Cox7a2 mediates steroidogenesis in TM3 mouse Leydig cells.

AIM: To investigate the regulatory function of Cox7a2 on steroidogenesis and the mechanism involved in TM3 mouse Leydig cells. METHODS: The cDNA of Cox7a2 was cloned from TM3 mouse Leydig cells. It was subcloned to pDsRed-Express-N1 and transfected back into TM3 mouse Leydig cells for Cox7a2 overexpression by transient gene transfection. Steroidogenesis affected by overexpressed Cox7a2 was studied by ELISA. To elicit the mechanism of this effect, expression of steroidogenic acute regulatory (StAR) protein and reactive oxygen species (ROS) were examined by Western blot and fluorometer, respectively. RESULTS: The cDNA of Cox7a2 (249 bp) was cloned from Leydig cells and confirmed by DNA sequencing. After constructed pDsRed-Express-N1-Cox7a2 was transfected back into TM3 mouse Leydig cells, Cox7a2 inhibited not only luteinizing hormone (LH)-induced secretion of testosterone but also the expression of StAR protein. At the same time, Cox7a2 increased the activity of ROS in TM3 mouse Leydig cells. CONCLUSION: Cox7a2 inhibited LH-induced StAR protein expression, and consequent testosterone production, at least in part, by increasing ROS activity in TM3 mouse Leydig cells.

The impact of human EGFR kinase domain mutations on lung tumorigenesis and in vivo sensitivity to EGFR-targeted therapies.

To understand the role of human epidermal growth factor receptor (hEGFR) kinase domain mutations in lung tumorigenesis and response to EGFR-targeted therapies, we generated bitransgenic mice with inducible expression in type II pneumocytes of two common hEGFR mutants seen in human lung cancer. Both bitransgenic lines developed lung adenocarcinoma after sustained hEGFR mutant expression, confirming their oncogenic potential. Maintenance of these lung tumors was dependent on continued expression of the EGFR mutants. Treatment with small molecule inhibitors (erlotinib or HKI-272) as well as prolonged treatment with a humanized anti-hEGFR antibody (cetuximab) led to dramatic tumor regression. These data suggest that persistent EGFR signaling is required for tumor maintenance in human lung adenocarcinomas expressing EGFR mutants.

Distributed Brillouin fiber sensor for detecting pipeline buckling in an energy pipe under internal pressure.

A distributed Brillouin fiber sensor has been employed to detect localized pipe-wall buckling in an energy pipe by measuring the longitudinal and hoop strain distributions along the outer surface of the pipe for the first time. The locations of the localized pipe-wall buckling are found and distinguished using their corresponding strain-load data. The formation of the buckling process for the compression and tension characters is studied in the longitudinal and hoop directions. For the pipe with internal pressure, concentric load, and bending load, a localized pipe-wall buckling takes place away from the middle of the pipe on the compressive side and a strain peak with an overall buckling occurs on the tensile side according to the longitudinal strain distributions along the pipe. Different strains on two neutral lines are also observed in the hoop strain distribution, which should be caused by the pipe weld joint.

[Long-term impact of transfer of phrenic nerve on respiratory system of children: a clinical study of 34 cases].

