Transformation and regeneration of mung bean (Vigna radiata).

The procedure relied on a protocol in which shoot organogenesis was induced on cotyledons of mung bean genotypes selected for susceptibility to agrobacterium seems to work reproducibly if not efficiently. Approximately 4-5% of the shoots produced on the kanamycin selected cotyledons are transgenic based on assays on kanamycin resistance and GUS activity. This demonstrated that transformation and regeneration in mung bean are possible. However, raising the transformed plants in field condition is yet to be perfected.

The efflux of nor-epinephrine from platelets in essential hypertension.

The precise role of sympathetic nervous system in initiation and/or maintenance of essential hypertension is unclear even today. Platelets have been used as a suitable model for studying neuronal turnover of biogenic amines. The present study comprised of hypertensive subjects (23) and normotensive controls (10). Hypertensive subjects exhibited significantly enhanced norepinephrine efflux from platelets at both 30 minutes and 60 minutes (p less than 0.001). The percent norepinephrine efflux from platelets correlated with diastolic (r = 0.66 and 0.76) and mean arterial blood pressure (r = 0.54 and 0.65) but not with systolic blood pressure. The norepinephrine efflux rate (K) similarly correlated with diastolic and mean arterial blood pressure in hypertensive subjects studied. From the above findings it appears that operative sympathetic nervous system activity is enhanced in essential hypertension. The enhanced efflux of norepinephrine from platelets may also indicate activated state of platelets in hypertension. Both could be important in genesis and complications of essential hypertension.

Reliability and limitations of fine needle aspiration cytology of lymphadenopathies. An analysis of 1,261 cases.

In a study spanning over two years, 1,261 cases with clinically significant lymphadenopathies were subjected to fine needle aspiration cytology (FNAC) to evaluate the reliability and limitations of the procedure. In the first phase of the study, consisting of 100 cases, the cytologic diagnosis was compared with histopathology, and the results were analyzed with a view to standardizing the procedure. In the second phase of the study, consisting of 1,161 patients, FNAC was used as the primary modality of diagnosis. Excision biopsy was done only when there was a clinical-cytologic discrepancy, failure to respond to treatment or FNAC diagnosis of non-Hodgkin's lymphoma. The highest diagnostic accuracy with FNAC was observed in metastatic carcinoma. Though tuberculous lymphadenitis, reactive hyperplasia and lymphoma were diagnosed with almost similar accuracy, reactive hyperplasia and non-Hodgkin's lymphoma proved difficult to diagnose. Although FNAC has proven to be a simple, safe, reliable and cost-effective diagnostic tool for lymphadenopathies, the limitations of the procedure should be kept in mind and excision biopsy used whenever required.

Q-T interval changes in acute transmural myocardial infarction.

The serial Q-T interval changes were studied in 29 survivors of acute transmural anteroseptal (11 patients), extensive anterior (10 patients), and inferior (8 patients) myocardial infarctions admitted 4 to 48 hours after the acute episode. Q-T prolongation evidenced by abnormal Q-T ratio was a constant and almost universal feature detected in 28 (97.06%) patients. The maximum Q-T prolongation was observed on an average about 36 hours after the onset of acute episode. Patients with anterior myocardial infarction had significantly higher Q-T ratios than inferior myocardial infarction group. There was a rapid decline towards normal in anteroseptal and inferior myocardial infarction groups in which it settled during initial six days, whereas, in extensive anterior myocardial infarction group, it took a longer time beyond six days to settle. The normalization of Q-T interval did not correspond to settling down of the elevated ST segment. Patients having ventricular tachyarrhythmias (VT and frequent VPBS) (14) had significantly higher Q-T ratios than those without arrhythmias. Further, the Q-T ratio was significantly higher in patients with VT (4) than in those with frequent VPBs (10). Alterations in Q-T ratio were not related to severity or extent of infarction and occurrence of heart failure. It is concluded that prolongation of electrical systole (Q-T interval) is a constant phenomenon after acute transmural myocardial infarction, magnitude and time course of its alterations being related to location of infarct and its electrical complications. It does not seem to have any correlation with mechanical complications of infarction.(ABSTRACT TRUNCATED AT 250 WORDS)

Incidence of actinomycetes infection in women using intrauterine contraceptive devices.

