Measures of bioavailable serum sex hormone levels in aging Chinese by protein chip.

The purpose of this study was to develop a protein chip technique based on receptor binding assays to measure bioavailable serum sex hormone levels (BSSHL). 224 aging healthy Chinese were investigated to get the referenced values of BSSHL for the first time. In the assays recombined sex hormone receptor proteins were jointed to polysaccharide coated slides to make protein chip, and the dose-dependence curves of sex hormone on chip were prepared. The data showed that this method had good precision (CV < 16%) and accuracy (Bias < 10%), and the sensitivity could reach 1 pmol/L. From the results, BSSHL of men and women declined with aging, but no significant differences were observed. The BSSHL of aging men were higher than those of women. The bioavailable serum androgen level of men was 52-112 pmol/L, women's was 3-70 pmol/L and the whole group was 41.9-81.4 pmol/L. The bioavailable serum estrogen level of men was 0.8-3.0 pmol/L, women's was 1.2-2.5 pmol/L and the whole group was 0.6-2.64 pmol/L. Based on the assays, BSSHL measurement by protein chip can meet the needs of epidemiological studies in terms of speed, accuracy and sample volume required, and was helpful in quantitative assessment of aging people's health.

Distinctive conflict processes associated with different stimulus presentation patterns: an event-related potential study.

Twelve subjects were asked to perform visual stimulus presentation tasks. Two figures were presented either simultaneously or sequentially. They were either in congruity (same shapes) or in conflict (different shapes) with each other. Conflicting stimulus pairs presented sequentially evoked a negative event-related component with a fronto-central and bilateral posterior scalp distribution, referred to as N270. In contrast, stimuli displayed simultaneously elicited a N220 with fronto-central prominence, irrespective of whether they were in conflict or in congruity. The results suggest that the N270 may be associated with the processing of a conflict with working memory information whereas the N220 might reflect the detection and evaluation of attention to visual information in the human brain.

D-galactose-caused life shortening in Drosophila melanogaster and Musca domestica is associated with oxidative stress.

D-galactose causes aging acceleration in different animal models but the mechanism is unclear. In the present study, we investigated the effects of D-galactose on lifespan and oxidative stress biomarkers in the fruit fly (Drosophila melanogaster) and housefly (Musca domestica). D-galactose was added to drinking water (20 mg/ml) for housefly and to culture medium (6.5%) for fruit fly from 24 h after emergence. Oxidative stress was estimated by measuring the activity of Cu-Zn-superoxide dismutase (SOD) and the levels of lipid peroxidation products, namely malondialdehyde (MDA) and lipofuscin in housefly brain (male) and in fruit fly (male and female). D-galactose caused a significant decrease in mean lifespan (by 12.6% of male and 15.9% of female) and maximum lifespan (by 12.9% of male and 17.1% of female) in fruit fly, and also a significant decrease in mean lifespan (by 27.1% of male, 19.8% of female) and maximum lifespan (by 27.1% of male, 21.9% of female) in housefly. MDA and lipofuscin increased with age in fruit fly and in housefly brains while change of the SOD activity showed a biphasic shape with age. D-galactose caused a significant increase in MDA and lipofuscin and decrease in SOD activity in the age-matched fruit flies and houseflies. These data indicate that D-galactose shortens the lifespan of the two different fly species and that the life shortening effect is associated with an increase in oxidative stress.

Cerebral amyloid angiopathy in aged Chinese: a clinico-neuropathological study.

To investigate the prevalence and clinico-neuropathological characteristics of cerebral amyloid angiopathy (CAA) in aged Chinese and its relationship to dementia and cerebrovascular lesions, we examined 362 archived brains of elderly with immunohistochemical staining for beta-amyloid peptide and Congo red, Bodian and Luxol fast blue stains. We found that: (1) CAA appeared in 31.7% examined brains without sexual preponderance, and the incidence increased with age; (2) the frontal lobe was most frequently involved in CAA, followed by occipital and parietal lobe; (3) subcortical white matter and cerebellum dentate nucleus areas may also be affected by CAA; (4) CAA has a close relationship to Alzheimer's disease and multiple cerebrovascular lesions; (5) CAA alone may result in dementia.