OBJECTIVE: To study the long-term impact of transfer of phrenic nerve on respiratory system of children. METHODS: Thirty-four children with brachial plexus injury, 25 boys and 9 girls, underwent transfer of phrenic nerve and were divided into 3 groups according to the age when they underwent operation: group of the age of 0 - 12 months (n = 17), group of 13 - 36 months (n = 11), and group of 37 - 60 months (n = 6). Thirty-four sex, height, and body weight-matched healthy children were used as controls. Follow-up, including physical examination, pulmonary function examination (tidal volume, ventilation, etc), blood gas analysis, and chest radiography, was conducted for 4.03 years (3 - 7 years). RESULTS: The values of maximum vital capacity of the group of 0 - 12 months and group of 13 - 36 months were 1.0 L +/- 0.2 L and 1.2 L +/- 0.4 L, both significantly lower than those of the corresponding control groups (1.3 L +/- 0.3 L and 1.4 L +/- 0.5 L, both P < 0.05). The values of one-second vital capacity of the group of 0 - 12 months and group of 13 - 36 months were 0.8 L +/- 0.1 L and 0.9 L +/- 0.1 L, both significantly lower than those of the corresponding control groups (1.0 L +/- 0.1 L and 1.0 L +/- 0.1 L, both P < 0.05). However, the values of the maximum vital capacity and one-second vital capacity of the group of 37 - 60 months were 1.6 L +/- 0.3 L and 1.8 L +/- 0.5 L respectively, both not significantly different from those of the controls (both P > 0.05). The results of blood gas analysis of the 3 operation groups were not significantly different from those of the corresponding controls. Chest radiograph showed that the diaphragm top was raised by 1.93 intercostal spaces (0.5 - 3.5 intercostal spaces) in comparison with the contralateral sides with significant differences between the group of 0 - 12 months and the group of 13 - 36 months and between the group of 0 - 12 months and the group of 37 - 60 months (both P < 0.05). The recurrent respiratory infection rate and of the groups of 0 - 12 months and 13 - 36 months were 47.1% and 27.3% respectively, both significantly higher than that of the group of 37 - 60 months (0%). The thorax deformity rate of the groups of 0 - 12 months and 13 - 36 months were 41.2% and 9.1% respectively, both significantly higher than that of the group of 37 - 60 months (0%). Three of the children in the group of 0 - 12 months (17.6%) had digestive system symptoms. CONCLUSION: Transfer of phrenic nerve operated on children younger than 3 years may cause abnormalities of respiratory system, thorax, and digestive system. The younger the patients the more severe the consequences of the operation. The children older than 3 years tolerate the operation better.

Influence of transient phonon relaxation on the Brillouin loss spectrum of nanosecond pulses.

For pump-probe stimulated Brillouin scattering with a probe pulse of a few nanoseconds duration and with a finite DC level, the acoustic wave relaxation time varies with the pump power and the DC level. For a pump power of 1-6 mW, the acoustic wave relaxation changes from approximately 9 to 90 ns for polarization-maintaining fiber at a temperature of -40 degrees C for a 2 ns pulse width. When the pulse DC ratio of the probe varies from 10 to 20 dB, the acoustic relaxation time changes from 24 to 45 ns for single-mode fiber at 25 degrees C. This induces a power-increment spectral feature in the detected AC pump signal in the Brillouin loss spectrum of two temperature or strain sections, where both spectral components appeared at positions far from those related to the natural phonon relaxation time (approximately 10 ns) equivalent length. The theoretical calculations confirm the prolonged phonon relaxation.

Critical exposure level of cadmium for elevated urinary metallothionein--an occupational population study in China.

Cadmium is a well-known nephrotoxic agent with extremely long biological half-time of 15-30 years in humans. To prevent nephrotoxicity induced by cadmium, it is necessary to identify specific and sensitive biomarkers of cadmium exposure and renal damage, and to define critical exposure levels related to minimal nephrotoxicity in humans. In this study, urinary cadmium (UCd) and blood cadmium (BCd) were used as cadmium exposure indicators, urinary beta(2)-microglobulin (UB2M), N-acetyl-beta-D-glucosaminidase (UNAG) and albumin (UALB) were applied as the effect biomarkers of tubular and glomerular dysfunction. The relationship between urinary metallothionein (UMT) and cadmium exposure biomarkers as well as effect biomarkers was examined. Significant correlations were found between the UMT and BCd, and UCd. At the same time, UB2M, UALB and UNAG showed positive correlation with UMT as well. According to this result, cadmium-exposed individuals with renal dysfunction excreted more metallothionein than those without. Dose-response relationships between UCd and urinary indicators of renal dysfunction were studied. The critical concentration of UCd was quantitatively estimated by the benchmark dose (BMD) method. The lower confidence limit of the BMD-10 (BMDL) of UCd (3.1 microg/g Cr) related to increased excretion of urinary metallothionein was slightly higher than that for UNAG (2.7 microg/g Cr), but lower than those of UB2M (3.4 microg/g Cr) and UALB (4.2 microg/g Cr). The results demonstrate that UMT may be used as a sensitive biomarker of renal tubular dysfunction in cadmium-exposed populations.

