitional cell carcinoma. CONCLUSION: Prostate cancer commonly develops in men over 65 years, and adenocarcinoma is the most common histological type. The disease has become a major malignant tumor to endanger elderly males.

Understanding the unique characteristics of suicide in China: national psychological autopsy study.

OBJECTIVE: To compare the characteristics of suicides in the four main demographic groups: urban males, urban females, rural males and rural females in order to help clarify the demographic pattern of suicides in China. METHODS: A detailed psychological autopsy survey instrument was independently administered to 895 suicide victims in family members and close associates from 23 geographically representative locations from around the country. RESULTS: Pesticide ingestion accounted for 58% (519) of all suicides and 61% (318/519) of deaths were due to unsuccessful medical resuscitation. A substantial proportion (37%) of suicide victims did not have a mental illness. Among the 563 victims with mental illness, only 13% (76/563) received psychiatric treatment. Compared to other demographic groups, young rural females who died from suicide had the highest rate of pesticide ingestion (79%), the lowest prevalence of mental illness (39%), and the highest acute stress from precipitating life events just prior to the suicide. CONCLUSION: Many suicides in China are impulsive acts of deliberate self-harm following acute interpersonal crises. Prevention of suicides in China must focus on improving awareness of psychological problems, improving mental health services, providing alternative social support networks for managing acute interpersonal conflicts, limiting access to pesticides, and improving the resuscitation skills of primary care providers.

[Quantitative analysis of factors affected mortality trend in Chinese, 2002].

OBJECTIVE: To explain trend of death in Chinese by quantitative analysis of demographic and non-demographic factors and estimate the proportion of contribution of non-demographic and demographic factors. METHODS: Using census data and death causes data of National Disease Surveillance Points at 1991 and 2000 to calculate the proportion of contribution of demographic and non-demographic factors and to change on various death causes from 1991 to 2000 by methods of decomposing the differences of death rates. RESULTS: The death rate showed a rapid decrease during 1950 - 1975, mainly owing to the contribution of non-demographic factors, including economic development, popularization of education and health service, especially the "patriotic hygiene movement". During 1991 - 2000, the death causes of lung cancer, liver cancer, breast cancer, chronic heart disease, stroke, diabetes and traffic accident had been increasing. The increase of deaths caused by these diseases were contributed to the non-demographic factors including 63% of the increase on lung cancer and 88% of increase on death rate of traffic accidents. CONCLUSION: The study showed that the risk factors had contributed to the increase of death rates, including behavioral risk factors described in the preceding 5 papers as smoking and passive smoking, unhealthy diet, sedentary life style, violating traffic regulation etc. In order to reduce the death rates on cancer, heart diseases, diabetes, traffic accidents, emphasis should be also laid on the change of unhealthy behaviors.

[Polymorphism of regulated upon activation, normal T cell expressed and secreted promoter region -28 position in Chinese allergic asthmatic children].

OBJECTIVE: To study the effect of regulated upon activation, normal T cell expressed and secreted (RANTES)-28 polymorphism on allergic asthma in Chinese children. METHODS: Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) was used to identify the polymorphism at position -28 of the RANTES promoter in 100 asthmatic children (group A). Levels of plasma IgE and RANTES were detected by chemiluminescence and ELISA respectively. The number of peripheral eosinophils was counted. Data were compared with those of the control group of 90 healthy children (group B). RESULTS: (1) Both C allele and G allele were detected at -28 of the RANTES promoter; the prevalence of the G allele in group A was 19.5%, as compared to 10.6% in group B; the prevalence of RANTES-28G allele was significantly different between the two groups (P < 0.05). (2) For genotypes C/C, C/G and G/G in asthmatics, levels of plasma RANTES were (289 +/- 199) ng/L, (515 +/- 119) ng/L and (1 071 +/- 138) ng/L, respectively, the difference being significant among the three genotypes (P < 0.01). (3) For genotypes C/C, C/G and G/G in asthmatics, total levels of plasma IgE (lg IgE) were 2.45 +/- 0.12, 2.77 +/- 0.07 and 3.16 +/- 0.09, respectively, the difference being not significant among the three genotypes (P > 0.05). (4) The numbers of peripheral eosinophils were (2.9 +/- 1.4) x 10(8)/L, (6.4 +/- 0.8) x 10(8)/L and (9.9 +/- 2.3) x 10(8)/L in genotypes C/C, C/G and G/G in asthmatics respectively, and the difference being significant among the three genotypes (P < 0.01). CONCLUSION: Polymorphism of RANTES C-28G was associated with susceptibility of asthma in children, and may aggravate the disease through increasing the level of RANTES and eosinophils.

[Expression of cytokeratin19, galectin-3 and HBME-1 in thyroid lesions and their differential diagnoses].

