Molecular cloning and nucleotide sequence of the gene encoding the major peptidoglycan hydrolase of Lactococcus lactis, a muramidase needed for cell separation.

A gene of Lactococcus lactis subsp. cremoris MG1363 encoding a peptidoglycan hydrolase was identified in a genomic library of the strain in pUC19 by screening Escherichia coli transformants for cell wall lysis activity on a medium containing autoclaved, lyophilized Micrococcus lysodeikticus cells. In cell extracts of L. lactis MG1363 and several halo-producing E. coli transformants, lytic bands of similar sizes were identified by denaturing sodium dodecyl sulfate (SDS)-polyacrylamide gels containing L. lactis or M. lysodeikticus cell walls. Of these clearing bands, corresponding to the presence of lytic enzymes with sizes of 46 and 41 kDa, the 41-kDa band was also present in the supernatant of an L. lactis culture. Deletion analysis of one of the recombinant plasmids showed that the information specifying lytic activity was contained within a 2,428-bp EcoRV-Sau3A fragment. Sequencing of part of this fragment revealed a gene (acmA) that could encode a polypeptide of 437 amino acid residues. The calculated molecular mass of AcmA (46,564 Da) corresponded to that of one of the lytic activities detected. Presumably, the enzyme is synthesized as a precursor protein which is processed by cleavage after the Ala at position 57, thus producing a mature protein with a size of 40,264 Da, which would correspond to the size of the enzyme whose lytic activity was present in culture supernatants of L. lactis. The N-terminal region of the mature protein showed 60% identity with the N-terminal region of the mature muramidase-2 of Enterococcus hirae and the autolysin of Streptococcus faecalis. Like the latter two enzymes, AcmA contains C-terminal repeated regions. In AcmA, these three repeats are separated by nonhomologous intervening sequences highly enriched in serine, threonine, and asparagine. Genes specifying identical activities were detected in various strains of L. lactis subsp. lactis and L. lactis subsp. cremoris by the SDS-polyacrylamide gel electrophoresis detection assay and PCR experiments. By replacement recombination, an acmA deletion mutant which grew as long chains was constructed, indicating that AcmA is required for cell separation.

Serum antithrombin III and alpha-2-antiplasmin concentrations in patients with Hodgkin's disease in the course of chemotherapy.

Disturbances in hemostasis in cancer disease frequently occur. This clinical situation may contribute to the spread and metastasis of cancer. The determination of inhibitors controlling hemostasis especially during treatment may be of value for the evaluation of effectiveness of treatment and balancing of hemostasis. Twenty four patients diagnosed with Hodgkin's disease were introduced to our studies. The level of antithrombin III (AT-III) and alpha-2-antiplasmin (alpha-2-AP) in sera were determined before, during and after treatment with MOPP. We found decrease of AT-III concentration before treatment, and chemotherapy increased the level of this inhibitor but still below the level of control. Alpha-2-AP concentration was higher in patients and MOPP application decreased the level below the values found in controls. The determination of concentration of both inhibitors may be important for the evaluation of the effect of treatment in understanding how the disturbances of hemostasis may be equilibrated.

Effect of dimethylsulfoxide-hydroxyl radical scavenger on cerulein-induced acute pancreatitis in rats.

Acute oedematous pancreatitis was induced in Wistar male rats by intravenous (i.v.) infusion of cerulein at the rate of 5.10(-6)g.kg-1.h-1 for 3 or 6 hrs. The effects of dimethylsulfoxide (DMSO)-hydroxyl radical scavenger in this model of disease were examined. DMSO was injected i.v., 0.75 g.kg-1, at 0 and 3 hrs during cerulein infusion. Treatment with this agent exerted a protective effect on the rat pancreas in the cerulein-induced acute pancreatitis. This effect is expressed by inhibition of lipid peroxidation in pancreatic tissue, less oedema formation, diminution of hyperlipasemia and a significant reduction in acinar cell vacuolisation. Our data suggest that besides the hydroxyl radical, other oxidants (secondary?) may cause cytotoxicity in this model of disease. We suggest that phagocytic inflammatory cells provide such oxidants.

[Granulocyte acid phosphatase activity in patients with Hodgkin's disease].

In 24 patients with Hodgkin's disease the activity of acid phosphatase was determined in granulocytes. Its level was only half that in the control group. The causes of this low activity of this enzyme may be increased number of cells without it, exocytosis, and possible influence of substances secreted by the neoplasm. It is suggested that the determination of this activity may be indirectly useful for verification of the efficacy of the antibacterial mechanisms, and also of antitumour resistance of the host.

The activity of neutrophil myeloperoxidase in patients with Hodgkin's disease.

Neutrophil myeloperoxidase activity has been studied in twenty one patients diagnosed with Hodgkin's disease. The presented data indicate no differences in total MPO activity, whereas we observed some differences in the percentage of granulocytes with different degree of scores. Changes in the intensity of reaction may indicate the possibility of exocytosis as a mechanism releasing MPO from the cell to the surrounding area. In peripheral, circulating neutrophils, such a phenomenon seems to be of no avail and may disorganise anti-cancer defence.

