Refined solution structure of the LpxC-TU-514 complex and pKa analysis of an active site histidine: insights into the mechanism and inhibitor design.

Lipopolysaccharide, the major constituent of the outer monolayer of the outer membrane of Gram-negative bacteria, is anchored into the membrane through the hydrophobic moiety lipid A, a hexaacylated disaccharide. The zinc-dependent metalloamidase UDP-3-O-acyl-N-acetylglucosamine deacetylase (LpxC) catalyzes the second and committed step in the biosynthesis of lipid A. LpxC shows no homology to mammalian metalloamidases and is essential for cell viability, making it an important target for the development of novel antibacterial compounds. Recent NMR and X-ray studies of the LpxC from Aquifex aeolicus have provided the first structural information about this family of proteins. Insight into the catalytic mechanism and the design of effective inhibitors could be facilitated by more detailed structural and biochemical studies that define substrate-protein interactions and the roles of specific residues in the active site. Here, we report the synthesis of the (13)C-labeled substrate-analogue inhibitor TU-514, and the subsequent refinement of the solution structure of the A. aeolicus LpxC-TU-514 complex using residual dipolar couplings. We also reevaluate the catalytic role of an active site histidine, H253, on the basis of both its pK(a) as determined by NMR titration and pH-dependent kinetic analyses. These results provide a structural basis for the design of more potent LpxC inhibitors than those that are currently available.

Observation of the lead carbonyls, PbnCO(n=1-4): reactions of lead atoms and small clusters with carbon monoxide in solid argon.

Reactions of laser-ablated Pb atoms with CO molecules in solid argon lead to the formation of the lead carbonyls, PbnCO (n=1-4), using matrix-isolation infrared spectroscopy. Absorption at 2027.7 cm(-1) is assigned to C-O stretching mode of the PbCO product, which appears and increases on annealing, disappears on broadband irradiation, and recovers on further annealing. Small lead cluster mono-carbonyls PbnCO (n=2-4) are also observed in the present infrared spectra. Based on the results of stepwise annealing and the comparison with theoretical predictions, the absorptions at 1915.5, 1923.8, and 2042.8 cm(-1) are assigned to Pb2CO, Pb3CO, and Pb4CO, respectively. Bridging CO is found in Pb2CO or Pb3CO, whereas terminal CO in Pb4CO. The density functional theory calculations have been performed on these molecules and small naked lead clusters. The good agreement between experimental and calculated vibrational frequencies, relative absorption intensities, and isotopic shifts provides strong support for the identifications of these lead mono-carbonyls PbnCO (n=1-4). Furthermore, energetic analysis for the possible reactions of lead atoms with CO molecules is also given.

Observation of anomalous C-O bond weakening on discandium and activation process to CO dissociation.

Sc2[eta2(mu2-C,O)], the first homoleptic dinuclear metal carbonyl with an unprecedented bridging and side-on-bonded CO, generated from the reaction of laser-ablated Sc atoms with CO in a solid argon matrix, exhibits an unusually low C-O stretching frequency at 1193.4 cm-1, characteristic of an anomalously weakened C-O bond. This CO-activated molecule undergoes ultraviolet-visible photoinduced rearrangement to the CO-dissociated molecule, c-Sc2(mu-C)(mu-O). The infrared absorptions of the new molecules are accurately predicted by quantum chemical calculations, and the activation energy for the isomerization of Sc2[eta2(mu2-C, O)] to c-Sc2(mu-C)(mu-O) is calculated to be 15.10 kcal/mol. Our experimental and theoretical results schematically depict an activation process to CO dissociation.

Localization of P2X7 receptor immunoreactivity in the dorsal root ganglia of guinea pig.