Detection of Escherichia coli O157 using equal-length double-stranded fluorescence probe in a real-time polymerase chain reaction assay.

BACKGROUND: Enterohemorrhagic Escherichia coli (E. coli) O157 is a dangerous pathogen, which causes bloody diarrhea and severe hemolytic uremic syndrome (HUS). Although several assay systems based on real-time polymerase chain reaction (PCR) have been integrated to detect this pathogen, most of them are not specific. We report a real-time quantitative PCR method targeting rfbE, a gene specifically expressed in E. coli O157. This method can therefore be used to diagnose enterohemorrhagic Escherichia coli (E. coli) O157. METHODS: A nucleic acid based diagnostic assay system, combining equal-length double-stranded fluorescence probe technique and real-time PCR, was developed to detect E. coli O157. This assay system take advantage of the highly conserved rfbE O-antigen synthesis gene, and a pair of fluorescence-quenching probes complementary to rfbE gene were used in a real-time PCR to quantify the presence of the pathogen. RESULTS: The specificity of the diagnostic method was assessed by comparing test results on 14 different related pathogens including common E. coli, enteroinvasive Escherichia coli (EIEC), Salmonella, Shigella and E. coli O157. The detection limit of the method was determined using 10-fold serial dilutions of an E. coli O157 standard sample, and as few as 1.49 x 10(3) CFU/ml could be detected. All E. coli with serotype O157, which expresses rfbE gene, were positive in this assay, while all other species without rfbE gene expression were negative. CONCLUSIONS: By combining equal-length double-stranded fluorescence probe technique and real-time PCR, we have developed a simple, rapid, specific and sensitive method to detect E. coli O157.

First principles study of adsorption and dissociation of CO on W(111).

The adsorption and dissociation of carbon monoxide on the W(111) surface is studied with density functional theory. The CO molecule is found to adsorb in end-on configurations (alpha states) and inclined configurations (beta states). The dissociation of the most strongly bound beta state CO is found to have an activation energy of about 0.8 eV, which is lower than the energy required to desorb CO molecularly from the surface. The diffusion of CO and O on W(111) is predicted to be facile at room temperature, whereas C atoms are virtually immobile up to approximately 600 K, according to our calculations. Preadsorbed carbon atoms are shown to prevent the dissociation of CO by blocking the most strongly bound beta state adsorption site and by blocking the dissociation pathway. We predict that dissociation of CO on W(111) is a self-poisoning process.

Rational synthesis of meso- or beta- fluoroalkylporphyrin derivatives via halo-fluoroalkylporphyrin precursors: electronic and steric effects on regioselective electrophilic substitution in 5-fluoroalkyl-10,20-diarylporphyrins.

[reaction: see text] Electrophilic nitration, formylation or bromination of metalated 5-fluoroalkyl-10,20-diphenylporphyrin (fluoroalkyl = CF3, ClCF2CF2, n-C6F13) proceeded with high regioselectivity, exclusively affording corresponding meso-substituted porphyrins, while the iodination reaction mainly took place at the adjacent beta site giving 2-iodo-10-fluoroalkyl-5,15-diphenylporphyrin. Suzuki, Sonogashira, and trifluoromethylation reactions of the obtained 5-bromo-15-fluoroalkyl-10,20-diphenylporphyrins or 2-iodo-10-fluoroalkyl-5,15-diphenylporphyrins could perform smoothly to give the corresponding various meso- or beta-functionalized fluoroalkylated porphyrin derivatives. Accordingly, two meso-to-meso butadiyne-bridged bisporphyrin dimers and two beta-to-beta butadiyne-linked dimeric porphyrins were prepared by the coupling reactions of 5-ethynyl-15-fluoroalkyl-10,20-diphenylporphyrins and 2-ethynyl-10-fluoroalkyl-5,15-diphenylporphyrins, respectively.

Genome-wide analysis of basic/helix-loop-helix transcription factor family in rice and Arabidopsis.

