Comparison of lectin-staining pattern in testis and epididymis of gerbil, guinea pig, mouse, and nutria.

The testis and epididymis of gerbil, guinea pig, nutria, and mouse were studied after staining with seven rhodamine-conjugated lectins to disclose the distribution of glycoproteins with different sugar residues. In the testis, the lectins showed a variable affinity for Leydig cells, tubular basement membrane, cytoplasm, acrosome, and plasma membrane of maturing spermatids as well as for Sertoli cell extensions. During acrosomal development, the staining pattern showed characteristic changes with different lectins indicating a gradual processing of the glycoprotein components. The staining in the Sertoli cell extensions displayed a cyclic change linked with the release of spermatozoa. A nuclear staining was prominent in zygotene and pachytene spermatocytes in the mouse, weak in the nutria, but absent in gerbil and guinea pig. The principal cells of epididymis showed a lectin-stained Golgi region as well as a similar staining in the apical surface, microvilli, and tubular contents. This staining was most prominent in the caput/corpus regions with some interspecies differences indicating the epididymal areas active in secretion. Narrow cells active in absorption of testis-derived material were lectin-positive in the initial segment of mouse, gerbil, and nutria epididymis. Large light cells with a strong affinity for some lectins were found in the proximal cauda of gerbil and guinea-pig epididymis. In the nutria, corresponding cells were arranged as islands within the low epithelium. The distal cauda of mouse, gerbil, and nutria was the site for lectin-stained light cells interspersed among the low principal cells. It is concluded that the high and low light cells may be active in the absorption and phagocytosis of residual bodies/cytoplasmic droplets and surplus epididymal secretory material, respectively. Thus, labeled lectins formed a useful tool in the analysis of glycoprotein distribution, processing, secretion, absorption, and degradation in the male reproductive tissues.

Postnatal development of lectin-binding pattern in the rat testis and epididymis.

Seven rhodamine-conjugated lectins were utilized to study the distribution of glycoproteins in the developing rat testis and epididymis. In the testis a clear developmental pattern was found in Leydig cells and the cell boundaries between Sertoli and spermatogenic cells, as well as during acrosome formation. Some of the first degenerating meiotic cells and the apical extensions of the Sertoli cells at the time of spermiation also displayed a characteristic lectin binding. The epididymal differentiation was characterized by an increasing lectin binding of the subapical Golgi zone and apical surface, and intratubular secretion prior to the arrival of sperm. After the accumulation of tubular secretion and sperm some epithelial cells were transformed into narrow (initial segment) and light cells (distal caput, cauda) with a strong affinity for some lectins. These cells appeared to be responsible for the absorption and digestion of tubular material derived from the testis and of surplus secretion and/or sperm structures.

Lectin staining of rat testis and epididymis after ligation of excurrent ducts at different levels.

Seven rhodamine-conjugated lectins (PNA, RCA I, SBA, Con A, WGA, UEA I, and DBA; see Table 1) were utilized in studying the staining pattern of glycoproteins in rat testis and epididymis after ligation of ductuli efferentes (DE), corpus epididymidis (CE), and vas deferens (VD) for various time periods. Ductuli efferentes ligation caused a widening of seminferous tubules and detachment of spermatids with formation of multinuclear cells. These cells acquired a strong affinity for all lectins. Corpus epididymidis ligation also caused degeneration of spermatids with increased lectin staining in some tubules, but after 7 days another cell population close to the periphery of seminiferous tubules showed an increased nuclear affinity for some lectins followed by a clear degeneration and strong cytoplasmic staining with all lectins. Vas deferens ligation caused no degenerative changes in testicular spermatids. However, the peripheral cell population showed degenerative changes similar to those found after CE ligation. In both cases this was coincident with the formation of spermatic granulomas at the site of ligation. Ductuli efferentes ligation caused a gradual decrease of intratubular content in caput epididymidis, while the contrary was true after CE ligation. The latter was associated with intratubular accumulation of lectin-positive swollen cells and sperm aggregates as well as an increased lectin staining of narrow cells in initial segment and light cells in distal caput. After VD ligation an increased staining of light cells was initially found in distal cauda and distal caput, but, concomitant with distension of the tubules this reaction decreased.(ABSTRACT TRUNCATED AT 250 WORDS)

Effect of cyproterone acetate on the reproductive system of the female rat. A histological review.