The P2X7 receptor mRNA and proteins in guinea-pig dorsal root ganglia (DRG) were studied by using RT-PCR and immunohistochemistry. The co-localization of P2X7 receptor with four cytochemical markers, the neurofilament protein NF200, S100, substance P and isolectin B4 (IB4) binding glyco-conjugates, were also examined. It was found that P2X7 receptor immunoreactivity (P2X7 R-IR) was present mostly in large- and medium-sized DRG neurons (62% +/- 9% and 36% +/- 6% respectively in all P2X7 R-IR neurons). All the P2X7R-IR neurons were also NF200 and S100 immunopositive. However, in a small number of NF200 or S100 immunopositive neurons no P2X7R-IR was detectable. All the IB4-positive or substance P-immunopositive neurons had no P2X7R-IR. These results demonstrate that P2X7 receptors are expressed in a large subpopulation of DRG neurons and they may play a role in the transduction of specific peripheral sensory signals.

Rapid assignment of protein side chain resonances using projection-reconstruction of (4,3)D HC(CCO)NH and intra-HC(C)NH experiments.

The reconstruction of higher-dimensional NMR spectra from projections can provide significant savings in instrument time. Here, we demonstrate its application to the (4,3)D HC(CCO)NH and intra-HC(C)NH experiments. The latter experiment contains a novel intra-residue filter element, which selectively correlates the side chain resonances with the corresponding intra-residue amide resonances. Compared to the conventional HC(C)NH experiment, the intra-HC(C)NH experiment reduces the spectral complexity and thus the minimum number of projections required for artifact-free reconstruction by half. The use of the projection-reconstruction technique allows rapid data collection and unambiguous assignment of aliphatic side chain nuclei at high resolution.

Fine mapping and marker-assisted selection (MAS) of a low glutelin content gene in rice.

Rice with low glutelin content is suitable as functional food for patients affected with diabetes and kidney failure. The fine mapping of the gene(s) responsible for low glutelin content will provide information regarding the distribution of glutelin related genes in rice genome and will generate markers for the selection of low glutelin rice varieties. Following an SDS-PAGE screen of rice germplasm from Taihu Valley of China, Japonica selection W3660 is identified to be a novel mutant characterized with low glutelin content. For fine mapping the mutant gene for low glutelin content, F2 and F3 populations were derived from a cross between W3660 and Jingrennuo. SDS-PAGE analysis of the total endosperm protein showed that the low glutelin content trait was controlled by a single dominant nuclear gene. Genetic mapping, using SSRs, located this gene to chromosome 2, in the region between SSR2-001/SSR2-004 and RM1358. The distances of the two markers to the target gene were 1.1 cM and 3.8 cM respectively. By semi-quantitative RT-PCR analysis, the transcripts of GluB4/GluB5 genes located within the region do not change. However, GluB5 gene located proximal to SSR2-001/SSR2-004 was specifically reduced. SSR profiles of seven Japonica varieties were compared with that of W3660 for loci in the relevant genetic region. The markers SSR2-004 and RM1358 were used for marker-assisted selection. The selection efficiencies of SSR2-004 and RM1358 were 96.8% and 92.7% respectively. This provides a standard starting point for the breeding of low glutelin content rice varieties in China.

Low-dose anticoagulation in Chinese patients with mechanical heart valves.

The aim of this study was to provide guidelines for optimal anticoagulation in Chinese patients after mechanical heart valve replacement. A Carbomedics valve was implanted in 178 patients between July 2000 and July 2003. During follow-up, 22 bleeding events and 1 thromboembolic complication occurred. The linearized rates of bleeding and thromboembolism were 5.83% and 0.26% per patient-year, respectively. The linearized mortality rate was 0.79% per patient-year. The final mean international normalized ratio (INR) was 1.68+/-0.38, however there was a significant variation between the early and late periods of follow-up. For Chinese patients with mechanical heart valves, bleeding was the major complication rather than thromboembolism. Low-dose anticoagulation (international normalized ratio 1.4-2.0) could markedly decrease bleeding and effectively prevent thromboembolism. As the INR was most unstable in the first postoperative month, re-examination of patients in this period is critical.

