Fate of anthracene in an artificial stream: a case study.

The fate of anthracene, a representative polycyclic aromatic hydrocarbon, was followed in a large outdoor stream microcosm . The major nonadvective route for the removal of anthracene was photolytic degradation to anthraquinone (half-life 43 min). The anthraquinone also photolyzed rapidly in this shallow stream system. Excluding the plastic channel liner, the sediment acts as the major sink for anthracene, absorbing 0.2% of the 14-day input dose. The periphyton community was the second most important sink, absorbing 0.04% of the input dose. All other compartments were of significantly less importance on a mass basis. Anthracene (11 micrograms liter-1) caused photo-induced 100% mortality of the bluegill sunfish in 9 hr in the upstream reach. Fish at the downstream station survived for approximately 26 hr and all died within 1 hr of each other. Other organisms, clams and dragonfly larvae, started to die off toward the end of the 14-day input period.

Translocation and rearrangement of c-myc into immunoglobulin alpha heavy chain locus in primary cells from acute lymphocytic leukemia.

We report the molecular analysis of an 8;14 reciprocal chromosome translocation in a case of acute lymphocytic leukemia (L3 type). DNA from primary leukemic cells was analyzed on the basis of restriction endonuclease mapping by hybridization with various human c-myc and Ig heavy chain probes. The breakpoint of the translocation is within an approximately equal to 200-base-pair region in the first intron of the c-myc gene. The first, untranslated exon thereby remains on chromosome 8q-, whereas the whole protein-coding region is rearranged in the C alpha 1 locus on chromosome 14q+. RNA transfer blot analysis showed high levels of at least two different c-myc transcripts originated from the translocated gene. Both differ in size from the normal 2.2- and 2.4-kilobase transcripts. Both c-myc structure and expression were apparently normalized in remission phase. These studies demonstrate rearrangement and abnormal expression of c-myc in primary cells from an acute leukemia patient, thus adding to the concept of a key role for c-onc in human oncogenesis.

Critical evaluation of various forms of therapy for idiopathic calcium stone disease.

The results are presented of the dietary management, alone or in association with thiazides and/or allopurinol, evaluated in 143 idiopathic calcium stone formers after a mean follow-up of 18 months. Diet alone proved to be effective in the prevention of stone relapses. The addition of thiazide and/or allopurinol provided mild improvements of urine environment but seemed to give no further clinical benefits irrespective of underlying metabolic abnormalities.

Hyperoxaluria in idiopathic calcium stone disease: further evidence of intestinal hyperabsorption of oxalate.

1. Seventeen healthy controls and 63 patients with idiopathic calcium stone disease of the urinary tract were investigated for urinary calcium and oxalate excretion and for [14C]oxalate intestinal absorption. 2. Under comparable controlled dietary intake a significant increase in calcium excretion as found in patients with stone disease. Oxalate excretion and [14C]oxalate intestinal absorption were mildly but not significantly increased. When patients with stone disease were subdivided into normocalciuric and hypercalciuric subjects, oxalate excretion and [14C]oxalate absorption were significantly increased in the latter. There was a significant direct relationship between calcium excretion and both oxalate excretion and [14C]oxalate absorption. 3. [14C]Oxalate absorption increased significantly in 22 stone-formers when dietary calcium was changed from normal to low. 4. The kinetics of [14C]oxalate intestinal absorption showed that the main difference between normocalciuric and hypercalciuric subjects occurred within the first 6 h after the oxalate-labelled meal. 5. These results confirm that mild hyperoxaluria is a frequent feature of idiopathic calcium stone disease even when patients and controls are studied under controlled dietary conditions. Our data are consistent with the hypothesis that hyperoxaluria is secondary to calcium hyperabsorption and is upper intestinal in origin.

Hormone patterns during bromocriptine-induced pregnancy in hyperprolactinemic patients.

Plasma prolactin (PRL) and human placental lactogen (HPL), and urinary estriol and pregnanediol were studied during pregnancies induced with bromocriptine (Parlodel, Sandoz) in 10 cases of hyperprolactinemia. Previous selective adenomectomy or intrasellar implantation of radioactive gold (198Au) failed to induce a complete remission in 3 of these subjects. PRL rapidly increases after bromocriptine withdrawal, reaching values higher than those in normal women in the same stage of pregnancy within a few weeks. At term, pathological PRL levels occurred in 3 subjects only (with distinct alterations of the sella turcica). Estriol, pregnanediol and HPL were normal in all cases. These findings suggest that PRL levels higher than those normally observed during pregnancy do not alter fetoplacental endocrine function.

RDT with acetate-free bicarbonate buffered dialysis fluid: long-term effects on lipid pattern, acid-base balance and oxygen delivery.

Thirteen uraemic patients with hypertriglyceridaemia were treated for 9 months with acetate-free, bicarbonate containing dialysis fluid. With this treatment more physiological correction of acid-base balance and better tissue oxygenation were obtained. This fact can explain the better tolerance to treatment we have seen. In 9 of these patients triglyceride levels fell significantly on bicarbonate treatment; they shifted back to higher values after return to acetate dialysis. No changes were found in the other 4 patients.
