Binding of ouabain to normal and tumoral pancreatic islet cells.

The binding of [3H(G)]ouabain was examined in both rat pancreatic islets and tumoral islet cells (RINm5F line). In the islets, the specific binding was considered too low to allow for extensive investigations, taking into account the limited number of islets readily available. In the RINm5F cells, which bound more ouabain than normal islet cells, the time course for binding, its temperature dependency, its competitive inhibition by K+, the number of binding sites, their affinity, and the turnover of ATP per binding site were all similar to those found in other cell types. An incomplete dissociation of [3H(G)]ouabain by unlabelled glycoside and the stimulation of binding by D-glucose were noticed, both phenomena being possibly attributable, in part at least, to the functional response of the RINm5F cells to D-glucose with resulting activation of exocytosis-endocytosis coupling.

Inhibition by a nonmetabolized analogue of L-leucine of protein biosynthesis in tumoral pancreatic islet cells.

The effect of L-leucine, its deaminated metabolite 2-ketoisocaproate and its nonmetabolized analogue b(+/-)2-aminobicyclo[2,2,1]-heptane-2-carboxylic acid (BCH) upon protein labelling was examined in tumoral islet cells (RINm5F line) exposed to L-[4-3H]phenylalanine or L-[3-3H]serine. The interpretation of the results, in terms of changes in biosynthetic activity, was obscured by a possible interference of the tested nutrients with the uptake and further metabolism of the tracer tritiated amino acids. Nevertheless, when the cells were preincubated with the nutrient secretagogues and then incubated in the sole presence of L-[3-3H]serine, BCH, but not L-leucine or 2-ketoisocaproate, still inhibited protein labelling, this coinciding with a decrease in the ratio between TCA-precipitable and total radioactivity in the RINm5F cells. The inhibitory action of BCH was antagonized, to a limited extent, by D-glucose. It is proposed that BCH could be used as a tool to interfere with the function and growth of insulinoma cells.

Glucagon storage, release and degradation by tumoral islet cells (RINm5F line).

Tumoral islet cells of the RINm5F line contain and release glucagon. Over 90 min incubation, the secreted hormone represents about 50% of the final cell content (1.4 +/- 03 fg/cell). L-arginine augments modestly glucagon release, the relative magnitude of such an effect being inversely related to the concentration of D-glucose. The immunoreactive material secreted by the cells consists of both true glucagon (59-64%) and larger peptides. The RINm5F cells also release a proteolytic factor able to degrade exogenous glucagon or insulin. These results reveal that glucagon producing tumoral cells display several abnormal features in their secretory behaviour.

Stimulation by D-glucose of protein biosynthesis in tumoral insulin-producing cells (RINm5F line).

Tumoral insulin-producing cells of the RINm5F line display a poor secretory response to D-glucose. Relative to basal insulin output, which corresponds to a fractional release of stored hormone close to 25%/90 min, the glucose-induced increment in insulin release does not exceed 28 +/- 7%. The low efficiency of D-glucose as an insulin secretagogue contrasts with a marked stimulant action of the hexose on peptide biosynthesis. Thus, D-glucose increases in a rapid, sustained, and dose-related manner the incorporation of [3H]leucine or [3H]phenylalanine into trichloroacetic acid-precipitable material; there is a 7-fold difference between basal and glucose-stimulated protein biosynthesis. Although the low proinsulin and insulin content of the tumoral cells, relative to their total protein content, hampers the quantification of newly synthesized hormonal products, the ratio of immunoreactive proinsulin to insulin exceeded the value in normal islet cells; this difference was more marked in secreted than stored material. It is concluded that despite their poor secretory response to D-glucose, the tumoral cells are, in fact, quite sensitive to this hexose, as documented by its effect on biosynthetic activity. Although RINm5F cells are known to display an acute secretory response to nonnutrient secretagogues, the apparent discrepancy between the biosynthetic and secretory responses to D-glucose may be accounted for in part by a severe perturbation in the capacity of the tumoral cells to store proinsulin and insulin.

Epidemiology of Trypanosoma cruzi in the oriental plains of Colombia.

Epidemiologic studies to define the domiciliary and extradomiciliary transmission cycles of Trypanosoma cruzi and Trypanosoma rangeli in the Oriental Plains of Colombia were conducted in the gallery forests near Carimagua and El Porvenir. One-hundred and seven palm trees belonging to nine genera were examined; triatomines were found in only three palm species, the leaves of which are locally used for roof thatching: 2/29 Maximiliana elegans, 1/7 Mauritia flexuosa and 7/7 Scheelea sp. Bugs were also found in 5/14 hollow Mauritia inhabited by bats, 4/21 bird nests and 1/4 armadillo burrows. Five species of triatomines were collected: Rhodnius prolixus was the most abundant, 192 of the total 207 (92%) collected; the bugs were found in Maximiliana and Mauritia but especially in Scheelea, and 8% were infected with T. cruzi and T. rangeli; Cavenicola pilosa and Triatoma maculata were found associated with bats; Psammolestes arthuri and Panstrongylus lignarius with bird nests and Panstrongylus geniculatus with armadillos. Although triatomine colonies were not found in human dwellings, flying adults of R. prolixus occasionally reached houses by their own locomotion and fed on man, but did not become established. Only 12 of 199 persons (6%) tested serologically were reactors to T. cruzi antigens and all 12 had lived in areas of domiciliary transmission elsewhere in the country, indicating that domiciliary transmission is not occurring in this region. Whether the presence of domiciliary R. prolixus in houses located in the ecologically altered piedmont of the oriental plains, a known area of domiciliary transmission of T. cruzi, is due to importation of domiciliary bugs from endemic areas or to the domiciliarization of wild R. prolixus remains to be determined.

