Effects of whole-body gamma irradiation on oxygen transport by rat erythrocytes.

In this work, we studied the influence of whole-body gamma irradiation (8 Gy) upon oxygen transport by erythrocytes, through the erythrocyte count and related parameters, and through the factors affecting the oxygen affinity of hemoglobin. The oxygen affinity of hemoglobin is increased from day D + 5 after irradiation, and a severe erythropenia develops from day D + 8. These modifications probably result in tissue hypoxia via diminished oxygen transport from lungs to tissues, and decreased oxygen release from oxyhemoglobin in tissues.

[Comparative morphological study of erythrocytes in various types of whole blood and packed cell preparations stored in liquid state].

Morphological comparative study observed by scanning electron microscopy have been made on three blood stored in PVC plastic bags at 4 degrees C: 1. Whole blood in CPD, CPD-A,D (CPD added with 0,4 mM adenine and enriched with glucose 1,5 time the normal concentration) and ACD-Ad,Gua (ACD with 0,5 mM adenine and 0,5 mM guanosine); 2. Red blood cells obtained by centrifugation from blood collected in CPD, CPD-A,D and ACD-Ad, Gua; 3. Red blood cells from blood collected in CPD or ACD and resuspended in balanced, nutritious and enriched media: SAG (Saline, Adenine, Glucose) or PAGGS (Phosphate, Adenine, Guanosine, Glucose, Saline) (CPD/SAG, ACD and CPD/PAGGS). The results of statistical analysis show that the addition of adenine and guanosine in the preservative solutions improves the morphological quality of erythrocytes. On this view the resuspended media are the least traumatic. For example, in the concentrates CPD/PAGGS the level of normal or practically shape cells (discocytes and echinocytes I) nears 70 p. cent at the 6th week of storage. Thus the PAGGS system appears superior to SAG system.

[Valve of a freeze-dried gelatin-agarose combination in selective extraction of fibronectin from human plasma].

We report here a simple procedure for the isolation of human plasma fibronectin by affinity adsorption on a new lyophilized adsorbent which is obtained by mixing and polymerizing agarose with gelatin. This method permits to obtain fibronectin and a fibronectin-depleted plasma without dilution. The yield of fibronectin is about 20 to 25 per cent. Fibronectin was identified by cross-reactions against specific antibodies. Its purity (greater than or equal to 99 per cent) was controlled by immunoelectrophoresis and SDS-PAGE. Furthermore, its biological activity was demonstrated by a spreading test performed on BHK cells. Such a test can be used to determine the specific activity of extracted fibronectin considering that the number of spread cells placed in presence of exogenous fibronectin (5 microgram/ml of culture medium) was 4.5 to 5 fold-superior to the control, after 30 minutes of incubation at 37 degrees C.

[Technics for the study of a total leukocyte dialysate and its fractions].

The crude dialysate extracted from human white blood cells from a thousand unselected donors were studied by leukocyte migration inhibition test with measles antigen. This dialysate and its fractions eluted from Biogel P4 and P6 chromatography were compared towards rosette restoration test and nucleotide composition.

The influence of urea and guanidine chloride on the binding of the binding of the bacterial substrate and inhibitors to hen lysozyme at physiological temperature (40 degrees) (*).

The influence of urea and of guanidine chloride on the binding of the bacterial substrate and of inhibitors such as N-acetylglucosamine or chitotetraose to hen lysozyme were studied at 20 degrees and at 40 degrees C (physiological temperature). The action of urea did not prevent a certain degree of organization of the enzyme compatible with its usual behaviour in the presence of some inhibitors and with its crystallization ; guanidine chloride, already at low concentrations, seemed to have a more severe effect on lysozyme.
