CD44 gene rs9666607 polymorphism is associated with papillary thyroid carcinoma and interacts with CREB3L1.

BACKGROUND: The incidence of papillary thyroid carcinoma (PTC) has been rising. CD44 is involved in cell adhesion and migration, but the role of its genetic variation in PTC remains unclear. METHODS: This study aimed to investigate the association of CD44 gene polymorphisms with PTC and to examine the interaction between CD44 and CREB3L1. This study enrolled 354 patients with PTC, and the genotype distribution of the CD44 polymorphism (rs9666607) was analyzed. Key PTC genes were screened using the Gene Expression Omnibus (GEO) database (GSE33630). Gene Ontology (GO) functional enrichment and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analyses were performed on these key genes. CD44 expression was validated using TCGA database, ELISA, qRTPCR, Western blot, and IHC in patient tissues, PTC mouse model, and human cell lines. The direct interactive molecules were screened through a bioinformatics method. RESULTS: The GSE33630 dataset identified a total of 124 upregulated and 85 downregulated differentially expressed genes. Enrichment analysis revealed 12 key PTC genes, including CD44. TCGA database validation revealed that CD44 was significantly overexpressed in PTC patients. The rs9666607‑A allele was associated with an increased risk of PTC and lymph node metastasis under a dominant model. CD44 mRNA and protein levels were significantly higher in PTC tissues versus adjacent tissue and further elevated in metastatic cases. Bioinformatic analysis predicted CD44 interaction with the transcription factor CREB3L1, and this was confirmed by molecular docking. CREB3L1 expression was synchronously upregulated with CD44 in PTC. CONCLUSION: CD44 polymorphisms, particularly the rs9666607-A allele, are significantly associated with PTC risk and metastasis in the studied population. CD44 is overexpressed in PTC, and its interaction with CREB3L1 suggests a potential novel interaction in PTC pathogenesis.

The RODS Open Source Project: removing a barrier to syndromic surveillance.

The goal of the Real-time Outbreak and Disease Surveillance (RODS) Open Source Project is to accelerate deployment of computer-based syndromic surveillance. To this end, the project has released the RODS software under the GNU General Public License and created an organizational structure to catalyze its development. This paper describes the design of the software, requested extensions, and the structure of the development effort.

[Photographic face recognition of cooperators vs. defectors].

Results of three experiments, in which a total of 167 students participated, consistently indicated that participants performed recognition tasks better for face photographs of defectors than those of cooperators. The face photographs used in the experiments were those of participants taken during prisoner's dilemma (PD) experiments a few years prior to the present study. In Experiments 1 and 2, photographs of cooperators and defectors in a one-shot PD experiment, taken after they had filled out a lengthy post-experimental questionnaire, were used. In Experiment 3, the photographs were those of high and low cooperators, taken at the moment of a cooperation or defection choice, respectively. Recognition was better for photographs that were judged unattractive than attractive. At the same time, it was better for photographs of less cooperative participants in the PD studies than those of more cooperative participants. Implications of the findings for Cosmides & Tooby's (1992) 'cheater-detection' module for social exchange domain were discussed.

A framework for infection control surveillance using association rules.

Surveillance of antibiotic resistance and nosocomial infections is one of the most important functions of a hospital infection control program. We employed the association rule method for automatically identifying new, unexpected, and potentially interesting patterns in hospital infection control. We hypothesized that mining for low-support, low-confidence rules would detect unexpected outbreaks caused by a small number of cases. To build a framework, we preprocessed the data and added new templates to eliminate uninteresting patterns. We applied our method to the culture data collected over 3 months from 10 hospitals in the UPMC Health System. We found that the new process and system are efficient and effective in identifying new, unexpected, and potentially interesting patterns in surveillance data. The clinical relevance and utility of this process await the results of prospective studies.

Where is the real cheese? Young children's ability to discriminate between real and pretend Acts.

This study examined 2- to 3-year-olds' ability to make a pretend-real distinction in the absence of content cues. Children watched two actors side by side. One was really eating, and the other was pretending to eat, but in neither case was information about content available. Following the displays, children were asked to retrieve the real food (Experiment 1) or point to the container with the real food (Experiments 2 and 3). 3- and 2.5-year-olds distinguished between the real and pretend acts based on behavioral cues alone. Two-year-olds chose the containers at random, but their spontaneous reactions suggested that they discriminated the real acts from pretense to some degree. Possible accounts for the discrepancy between the different behavioral measures are discussed.