[Analysis of heading time genotype for a rice photoperiod and thermo--sensitive male sterile line PeiAi64S].

PeiAi64S, a photoperiod and thermo-sensitive genic male sterile, has been wildly applied to hybrid rice seed production in China, but its photoperiod-sensitivity gene for heading date in this sterile line was still unknown. This definitely limited the further use of this sterile line in breeding practice and re-production of hybrid seeds. To solve this problem, using heading time nearly isogenic lines EGO - EG7, ER - LR, T65 - T65m and six heading date QTL-isogenic lines, NIL (Hd1) - NIL (Hd6) with the genes of Nipponbare but Hd1 - Hd6 genes from Kasalath respectively, we performed a genetic analysis of PeiAi64S with special reference to photoperiod-sensitivity loci, in natural long days at Nanjing (32 degrees N) where the average day-length is about 14 h and in natural short days at Linshui county, Hainan province(18 degrees 29'N), where the average day-length is about 11.6 h during the course of rice growing respectively. The F1 and F2 generations from the crosses 'PeiAi64S x heading time nearly isogenic lines' were subjected to genetic analyses. Experimental results showed that PeiAi64S carries photoperiod-sensitivity allele gene E1 and E3 and dominant earliness gene Ef-1 in E1 and E3 and Ef-1 loci, respectively, and a photoperiod insensitivity allele Se-1 degrees in Se-1 locus. Meanwhile, the photoperiod-sensitivity gene E1 and photoperiod-insensitivity gene Se-1e in PeiAi64S were also identified by crossing with the NIL(Hd1) and NIL(Hd4). In addition, a recessive inhibitor for photoperiod-sensitive gene E1 or Se-1(n) and other modified photoperiod-sensitive genes: i-Se-1, E3, Hd3 (En-Se-1), Hd5 and Hd6, were identified in PeiAi64S by crossing with QTL nearly isogenic lines: NIL(Hd2), NIL (Hd3), NIL (Hd5) and NIL( Hd6), The results indicated that the genotype of PeiAi64S's heading date was: E1E1e2e2E3E3Se-1(e)Se-1(e)Ef-1 Ef-1 i-Se-1 i-Se-1.

[Genomic imprinting and seed development].

The endosperm, a seed tissue that mediates the transfer of nutrients from the maternal parent to the embryo, is an important site of imprinting in flowering plants. In Arabidopsis thaliana, three genes were identified that prevent fertilization-independent seed development: FIS1/MEDEA, FIS2 and FIS3/FIE. MEDEA (MEA), a master regulator of endosperm development, is known to be imprinted in the endosperm. FWA is also imprinted in the endosperm of the model plant Arabidopsis. The following aspects were included in the present review: the imprinting mechanism in angiosperms, the latest progress in the control of MEA and FWA imprinting, the parental conflict theory to explain imprinting, the imprinting methods and other imprinted genes found in plants.

[QTLs mapping and genetic analysis of tiller angle in rice (Oryza sativa L.)].

Tiller angle is one of the most important morphological characters that has a significant effect on the formation of rice high-yield population. The tiller angles were measured in a japonica/indica RIL population with 71 lines and a genome-wide chromosome segment substitution line (CSSL) population with 65 lines at two experimental sites. A transgressive segregation was observed in both populations. QTL analysis of tiller angle was conducted based on the saturated RFLP marker linkage map and the CSSL graphical genetype. Five main-effect QTLs and three pairs of epstatic loci were detected in the RIL population. A main QTL, qTA-9, located on chromosome 9 at XNpb108 - C506, was identified at both experiment sites. The positive allele TA-9(I) on qTA-9 was contributed by indica rice IR24 with 28.6% average contribution to variance. Meanwhile, an analysis of CSSL graphical genetypes also showed that there was a positive allele on the IR24 chromosome substitution segment delimited by RFLP marker C609 and C506 with approximate 15 cM interval, which proved the existence of qTA-9. The TA-9(I) could increase tiller angle by about 15 degrees in japonica Asominori background under the two environments. The measurement of the F1 from the cross between background parent and CSSL AIS68 with TA-9(I) and the analysis of F2 population indicated that the TA-9(I) was an incomplete dominant gene. Genetype x environment interaction(G xE) was not widely present except a pair of epistatic loci with 5.32% contribution to variance of tiller angles and a relatively small additive effect. The combining action of the additive effect of the genes from both parents and the two-loci epistasis-effect may be responsible for the transgressive segregation of tiller angle in rice population. The value and approach of application of TA-9(I) in hybrid rice breeding program were discussed.

