A practical approach to crosslinking.

The various aspects of chemical crosslinking are addressed. Crosslinker reactivity, specificity, spacer arm length and solubility characteristics are detailed. Considerations for choosing one of these crosslinkers for a particular application are given as well as reaction conditions and practical tips for use of each category of crosslinkers.

Carbon monoxide-dependent chemolithotrophic growth of Clostridium thermoautotrophicum.

The acetogen Clostridium thermoautotrophicum was cultivated under CO-dependent chemolithotrophic conditions. CO-dependent growth profiles and energetics indicated that supplemental CO2 was fundamental to efficient growth at the expense of CO. Overall product stoichiometry approximated 6.5CO----CH3CO2H + 3.5CO2 + 0.6 cell C + 0.5 unrecovered C. Initial CO/CO2 ratios of 2 to 4 yielded optimal doubling times and cell yields. Maximal YCO values approximated 2.5 g of cell dry weight per mol of CO consumed; YH2 was considerably lower than YCO. Cross-transfer growth experiments and protein profiles indicated differential expression of genes between CO and methanol cultures.

Carbon Dioxide

Adaptation of the acetogen Clostridium thermoautotrophicum to minimal medium.

Clostridium thermoautotrophicum was adapted to minimal medium and cultivated at the expense of glucose, methanol, or H2-CO2. No supplemental amino acids were required for growth of the adapted strain, and nicotinic acid was the sole essential vitamin. Neither N2 nor nitrate could replace ammonium as the nitrogen source, and biotin was preferentially stimulatory for glucose cell lines. Growth in minimal medium yielded substantially higher acetate concentrations per unit of biomass formed than did growth in undefined medium.
