[Effect of surface property of different polyether-ester copolymers on growth of smooth muscle cells and endothelial cells].

OBJECTIVE: To investigate the effect of surface property of different polyether-ester block copolymers [poly(ethylene glycol-terephthalate)/poly(butylene terephthalate), PEGT/PBT] on the growth of smooth muscle cells (SMCs) and endothelial cells(ECs). METHODS: Three kinds of copolymers were synthesized, which were 1000-T20 (group A), 1000PEGT70/PBT30 (group B) and 600PEGT70/PBT30 (group C). The water-uptake and contact angle of three polyether-ester membranes were determined. The canine aorta smooth muscle cells and external jugular vein endothelial cells were primarily harvested, subcultured, and then identified. The proliferation of SMCs and ECs on the different polyether-ester membranes were investigated. RESULTS: The water-uptake of three copolymers arranged as the sequence of group C group A> group B, indicating group B being more hydrophilic. However, smooth muscle cells and endothelial cells grew poorly on the membrane of group B after low density seeding, but proliferated well on the membranes of group A and group C. CONCLUSION: In contrast with more hydrophilic 1000PEGT70/PBT30, moderately hydrophilic 1000-T20 and 600PEGT70/PBT30 has better compatibility with vascular cells. The above results indicate that the vascular cells can grow well on moderately hydrophilic PEGT/PBT and that PEGT/PBT can be used in vascular tissue engineering.

[A case-control study on the risk factors related to obstetric brachial plexus palsy].

OBJECTIVE: To identify the risk factors and related degrees associated to obstetric brachial plexus palsy(OBPP). METHODS: A case-control study was performed. Neonatal records of thirty-one cases with OBPP and their corresponding maternal records from the Department of Gynecology and Obstetrics of eight hospitals in Shanghai city from 1988 to 2002 were reviewed. Four controls, all living in Shanghai were selected to match each case and were born within the same year at the same hospital. The control group also included 124 cases without OBPP. According to the uniformed data and tables used were from medical records and from pregnant women. Epidemiological study was carried out on both case group and control group. Variables for analyses would include: (1) race, age, height, family history, pre-pregnancy weight, body mass index at the pre-pregnancy (weight/height2) on those pregnant women as well as on parity of their mothers; (2) the process of delivery which includeing clinic pelvis evaluation, height of uterus, abdomen circumference, antepartum weight, body mass index before delivery, mode of delivery, the duration of active phase and 2nd stage of labor, shoulder dystocia; (3) on neonates: sex, gestational age, birth weight, affected limb, Apgar scores of 1 and 5 minutes, other birth trauma and resuscitation of infant. Statistical tests applied to these data would include Student's T test for continuous variables and chi2 analysis for discrete data. Risk calculation of OBPP was performed by univariable and multivariable conditional logistic regression analysis. RESULTS: 12 factors related to expsure were identified for OBPP through univariable conditional logistic regression analysis. When multivariable conditional logistic regression model at P = 0.1 was applied, four factors such as cesarean (OR = 0.060), forceps (OR = 65.237), birth weight (OR = 35.468), and pre-pregnancy body mass index (OR = 23.901) were selected. CONCLUSION: Forceps delivery, macrosomia, and increase of pre-pregnancy body mass index (> or = 21) were risk factors of OBPP in the order of degrees to risk while cesarean seemed to serve as a protective factor.

Water resources: groundwater maintains dune landscape.

The Badain Jaran desert in western Inner Mongolia in China has a unique landscape that contains 72 lakes, with a total water surface area of 23 km2, and the world's highest stationary sand dunes, which are up to 500 m tall--despite the prevailing dry and windy weather conditions. Here we present evidence of a major groundwater system that underpins the factors leading to this landscape. Our finding could transform plans for the region's water resources.

[The contrast study between single and double balloon bilateral dilatation of kyphoplasty].