[Investigation on prevalence and influencing factors of three chronic diseases in elderly adults].

OBJECTIVE: To find out the prevalence of hyperlipemia, hypertension and diabetes, and explore the primary influence factors. METHODS: The survey consists of questionnaire and health examination on 2120 elderly adults aged above 45 year old. The possible influence factors and the primary factors in 19 factors were analyzed and screened by case-control study and logistic regression analysis. RESULTS: This investigation showed that the prevalence rates of hyperlipemia, hypertension, and diabetes were 50.12%, 44.26% and 8.74% respectively. For hyperlipemia: monthly income, BMI, WHR, coarse food grain were primary factors. For hypertension: age, smoke, intensity, BMI, WHR were primary factors. For diabetes: family history of disease, BMI, WHR, coarse food grain, exercises were primary factors. CONCLUSION: The risk factors of the three chronic diseases were obesity and unhealthy life styles, and the protective factors were coarse food grian and exercises. It's important to decrease prevalence of chronic diseases in community by controlling these risk factors.

[Changes in some defensive enzyme activity induced by the piercing-sucking of brown planthopper in rice].

The effects of brown planthopper on the activities of some key enzymes involved in defense response and the H(2)O(2) content were investigated in rice. From the fact that after piercing-sucking of brown planthopper, malondialdehyde (MDA) content of rice plant was increased rapidly (Fig.3), one can conclude that it induced lipid peroxidation; lipoxygenase (LOX), hydroperoxide lyase (HPL) and phenylalanine ammonia-lyase (PAL) activities were increased significantly in plants of resistant and susceptible varieties by piercing-sucking of brown planthopper (Figs.1,2,4); whereas the catalase activity and the H(2)O(2) content were significantly different between resistant and susceptible plants by piercing-sucking of brown planthopper, the former being decreased and the latter increased in resistance plants of variety RH (Oryza sativa L. cv. Rathu Heenati), while the reverse was true in the susceptible plants of variety TN1 (Oryza sotiva L. cv. Taichung Native 1) (Figs.5,6). The effect of brown planthopper on the PAL activity and the H(2)O(2) content were systemic (Figs.4,6), but the effect on the LOX activity occurred only in stem, the organ which brown planthopper sucking (Fig.1).

[Study of the impact of candidate genes on bone mineral density in postmenopausal women].

