[Comparative evaluation of various direct and culture methods for the diagnosis of genital Chlamydia trachomatis infections].

Commercially produced fluorescein labelled monoclonal antibodies for Chlamydia trachomatis detection have been recently become available. We have compared the data obtained using two monoclonal antibodies, one for detecting inclusion on cell cultures (culture confirmation) and the other for detecting C. trachomatis in smears from urethral and cervical swabs, with our routine isolation method which utilizes Giemsa staining of cycloheximide treated McCoy cell cultures. We also evaluated an enzyme immunoassay for detecting C. trachomatis antigens in urethral and cervical specimens. The culture confirmation system was slightly more sensitive and simpler than Giemsa staining. Between the results of immunofluorescence direct test and culture there was 96.3% agreement. Sensitivity, specificity and predictive positive and negative value were 72.2, 98.4, 80 and 97.6%. Between results of culture and enzyme immunoassay there was 97.2% agreement. The immunoassay sensitivity, specificity predictive positive and negative value were, in women, 100, 97.1, 63.6 and 100%; in men, 100, 95.7, 81.8, 100%.

[Phenotypic relations with regard to pyocin sensitivity, of 191 strains of Pseudomonas aeruginosa isolated in a hospital environment].

191 strains of Pseudomonas aeruginosa, isolated from clinical specimens, were tested by passive and active pyocin typing. The results of passive typing were computerized and a final dendrogram, with its peculiar similarity levels between the single strains, was obtains. This study will permit to build a system, to mark out the Pseudomonas strains, through active pyocin typing or through passive pyocin typing.

[Evaluation of two miniaturized systems widely used in the laboratories for the identification of the "Enterobacteriaceae" (author's transl)].

We have attended to a comparative research between two commercial microsystems: Enterotube and Minitek in order identify the Enterobacteriaceae and a reference system given by the combination of the usual macromethods already used in our laboratory. We have examined 401 bacterial cultures of Gram bacillus which we trought to belong to Enterobacteriums, coming from clinical material (excrements, urine, pharyngeal swabs, vaginal swabs, urethral swabs and espectoration) we have received for the bacteriological diagnosis. 390 of 401 cultures have shown to be Enterobacteriums. The biochemical reactions they have given show that the Enterotube and the Minitek have, with the usual system a good accordance for the following tests: dextrose (acid and gaz) lysin and ornithine decarboxylase, production of H2S and indole, phenylalanine deaminase and urease; while we have some statistically significant discordances for the fermenting of lactose and the use of citrate. We have also significant discordances E/C for the fermenting of dulcitole while the ones of Minitek are acceptables. The notes recommend the use, in the specialized bacteriological laboratories, of the conventional tests.

Cryoglobulinaemic membranoproliferative glomerulonephritis and hepatitis C virus infection.

BACKGROUND/AIM: A striking correlation between mixed cryoglobulinaemia and chronic hepatitis C virus infection has recently been described. Since membrano-proliferative glomerulonephritis is a rare complication of mixed cryoglobulinaemia, this study was undertaken to determine the prevalence of Hepatitis C virus infection in membrano-proliferative glomerulonephritis. PATIENTS: Eighteen patients, selected among a group of 121 affected by mixed cryoglobulinaemia, with renal involvement were included in the present study. A group of 148 patients affected by renal disease of different aetiology and the general population (6,917 people) were used as control groups. METHODS: The presence of anti-hepatitis C virus antibodies was determined by a commercial kit. The hepatitis C virus genotype was determined according to Okamoto. All patients underwent kidney and bone marrow biopsy, while the hepatic biopsy was performed in those showing signs of chronic liver disease. RESULTS: In patients with renal involvement, the kidney biopsy showed the presence of membrano-proliferative glomerulonephritis Type I in all cases. Chronic liver disease was present in eleven patients (61%). All patients were positive for serum hepatitis C virus-RNA. Bone marrow biopsy was normal in five cases, while in the others paratrabecular foci of infiltration by small lymphocytes were present. In six of these, the massive bone marrow infiltration by lymphoplas-macytoid lymphocytes suggested the diagnosis of low grade non-Hodgkin's lymphoma. In the group of patients affected by other chronic renal disease, the prevalence of hepatitis C virus infection (3.1%) was not different from that of the general population (3.2%). CONCLUSIONS: Hepatitis C virus seems to be the aetiologic agent of mixed cryoglobulinaemia and, consequently, of membrano-proliferative glomerulonephritis.

[Comparative evaluation of 3 tests for the determination of antistreptolysin titer].

We have evaluated three commercial assays for ASLO titer determination Actually a hemolysis inhibition plate micromethod (ASLO Pasteur), an automated method based on kinetic measurement of an haemolytic reaction (TASOMATIC/TASOTEC, Diagnostica Senese), and another automated assays using the nephelometric quantitation of a latex particle agglutination.