The basic/helix-loop-helix (bHLH) transcription factors and their homologs form a large family in plant and animal genomes. They are known to play important roles in the specification of tissue types in animals. On the other hand, few plant bHLH proteins have been studied functionally. Recent completion of whole genome sequences of model plants Arabidopsis (Arabidopsis thaliana) and rice (Oryza sativa) allows genome-wide analysis and comparison of the bHLH family in flowering plants. We have identified 167 bHLH genes in the rice genome, and their phylogenetic analysis indicates that they form well-supported clades, which are defined as subfamilies. In addition, sequence analysis of potential DNA-binding activity, the sequence motifs outside the bHLH domain, and the conservation of intron/exon structural patterns further support the evolutionary relationships among these proteins. The genome distribution of rice bHLH genes strongly supports the hypothesis that genome-wide and tandem duplication contributed to the expansion of the bHLH gene family, consistent with the birth-and-death theory of gene family evolution. Bioinformatics analysis suggests that rice bHLH proteins can potentially participate in a variety of combinatorial interactions, endowing them with the capacity to regulate a multitude of transcriptional programs. In addition, similar expression patterns suggest functional conservation between some rice bHLH genes and their close Arabidopsis homologs.

Oligonucleotide cleavage and rejoining by topoisomerase III from the hyperthermophilic archaeon Sulfolobus solfataricus: temperature dependence and strand annealing-promoted DNA religation.

Topoisomerase III from the hyperthermophilic archaeon Sulfolobus solfataricus (Sso topo III) is optimally active in DNA relaxation at 75 degrees C. We report here that Sso topo III-catalysed DNA cleavage and religation differed significantly in temperature dependence: the enzyme was most active in cleaving ssDNA containing a cleavage site at 25-50 degrees C, but was efficient in rejoining the cleaved DNA strand only at higher temperatures (e.g. > or = 45 degrees C). The failure of Sso topo III to rejoin the cleaved DNA strand efficiently appeared to be responsible for the inability of the enzyme to relax negatively supercoiled DNA at low temperature (e.g. 25 degrees C). Intriguingly, Sso topo III facilitated DNA annealing although it showed higher affinity for ssDNA than for dsDNA. Religation of the DNA strand cleaved by Sso topo III was drastically enhanced when the DNA was allowed to anneal to a complementary non-cleaved oligonucleotide, presumably as a result of destabilization of the interaction between the enzyme and the cleaved strand through the formation of duplex DNA. A region in the non-cleaved strand corresponding to a sequence containing six bases on the 5' side and two bases on the 3' side of the cleavage site in the cleaved strand was crucial to the annealing-promoted religation. However, the annealing-promoted religation was relatively insensitive to mismatches in this region and the region conserved for oligonucleotide cleavage, except for that at the 5' end of the broken strand. These results suggest that Sso topo III is well suited for a role in DNA rewinding, whether it leads to homoduplex or heteroduplex formation.

Plasma metallothionein antibody, urinary cadmium, and renal dysfunction in a Chinese type 2 diabetic population.

OBJECTIVE: It has been reported that diabetes may increase the risk of cadmium-induced kidney damage. The presence of metallothionein antibody (MT-Ab) increased the susceptibility for tubular damage among cadmium workers. This study focused on the relationships between levels of MT-Ab, urinary cadmium, and kidney function in a Chinese type 2 diabetic population. RESEARCH DESIGN AND METHODS: A cross-sectional study was performed on 229 type 2 diabetic patients (92 men and 137 women) who were recruited from two community centers in one district of Shanghai City in China. Information was obtained from interviews, health records, and blood and urine samples. RESULTS: Levels of the tubular biomarker beta2-microglobulin increased significantly when the levels of MT-Ab and urinary cadmium were elevated in male and female subjects; in contrast, the levels of urinary albumin, a glomerular biomarker, did not display such a pattern. After adjusting for potential confounding covariates, logistic regression showed that the odds ratios (ORs) of tubular dysfunction increased upon 1) increasing the MT-Ab concentration from a low to high level (OR 5.56 [95% CI 2.25-13.73]) and 2) increasing the level of urinary cadmium from <1 to >or=1 microg/g creatinine (3.34 [1.17-9.53]); the OR of patients currently smoking was 3.51 (1.14-10.80) relative to that of those who had never smoked. CONCLUSIONS: This study proves that the presence of MT-Ab can potentiate tubular dysfunction among diabetic subjects and that patients with high MT-Ab levels are more prone to development of tubular damage.

