Aliphatic ketones are acetylcholinesterase inhibitors but not transition state analogs.

A number of C4--C9 aliphatic ketones are acetylcholinesterase (acetylcholine hydrolase, EC 3.1.1.7) inhibitors, with Ki values in the 0.7--5 mM range. Comparison to analogous substrates would suggest that these ketones are transition state analogs; e.g. 2-pentanone binds to the enzyme approx. 550 times more tightly than ethylacetate. However, a number of other criteria contradict this conclusions: (1) the binding is insensitive to ketone structure: isomeric ketones, cycloalkanones, and sterically hindered ketones have similar inhibitory potencies. (2) Analogous alcohols are also good inhibitors even though they cannot form hemiketals with the enzyme. (3) Representative ketones are relatively ineffective at blocking inactivation of the enzyme by methylsulfonyl fluoride, indicating that ketones do not bind principally at the hydrolytic site. (4) A competition experiment shows that binding of tetramethylammonium chloride excludes binding of 2-pentanone, suggesting that ketones bind to the anionic rather than the hydrolytic site. Thus, observation of tight binding relative to a substrate is not a sufficient criterion to establish that an inhibitor is a transition state analog.

Ultrastructure of the antennal sensilla of aphids. I. Coeloconic and placoid sensilla.

An electron microscopical study was made of the coeloconic and placoid sensilla on the antennae of the aphids Aphis pomi, Macrosiphum euphorbiae, Nasonovia ribis-nigri, and Pemphigus bursarius. Scanning electron microscopy revealed some variation in morphology which may be functionally important but is more likely to reflect the evolution of these species. The placoid sensilla were shown by tranwo or three neurons is surrounded by two ensheathing cells. The ciliary regions of the dendrites pass through a vacuole into a cavity between an outer and an inner cuticle where they may be connected to the dendritic branches although such connections were no seen. Small pores (8 nm diameter) partially penetrate the cuticle implying that these sensilla have an olfactory function. They are suggested to be important in host selection by alate aphids. The coeloconic sensilla are poreless pegs with nonsensory cuticular projections at their tips. The distal portions of their dendrites contain densely packed microtubules and the cellular arrangement of the sensila is similar to that of the placoid sensilla. It is suggest that they may function as thermoreceptors.

Ultrastructural evidence for disuse atrophy in insect skeletal muscle.

The changes which occur in the ultrastruct of the mesothoracic retractor unguis muscle of the cockroach Periplaneta americana as a result of disuse are described. Breakdown of myofibrils, sarcoplasmic reticulum, and mitochondria are all marked, this degeneration only being apparent 9 weeks after the operation.

Comparison of four weeks' treatment with fenoterol and terbutaline aerosols in adult asthmatics. A double-blind crossover study.

Fenoterol and terbutaline, two long-acting beta 2-adrenoceptor agonists in aerosol form, were compared in an 8-wk randomized double-blind crossover study in 22 mild to moderately severe asthmatics. Patients completed diary cards, recorded peak expiratory flow rate (PEFR) twice daily, and attended a clinic for measurement of PEFR, 1-sec forced expiratory volume (FEV1), and forced vital capacity (FVC) twice during each treatment period. Fifteen patients completed the study; 5 dropped out while using fenoterol, and 2 while using terbutaline. At clinic attendances, the patients had a significantly higher mean PEFR after 4 wk on terbutaline (385 L/min) than after fenoterol (316 L/min) (p less than 0.001). Similar results were found on analysis of the morning and evening PEFR recordings. On comparing each individual's PEFR recordings during the 2 treatments, it was found that there was no significant difference among the treatments in 3 patients, while 9 patients had a better response to terbutaline, and 3 patients had a better response to fenoterol. While similar number expressed a subjective preference for each treatment, the lung function data suggested that the effectiveness of fenoterol appeared to decline during the trial period.

Samaritan contact among 325 parasuicide patients.

With the aim of aiding primary prevention of parasuicide by Samaritans, a study of 325 parasuicide patients admitted to hospital was undertaken to discover the extent of knowledge about Samaritans, the proportion of patients who had contacted Samaritans and the reasons why the remainder had not done so. Ignorance about Samaritans was found in 28 per cent, such ignorance being significantly more common among teenagers than in other age groups. Only 1.4 per cent had sought help from Samaritans immediately before the present parasuicidal act, though a further 13 per cent had contacted Samaritans previously. Suggestions are made for future public education programmes about Samaritans.

Enzyme immunoassay for measuring lipoprotein lipase activator in milk.

A method for determining the relative amount of lipoprotein lipase activator in milk was developed. The activator was measured in arbitrary units which were based on a standard curve for high density lipoprotein. Activator varied between cows not only in whole milk but also in skim milk and milk serum. Most of the activator was in the skim milk, and the amount of activator in milk serum varied between 32 and 91% of that in skim milk. In seven of eight cows, free fatty acids of milk increased with increasing units of activator.

Treatment in an alcoholism and drug addiction unit.

Estimates of the number of alcoholics in New Zealand indicate that a highly selected minority actually receive treatment for their alcoholism. This report examines who refers these patients to an alcoholism and drug dependence unit, the utilisation of compulsory treatment, and the demographic and social characteristics of the patients. A one year sample of these patients was followed for 12 months to assess treatment outcome. One year after discharge 11 percent had adopted an abstinent life-style. Forty-two percent had been rehospitalised.

A microscopical study of the innervation of flight muscles in the tsetse fly.

The gross morphology of the nervous system supplying the dorsal longitudinal flight muscles of tsetse flies is described. Electron microscopical investigation of the nerves reveals that the dorsal longitudinal muscles are innervated by branches from four main axons. A detailed description of the neuromuscular junctions in the flight muscle is presented.

Posterior spinal fusion with Harrington instrumentation using "balanced" anesthesia.

In our experience nitrous oxide-oxygen-morphine-curare anesthesia has proved preferable because of the need for swifly awakening patients after instrumentation to ascertain neurologic status. Adjunctively, doxapram has been used to awaken patients in a predictably short time span. To prevent massive blood loss, hypotension has been attained by pentolinium, trimethaphan camsylate, and sodium nitroprusside.

Anesthesia

Coagulation and fibrinolytic activity of cerebrospinal fluid.

Fibrin/fibrinogen degradation products (fragments D and E) were detected in cerebrospinal fluid in 23.4% of 252 patients admitted to a neurological/neurosurgical unit. Other coagulation proteins of low molecular weight (plasminogen and factor IX) were also present but larger proteins (fibrinogen and factor V) were not. These findings are consistent with protein leakage across a blood-CSF barrier damaged by inflammatory, vascular, or neoplastic disease. Fibrin/fibrinogen degradation products in cerebrospinal fluid after subarachnoid haemorrhage may not, therefore, be a reliable index of increased fibrinolytic activity in the subarachnoid space and may be misleading when selecting patients for fibrinolytic blockade.