OBJECTIVE: To contrast single and double balloon-inflated kyphoplasty for vertebral compression fractures (VCFs) and evaluate its clinical efficacy. METHODS: From May 2000 to May 2004, 90 consecutive procedures were performed in 58 patients who suffered from painful vertebral compression fractures, transferring tumour and angioma. Ninety vertebrae were inflated while 62 as A group were double balloon and 28 as B group were single balloon, fracture reduction and bone cement augmentation. Preoperative and postoperative symptom levels, variables, complications were recorded and the vertebral height and Cobb angle were measured and analyzed. RESULTS: All patients' pain was alleviated or disappeared without syndrome, and the vertebral height and Cobb angle of both groups were improved. The average recovery rate was 72.6% (22.9% approximately 100%), Cobb angle from 17.9 degrees (3.1 degrees approximately 31.6 degrees ) were corrected to 9.6 degrees (0.6 degrees approximately 28.2 degrees ), the average angle was 8.7 degrees (0.3 degrees approximately 27.2 degrees ), and the contrast between preoperative and postoperative showed obvious differences (P <0.001). The average recovery rate of A group was 77.6% (55.3% approximately 100%), B group was 64.3% (22.9% approximately 100%). The average postoperative Cobb angle of A group was 9.9 degrees (0.3 degrees approximately 27.2 degrees ), B group was 8.6 degrees (0.6 degrees approximately 19.8 degrees ) (P >0.05). CONCLUSIONS: As a promising minimally invasive surgery, balloon kyphoplasty can provide early relief of pain and improve the function as well as spinal alignment in treatment of painful compression fracture owing to recovering the vertebral height and Cobb angle of the vertebral body. Single balloon-inflated kyphoplasty can improve VCFs as double balloon.

Gene-selective modulation by a synthetic oxysterol ligand of the liver X receptor.

Liver X receptors (LXRs) play key roles in the regulation of cholesterol homeostasis by limiting cholesterol accumulation in macrophages within arterial wall lesion sites by a mechanism that includes the upregulation of ATP binding cassette transporters. These atheroprotective properties distinguish LXRs as potential targets for pharmaceutical intervention in cardiovascular disease. Their associated activity for promoting lipogenesis and triglyceride accretion through the activation of sterol-response element binding protein 1c (SREBP-1c) expression, however, represents a potential proatherogenic liability. A newly characterized synthetic oxysterol, N,N-dimethyl-3beta-hydroxycholenamide (DMHCA), represents a gene-selective LXR modulator that mediates potent transcriptional activation of ABCA1 gene expression while exhibiting minimal effects on SREBP-1c both in vitro and in vivo in mice. DMHCA has the potential to stimulate cholesterol transport through the upregulation of LXR target genes, including ABCA1, in liver, small intestine, and peritoneal macrophages. Compared with known nonsteroidal LXR agonists, however, DMHCA exhibits only limited activity for increasing hepatic SREBP-1c mRNA and does not alter circulating plasma triglycerides. Cell-based studies also indicate that DMHCA enhances cholesterol efflux in macrophages and suggest a mechanism whereby this selective modulator can potentially inhibit cholesterol accumulation. DMHCA and related gene-selective ligands of LXR may have application to the study and treatment of atherosclerosis.

Strain dependence of Brillouin frequency, intensity, and bandwidth in polarization-maintaining fibers.

The strain dependence of the Brillouin gain-loss spectrum for PANDA, bow-tie, and tiger polarization-maintaining fibers has been studied in the range 0 to 50 degrees C. We found a linear relationship between the strain and the Brillouin frequency, intensity, and bandwidth for PANDA and bow-tie fibers. For PANDA fiber at 20-cm spatial resolution, Brillouin frequency gives 7-micro epsilon uncertainty, which is the highest accuracy among three parameters, whereas the Brillouin bandwidth gives 19-micro epsilon uncertainty.

[Study of expression of slit2 mRNA in rats of different age after sciatic nerve injury].

OBJECTIVE: To determine the age-related expression features of slit2 mRNA at various intervals at the proximal and distal parts of sciatic nerve during re-innervation after severance injury, and to investigate the molecular biological reason why co-contraction occurs more commonly in the young rats than in the adults after peripheral nerve injury. METHODS: The right sciatic nerve was transected sharply at the midthigh in the young and adult rats, 28 in each group. The proximal and distal stumps were inserted into a medical silicone tube with a gap of 3 mm. Before the injury, and 1, 3 and 7 days (6 rats at each interval) after sciatic nerve severance injury, both stumps of the sciatic nerve were harvested. Reverse transcription polymerase chain reaction (RT-PCR) was use to examine the expression of slit2 mRNA in the young and adult group rats. In situ hybridization was done to detect the cell location of slit2 mRNA expression in young and adult rats, 4 rats in each group, 2 rats used before the injury and 2 rats used on the postoperative third day. RESULTS: RT-PCR results suggested that slit2 mRNA at both stumps was significantly lower in the young group of rats than in the adult except that at the proximal part on the postoperative 7th day. There was statistically significant difference (P < 0.01). In situ hybridization studies indicated that the slit2 mRNA was specifically expressed in the cytoplasm of Schwann cells both in the young and adult rats before injury and 3 days after injury. CONCLUSION: The expression of slit2 mRNA by Schwann cells in the young rats is lower than that in the adult rats after peripheral nerve injury. These findings suggests the lower expression of chemotactic factor (slit2) by Schwann cells in the young group is likely to be the molecular biological reason for the higher incidence of co-contraction between agonists and antagonists in the younger rats.