OBJECTIVE: To investigate the impact of multiple candidate genes on bone mineral density in postmenopausal women. METHODS: Bone mineral density (BMD) at lumbar spine and femoral neck were measured by dual-energy X-ray absorptiometry (DEXA) in 205 postmenopausal women. Polymerase chain reaction (PCR) and direct sequencing technique were used to identify osteoprotegerin gene polymorphism. Parathyroid hormone, calcitonin receptor, osteocalcin and leptin receptor genes were evaluated through PCR and restriction fragment length polymorphism. Leptin gene was genotyped by PCR and agarose electrophoresis. RESULTS: One G-1181C single nucleotide polymorphism was found in the first exon of the osteoprotegerin gene. After adjustment for age and body mass index, women who were with the CC genotype of osteoprotegerin gene or the bb genotype of parathyroid hormone gene had the highest BMD value at the lumbar spine, which was (1.042 +/- 0.142) g/cm(2) and (1.196 +/- 0.133) g/cm(2), respectively. No association was found among calcitonin receptor, osteocalcin, leptin and leptin receptor genes with BMD in postmenopausal women. Multivariate regression analysis found that the osteoprotegerin, parathyroid hormone and osteocalcin genes associated with the variance of BMD at the lumbar spine, and the parathyroid gene associated with the variance of BMD at the femoral neck. CONCLUSIONS: There is some association between osteoprotegerin, parathyroid hormone genes and BMD in postmenopausal women. However, no relationship has been found among calcitonin receptor, osteocalcin, leptin and leptin receptor genes with BMD in postmenopausal women. The osteoprotegerin gene may be the useful genetic marker for osteopenia in postmenopausal women.

Reactions of silicon atoms and small clusters with CO: experimental and theoretical characterization of SinCO (n=1-5), Si2(CO)2, c-Si2(mu-O)(mu-CSi), and c-Si2(mu-O)(mu-CCO) in solid argon.

Reactions of silicon atoms and small clusters with carbon monoxide molecules in solid argon have been studied using matrix isolation infrared absorption spectroscopy. In addition to the previously reported SiCO monocarbonyl, Si(2)(CO)(2) and Si(n)CO (n=2-5) carbonyl molecules were formed spontaneously on annealing and were characterized on the basis of isotopic substitution and theoretical calculations. It was found that Si(2)CO, Si(3)CO, and Si(5)CO are bridge-bonded carbonyl compounds, whereas Si(4)CO is a terminal-bonded carbonyl molecule. The Si(2)(CO)(2) and Si(3)CO molecules photochemically rearranged to the more stable c-Si(2)(mu-O)(mu-CCO) and c-Si(2)(mu-O) (mu-CSi) isomers where Si(2) is inserted into the CO triple bond.

C-C double- and triple-bond formation from reactions of B atoms with CO: experimental and theoretical characterization of OBBCCO and OBCCBO molecules in solid argon.

Reactions of boron atoms with CO molecules in solid argon form the following boron carbonyl species (which have been reported earlier): BCO, BBCO, OCBBCO, B(CO)2, and B4(CO)2. The OCBBCO molecule underwent a photochemical rearrangement where CO was activated to form the OBBCCO and OBCCBO molecules. The new molecules were identified on the basis of isotopic IR studies with 10B, 11B, 13C16O, 12C18O, and carbon dioxide mixtures in addition to comparison with quantum-chemical calculations of isotopic frequencies. Theoretical analyses showed that the OBBCCO and OBCCBO molecules are linear with C-C double and triple bonding, respectively, and lie at a much lower energy than the linear OCBBCO structure.

Simultaneous EQCM and diffuse reflectance UV-visible spectroelectrochemical measurements: poly(aniline-co-o-anthranilic acid) growth and property characterization.

Piezoelectric diffuse reflectance spectroelectrochemistry (PDRSEC), a new technique of diffuse reflectance spectroelectrochemistry (DRSEC) in combination with electrochemical quartz crystal microbalance (EQCM), was developed to study the electrochemical copolymerization of aniline and o-anthranilic acid in 1.0 mol l(-1) HClO4 and the properties of these copolymers. The DRSEC using an integral sphere was proven to possess a higher optical sensitivity at the unpolished piezoelectric quartz crystal electrodes used than the mirror reflectance spectroelectrochemistry mode. The copolymers grown from the copolymerization bath of different molar fractions of o-anthranilic acid (F1, relative to the total amount of the two monomers) showed intermediate properties between those of the homopolymers, which varied gradually with F1. The swelling/dissolution behavior of the copolymers vs solution pH was traced via the EQCM frequency and resistance signals, and its large dependence on F1 was found and discussed. In a HAc-NaAc buffer solution at pH 5.6, the amount of adsorbed lysozyme was found to be positively correlated with F1, via an EQCM impedance investigation, demonstrating the feasibility of using poly(aniline-co-o-anthranilic acid) as a load-adjustable immobilization matrix for cationic proteins. The novel PDRSEC method proposed is highly recommended for surface electrochemistry studies at relatively rough electrodes.

