[Apoptosis and expression of caspase-3 protein in germ cells of varicocele rats].

OBJECTIVE: To explore the relationship between the expression of caspase-3 in testicular germ cells of rats with experimental left varicocele (ELV) and apoptosis of germ cells. METHODS: Twenty-four male SD rats were randomly divided into three groups with eight animals each: sham-operation group (SOG), 30-day post-operation group (PG1) and 60-day psot-operation group (PG2). ELV model was established by the partial ligation of the left renal vein. To detect apoptosis of germ cells and expression of caspase-3, TUNEL assay and immunohistochemistry (SABC) were used respectively. RESULTS: The number of caspase-3 positive germ cells per tubular cross section in left and right testes of rats in SOG, PG1, PG2 were 0.1175 +/- 0.0129, 0.2463 +/- 0.0421, 0.2938 +/- 0.0511 and 0.1650 +/- 0.0192, 0.2538 +/- 0.0219, 0.2775 +/- 0.0343, respectively. Compared with SOG, the expression of caspase-3 in bilateral testes of rats in PG1 and PG2 were increased, and the differences were statistically significant(P = 0.0115 and P = 0.0144). CONCLUSION: Expression of caspase-3 protein increased in germ cells of rats with ELV, which may be one of the molecular mechanisms related to excessive testicular germ cell apoptosis.

[Detection of peripheral blood Th1/Th2 cell ratio in patients with chronic abacterial prostatitis/chronic pelvic pain syndrome].

OBJECTIVE: To evaluate the change of peripheral Th1/Th2 cells in patients with CAP/CPPS and its significance in various clinical types of CAP/CPPS. METHODS: Interferon-gamma (IFN-gamma) and interleukin-4 (IL-4) secreted by CD3+ CD8(-)T cells were detected by flow cytometry in 35 patients with CAP/CPPS and 12 healthy volunteers, and significance of Th1/Th2 cells ratio in the etiology of CAP/CPPS was analyzed. RESULTS: Compared with control group, peripheral Th1 cells were significantly increased in patients with IIIA and III B CAP/CPPS, the Th1/Th2 cell ratio was apparently increased (P <0.05), while Th2 cells was not different from that in the control(P>0.05 ); Th1, Th2 cells and Th1/Th2 in patients with IIIA CPPS were not statistically different from those in III B CPPS respectively (P > 0.05) . CONCLUSION: There is a Th1/Th2 imbalance in CAP/CPPS patients with an increase of Th1 type cytokines. Th1 cell may play an important role in the pathogenesis of CAP/CPPS.

[A list and analysis of reproductive medicine periodicals indexed in MEDLINE in 2006].

MEDLINE established by National Library of Medicine (NLM), USA, is the most commonly used biomedical literature retrieval system, whose indexed literature generally represents the top of the medical world. Of the total number of 4959 journals listed in the Lists of Journals Indexed in MEDLINE (2006) published by NLM, 46 are periodicals of reproductive medicine, mainly including andrology, fertility, and sterility. Researchers of reproductive medicine should be familiar with the journals indexed in MEDLINE, closely follow their published frontier researches in this field, and consciously contribute to them in English. Editors of reproductive medicine journals should endeavour to have their journals included in such internationally authoritative systems as MEDLINE, SCI, etc by publishing them in English in compliance with international standards. This article presents a list of the reproductive medicine periodicals indexed in MEDLINE (2006) together with a brief analysis, which is meant to help researchers in reproductive medicine, andrology and other relevant fields with their contribution, subscription, reading and retrieving, as well as to share some more knowledge about MEDLINE and its indexed journals with Chinese editors of reproductive medicine journals.

[Evaluation of sperm mitochondrial function using Rh123/PI dual fluorescent staining].

OBJECTIVE: To investigate the feasibility and clinical significance of detecting sperm mitochondrial function by using Rh123/PI dual fluorescent staining and flow cytometry analysis, and to explore the relationship between the results of Rh123/PI dual fluorescent staining and seminal parameters. METHODS: Sixty-three semen samples were classified as normal (n=31) and abnormal (n=32) according to the World Health Organization guidelines. Rh123/PI dual fluorescent staining was then carried out to evaluate sperm mitochondrial function by flow cytometry analysis. RESULTS: Significant differences in Rh123+ PI-, Rh123- /PI+ and Rh123- /PI- were detected between the normal and abnormal semen samples (P < 0.05). There was a significant positive correlation between the Rh123+ PI- sperm and sperm motility and a significant inverse correlation between Rh123+ PI- and immotile sperm. But the Rh123- PI+ sperm showed a contrary relationship with Rh123+ PI-. A significant inverse correlation was also observed between the Rhl23- /PI- sperm and sperm concentration in the abnormal group. CONCLUSION: Rh123/PI dual fluorescent staining and flow cytometry analysis can readily and quickly detect sperm mitochondrial function and be used to evaluate semen quality.