Comparison of filtration staining (Bell) and thick smear (Kato) for the detection of quantitation of Schistosoma mansoni eggs in faeces.

We compare results of one Bell and one Kato-Katz examination performed on each of 315 stool specimens from residents in an area in north-eastern Brazil endemic for schistosomiasis mansoni. The prevalence of Schistosome infection detected by the Bell technique was 76% and by the Kato-Katz technique was 63%. 81% (44/54) of the infections missed by a Kato-Katz smear were light infections (one of 50 epg range by Bell examination). Over, all, 55% (44/80) of stools in this egg count range by the Bell technique were negative on a single Kato-Katz smear. This implies that five Kato-Katz smears per stool would ensure a 95% probability (0.55(5) X 100) of detecting such light infections. However, a single Kato-Katz smear detected eggs in 97% (124/128) of stools with a Bell count greater than 100 epg. For stools positive by both methods the egg counts per gram of stool were higher (p less than 0.001) by Kato-Katz examination. Geometric mean egg counts for the infected population were 199 epg by the Kato-Katz and 92 epg by the Bell methods. 64% (59 v. 36) more persons were classified as heavily infected (greater than 400 epg) by the Kato-Katz method than by the Bell method. The differing measurements of schistosome infection obtained with the Bell and Kato-Katz methods must be considered when comparing data on morbidity-infection relationships.

Non-invasive assessment of airway inflammation in ship-engine workers.

Smoking is harmful for respiratory function. In young to middle-aged men the damage is insidious and difficult to demonstrate. The respiratory impairment could increase under specific stressful conditions in the professional environment. On the hypothesis that exhaled markers are useful for assessing airway susceptibility to inhaled irritants, we measured exhaled markers and lung function in smoking and non-smoking engine-driver military coastguards before and after a patrol at sea. Eighteen men, mean age 39 yrs (range 23-58 yrs), 8 smokers, underwent spirometry, exhaled and nasal nitric oxide (eNO, nNO), exhaled carbon monoxide (CO) and exhaled breath condensate (EBC) for measures of hydrogen peroxide (H2O2), leukotriene B4 (LTB4), proteins (Prots), 8-isoprostanes (8-IsoPs), nitrite (NO2-) and nitrosothiols (RS-NOs) at baseline and after an 8-hour patrol navigation on board small, high-speed diesel-powered ships. At baseline, the smokers showed higher middle flows and CO levels, lower eNO and nNO than non-smokers, but similar levels of EBC markers; geometric means (95% confidence interval), CO: 23.6 (14.5 to 38.3) vs. 3.5 (2.5 to 5.3) ppm; eNO: 7.9 (4.8 to 12.9) vs. 26.7 (15.7 to 45.5) ppb, p=0.000. After navigation, Prots, 8-IsoPs and RS-NOs (but not lung function variables or other markers) significantly increased only in smokers; baseline vs post-navigation RS-NOs: 0.27 (0.11 to 0.65) vs. 1.30 (0.58 to 2.89) micromol, p=0.012. The respiratory consequences of a stressing environment in engine-driver military coastguards who actively smoke are better assessed by measuring EBC markers than by eNO, nNO or lung function. By increasing airway inflammation from oxidative-stress, tobacco smoking appears to interact with other chemical or physical factors elicited during sea navigation. Precisely what these factors are deserves further investigation.

Lutzomyia sand flies (Diptera: Psychodidae) from middle and lower Putumayo Department, Colombia, with new records to the country.

A total of 4,840 phlebotomine sand flies from 54 localities in Putumayo department (=state), in the Colombian Amazon region, were collected in Shannon traps, CDC light traps, resting places and from human baits. At least 42 Lutzomyia species were registered for the first time to the department. Psychodopygus and Nyssomyia were the subgenera with the greatest number of taxa, the most common species being L. (N.) yuilli and L. (N.) pajoti. They were sympatric in a wide zone of Putumayo, indicating that they should be treated as full species (new status). Among the anthropophilic sand flies, L. gomezi and L. yuilli were found in intradomiciliar, peridomestic, urban or forest habitats. L. richardwardi, L. claustrei, L. nocticola and L. micropyga are reported for the first time in the Colombian Amazon basin. L. pajoti, L. sipani and L. yucumensis are new records for Colombia.