[Relationship between plasma anti-metallothionein antibody and renal dysfunction in cadmium exposed workers].

OBJECTIVE: To investigate whether renal dysfunction induced by cadmium is related to plasma anti-metallothionein antibody (anti-MT Ab) in workers occupationally exposed to cadmium. METHODS: The male workers in a smeltery were selected as the subjects for the exposure and effect assessment. The urine cadmium (UCd), the blood cadmium (BCd) and the occupational cadmium intake (TTCd) served as the exposure indexes while the urine beta(2) microglobulin (Ubeta(2)-MG), the N-acetyl-beta-D-glucosaminidase (UNAG) and the urine albumin concentration (UALB) served as the effect markers for the renal dysfunction caused by the cadmium. The titer of the plasma anti-metallothionein antibody was determined with the enzyme linked immunosorbent assay (ELISA). RESULTS: The UCd (3.16 microg/g Cr), BCd (9.28 microg/L), Ubeta(2)-MG (81.17 microg/g Cr) and UALB (7.03 mg/g Cr) in the occupational cadmium exposure group were significantly higher than those in the control group and the Ubeta(2)-MG, UNAG and UALB as well as the occurrence rate of abnormality would be increased with the increase of the level of the occupational cadmium exposure. There was no significant difference in the titer of anti-MT Ab between the exposure group and the control group (P > 0.05). The titer of the anti-MT Ab would not be increased with the increase of the dosage of the exposure and had no significant correlation with BCd, UCd and TTCd (P > 0.05). The positive correlation were found between anti-MT Ab and UNAG as well as between anti-MT Ab and Ubeta(2)-MG in the exposure group with the correlation coefficient of 0.302 and 0.218 respectively. The workers with high level anti-MT Ab are more susceptible to cadmium nephrotoxicity than those with low anti-MT Ab with the odds ratio (OR) value of 4.200 and the 95% CI between 1.213 and 14.541 (P < 0.05). CONCLUSION: There is a dose-effect relationship between cadmium exposure and renal dysfunction in workers occupationally exposed to cadmium, but no correlation is found between cadmium exposure and plasma anti-MT Ab. The workers occupationally exposed to the cadmium with higher level of anti-MT Ab are easier to suffer from renal dysfunction caused by cadmium. Plasma anti-MT Ab could be used as a biomarker of susceptibility in the workers exposed to cadmium.

[Application of metallothionein gene isoforms expression as biomarkers in cadmium exposure].

OBJECTIVE: To investigate the feasibility of metallothionein (MT) gene expression level in human peripheral blood lymphocytes (HPBLs) as a biomarker in cadmium exposure. METHODS: The MT gene expression level in HPBLs of workers exposed to cadmium was examined using RT-PCR technique, and the exposure assessment and effect assessment were conducted in exposed workers. RESULTS: The basal MT-1A, IE, IF, IX and MT-2A expression level in workers exposed to cadmium were significantly higher than those in the control group (P < 0.05). The basal MT-1A, IE, IF, IX and MT-2A expression level would be significantly increased with the increase of the blood cadmium (BCd) level (P < 0.05). There was a trend of increase for the mRNA expression of the basal MT-1A, 1E, IF, IX, MT-2A, especially for the mRNA expression of MT-1A and MT-2A (P < 0.05) with the increase of the level of the urine cadmium (UCd). There was a good dose-response relationship between basal MT-1A expression and UCd. The basal MT-1A, IE, IF, IX and MT-2A expression level were significantly correlated with BCd (P < 0.05) while the basal MT-1A, IF and MT-2A expression level were significantly correlated with UCd (P < 0.05). There were dose-effect relationships of BCd to the basal MT-1E, MT-1F, MT-1X and MT-2X expression level respectively and there were also dose-effect relationships of UCd, beta(2)-MG and the urine metallothionein to the basal MT-1A expression. CONCLUSION: The expression of the MT gene isoforms in HPBLs can serve as the biomarker for the cadmium exposure and MT-1A can also serve as the effective biomarkers for the cadmium-induced renal toxicity.

