[Cryopreservation of spermatozoa of a transgenic mouse].

Spermatozoa of a homozygous transgenic mouse, in which the firefly luciferase gene was expressed under the control of beta-actin promoter, were frozen at -196 degrees C. One fourth of the frozen sperm was later thawed and used for in vitro fertilization. Thirty-six of 65 oocytes (55.4%) developed to the 2-cell stage. All the 2-cell embryos were transferred to the oviducts of pseudopregnant recipients and 23 young (63.9%, 23/36) were born. All of young analyzed carried the transgene and showed the luciferase gene expression.

Synthesis of radiopaque cyclophosphazene monomers, properties of bulk polymers and their application to composite resin.

A series of studies was conducted on the synthesis of polyfunctional cyclophosphazene monomers having radiopacity and a polymerization group in the same molecule, and their properties and applicability to composite resin were examined. Using octachlorocyclotetraphosphazene, P4N4Cl8 (4PNC), monomers were synthesized by replacing the 1-4 of chlorine (Cl) with p-bromophenol (BrC6H4OH, BrPh), and replacing the residual number of Cl, 7-4, with 2-hydroxyethyl methacrylate [CH2:C(CH3)COOCH2CH2OH](HEMA), so as to obtain four kinds of transparent monomer having radiopacity and a polymerization group in the same molecule. We then analyzed these monomers and examined their physical properties after bulk-polymerization. Next, we prepared an organic composite filler using 4PN-(BrPh)3-(EMA)5 monomer, which showed comparatively good radiopacity, to produce a new experimental radiopaque composite resin. Although radiopacity improved in accordance with the increase in the number of BrPh molecules replaced, the mechanical properties of the polymer became poorer. Similarly it was proved that the radiopacity of composite resin made with 4PN-(BrPh)3-(EMA)5 monomer was equivalent or even superior, compared with the radiopacity of the front tooth. Consequently, it was shown that these synthesized monomers can be applied to visible light-cured radiopaque composite resin.

cAMP activates Cl-/HCO-3 exchange for regulation of intracellular pH in renal epithelial cells.

The role of cAMP in regulation of intracellular pH in the confluent LLC-PK1 cells was investigated. DibutyrylcAMP and forskolin induce intracellular acidification. This acidification is inhibited by DIDS and ethacrynic acid, inhibitors of Na(+)-independent Cl-/HCO3- exchange, and by removal of extracellular Cl-. In addition, Bt2 cAMP causes Cl- entry into LLC-PK1 cells. These results suggest that cAMP activates Cl- transport, namely Na(+)-independent Cl-/HCO3- exchange, which participates in pHi regulation.

Anti-murine IL-6 receptor antibody inhibits IL-6 effects in vivo.

Thrombopoiesis, as well as antibody production, is one of the major events in which interleukin-6 (IL-6) has been reported to be involved. Polyclonal anti-murine IL-6 receptor antibody was prepared to examine the effect of the antibody on these events in IL-6-treated mice. Administration of the anti-mIL-6R antibody inhibited the IL-6-induced increase in the number of platelets. Enhancement of the serum level of DNP-specific antibody by intraperitoneal injection of IL-6 was inhibited completely with simultaneous administration of the anti-mIL-6R antibody. The level of DNP-specific antibody was decreased, even below the basal value, by the higher dose of anti-mIL-6R antibody, indicating its effect also on endogenous IL-6. This work provides evidence that anti-IL-6R antibody inhibits IL-6 function in vivo, and provides an animal model of the therapeutic use of anti-IL-6R antibody for IL-6-related disease.

Survey of Hanganutziu and Deicher antibodies in operated patients.

The appearance of Hanganutziu and Deicher (HD) antibody in the sera of patients suffering from various diseases, including malignancies of some organs and liver disorders, was investigated by enzyme-linked immunosorbent assay using N-glycolylneuraminyl-lactosylceramide (HD3) and 4-O-acetyl-HD3 as the antigenic molecules. More than 25% of sera from patients suffering from malignancies, cholelithiasis and liver cirrhosis had HD antibody, whereas none of 41 sera from healthy persons had HD antibody. The percentage of HD antibody-positive patients was similar in stages I, II and III of gastric cancer and recurrence cases. Antibody titers of the positive patients in each stage were also not different from those in each other stage. These results indicated that HD antigenic expression on cancerous tissue is not dependent on the cancerous malignancy. The HD antibody level was elevated after surgical removal of cancerous tissues in 5 of 6 patients examined, indicating that tumor growth absorbed the serum antibody. Serum antibody against 4-O-acetyl-HD3 was detected independently of HD3 antibody in some cases; however, in most cases, correlation between the two antibody titers was observed.

Root canal system of the maxillary central incisor.

To better assess the efficacy of mechanical preparation of root canals, transparent specimens of 510 extracted maxillary central incisors were investigated for thickness and curvature of the root canal, condition of any accessory canals, and location of the apical foramen. Over 60% of the specimens showed accessory canals that are impossible to clean mechanically. Most lateral branches were small, 80% were the size of a #10 reamer or less, and only 3% were thicker than a #40 reamer. Apical foramina located away from the apex were observed in 45% of the teeth, although nearly 80% of all foramina were within approximately 0.5 mm of the apex, and 95% were within approximately 1.0 mm. Data on the thickness and curvature of the main canal showed that normally it is adequately prepared when reached with a #60 reamer to the apical constriction and supplemented by flare preparation.