Effects of cyproterone acetate, a synthetic steroidal compound, on the reproductive organs of female rats have been investigated. This agent caused reduction of ovarian weights indicative of suppression of pituitary gonadotrophins. Oestrogenic nature of cyproterone acetate was investigated in intact and ovariectomized rats taking uterine weight and vaginal keratinization as an index of oestrogenicity. Cyproterone acetate in ovariectomized animals induced vaginal keratinization and increased the uterine weights. These effects were parallel to the effect of oestradiol dipropionate in ovariectomized animals, thus indicating oestrogenic activity of cyproterone acetate. We may conclude that the above compound caused antifertility effects due to its oestrogenic nature at the dose level of 2 mg/alternate day in rats when the compound was administered subcutaneously.

Effect of short-term administration of cyproterone acetate on the reproductive tract histology, ovarian and serum cholesterol contents of female monkey (Presbytis entellus entellus).

Short courses of cyproterone acetate, a compound with progestational and antiandrogenic activities were administered to normally cycling female monkeys to investigate into the effects of cyproterone acetate on the reproductive tract. In little earlier follicular stage 20 mg of the drug daily for 30 days increased the cholesterol contents in the gonads. Serum cholesterol contents were slightly increased. When the reproductive tract was examined histologically, the gonads showed reduction in size, along with widespread degeneration of the young follicles as well as other growing follicles. Uterus showed progestational endometrial proliferation and sloughing of the vaginal productive tract.

Effects of methallibure (I.C.I. 33828) on the female reproductive tract of the Indian Desert gerbil (Meriones hurrianae Jerdon).

Effects of methallibure (I.C.I. 33828) have been studied on the reproductive organs of female gerbils, Meriones hurrianae. Methallibure administration caused reduction of gonadal weight indicating suppression of pituitary gonadotrophins. In the ovaries of the treated females extensive atresia of follicles and of corpora lutea was observed. In addition methallibure interrupted the sex cycle, and a dioestrous vaginal pattern was retained throughout medication. A significant decrease in the level of RNA, protein and sialic acid in the uterus and vagina and of glycogen in the uterus was found. Histological and biochemical changes in the ovaries and uteri suggest a possible direct antioestrogenic action of the compound.

Antifertility activity of alpha-chlorohydrin (3-chloro-1, 2 propanediol, U-5897) on the female rats.

alpha-chlorohydrin administration brings about regression in the corpus luteum and shedding of the uterine endometrium, indicating its luteolytic activity, alpha-chlorohydrin inhibited vaginal keratinization and interrupted the sex cycle. A dioestrous leucocytic pattern of the vaginal smear is retained during medication. Decreased synthesis of RNA, protein, sialic acid and glycogen concentration in the uterus and vagina suggest possible antioestrogenic action of the compound. The antifertility action of the drug may be due to its antioestrogenic property.

Effect of cyproterone acetate on the genital organs of female rats.

Effects of cyproterone acetate, a synthetic steroidal compound, on the reproductive organs of female rats have been investigated. This agent causes reduction in ovarian weights indicative of suppression of pituitary gonadotrophins. Oestrogenic nature of cyproterone acetate was investigated in intact and ovariectomized animals taking uterine weight, vaginal keratinization and other oestrogen sensitive biochemical parameters. Cyproterone acetate in ovariectomized animals induced vaginal keratinization, increase in uterine weight and uterine protein, RNA, glycogen and sialic acid contents. These effects were parallel to the effect of oestradiol dipropionate in ovariectomized animals, thus indicating oestrogenic activity cyproterone acetate. The histological and biochemical parameters lead to the conclusion that cyproterone acetate possessed antifertility property due to its inherent oestrogenic nature.

Estrogenic and antifertility effect of cyproterone acetate in female gerbils, meriones hurriane Jerdon.

This agent causes reduction of ovarian weights indicative of suppression of pituitary gonadotrophins. Estrogenic nature of cyproterone acetate was investigated in intact and ovariectomized gerbils taking uterine weight, vaginal keratinization and glycogen contents are parameters of estrogenic action. Cyproterone acetate in ovariectomized gerbils induced vaginal keratinization, increase in uterine weight, protein, RNA, glycogen and sialic acid contents of uterus, thus indicating an estrogenic activity. The histological and biochemical parameters lead to the conclusion that cyproterone acetate possesses estrogenic properties.

