The investigation of influent of antiatherosclerotic diets with chitosan on clinical and metabolic parameters in patients with cardiovascular diseases. Results of the study show that enrichment of a diet with chitosan in patients with ishemic heart disease and hypertension improved clinic, immune status, antropometric levels and lipid spectrum of blood. The research has shown, that the use in the treat-preventive purposes chitosan is rather perspective.
Research of natural antibodies (NAA) is a new trend in immunohistochemistry. NAA shape up, in biological fluids, as normal human endogenous low- and high-weight substances, e.g. angiotensin 2, bradykinin and others, and play the regulatory role in homeostasis. Endothelial NO-synthase (e-NOS) is a key enzyme in the path of NO shaping. An e-NOS alteration is important in the pathogenesis of diabetic microangiopathy including retinopathy. We determined, within our case study, the level of NAA to e-NOS in blood and tears of patients with diabetic retinopathy and in donors. The level of NAA to e-NOS was found to be twice higher in tears versus blood in all groups of patients. It depended on specific features and complications of diabetes mellitus.
The results of the analysis of diagnostic significance of examination of natural antibodies (Nab) to agiotensin-converting enzyme (ACE) and its substrates in the serum of hypertensive patients indicate that concentration of Nab to ACE differ from this mean concentration in donors. An elevated level of Nab to ACE may be considered as a compensatory reaction to increased content of the enzyme in vascular endothelium and blood flow. The same patients were examined for antibodies to peptide angiotensin II (AT-II). Enzyme immunoassay has shown that a significantly elevated level of antibodies to AT-II was only in 5 examinees. The same patients had also high Nab to ACE. The study of a group of ischemic heart disease patients with adverse effects attributed to bronchial affection treated with enalapril and diroton (ACE inhibitors) demonstrates that deterioration of cough and external respiration function is not related to exacerbation of existing chronic pulmonary inflammation. None of the patients had elevated body temperature or inflammatory changes in the blood, other signs of inflammation. Enzyme immunoassay also proved that the initial level of Nab equaled mean value for donors or was insignificantly lower. Blood serum patients with side effects contained a significantly (p < 0.02) elevated quantities of Nab to bradikinin vs initial values. Thus, the proposed method of solid phase enzyme immunoassay quantifies Nab to ACE and its substrates in the patients.
Serum levels of autoantibodies to endogenous bioregulators (prostaglandin F2 alpha, angiotensin II, epinephrine, bovine serum albumin, dinitrophenol) were measured in patients with systemic and integumental lupus erythematosus and donors and the diagnostic significance of deviations of these levels from the norm was evaluated. A total of 75 patients with lupus erythematosus aged 19-54 years with disease lasting for 0.5 to 18 months were examined. Significant differences between patients and donors were observed as regards virtually all parameters except IgG to angiotensin II.
The dynamic of level of natural antibodies to factors of blood coagulation system (thrombin, antithrombin III, alpha 2-macroglobulin), to angiothensinogen and to noradrenaline in 95 patients with ischemic heart disease and hypertension was studied in antiatherosclerotic diet and diet with soy-protein (soy protein isolate and soybean flour). Universal normalizing effect of the diet with soy-protein consist in increase of levels of natural antibodies to thrombin, antithrombin III, alpha 2-macroglobulin, angiothensinogen and to noradrenaline.
The aim of the study was assessment of bronchial capacity in the treatment with ACE inhibitors. 92 patients with essential hypertension stage I-II (mean age 52 years), ischemic heart disease and bronchial asthma (a mild, moderate and severe course) in a relative remission were treated for arterial hypertension with ACE inhibitors enalapril and diroton. Enalapril improved cough indices by 38.3%, diroton--by 13.3%. This was accompanied by a rise in the level of natural antibodies to bradikinin.

A method for detecting natural antibodies to bradykinin by ELISA has been developed. A polymeric matrix model of peptide antigen has been synthesized for this purpose. Optimal conditions for detecting antigen and antibodies in the sera of donors and cardiovascular patients have been chosen. The level of antibodies to bradykinin is constant in donors and shifted in patients.