[Preparation and identification of anti-human epididymal sperm protein P34H monoclonal antibody].

OBJECTIVE: To prepare and identify anti-human epididymal sperm protein P34H monoclonal antibody (McAb). METHODS: The previously purified recombinant P34H protein was used as an antigen to immunize BALB/c mouse. Cell strains secreting anti-P34H McAb were established by hybridoma technique and then ascitic fluid-type McAb prepared. The sensitivity and specificity of McAb were detected by ELISA and Western blot, respectively. RESULTS: One strain (2C4) of IgG1 Kappa anti-P34H McAb was harvested. The titer detected by ELISA technique was 1:10(3) - 1:10(5). Western blot of healthy human sperm samples and purified recombinant P34H antigen probed with the prepared McAb were immunoreactive at 34,000 and 27,000, respectively. CONCLUSION: Anti-P34H McAb has been prepared successfully by the above methods, which may provide a powerful tool for exploring the relationship between P34H and male fertility.

[Effects of vasectomy on prostatic histomorphology and cell apoptosis in rats].

OBJECTIVE: To detect the effects of vasectomy on the growth and apoptosis of prostatic tissue cells. METHODS: Forty-five SD rats were divided at random into three groups of fifteen each. Vasectomy (Vsm) model was established by ligating the bilateral vas deferens of the rats in Group A and testosterone propionate and normal saline were subcutaneously injected in those of Groups B and C, respectively. The area percentage of each part in the prostatic tissue was measured with computer-assisted image analysis system. The apoptotic rate was examined with TUNEL. RESULTS: The ratio of stromal area to epithelial plus lumen area in the prostatic tissues in Groups A, B and C were (29.20 +/- 6.85), (39.77 +/- 7.58) and (48.90 +/- 6.49), respectively, and the differences were significant statistically (P all < 0.05). The apoptosis rates in Groups A, B and C were (6.39 +/- 0.84)%, (2.62 +/- 0.57)% and (4.58 +/- 0.93)%, respectively, significantly higher in Group A than in Groups B and C (P all < 0.05). CONCLUSION: Vasectomy may induce interstitium reduction and cell apoptosis in the prostatic tissues of rats, which may help prevent benign prostatic hyperplasia.

[Androgen deficiency and erectile dysfunction].

Androgen not only indirectly affects penile erection by enhancing sexual desire and arousal, but also play an important role in maintaining the normal structure of the erectile tissue. Testosterone deficiency may result in a reduction of trabecular smooth muscle content concomitant with increased accumulation of extracellular matrix and deposition of adipocytes in the subtunical region. All this can lead to corporal veno-occlusive dysfunction. In addition, testosterone can affect the expression and activity of nitric oxide synthases (NOS) and RhoA/Rho kinase and influence penile erection directly. Androgen administration may restore erectile function for hypogonadal patients with erectile dysfunction who fail to respond to PDE5 inhibitors alone.

[The effects of dietetic hyperlipidemia on the development of testes and penises in male New Zealand rabbits].

OBJECTIVE: To study the effects of dietetic hyperlipidemia on the development of testes and penises in male New Zealand rabbits. METHODS: The animal model of dietetic hyperlipidemia was established by feeding 36 male New Zealand rabbits which were divided into the test group (n = 20) and the control group (n = 16) at random with rich fat. The concentration of TC, TG, HDL-C as well as LDL-C was determined by using automatic biochemistry analyzer. Radioimmunoassay methods were used to measure the level of T, LH and FSH as well. Alterations of testicular and penial development in morphology were observed by HE staining. RESULTS: Compared with the control group, concentrations of TC, TG and LDL-C increased dramatically as well as the levels of T, LH and FSH decreased obviously. There are significant differences between the two groups (P <0.01). Compared with the control group, the penial length was short (P < 0.05) and testicular coefficient declined (P < 0.01) in the test group. With light microscope, the following phenomenon was manifestly detected: the sabotage of spermatogenic epithelium and the adipocyte sedimentation of the penial tissue. CONCLUSION: It is obvious that intaking a large quantity of food with rich fat from juvenile would induce dietetic hyperlipidemia, result in the functional disorder of hypothalamo-pituitary-gonadal axis, and leads to short penis in development and the damage of spermatogenesis function as well.
