L3L4ES antigen and secretagogues induce histamine release from porcine peripheral blood basophils after Ascaris suum infection.

The aim of this paper was to investigate the role of porcine basophils in protective immunity. Experimental pigs were infected with 10(3) Ascaris suum eggs daily for 21 days. Control pigs were maintained helminth-free. Circulating porcine basophils were isolated from the anticoagulated whole blood of A. suum-infected and noninfected pigs by dextran (4.5%) sedimentation of erythrocytes or by the centrifugation of dextran-isolated leukocytes through discontinuous Percoll gradients. Results showed that 2.2% of the isolated leukocytes, stained with May-Grunwald Giemsa, were basophils. Each basophil from infected pigs contained 1.30 x 10(-2) to 1.20 x 10(-1) pg of histamine. Peripheral blood basophils (PBBs) from infected swine released 49% specific histamine when induced with A. suum-derived antigen (L3L4ES), 55% with anti-immunoglobulin G, and 62% with calcium ionophore A23l87. During A. suum infection, the number of isolated basophils and histamine levels peaked at 14 to 21 days postinfection and then showed a significant decrease. Percent-specific histamine released from PBBs by infected swine was significantly greater than that released by control pigs. The L3L4ES antigen and secretagogues effectively induced specific/nonspecific histamine release from PBBs and should facilitate future investigations of porcine basophils.

Sodium and potassium excretion in normotensive and hypertensive population in Kashmir.

One hundred thirty five hypertensive patients and equal number of age and sex matched healthy controls were taken up for studying the relationship of 24 hour urinary sodium and potassium excretion, sodium-potassium molar ratio and body mass index (BMI) with blood pressure in normotensive and hypertensive population in Kashmir. There was statistically significant elevated 24 hour urinary sodium excretion (p < .001), increased Na+-K+ molar ratio, significantly higher BMI in hypertensive population as compared to controls whereas there was a lower 24 hour urinary excretion of potassium (p > .20) in patients with hypertension. Thus sodium and potassium excretion, Na+-K+ molar ratio and body mass index has direct bearing in perpetuation or causation of hypertension in Kashmir which may be related to intake of salt tea.

Seasonal variation in selenium status of different classes of grazing goats in a semi-arid region of Pakistan.

The selenium status of three different classes of goats ((i) female lactating, (ii) female non-lactating, and (iii) male goats) grazing semi-arid pasture in the southern part of the Punjab province, Pakistan and that of selenium concentration of soil and dietary sources, ingested by those animals were investigated during two different seasons of the year (winter and summer). Soil, forage, feed, water from the pasture and blood plasma, urine, faeces, and (if applicable) milk from these goats were collected fortnightly. The samples were analyzed for selenium concentrations. Soil selenium showed both seasonal and sampling periods effect on its concentration while forage selenium was affected only by the seasonal changes. No significant effect of seasons or fortnights on feed selenium level was observed. In fecal samples selenium concentration in lactating and non-lactating and plasma of male goats were affected by sampling periods. While fecal selenium in male goats showed significant effect on its concentration both seasonal and within fortnights. Severe deficient level of soil selenium during both seasons and marginal deficient level of forage selenium during summer were observed. Selenium concentrations in feed slightly exceeded the requirements of ruminants in feed during both seasons of the year. Plasma selenium concentrations in all goat classes were higher in winter than that in summer showing no seasonal or fortnight variation and its concentration was slightly lower in lactating goats as compared to other classes. On the bases of these results, it is concluded that overall selenium status of the goats based on plasma selenium concentration may be considered adequate mainly due to the mineral supplement provided all over the year, since soil and forage selenium concentrations were low to deficient.

Complete liver duplication with right central diaphragmatic defect.

An 11 years old boy was admitted with history of repeated chest infections, constipation and breathing difficulty especially on eating solid diet since birth. CT scan chest showed diaphragmatic hernia. Complete liver duplication in the right chest with its independent vascular and biliary systems associated with right central diaphragmatic defect leading to diaphragmatic hernia was found during surgery.

Fine needle aspiration of unilocular ovarian cysts--a cytohistological correlation.