Event-related potential N270, a negative component to identification of conflicting information following memory retrieval.

OBJECTIVE: N270, an endogenous ERP component of conflict effect, was evoked in previous studies with S1-S2 paradigm. The present experiment is designed to confirm the speculation that this conflict-related negativity could also be elicited by stimulus probes having conflict with a memorized item in a visual post-retrieval comparison task. METHODS: A Sternberg probe-matching paradigm was modified in the present study. The stimuli consisted of a memory set of 3 different items (simple figures) and a retrieval set of 3 probe figures. Subjects matched each probe to its corresponding item in the memory set. The tasks were designed with different conflict loads of no-conflict, low-conflict and high-conflict in the probe retrieval test. RESULTS: Probes of no-conflict elicited a major positive going component, P300, with bilateral parietal distribution. Probes of low- and high-conflict evoked N270, while N430 was elicited only in high-conflict condition. N270 was more negative in high-conflict condition than in low-conflict condition. The N270 was right hemispheric prominent in the low-conflict task and remarkably distributed over the right prefrontal areas. On the other hand, both N270 and N430 were distributed bilaterally on the scalp in the high-conflict task. CONCLUSIONS: The present results demonstrate that the N270 is an index to the conflict identification, while the N430 of the high-conflict task reflects the processing for complex conflicts following probe retrieval. These negativities are related to the processing of conflicts.

[The application of Gallyas-Braak stainings in pathologic diagnosis of neurodegenerative diseases].

OBJECTIVE: To evaluate the role of Gallyas silver staining in the diagnosis of neurodegenerative diseases. METHOD: Modified Gallyas-Braak staining method was used to investigate samples of the brain and spinal cord of 22 cases with neurodegenerative disease including Alzheimer's disease (AD), Parkinson's diseas (PD), Pick's disease, diffuse Lewy body disease (DLBD), progressive supranuclear palsy (PSP), diagnosed by clinical and routine pathologic method. 10 cases without clinical symptoms and pathologic abnormalities of the nervous system served as control. RESULT: As compared with Bodian staining, Gallyas-Braak staining demonstrated clearly neurofibrillary tangles in the hippocampus and the cortex of frontal and temperal lobe in all the cases with Alzheimer's disease, 6 cases with dementia of other causes and 3 normal aged. However, global neurofibrillary tangles in the midbrain and the basal ganglia were found only with Gallyas-Braak staining in 4 cases with both dementia and extrapyramidal features. In addition, tuft-shaped astrocytes were shown with this method in the motor cortex, basal ganglia, midbrain of the above 4 cases and astrocytic plaques in the same area in 2 cases of the 4 cases. In this connexion, pathologic findings in 2 of the 4 cases corresponded to PSP and those of the other two cases fufiled the diagnostic criteria of corticobasal degeneration (CBD) Oligodendroglial cytoplasmic inclusions in the white matter of the brain and the spinal cord were founded in 3 of the 4 cases with multiple system atrophy (MSA). This silver staining demonstrated as well a lot of argyrophilic grains in the neuropil of the temporal lobe and the hippocampus in one case with AD. CONCLUSION: Gallyas silver staining could better reveal not only Alzheimer-like neurofibrillary tangles but also different glial inclusions in other neurodegenerative diseases such as PSP, CBD and MSA. Consequently, it is of great value in the pathologic diagnosis and study of such degenerative diseases.

Deep cerebral venous thrombosis in adults.