[Effect of alcohol on the lymphocyte system. V. Evaluation of blast transformation and rosette formation of peripheral blood lymphocytes in alcoholics].

Transformation and rosette formation of peripheral blood lymphocytes were studied. We have observed decrease in lymphocyte response to the mitogen as well as in spontaneous ability to the transformation. The percent of B-rosettes was diminished, while T-rosettes were in normal ranges. Lower index of completely transformed ones in men indulged to freely in alcohol may be consequence of altered cell membrane fluidity of shifts in T lymphocyte subpopulations.

Isotopic composition of high-activity particles released in the Chernobyl accident.

Gamma spectra were measured and activities of the detected isotopes were analyzed for 206 high-activity particles (hot particles, HPs) found in northeastern Poland after the Chernobyl accident. The isotopic composition of HPs observed in gamma-activity is compared with that of the general fallout and core inventory calculations. Particle formation and a process of depletion in Ru and Cs isotopes are discussed. On the basis of a search performed a year later, some comments on the behavior of HPs in the soil are made.

The activity of neutrophil myeloperoxidase in patients with lung carcinoma.

MPO activity in the neutrophils has been evaluated in 25 patients with lung carcinoma. Surprisingly for us we observed no differences between patients and controls in the total MPO activity. However, we noted the increase in the percentage granulocytes with the first and fourth degree of scores in cancer patients in comparison to normal adults. The highest activity was characterized by the distribution of this enzyme not only in granules but also outside them. Any changes in the total activity of MPO may reflect the balance between the factors which may stimulate it as tumor promoters enhancing the production of superoxide anion or the inhibitory effect of plasma. Diffuse enzymatic reaction may be due to some functional abnormalities of phagocytosis or membrane damage by the MPO system. The role of MPO in anticancer mechanisms is briefly discussed.

Serum antithrombin III and alpha-2-antiplasmin concentration in patients with lung carcinoma.

Serum antithrombin III and alpha-2-plasmin inhibitor concentrations has been evaluated in 26 patients with lung carcinoma. We observed a twofold decrease in antithrombin III level and no differences between test and control groups in alpha-2-plasmin inhibitor concentrations. Evidently, the decreased antithrombin III level may reflect its consumption because of enhanced plasma thrombin activity, whereas normal alpha-2-plasmin inhibitor may result in no induction of secondary fibrinolysis followed by the stimulation of the coagulation system or no conditions for primary increased fibrinolysis. It seems possible, that the antithrombin III level in the serum may at least partly reflect the tendency for hypercoagulability and spreading of cancer.

Serum concentration of alpha-2-macroglobulin, alpha-1-antitrypsin and alpha-2-antichymotrypsin in patients with lung carcinoma.

Serum concentration of alpha-2-macroglobulin, alpha-1-antitrypsin and alpha-2-antichymotrypsin was evaluated in 26 patients with lung carcinoma. We observed an evident decrease in alpha-2-M and alpha-1-antitrypsin level and no differences between tested and control groups in alpha-1-antichymotrypsin concentration. The deficiency of protease inhibitors may be due to the increased level of protease activity in malignant cells. Infiltration of granulocytes near tumor and released enzymes from them may exhaust proteolytic inhibitory capacity, too. Increased protease activity is associated with transformation and uncontrolled proliferation, therefore antiproteases may be accepted as anticancerogenic factors. Further investigations are needed to bring us closer to understanding this question.

Does ceftazidime or latamoxef affect the phagocytic capabilities of human granulocytes?

In comparative studies made in patients with pneumonia we have treated 14 cases with Latamoxef and 16 cases with Ceftazidime. We have focused attention on the side effects of these drugs and on the phagocytic capacity of granulocytes. In our patients we have noted a decrease of bactericidal activity of plasma and granulocytes and an elevated NBT reduction capability of resting granulocytes. The other tested function of the granulocytes exhibited only small variations. The applied therapy caused the bactericidal activity to be increased in granulocytes and plasma. Both drugs had no harmful effect on the phagocytic activities of granulocytes.

Changes in some hemostatic parameters in patients with infections treated with ceftazidime and latamoxef.

In comparative studies made in patients with pneumonia we have treated 14 cases with Latamoxef and 16 cases with Ceftazidime. We have concentrated our attention on the side effects of these drugs on hemostasis. In 8 patients treated with Latamoxef we have found a prolongation bleeding time, moderate hypoprothrombinemia, thrombocytopenia and nearly no normalization of APTT and plasma euglobulin fibrinolysis. In one patient treated with Ceftazidime only slight effect of this drug on hemostasis was observed. Crucial reason for coagulopathy is probably due to impaired synthesis of vitamin K by diminished gut flora conditioned by antibiotics treatment. In the case of Ceftazidime mentioned there seem to be minimal side effects.