Pancervicovaginal smears taken from 350 women using an intrauterine contraceptive device (IUD) were screened for the presence of actinomycetes organisms. Of the 12 cases in which actinomycetes-like organisms were seen in Papanicolaou-stained smears, the presence of actinomycetes organisms was confirmed by immunofluorescence in 10 cases. The prevalence of actinomycetes infection was thus 2.8% (10 of 350 cases) in the IUD users. Eight (4.3%) of 173 symptomatic subjects had actinomycetes infections. Two of the positive cases were asymptomatic. Eight of the ten patients with confirmed actinomycetes infection were using the Cu T device while two were wearing the Lippes Loop IUD. Seven of the ten patients had been using an IUD for more than two years. The time of insertion of the IUD (postpuerperal, postmenstrual or after medical termination of pregnancy) did not show any correlation with the presence of actinomycetes infection. Actinomyces israelii was responsible for the infection in eight cases while Arachnia propionica was seen in two cases. The organisms could not be grown in culture.

Rapid onset of co-trimoxazole induced interstitial nephritis.

An infant developed anuric renal failure within 18 hours of starting therapy with Co-trimoxazole for otitis media. There was no prior exposure to Co-trimoxazole, sulfonamides or trimethoprim. A renal biopsy revealed acute interstitial nephritis with eosinophilic infiltration (AIN). The lymphocyte blast transformation test revealed increased proliferation of the patient's lymphocytes on exposure to Co-trimoxazole (Bactrim). Both parents have clinically demonstrated hypersensitivity to sulfonamides. The extremely short latent period between ingestion of the offending drug and the onset of AIN in the absence of prior exposure to the drug has been reported previously. It suggests that drug induced AIN may develop more rapidly in patients with a strong genetic hypersensitivity to the drug.

Clinicopathological, hematological, and biochemical studies in some outbreaks of nephritis in poultry.

Hematological, biochemical, histoenzymological, and histopathological changes in serum and tissues were studied in chickens during outbreaks of nephritis. Hematological studies revealed normocytic-normochromic anemia characterized by increased total erythrocyte counts, hemoglobin, packed cell volume, and erythrocyte sedimentation rate. Albumin-to-globulin ratio and sodium levels in serum, glucose in blood, and alkaline phosphatase and glucose-6-phosphatase in liver and kidneys were decreased. Glutamate pyruvate transaminase, uric acid, non-protein-nitrogen, and potassium levels in serum were increased. No significant change in the calcium, phosphorus, and total protein levels in serum was observed. These changes were directly related to the severity of the nephritis.

Renal and biochemical changes produced in broilers by high-protein, high-calcium, urea-containing, and vitamin-A-deficient diets.

Three hundred 18-day-old male chicks (Arbor Acre) were divided into five groups of 60 each and given high-protein (42.28%), high-calcium (3.37%), urea-containing (5%), vitamin-A-deficient, or control diets to study the effect of nutritional imbalances on the development of nephritis and related biochemical changes over 15 weeks. The first four diets increased the levels of glutamate oxaloacetate transaminase, glutamate pyruvate transaminase, uric acid, and nonprotein nitrogen in serum. Blood urea was increased by only the urea diet. Hypoglycemia and a decrease in hepatic glucose-6-phosphatase were also observed in chicks fed the first four diets. The vitamin-A-deficient diet resulted in a depletion of vitamin A in the liver and kidneys. These changes were directly correlated with the prolonged feeding of experimental diets and also with the severity of nephritis and degenerative changes in various organs. It was concluded that increasing the intake of nitrogen or calcium in order to increase production may in fact have the opposite effect, leading to degenerative changes in various tissues and to nephritis.