OBJECTIVE: To investigate the pathogenesis, clinical features, radiographic findings and therapeutic outcomes of non-acute intracranial deep venous thrombosis in adults. METHODS: Five patients who presented with increased intracranial pressure were examined with computed tomography, magnetic resonance and angiography, diagnosed as having non-acute intracranial deep venous thrombosis, and treated with thrombolytic therapy. They were reviewed retrospectively. RESULTS: There were 3 men and 2 women, aged from 22 to 49 years. Symptom duration ranged from 1 month to 7 months, and 4 of the 5 patients were associated with venous sinus thrombosis. Two patients developed cold and fever before the onset of disease, and 3 patients had no evident predisposing factors. After the infusion of thrombolytic and systemic anti-coagulant therapy, the neurological symptoms and signs of the patients were alleviated. CONCLUSIONS: Digital subtraction angiography (DSA) is more sensitive and accurate than MRI on diagnosing intracranial deep venous thrombosis. It may play an important role in the assessment of the treatment of intracranial deep venous thrombosis. Thrombolysis and anticoagulation of intracranial deep venous thrombosis appears to be a safe and efficacious treatment not only in the acute stage but also in the non-acute stage.

Contrast study on cognitive function with MRI and positron emission tomography imaging in transient global amnesia.

OBJECTIVE: To study cognitive function and cerebral metabolic changes in patients with transient global amnesia (TGA). METHODS: Three patients with TGA were given mini-mental state examination (MMSE), revised Wechsler memory scale (WMS-R) examination and magnetic resonance imaging (MRI) scans after they had been diagnosed as TGA. Using (18)F labelled deoxyglucose as tracer, patients were given a positron emission tomography (PET) examination at different periods during recovery. RESULTS: No obvious abnormality was found in MMSE and MRI scans in the three patients. However, WMS-R examination and cerebral PET imaging displayed cognitive dysfunction of varying degrees and low metabolism in local areas related to memory in 2 of 3 patients. CONCLUSIONS: In TGA patients, cognitive function and cerebral metabolic levels are closely correlated with duration of symptoms. It is necessary to stop the TGA attack as quickly as possible early time.

[A clinicopathological study of corticobasal degeneration].

OBJECTIVE: To elucidate clinical and pathological features of corticobasal degeneration. METHODS: Clinical and image materials of 2 the aged with movement disturbances and dementia were reviewed, their brain tissues were investigated by routine pathological methods, Gallyas-Braak staining and Tau immunohistochemical staining. RESULTS: Both cases early presented movement disturbance on unilateral extremity including akinesia, rest tremor, postural instability, and then gradually developed rigidity of axial and four extremities, myoclonus, impaired ocular movement, cognitive disturbance from moderate dementia to decorticate state. On SPECT, one case showed hypoperfusion of left frontal-temporal cortices, another presented asymmetric atrophy of frontal-parietal cortices on CT. Main findings on routine pathological examination were prominent atrophy of the frontal-parietal lobe, the basal ganglia, the midbrain and upper pons. The cortical layers of the frontal, parietal and insular lobe showed mild neuronal loss with gliosis and scattered ballooned neurons. Neuron losses and extensive gliosis of basal ganglia and substantia nigra were remarkable. Gallyas-Braak and tau staining showed globous inclusion in remaining neurons of basal ganglia, substantia nigra and locus ceruleus et at, substantive glial cytoskeletol degeneration including astrocyte plaques, tuft-shaped astrocyte were also found in cortex on frontal and parietal lobe and in basal ganglia. CONCLUSION: Based on the above-mentioned clinical and pathological features, it may be not difficult to diagnosis corticobasal degeneration.

[The cognitive variations among non-dementia elders: from successful aging to mild cognitive impairment].

OBJECTIVE: To study the heterogeneity of cognitive function in the non-demented elderly population with complaint of memory decline. METHODS: Neuropsychological tests were carried out in 34 subjects by using clinical memory scales including 17 variables and Q-type cluster analysis was performed. RESULTS: Fifteen subjects (44%) were diagnosed as mild cognitive impairment (MCI). Six new clusters were produced by cluster analysis. CONCLUSION: In the transitional period from normal aging to early Alzheimer's disease, the cognitive function of elders are heterogeneous, there may be cognition of successful aging, cognition of normal aging and even cognitive impairment with potential risk of dementia.
