[Isolation and characteristics of IKO-GM-1 monoclonal antibodies].

The hybridoma clone IKO-GM-1 was obtained as a result of fusion of P3X63Ag8.653 cells with splenocytes of BALB/c mice with the aid of 50% polyethyleneglycol. Antigen demonstrated by antibodies IKO-GM-1 expressed on 100% polymorphonuclear neutrophils, 54.4 +/- 3.1% monocytes, 25.9 +/- 2.4% mononuclears of healthy subjects' blood, on 11.1 +/- 1.0% T lymphocytes, on 14.6 +/- 1.6% T lymphocytes bearing Fc-receptor for IgG, on 49.3 +/- 8.2% enriched population of B lymphocytes and O cells. The treatment of healthy donors' mononuclears with antibodies IKO-GM-1 and complement blocked EK cellular activity against Molt-2 cells but not against K-562 cells. Antigen demonstrated by MAT IKO-GM-1 did not express on the colony-forming granulocyte or macrophagal cells. Antigen expressed on blast cells of patients with AMonoL, on those in part of patients with AML and AMML, on leukocytes of patients with chronic ML, on monocytes of a patient with chronic MonoL. Antigen was absent from blast cells of patients with ALL, LSA, on lymphocytes of patients with ChLL.

[Immunologic phenotype of blast cells in the diagnosis of myeloblastic leukemias].

A study was made of immunologic cell markers in 32 patients with acute myeloid leukemia by indirect immunofluorescence with the aid of xenogeneic antisera and monoclonal antibodies obtained by the authors. Ia-like antigen demonstrated by monoclonal antisera IKO-I expressed in myeloblastic leukemia with maturation in myelomonoblastic leukemia and in all patients with monoblastic leukemia. Antigen of EK-cells demonstrated by monoclonal antibodies IKO-II was found in 10 out of 18 patients. The number of antigen positive cells correlated with the number of monoblasts. One out of 22 patients showed 19% of cells which express with OLL antigen. In 2 out of 18 cases, the blast cells carried T-cell antigens. A remission was attained in 1 out of these 2 patients treated similarly to patients with OLL.

[Changes in the variant of acute leukemia].

The authors describe a female patient with acute lymphoblastic leukemia. After the first chemotherapy cycle according to the VAMP program the patient developed a complete remission. Further on, repeated extramedullary relapses (neuroleukemia, involvement of the mammary gland tissue and lymph nodes of the mesentery) were observed over 7 years, the data of blood and bone marrow analyses being within normal. At the end of the disease the patient developed a relapse with changes in the blood and bone marrow, however the blast cells were sudan and peroxidase-positive i.e. could be classified as myeloblasts.

[Immunologic phenotype of blast cells in patients with myelocytic leukemia and its clinical significance].

The immunologic phenotype of blast cells was studied in 66 patients with chronic myeloleukemia (CML) in the stage of blast crisis. cALL antigen was determined with the aid of rabbit antiserum, Ia-like antigen with the aid of monoclonal antibodies IKO-I; part of the patients showed cytoplasmic immunoglobulins. As regards the expression of the immunologic markers, the patients were divided in 2 subgroups: the lymphoid one having cALL antigen and/or the cytoplasmic chain of immunoglobulins, and the nonlymphoid having no such markers. The presence of the immunologic markers in blast cells correlated with the cytochemical traits of lymphoblasts (PAS-substance in the granular form). Nine out of the 11 patients belonging to the lymphoid group and 3 out of the 10 non-lymphoid group patients responded by short-term remissions during the treatment according to the AVAMP program.