Histological and biochemical changes in the female genital tract of rats and mice after methallibure treatment.

Methallibure administration caused reduction of gonadal weights in female rats and mice indicative of suppression of pituitary gonadotrophins. In the ovaries of the methallibure treated females, there was a great atresia of the follicles and of the corpora lutea. Methalbure caused atrophic changes in the endometrial lining of the vagina. Uterine horns became thin after methallibure treatment. A significant decrease in protein, RNA and sialic acid concentration in uterus and vagina was observed following methallibure treatment. Uterus of treated animals showed decreased glycogen contents. Histological and biochemical changes in the female genital tract of rats and mice suggested that the drug caused antifertility effect due to its possible antiestrogenic action.

The oestrogenic activity of danazol in the female gerbil (Meriones hurrianae Jerdon).

Antifertility and estrogenic effects of Danazol have been studied in female gerbils. Danazol brings about supression of ovarian weight, presumably by inhibiting gonadotrophin secretion. Enlargement of uterine horns and well defined vaginal keratinization was seen in Danazol treated ovariectomized animals. The results were similar to that of oestradiol dipropionate confirming the oestrogenic activity of Danazol.

Observations on the effects of danazol on the female reproductive tract of mice.

Enlargement of uterine horns and well defined vaginal keratinization was seen in Danazol treated ovariectomized animals. The results were similar to that of oestradiol dipropionate confirming the estrogenic activity of Danazol.

The effect of hypothyroidism on the female genital tract of gerbils (Meriones hurrianae jerdon).

The effects of hypothyroidism on the female genital tract of gerbils have been studied. Hypothyroidism was produced by (a) surgical ablation, and (b) pharmacological suppression of the gland. Hypothyroidism resulted in atrophic ovaries. Follivular development was severely arrested, with most of the follicles showing atresia. Distinct effects were produced upon the uterine physiology. Uterine regression was conspicuous in thyroidectomized females. Hypothyroidism resulted in a decreased RNA, protein, sialic acid and glycogen concentration of the uterus. Vaginal RNA, protein and sialic acid contents were low after thyroidectomy. The vaginal cytology showed a constant pattern of its cells, i.e. dioestrous. L-thyroxine treatment restores the biochemical changes of uterus and vagina to subnormal levels in thyroidectomized animals. It is concluded that hypothyroidism affected the weight, cytology and biochemistry of the female genital tract of gerbils.

The effects of cadmium chloride on testis and epididymis of the Indian Hanuman langur, Presbytis entellus entellus Dufresne.

Administration of single sub-lethal dose (12 mg/kg body wt.) of cadmium chloride neutralizes endogenous hormonal activity in th male Indian Hanuman langur (Presbytis entellus entellus), resulting in a marked decrease in the production of androgens and subsequent reduction in the weights of sex accessory glands. Cadmium chloride in low dose (4 mg/kg body wt.) failed to bring about any significant change in the weights of excised tissue but did change biochemical composition and histo-architecture of testes. A significant increase in the concentration of testicular cholesterol was observed after the treatment. Decrease in the sialic acid concentration, total lipids and acid phosphatase activity of testis was associated with the degenerative changes of the spermatogenic elements. Reduced fructose and sialic acid concentration in the seminal vesicle of cadmium chloride treated animals indicate an inhibition of androgen production. Our results show that sialic acid epididymis is not androgen dependent in langurs. It is suggested that cadmium induced biochemical and histological changes in the testes and epididymides are dose dependent.

Long-term effects of vasectomy on the pituitary-gonadal system of rat and mouse.

A long-term vasectomy operation in rat and mouse did not reveal any consistent change in the weights of testis and androgen dependent organs. Histological structure of the testis and caput epididymis remains normal after vasectomy. The androgenic activity of the testis remains normal as reflected by fructose content of seminal vesicles. Pituitary cytology remains unaltered. Yellowish cysts were observed frequently in the cauda epididymis of experimental animals, associated with increased epididymal weights. The protein and RNA content of the testis did not show appreciable change. A significant increase was observed in the protein and RNA content of epididymis after bilateral vasectomy. There was no effect of bilateral vasectomy on sialic acid contents of testis and epididymis of rats and seminal vesicular fructose contents of mouse.