A simplified approach for evaluation of the polarity parameters for polymer using the K coefficient of the Mark-Houwink-Sakurada equation.

Since two solvent polarity parameters, e.g., the Et(30) defined by Reichardt and the pi* defined by Kamlet and Taft, respectively, plotted with the K coefficient of the Mark-Houwink-Sakurada equation, presented good linear fits, this paper suggests applying the extrapolating method in the same way as the Zisman plot to obtain similar polarity parameters for polymers. In order to verify those obtained polarity parameter values for various polymers, literature-reported polarity values determined using other techniques are referenced and indicate that this paper introduced a simplified approach that can be applied to evaluate the polarity parameters for polymers. In this paper, since we observe that various polymers are of interest to present absolutely different slope tendencies, e.g., positive and negative, it is also suggested that polarity properties of polymers may be reinterpreted.

A statistical method for identifying differential gene-gene co-expression patterns.

MOTIVATION: To understand cancer etiology, it is important to explore molecular changes in cellular processes from normal state to cancerous state. Because genes interact with each other during cellular processes, carcinogenesis related genes may form differential co-expression patterns with other genes in different cell states. In this study, we develop a statistical method for identifying differential gene-gene co-expression patterns in different cell states. RESULTS: For efficient pattern recognition, we extend the traditional F-statistic and obtain an Expected Conditional F-statistic (ECF-statistic), which incorporates statistical information of location and correlation. We also propose a statistical method for data transformation. Our approach is applied to a microarray gene expression dataset for prostate cancer study. For a gene of interest, our method can select other genes that have differential gene-gene co-expression patterns with this gene in different cell states. The 10 most frequently selected genes, include hepsin, GSTP1 and AMACR, which have recently been proposed to be associated with prostate carcinogenesis. However, genes GSTP1 and AMACR cannot be identified by studying differential gene expression alone. By using tumor suppressor genes TP53, PTEN and RB1, we identify seven genes that also include hepsin, GSTP1 and AMACR. We show that genes associated with cancer may have differential gene-gene expression patterns with many other genes in different cell states. By discovering such patterns, we may be able to identify carcinogenesis related genes.

Dependence of the brillouin frequency shift on strain and temperature in a photonic crystal fiber.

The dependence of the Brillouin frequency shift on strain in a photonic crystal fiber (PCF) was measured at a wavelength of 1320 nm for the first time to the authors' knowledge. Together with measurements of the dependence of the Brillouin frequency shift on temperature in the PCF, we demonstrate the feasibility of the highly precise simultaneous measurement of temperature and strain by use of the PCF in a distributed Brillouin sensing system with a spatial resolution of 15 cm.

Simultaneous strain and temperature measurements with polarization-maintaining fibers and their error analysis by use of a distributed Brillouin loss system.

Simultaneous temperature and strain measurement with a distributed Brillouin loss system is proposed by use of the parameters Brillouin frequency, power, and bandwidth, for PANDA, bow-tie, and tiger polarization-maintaining fibers for the first time to our knowledge. The expressions for simultaneous temperature and strain sensing and the maximum errors and rms values of temperature and strain measurements are derived with three combinations of the parameters: (1) power and Brillouin frequency, (2) bandwidth and Brillouin frequency, and (3) bandwidth and Brillouin power. Our experiments demonstrate that simultaneous temperature and strain sensing at 20-cm spatial resolution for Brillouin frequency combined with bandwidth the strain/temperature resolutions are 39 microepsilon/2 degrees C (PANDA), 126 microepsilon/3 degrees C (bow tie), and 598 microepsilon/16 degrees C (tiger); for the Brillouin frequency combined with power the strain/temperature resolutions are 153 microepsilon/8 degrees C (PANDA) and 237 microepsilon/4 degrees C (bow tie); and for the bandwidth combined with power the strain/temperature resolutions are 135 microepsilon/38 degrees C (PANDA) and 195 microepsilon/38 degrees C (bow tie).

[Effects of icariin on the erectile function and expression of nitrogen oxide synthase isoforms in corpus cavernosum of arterigenic erectile dysfunction rat model].