OBJECTIVE: To assess the diagnostic efficacy of Fine Needle Aspiration Cytology in differentiating neoplastic and non-neoplastic dysfunctional ovarian cysts--cytological findings to be verified with histology of excised cyst. METHODS: In this prospective study fifty-three cases with unilocular nonseptate ovarian cystic masses,detected on ultrasound examination were subjected to ultrasound guided fine needle aspiration of the cyst contents at Department of Pathology, Allama Iqbal Medical College Lahore, from January 1999 - January 2000. Aspirated fluid was examined cytologicaly using Giemsa stain on the smears prepared from centrifuged deposit. The same cyst removed surgically was examined histologicaly and cytohistological correlation was carried out. RESULTS: In this series of 53 cases, histologically confirmed break up of various types of cysts was follicular cysts (n=25), leuteal cysts (n=6), serous cysts (n=15), mucous cysts (N=4) and Endometriotic cysts (n=3). Non-diagnostic fine needle aspirate was obtained in 19/53 cases, majority being the follicular cysts. The cytohistological correlation revealed no false positive but 40% false negative results for follicular cysts on cytological examination of the aspirate. Hence the specificity and sensitivity for cytological diagnosis of follicular cyst was 100% and 60% respectively. For leuteal cysts, false positive and false negative results on cytological examination were 0% and 16.6% respectively with a specificity and sensitivity value of 100% and 83% respectively. For neoplastic serous cysts cytologically false positive and false negative diagnosis was 0% and 46.6% respectively with specificity and sensitivity of 100% and 53%. For mucinous cystadenomas sensitivity and specificity of cytological diagnosis was 100%. For endometriotic cysts a sensitivity of 67% and specificity of 100% was procured with cytological evaluation. CONCLUSION: Guided fine needle aspiration cytology may prove to be one of the most valuable and acceptable tools in the differential diagnosis of ovarian cystic lesions.

Pigtail catheter for the treatment of ascites associated with ovarian hyperstimulation syndrome.

BACKGROUND: Severe ovarian hyperstimulation syndrome (OHSS) is potentially dangerous. The study aim was to evaluate the efficacy and safety of percutaneous pigtail catheter drainage for the management of ascites complicating severe OHSS. METHODS: This was a prospective trial conducted at a private IVF centre and a tertiary teaching medical centre. A total of 26 patients with severe OHSS was recruited. Patients were divided into two groups. Patients in group 1 (n = 13) were hospitalized, while patients in group 2 (n = 13) were managed on an outpatient basis. A pigtail catheter was inserted under transabdominal ultrasound guidance and kept in place until drainage ceased. The main outcome measures were resolution of OHSS as determined by symptomatology and laboratory values, time to removal of catheter, patient tolerance of the procedure and complication rate. RESULTS: The catheter was successfully placed in all patients following one attempt and was kept in place for a mean +/- SD of 12.9 +/- 4.3 days (range 7-24). Average amount of fluid drained was 11.2 +/- 4.3 l (range: 3.35-18.5). An improvement of symptoms and signs was noted 24-48 h after catheter placement in all patients in both groups. Procedure was well tolerated and no complications reported. CONCLUSIONS: Percutaneous placement of a pigtail catheter is a safe and effective treatment modality for severe OHSS. It may represent an attractive alternative to multiple vaginal or abdominal paracentesis.

Effects of selenium deficiency in the development of trypanosomes and humoral immune responses in mice infected with Trypanosoma musculi.

The selenium-deficient mouse-trypanosome system was used to study the effects of selenium deficiency in Swiss Webster mice infected with Trypanosoma musculi. In selenium-deficient mice, a low parasitemia was observed and infection was cleared by day 16 post-inoculation (PI), whereas control mice sustained the parasitemia until day 24 PI. There were no significant differences in size variability of the trypanosomes; however the range of variability in the length of parasites differed significantly between the three groups. In comparison to mice on complete or pair-fed diets, the selenium-deficient mice produced lower concentrations of IgG(1), IgG(2b), IgG(3), and IgM. The levels of IgG(2a) and IgA were lower than normal controls. The results of the present study indicated that there was a severe depression in primary and secondary antibody responses to sheep red blood cells in all inoculated mice. However, these responses were significantly less depressed in selenium-deficient mice.

Genetic basis of plant height and its degree of indetermination in mungbean (Vigna radiata (L.) Wilczek).

The genetic basis of plant height at various growth stages and the degree of indetermination of plant height in six mungbean genotypes (NM 92, 6601, NM 89, VC 1560D and VC 3902A) were assessed through half diallel cross. Cultivars, 6601 and NM 92, were the best general combiner for pre-flowering dry matter accumulation and minimum increase in plant height from first flower to 90% pods maturity, respectively. For these traits, the combination NM 92 x NM 89 was the best specific combiner of all the crosses. Both additive and dominant gene effects controlled the inheritance of plant height at first pod and to 90% pods maturity, degree of indetermination of plant height (DDh) from first flower to first pod maturity (DDh1), DDh from first flower to 90% pods maturity (DDh2) and DDh from first pod maturity to 90% pods maturity (DDh3). Plant height at first flower was additively inherited. The additive gene action was predominant as compared to dominant gene action for all the traits examined. High narrow and broad sense heritability estimates for DDh2 showed that better response to selection is possible for the development of mungbean genotypes with minimum increase in plant height during post-flowering development.

Comparative positions of kinetoplasts in Trypanosoma musculi and Trypanosoma lewisi during development in vitro.