[Heme oxygenase-1 expression and apoptosis induced by cadmium in human embryon kidney cells].

OBJECTIVE: To investigate apoptosis and expression of heme oxygenase-1 (HO-1) induced by cadmium in human embryonic kidney cells (HEK239T). METHODS: The MTT method was used for determining the cell proliferation activity. The apoptosis was determined by the flow cytometry. The HO-l mRNA expression and protein level were detected by RT-PCR method and Western blot respectively. RESULTS: The ratios of apoptosis in HEK239T cells were 11.90% +/- 0.28%, 9.27% +/- 1.73%, 9.79% +/- 0.67% and 8 .97% +/- 1.60% at the concentration of 5.0, 10.0, 20.0 and 40.0 micromol/L CdCl(2) respectively, higher than those in the control group (6.69% +/- 0.46%) with the significant difference (P < 0.01). The CdCl(2) of between 10 and 40 micromol/L could highly induce the expression of HO-1 of the human embryonic kidney cells. The expression would increase slowly till the flat stage with the increase of the dosage and then would decrease slightly over time. CONCLUSION: The cadmium can induce the apoptosis of the human embryonic kidney cells and up-regulate the expression of HO-1.

[Canal laminoplasty with titanium miniplate re-implantation of vertebral lamina].

OBJECTIVE: To evaluate the reliability and the advantage of canal laminoplasty with laminoplasty over laminectomy for treatment of lumbar spinal stenosis. METHODS: From June 2000 to September 2004, the titanium miniplate fixation technique was applied to re-implantation of the vertebral lamina in the lumbar spine. The vertebral lamina was made with a specially-made osteotome and a special technique in 18 patients with lumbar spinal stenosis. RESULTS: The patients were followed up for 1 yr and 8 mon on average (range, 3 mon-4 yr and 3 mon) and were observed to have a bony fusion of the re-implanted lamina 3-9 months postoperatively. There was no nonunion, displacement of the re-implanted lamina, overgrowth of the anterior bone edge of osteotomy, recompression of the nerves or instability of the lumbar spine. CONCLUSION: The result demonstrates that canal laminoplasty with the titanium miniplate re-implantation of the vertebral lamina in lumbar spine can restore the normal anatomy, keep stability of the spine, and avoid adhesion and scar in the canal.

[Effects of deltamethrin on neurobehavioral development of offspring of intoxicated rats].

OBJECTIVE: To investigate the effects of deltamethrin on the filial brain nitric oxide synthase (NOS) activity and neurobehavioral development of the exposed lactational rats. METHODS: Pregnant rats were randomizedly divided into the treated group and the control group. The treated group was administered orally with 3.35, 6.70 mg/kg deltamethrin every other day from postnatal day (PND) 1 to PND 19 while the control group was administered with the corn oil of same amount in the same period. The activity of NOS of filial brain and neurobehavioral functions of the filial rats were observed. RESULTS: The lactational survival rate (81.80%:78.60%) in both treated groups was decreased significantly (P < 0.01) compared with that in the control group. The body weight of filial rats on PND 10, 21 in 6.70 mg/kg DM treated group [(16.62 +/- 2.2 8), (31.34 +/- 6.94) g] was less than those in the control group (P < 0.05). The delayed time in the filial rats in 6.70 mg/kg group was (3.05 +/- 1.20) s and the positive rates of passive escaping response in 3.35 and 6.70 mg/kg DM treated group were 22.5% and 21.5% respectively. There was the trend of the developmental increase of the activity of filial brain NOS between PND 5 and PND 21 and the NOS activity of rat brain on PND 5 in 6.70 mg/kg group [(0.60 +/- 0.07) U.mg pro(-1).h(-1)] was lower than that in the control group (P < 0.05). CONCLUSION: Exposure to high dose of deltamethrin in lactational female rats will decrease the activity of NOS of brain and retard the neurobehavioral development of their filial rats.