OBJECTIVE: The study the effects of oral administered icariin on intracavernosal pressure (ICP) and on expression of the nitrogen oxide synthase (NOS) isoforms in corpus cavernosum (CC) of arteriogenic erectile dysfunction (A-ED) rat model. METHODS: Forty adult male Wistar rats were randomly divided into 4 groups of 10 rats: shame operated group (group A) and three A-ED model groups (group B, C and D). The internal pudendal arteries were isolated and ligated with 7-O nylon thread at both the main trunk and the penile branches to establish the A-ED model. ICP were tested after the operation to make sure the successful model establishment. The groups A and B were treated with saline: and the groups C and D were treated with icariin (5 mg/kg/day and 10mg/kg/day respectively) orally for 30 days. Then the ICP was measured again. The tissues of corpus cavernosum were taken and RT-PCR was used to detect the mRNA expression of nNOS, iNOS and eNOS in CC, and Western-blot was used to detect the protein expression of these NOS isoforms. RESULTS: The ICP in the group B was significantly decreased compared to the group A (P < 0.01), but the ICP values in the groups C and D were both increased compared to those in the group B (both P < 0.01). The expressions of the mRNA and protein of nNOS, iNOS, and eNOS were all decreased in the group B, however, the mRNA and protein expressions of eNOS were increased a in the groups C and D. In the group C, iNOS also increased. The expression of nNOS showed no obvious changes in the group C and group D. CONCLUSION: Chronic oral treatment with Icariin increases the erectile function (ICP) and restores the eNOS expression in CC of A-ED rats. Icariin may have a long-term therapeutic effect on ischemia/hypoxia induced ED.

Distributed brillouin scattering sensor for discrimination of wall-thinning defects in steel pipe under internal pressure.

A distributed Brillouin scattering sensor has been employed to identify several inner wall cutouts in an end-capped steel pipe by measuring the axial and hoop strain distributions along the outer surface of the pipe. The locations of structural indentations that constitute 50-60% of the inner pipe wall are found and distinguished by use of their corresponding strain-pressure data. These results are quantified in terms of the fiber orientation, defect size and depth, and behavior relative to those of unperturbed pipe sections.

Supramolecular self-assembly of inclusion complexes of a multiarm hyperbranched polyether with cyclodextrins.

A new class of crystalline inclusion complexes of a multiarm hyperbranched polyether combined with various cyclodextrins (CDs) was successfully prepared. Using self-condensing ring-opening polymerization, a kind of multiarm polyether with a hyperbranched poly(3-ethyl-3-oxetanemethanol) core and multiple linear poly(ethylene glycol) (PEG) arms was obtained. It has been found that this kind of hyperbranched polyether can be dissolved in water. Adding alpha-CDs to the multiarm hyperbranched polyether solution, molecular recognition results in the formation of crystalline inclusion complexes based on the noncovalent interactions between the linear PEG arms of the polyether particles and the alpha-CDs. These multiarm polyether inclusion complexes have been well characterized. Interestingly, quite different from inclusion complexes of CDs and linear polymeric guests, the complexes of the multiarm hyperbranched polyether with alpha-CDs show a novel lamellar morphology. The experimental results validate that the resultant lamellar crystals have a juxtaposed structure. In addition, the formation mechanism of these inclusion complexes of a multiarm polyether with alpha-CDs has also been well described. Besides the role of displacement of associated water molecules and the presence of hydrogen bonding between CDs in channel structure CD inclusion complexes, the noncovalent intermolecular forces between CDs and polymers also play an important role in the formation of complex architectures.

Temperature dependence of Brillouin frequency, power, and bandwidth in panda, bow-tie, and tiger polarization-maintaining fibers.

We report a study of the temperature dependence of the Brillouin gain and loss for three different kinds of commercial polarization-maintaining fibers for the first time to our knowledge. The Brillouin frequency differences between the fast and slow axes are independent of the temperature, varying between 2.9 and 4.3 MHz. Using 2-ns pulses (equivalent to a spatial resolution of 20 cm), we find that the temperature coefficients for the relative Brillouin power at a wavelength of 1310 nm are 0.26%/degrees C (panda fiber), 0.23%/degrees C (bow-tie fiber), and 0.04%/degrees C (tiger fiber); the temperature coefficients for the Brillouin frequency are 1.37 MHz/degrees C (panda), 1.66 MHz/degrees C (tiger), and 2.30 MHz/degrees C (bow-tie). The temperature coefficients for the Brillouin gain bandwidth are 0.15 MHz/degrees C (panda), 0.20 MHz/degrees C (bow-tie), and 0.22 MHz/degrees C (tiger).