The development of Trypanosoma musculi and Trypanosoma lewisi were studied in vitro in the presence of adherent splenic cells. Both parasites developed only when attached by their flagellar tips to adherent splenic cells. During the proliferation of T. musculi, the kinetoplast migrated towards the nucleus, and once in the vicinity of the nucleus, the nuclear division was triggered. The kinetoplast of T. lewisi did not migrate towards the nucleus, but remained at its original location. The nucleus and kinetoplast divided at the same time in both parasites, and parasites started dividing from their flagellar ends and T. musculi and T. lewisi daughter cells were formed within 48 h. The unavailability of the adherent splenic cells in vitro led the parasites to transform into round/oval nonviable forms.

Downregulation of protein kinase C inhibits activation of mitochondrial K(ATP) channels by diazoxide.

BACKGROUND: The mitochondrial K(ATP) (mitoK(ATP)) channel has been shown to confer short- and long-term cardioprotection against prolonged ischemia via protein kinase C (PKC) signaling pathways. However, the exact association between PKC or its isoforms and mitoK(ATP) channels has not yet been clarified. The present study tested the hypothesis that the activity and translocation of PKC to the mitochondria are important for cardiac protection elicited by mitoK(ATP) channels. Methods and Results-PKC was downregulated by prolonged (24-hour) treatment with phorbol 12-myristate 13-acetate (4 microgram/kg body weight) before subsequent experiments in rats. Langendorff-perfused rat hearts were subjected to 40 minutes of ischemia followed by 30 minutes of reperfusion. Effects of PKC downregulation on the activation of mitoK(ATP) channels and other interventions on hemodynamic, biochemical, and pathological changes were assessed. Subcellular localization of PKC isoforms by Western blot analysis and immunocytochemistry demonstrated that PKC-alpha and PKC-delta were translocated to the sarcolemma and that PKC-delta was translocated to the mitochondria after diazoxide treatment. In hearts treated with diazoxide (80 micromol/L), a significant improvement in cardiac function and an attenuation of cell injury were observed. In PKC-downregulated hearts, protection was abolished because mitoK(ATP) channels could not be activated by diazoxide. CONCLUSIONS: These data suggest that PKC activation is required for the opening of mitoK(ATP) channels during protection against ischemia and that this effect is linked to isoform-specific translocation of PKC-delta to the mitochondria.

Effects by doxorubicin on the myocardium are mediated by oxygen free radicals.

We hypothesized that doxorubicin (DOX) induces cardiotoxicity of myocardium via oxygen radicals. The present study is aimed at examining the membrane alterations by oxygen radicals generated by DOX in adult rats and cultured neonatal myocytes. Our results showed that DOX 1) decreased beta-adrenoceptor (BAR) density in the cell membrane, 2) increased the membrane permeability of cultured neonatal rat myocytes and 3) altered the ultrastructure of myofibrils and subplasmalemmal actin networks. These effects were reproducible by exogenous hydrogen peroxide. The antioxidant melatonin (MLT) inhibited enzyme leakage and peroxidation in a concentration-dependent manner. It is concluded that DOX induces cardiotoxicity through lipid peroxidation and melatonin is an effective antioxidant against the reactive oxygen intermediates generated by DOX.

Mitochondrial K(ATP) channel as an end effector of cardioprotection during late preconditioning: triggering role of nitric oxide.

Nitric oxide (NO) has been implicated in the "second-window" of ischemic preconditioning (PC). However, the identity of the end effector after initiation of preconditioning by NO is not known. It is likely that NO is involved in opening of mitochondrial ATP-sensitive potassium (mitoK(ATP)) channels. We hypothesized that NO is an important trigger for the opening of mitoK(ATP) channels in the late phase of preconditioning and inducible nitric oxide synthase (iNOS) up-regulation via NF kappa B plays a critical role in diazoxide-induced cardioprotection. To examine this, diazoxide (7 mg/kg) was administered to wild-type (WT) mice and mice lacking the gene 24 hours before 40 minutes of global ischemia. Hearts were perfused in a Langendorff mode and effects of activation of mitoK(ATP) channel and other interventions on functional, biochemical and pathological changes in ischemic hearts were assessed. In hearts from WT mice treated diazoxide, left-ventricular-developed pressure, end-diastolic pressure and coronary flow were significantly improved after ischemia/reperfusion (I/R); lactate dehydrogenase (LDH) release was also significantly decreased, while ATP contents were significantly higher. Administration of 5-HD, a specific blocker of mitoK(ATP) channel or l -NAME, an inhibitor of iNOS before I/R, during diazoxide-pretreatment completely blocked the late cardioprotection against ischemia. Late cardioprotection was also blocked by inhibition of either PKC- delta by rottlerin or NF kappa B by DDTC before diazoxide pretreatment. Diazoxide pretreatment significantly increased nuclear translocation of p65 which was blocked by protein kinase C (PKC) or nitric oxide synthase (NOS) inhibition. Diazoxide was totally inefffective in iNOS knockout mice. These results suggest that diazoxide activates NF kappa B via PKC signaling pathway and that leads to iNOS up-regulation after 24 hours. NO which is generated upon ischemic stress triggers the opening of mitoK(ATP)channel as an end effector of cardioprotection during late PC.
