Solid-phase/solution-phase combinatorial synthesis of neuroimmunophilin ligands.

A novel solid-phase/solution-phase strategy for the synthesis of neuroimmunophilin ligands based on GPI 1046 was developed. The synthesis employs a solid-phase esterification strategy followed by a solution-phase pyruvic amide formation to produce multi-milligram quantities of discrete compounds for assay. The protocol was applied to a production library of 880 discrete compounds. A highlight of the strategy is an aqueous extractive purification of the final compounds using a novel liquid/ice extraction system developed for high throughput.

Stress, reactivity, and immune function in healthy men.

We examined the effects of acute psychological stress on lymphocyte proliferation and circulating levels of interleukin-1 and -2. Healthy men were exposed to two viewings of a gruesome surgery film and were asked to recall details of the film twice during a 30-min period. These subjects were compared to a nonstress control group. Lymphocyte proliferation to the mitogen concanavalin A (Con A; 5 micrograms/ml) was decreased during and after exposure to the stressor when compared to the control group. This decrease was more pronounced in subjects exhibiting greater blood pressure reactivity while viewing the film than in subjects showing smaller blood pressure responses. None of the other immunological measures was significantly affected by the stressor. Cortisol was not correlated with lymphocyte responsiveness. Possible explanations for these results and implications for further research are discussed.

Effect of prostaglandin E2 on oviductal adenosine 3':5'-monophosphate levels during estrus and pseudopregnancy.

The basal levels of adenosine 3':5'-monophosphate (cyclic AMP) in the oviductal isthmus were lower during pseudopregnancy than during estrus. No differences were observed in the ampulla. Prostaglandin E2(PGE2) significantly increased cyclic AMP levels in the isthmus during pseudopregnancy but not during estrus. In contrast, in the ampulla, PGE2 did not alter cyclic AMP levels in either hormonal condition. Theophylline increased cyclic AMP levels in the isthmus and in the ampulla during pseudopregnancy. These results show that PGE2 causes a specific increase in cyclic AMP levels in the isthmus after ovulation.