[The development of visible light-cured composite resin. Kinds of fillers to be mixed with cyclophosphazene system monomers and its amount and its physical properties].

A visible light-cured composite resin was developed. Cyclophosphazene monomer, 4 PN-(TF)1-(EMA)7 was prepared as a monomer. The ratio of brush abrasion, mechanical properties, water sorption, thermal expansion coefficient and the surface of abrasion were examined after mixing with fillers of different particle size (R-972, OX-50 and VL-30). The ratio of brush abrasion showed a tendency to be small when more than 50 wt% of VL-30 with a large particle size was mixed with 4 PN-(TF)1-(EMA)7 monomer. However its abrasion surface was rough compared with that of the microparticle filler. When the microparticle filler (50 wt% of OX-50) was mixed with the monomer, its mechanical properties were good for the mixture with 50 wt% OX-50. In that case, the ratio of brush abrasion was 0.268, compressive and transverse strength, 124.3 and 86.3 MPa respectively, hardness, 43.2 Hk, water absorption 14.2 micrograms/mm3 and thermal expansion coefficient, 47.4 x 10(-6)/degrees C.

[Durability of commercial composite resin. Chronological change in transverse strength and transverse elastic modulus of composite resin immersed in MeOH].

The durability of visible light-cured composite resin was examined. Five kinds of commercial redox type composite resin and 7 kinds of visible light-cured composite resin were used and their base monomers were analyzed by HPLC. After the set products were immersed in MeOH, transverse strength and transverse elastic modulus were measured. Furthermore, the MeOH sorption, solubility in MeOH and main soluble component were examined. The main component of the base monomer in all 5 kinds of redox type was Bis-GMA. Seven kinds of visible light-cured composite resin consisted of 4 kinds of Bis-GMA (including BMPEPP) and 3 kinds of UDMA. Both cases of redox type and visible light-cured type of composite resin, when they were immersed in MeOH, transverse strength and transverse elastic modulus decreased. In the case of redox type (Clearfil posterior new bond), the decrease of transverse strength and transverse elastic modulus was small, that is, its base monomer was Bis-GMA, and large quantities of hybrid type filler were mixed. In the case of redox type, transverse strength and transverse elastic modulus showed a tendency to decrease with the increase of MeOH sorption. Solubility of 12 kinds of these composite resins was 0.25-4.78% and its main component in Pyrofil light bond A was BMPEPP, the residual were coincident with the main component of base monomer.

[Water sorption, solubility, MeOH sorption and THF sorption of visible light cured resin--on resin using cyclophosphazene monomers and commercial monomers].

Studies were done to decrease both the water sorption and solubility of visible light cured resin. As monomer, three kinds of cyclophosphazene monomers and three kinds of commercial monomers were prepared to the visible light cured unfilled resins. When these set products were immersed in water, MeOH and THF, water sorption and solubility in water, MeOH sorption and solubility in MeOH, THF sorption, and mechanical properties were examined. Water sorption was increased by degrees for all the monomers. In the cases of 4 PN-(EMA)8, 4 PN-(TF)1-(EMA)7, 4 PN-(TF)2-(EMA)6, Tri-EDMA, BMPEPP and Bis-GMA + Tri-EDMA, the value of solubility in water after 30 days, was 0.16, 0.20, 0.51, 0.65, 0.17 and 0.81% respectively. MeOH sorption showed a tendency to increase, except for Bis-GMA + Tri-EDMA and 4 PN-(TF)2-(EMA)6. Solubility in MeOH after 30 days, for cyclophosphazene monomers and commercial monomers, was 0.09-0.36% and 1.36-5.40%. THF sorption after 30 days, for cyclophosphazene monomers, was small in comparison with that of commercial monomers. Compressive strength was 230-275 MPa in all cases. But, transverse strength, for all the cyclophosphazene monomers, was 45 MPa or below. In the cases of commercial monomers, the value of transverse strength was 55-90 MPa.

Heterogeneity in the myoglobin content of chicken heart Purkinje fibers.

The myoglobin content of chicken myocardial cells was studied using indirect-immunoperoxidase histochemistry. While ordinary myocardial cells exhibited a homogeneous reaction pattern, the reactions for ventricular Purkinje fibers were remarkably heterogeneous. On the basis of the degree of staining, three types of cells, i.e., dark, intermediate, and clear, were distinguishable. In addition, the cytological heterogeneity of Purkinje cells was confirmed using conventional and immunological electron microscopy. The dark cells contained more myofibrils, mitochondria, and other organelles (e.g., ribosomes) than the clear cells.

Effects of decalcification on immunoperoxidase staining.

One of the limitations of immunoperoxidase methods applied in surgical pathology is the loss of immunoreactivity during fixation and processing. In this study, the effects of decalcification on the immunoreactivity of various antigens in formalin fixed tissues were examined. The pieces of fixed tissue were placed in four different decalcifying solutions (EDTA, formic acid, nitric acid, and Plank-Rychlo solution) for various time periods and then processed routinely and embedded in paraffin. The paraffin sections of the tissues were stained with immunoperoxidase methods for immunoglobulins, hormones, or tissue/tumor-specific markers. Sections of nondecalcified tissues were used as a control. The immunohistochemical staining intensity and the number of positive cells were not reduced significantly in decalcified tissues within the usual time period of routine decalcification procedures. Many antigens survived even prolonged decalcification. The results of this study indicate that routinely decalcified tissues can be used for immunoperoxidase staining without significant loss of immunoreactivity.