OBJECTIVE: To study immunohistochemical expression of cytokeratin19 (CK19), galectin-3 (Gal-3) and HBME-1 in thyroid lesions and to assess their usefulness as markers in the differential diagnoses of thyroid nodular lesions. METHODS: Immunohistochemical staining was performed on formalin-fixed paraffin-embedded tissue of 21 cases of nodular goiters, 14 cases of toxic goiters, 15 cases of follicular adenomas (FA), 13 cases of follicular carcinomas (FC), 13 cases of follicular variant papillary carcinomas (FVPC) and 48 cases of classic papillary carcinomas (CPC). RESULTS: All three markers were expressed in the cytoplasm with no or weak expression in benign lesions and diffuse and strong in malignant cases. Positive expressions of CK19, Gal-3 and HBME-1 were present in 11of 21, two of 21, four of 21 in nodular goiters, seven of 14, one of 14, one of 14 in toxic goiters, nine of 15, two of 15, two of 15 in FA, 10 of 13, eight of 13, seven of 13 in FC, 13 of 13, 11 of 13, 12 of 13 in FVPC, and 48 of 48, 45 of 48, 46 of 48 in CPC. The expression rates of the three markers between benign lesions (nodular goiters, toxic goiters and FA) and malignant lesions (FA, FVPC and CPC) were statistically significant. Among the three follicular lesions (FA, FC and FVPC), the differences were statistically significant as well. Nine, seven and six cases were negative for all three markers in nodular goiters, toxic goiters and FA, respectively. Only one case in FC was negative for all three markers, no case was all negative in FVPC and CPC; the rate of one case with two or more positive marker expression in nodular goiters, toxic goiters, FA, FC, FVPC and PC was 14.2% (3/21), 21.43% (3/14), 20.0% (3/15), 69.2% (9/13), 92.3% (12/13), 100.0% (48/48), the differences between benign lesions and malignant lesions and between FA, FC and FVPC were also statistically significant. CONCLUSIONS: Immunohistochemical stains of CK19, Gal-3 and HBME-1, especially when used in combination, can be an important adjunct to the histopathological diagnoses of thyroid lesions.

Intramuscular injection of AAV-GDNF results in sustained expression of transgenic GDNF, and its delivery to spinal motoneurons by retrograde transport.

Adeno-associated virus (AAV) vector has been developed as an attractive gene delivery system with proven safety. Glial cell line-derived neurotrophic factor (GDNF) is proposed to be a promising therapeutic agent for amyotrophic lateral sclerosis (ALS) and other motor neuron diseases. The purpose of this report was to investigate transgenic GDNF expression at different time points post AAV mediated GDNF intramuscular delivery. An AAV vector was constructed to encode a recombinant fusion of GDNF tagged with a FLAG sequence at the C-terminal (AAV-GDNF) to distinguish it from its endogenous counterpart. A single intramuscular injection of AAV-GDNF led to substantial expression of transgenic GDNF which remained for at least 10 months in transduced gastrocnemius muscle. This transgenic GDNF was distributed in a large number of myofibers, mainly in the vicinity of the sarcolemma and predominantly concentrated at the sites of neuromuscular junctions (NMJs). Furthermore, transgenic GDNF, but not beta-galactosidase expressed as a control, was detected in the motoneurons that projected axons to the injected muscles, thus, indicating retrograde axonal transportation of the transgenic GDNF. This study provides a basis for a strategy of intramuscular AAV-GDNF delivery to protect motoneurons as a possible means of ALS treatment.

Field-dependent-independent cognitive style in solving dynamics problems.

251 senior middle school (Grade 11) students were tested on the Chinese Group Embedded Figures Test and Dynamic Problems Test. A 2 (cognitive style) x 2 (sex) analysis of variance indicated a nonsignificant effect with respect to cognitive style in solving easier Dynamics problems but a main significant effect in solving complex Dynamics problems. Multiple comparisons (post hoc t tests) indicated that cognitive style is a significant factor among boys but not girls. The difference between Field-independent students and Field-dependent students in solving complex Dynamics problems may be that the former students more easily form a clear map of motion than the latter students.

[The clinical significance of angiogenesis in the bone marrow of acute leukemia patients].

OBJECTIVE: To study the angiogenesis in bone marrow and the level of vascular endothelial growth factor (VEGF) in bone marrow fluid from acute leukemia (AL) patients and to explore their clinical significance. METHODS: Microvessels in each bone marrow specimen section were counted by immunohistochemical identification of microvascular endothelial cells with anti-human von Willebrand factor under light microscopy in field 400 times. The VEGF level in bone marrow fluid was measured by ELISA. RESULTS: Microvessel counts in untreated AL patients (22.82/x 400 field) were significantly higher than those in normal controls (7.17/x 400 field) and in the bone marrow remission (BMR) AL patients (8.57/x 400 field) (P < 0.001, both), while the controls in the latter were higher than those in controls (P < 0.05). There was no difference in microvessel counts between untreated AL patients and relapse/refractory AL patients (21.83/x 400 field) (P > 0.05). In the untreated AL group, the microvessel counts showed difference neither between 23 ALL patients (23.09/x 400 field) and 43 ANLL patients (22.37/x 400 field) (P > 0.05), nor between patients with M(1 - 3) (22.91/x 400 field) and M(4 - 5) (21.46/x 400 field) the two subtypes of FAB (P > 0.05). There was a positive correlation between the microvessel counts and the percentage of blast cells in the bone marrow (r = 0.311, P < 0.01). The VEGF level in the bone marrow fluid in untreated AL patients (188.88 ng/L) was significantly higher than that in BMR AL patients (78.74 ng/L) and controls (79.52 ng/L) (P < 0.01), while the latter two groups had no difference between each other (P > 0.05). The VEGF level in the bone marrow fluid from ANLL group (270.12 ng/L) was enhanced significantly compared with that from ALL group (101.70 ng/L) (P < 0.01) and associated with the microvessel counts (r = 0.464, P < 0.05). CONCLUSION: Microvessel counts increased in the bone marrow in patients with different types of AL, suggesting that angiogenesis might play an important role in the pathology of AL. Testing the VEGF level in the bone marrow fluid from ANLL patients could reflect the degree of angiogenesis and disease condition.