[QTL analysis for rice stripe disease resistance gene using recombinant inbred lines (RILs) derived from crossing of Kinmaze and DV85].

Rice stripe disease transmitted by small brown planthopper (Laodelphax striatellus Fall.) is one of the most serious viral diseases in East Asia. The disease is severely epidemic in most rice growing areas where the main cultivars are susceptible or moderately susceptible to rice stripe virus. In this research, a recombinant inbred lines (RILs) population of 81 lines derived from a cross of Kinmaze (japonica)/DV85(indica) by the single seed descent method was used to detect quantitative trait loci (QTL) conferring resistance to rice stripe virus(RSV). The response of the two parents and 81 RILs to RSV were investigated by inoculating seedlings with viruliferous small brown planthopper insects, and scored by the disease rate index. The quantitative trait loci for rice stripe disease resistance were analyzed by QTL Cartographer software. Three QTL controlling RSV resistance were detected on chromosomes 1, 7 and 11, respectively. Individual QTL accounted for 19.8%-30.9% of the phenotypic variance in the RILs population. The direction of the additive gene effects at two loci qStv7 and qStv11 coincided with that predicted by phenotypes of the parents. At these two loci, the DV85 alleles increased the resistance to RSV, while at qStv1, the Kinmaze alleles increased the resistance to RSV.

An efficient method for sonication assisted Agrobacterium-mediated transformation of coat protein (CP) coding genes into papaya (Carica papaya L.).

An efficient method for the production of transgenic papaya was developed via Sonication Assisted Agrobacterium-mediated Transformation (SAAT) of somatic embryos. The plasmid pGA482G was modified to contain gene PTi-Epj-TL-PLDMV with CP coding sequence of PLDMV Japan strain and chimeric gene PTi-NP-YKT with multiple CP coding sequences from PRSV Taiwan strain, PRSV Hawaii strain and PRSV Thailand strain, respectively. Disarmed Agrobacterium tumefaciens strain LBA4404 carrying the binary plasmid pGA482G with the CP genes and nptII gene was used to transform embryo calli of papaya variety Sunset to produce transgenic papaya plants. The experiment was focused on the screening of effective transformation method. The engineered Agrobacterium grown overnight was diluted with an infection media of high osmotic pressure (1/2 MS medium contain 6% sucrose and 1% glucose, pH 5.7) and adjusted to optical density OD600nm = 0.15-0.20, embryonic calli were immerged in it for 30 min and treated with 5 s, 15 s, and 20 s sonication respectively during the infection. Results indicated that 15 s sonication treatment improved the transformation efficiency dramatically. After 15 s sonication treatment on embryo calli loaded in 15 ml sterile plastic tubes, 21 putative transgenic lines were produced from 80 pieces embryonic calli (26.3%) transformed by Agrobacterium [pGA482G/CPG] and 8 putative transgenic lines was produced from 48 pieces embryonic calli (16.7%) transferred by Agrobacterium [pGA482G/CPB], while only a single line came out of 64 pieces embryonic calli (1.6%) transformed by Agrobacterium [pGA482G/CPG] and none from 25 pieces embryonic calli transformed by Agrobacterium [pGA482G/CPB] in the non-treatment control. Results also showed that the best concentration of selection antibiotic was 120 mg/L kanamycin. A total of 42 resistant shoots were produced from 421 pieces of original embryonic calli in 9 months. The presence of the CP genes in the transgenic plants and their integration into the papaya genome were confirmed by PCR and Southern hybridization respectively.
