Effect of triiodothyronine on cAMP-dependent and cAMP-independent protein kinase activities in developing chick embryo liver.

1. Changes in liver cytosol cAMP-dependent kinase and cAMP-independent growth-related quercetin-inhibited casein kinase activities during chick embryo development were studied. 2. Both kinase activities were found to increase continuously during the experimental period. 3. Upon treatment of embryos with triiodothyronine, an activation of cAMP-dependent kinase A and cAMP-independent casein kinase was observed which was most pronounced on days 12 and 14.

Microtubules and regulation of granulosa cell steroidogenesis by porcine granulosa cell conditioned medium.

Possible involvement of microtubules in the regulation of granulosa cell steroidogenesis by large follicle granulosa cell conditioned media (LGCCM) was assessed by monitoring the effect of agents that alter the cytoplasmic microtubule-tubulin equilibrium. The changes in microtubule organization and cell shape were examined by immunohistochemical procedure and morphometric analysis. Progesterone production stimulated by LGCCM was reduced by colchicine (agent that polymerizes microtubules) in a dose-dependent manner. In contrast, LGCCM stimulated progesterone secretion was significantly decreased by microtubule stabilizing agent (taxol: 1-10 microM). Cultured granulosa cells with a flattened appearance and projections after 24 h of incubation assumed a spherical configuration and were devoid of cytoplasmic processes when cultured with these agents. LGCCM stimulated GCs showed a reduction in the perimeter as compared with controls, although some cytoplasmic processes were observed. These findings suggest the involvement of microtubules in the regulation of cultured granulosa cell progesterone production by LGCCM, possibly through an effect on subcellular organelle distribution by altering the morphology of granulosa cells.

[New epidemiological study data on the manufacture of divinyl].

Complex labour-hygienic studies were carried out on the working environment in divinyl production as well as on the health state of the workers. The leading deleterious factor of the environment are the concentrations of divinyl, hydrocarbons and ammonia above the norms. The latter have been determined by stationary and personal sample collecting. More than half of the workers examined work under labour conditions characterized as "particularly unfavourable" and "dangerous". In comparison with the hygienic characteristic by the middle of 1978-1980, a considerable increase of ammonia concentrations was established. A total of 102 workers have been covered by a complete clinical-laboratory examination. The results have been statistically processed with a view to establishing the correlation between the effect of the deleterious factors of the working environment and the deviations established in the health state of the workers. The following cases could be associated with occupational risk: cases with "positive" and "very likely" chronic ischemic heart disease as well as those with arterial hypertension, gastrointestinal diseases, chronic diseases of upper respiratory pathways with loss of sense of smell to various degrees, the neurological diseases--autonomic-vasal and neurotic syndrome, the cases of contact dermatitis and anemic syndrome.

[Experimental study of the dynamics of alkaline phsophatase activity after a liver wound].

The investigation of alkaline phosphatase activity on 80 rabbits with an experimental model of liver wound showed the following results: 1. Alkaline phosphatase in blood serum was elevated considerably during the first postoperative days, but after the fifth day was close to the normal ranges. 2. Tissue alkaline phospatase sharply fell up to the third day. Its values began to raise after the 20th day and reached the normal level in the end of the experimental period (30th day). 3. Histochemical studies of liver wound showed a positive reaction for alkaline phosphatase in the cells of the inflammatory infiltrate during the first days. Increased phosphatase activity was demonstrated in fiber structures, epithelium of bile canals and capillary endothelium. The dynamics of alkaline phosphatase activity in blood serum is a reflection of the delicate biochemical and histochemical changes in the liver wound. The follow-up of serum alkaline phosphatase together with the clinical course of healing could be an useful index for evaluation of diet and working capacity of the patient.

[Biochemical characteristics of soluble collagen in glued and sewed liver wounds].

The author examined soluble fractions of collagenalkaline soluble, acid soluble and neutral soluble in sewed and glued with cyanacrylic glue experimental wounds of liever in rabbits. The operative wounds were treated as regenerating tissues with various metabolism in respect to the soluble collagen. There was higher percentage ratio of the fractions revealing the synthesis of the collagenous molecule and the formation of colagemous fibre in the sewed wounds.