AIM: To estimate the level of natural autoantibodies (NAAb) to angiotensin-converting enzyme (ACE) and endogenic mediators affecting vascular tone (bradykinin--BK, angiotensin II--AII, vasopressin--VP) as well as the activity of serum ACE in patients with systemic diseases of the connective tissue. MATERIAL AND METHODS: Levels of NAAb were measured by enzyme immunoassay in sera from 30 patients with SLE, 19 patients with rheumatoid arthritis (RA) and 36 patients with scleroderma systematica (SS). Serum from donors served control. IgM NAAb to ACE were measured by a new technique. Serum ACE activity was determined by the initial velocity of hydrolysis reaction using spectrofluometry. RESULTS: IgM NAAb were detected in the sera of both patients and donors. SS patients had the level of NAAb to ACE in diffuse form significantly higher than in limited (p < 0.05). In SLE and SS patients ACE activity was significantly lower (p < 0.05) than in healthy subjects and RA patients. Levels of NAAb to BK was significantly elevated (p < 0.01) in patients with SLE and RA vs donors while to AII in SS patients it was lowered (p < 0.001). Patients with diffuse SS had NAAb to BK higher than patients with SS limited form (p < 0.01). In SLE the lowest levels of NAAb to all the mediators studied were observed in patients with nephritis, for NAAb to VP the differences were significant (p < 0.05). In patients with urinary syndrome concentration of NAAb to BK was significantly higher (p < 0.01), differences between their levels in patients with nephritis and urinary syndrome were also significant (p < 0.05). CONCLUSION: Further studies are needed for specification of physiological or pathological role of NAAb to endogenic mediators.
The dynamic of natural antibodies against catecholamines and alpha-2-macroglobulin, thrombin, antithrombin III and parameters of cellular immunity in 92 patients with ischemic heart disease and ischemic heart disease complicated by impaired glucose tolerance was studied of influence of antiatherosclerotic diet with fish and vegetable PUFA omega-3 from "Eicolen". Besides favorable influence to a clinical picture of the disease universal normalizing influence of antiatherosclerotic diet with addition Eicolen on parameters of humoral a cellular immunity.
Solid-phase enzyme immunoassay (EIA) was developed for detecting natural antibodies to angiotensine-converting enzyme (ACE). Optimal conditions for detecting natural anti-ACE by EIA in the sera of donors and patients with disorders of arterial pressure are selected. The findings indicate that the level of natural anti-ACE is normally constant, while in the patients it is increased in 50% cases.
The dynamics of level of natural antibodies to catecholamines (adrenaline, noradrenaline and dopamine) and factors of blood coagulation system (thrombin, antithrombin III and alpha 2-macroglobulin) in 122 patients with hypertension and obesity was studied in hyposodium antiatherosclerotic diet and this diet enriched with phospholipid preparation "Vitol" during 4 weeks. Universal normalizing effect of the diet consists in increase of levels of natural antibodies to adrenaline, noradrenaline, dopamine, thrombin, antithrombin III and alpha 2-macroglobulin was found. The influence of the diet was more expressed during it enrichment by "Vitol".
A solid-phase enzyme immunoassay procedure for detecting natural antibodies to vasopressin (VP) is developed. For this purpose, a VP antigen is synthesized on polymer matrix. Optimal conditions of enzyme immunoassay for detecting antibodies to this antigen in the sera of donors and patients with systemic lupus erythematosus (SLE) are selected. The level of anti-VP antibodies is constant in donors and shifted in SLE patients. Changes in the level of natural antibodies to VP correlate with immunochemical parameters.
Significant disturbances of immune regulation of prostaglandine metabolism were found in patients with hypertension and obesity. Use of antisclerotic diet with low sodium contents promoted positive changes of clinical symptoms of diseases, in particular normalizations of process of immune regulation of prostaglandine metabolism connected with formation of natural antibodies.
AIM: Enzyme immunoassay (EIA) determination of the level of natural antibodies (n-Ab) to catecholamines in patients with metabolic disturbances and cardiovascular diseases, correlation between content of n-Ab to these bioregulators and clinical diagnosis, biochemical indices. MATERIALS AND METHODS: EIA with specially synthetized artificial antigens was used to determine n-Ab to catecholamines (adrenaline, noradrenaline, dopamin) in 41 patients with cardiovascular diseases and metabolic disturbances (ischemic heart disease, essential hypertension, hyperlipoproteinemia) and in 10 healthy subjects (donors, controls). RESULTS: Changes in the levels of antibodies to catecholamines correspond to the clinical diagnosis. Interrelations exist between n-Ab to catecholamines and biochemical parameters in the disease. CONCLUSION: The trends in the amount of n-Ab to catecholamines are a unique marker of metabolic disturbances of these endogenic substances and can be used in diagnosis.
A highly effective rapid method is developed for detecting the rheumatoid factor in human serum. Polystyrene suspension-based latex conjugate is synthesized, representing a styrene and methacrylic acid copolymer. The immunospecific reagent is human IgG. The resultant latex conjugate can be used in practical medicine for analysis of the rheumatoid factor.
